1A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 2Epidemiological Status of Chronic Diabetic Complications in China显示文摘With the booming development of industrialization and urbanization, popularization of high calorie diet and sedentary lifestyle, and the rapid growth of aging, the prevalence of diabetes increased nearly 10 times during the past three decades in China.[1-3] China has become the country with the largest number of people with diabetes and thus will face considerable challenge of chronic complications in the next 10-20 years.Diabetes-associated chronic complications, such as vision disorder, kidney disease, neuropathy, peripheral vascular disease, cardiopathy, and stroke, are the major causes of physical disability and mortality, making it a nonnegligible problem of social healthcare system.Therefore, it is of importance to summarize and update current prevalence and risk factors of diabetic complications in China, not only to establish the strategy of prevention and treatment to delay the development of chronic complications, but also to improve the living quality of patients and alleviate the burden of medical cost.Jian-Ping Weng Yan Bi 2015Chinese Medical Journal2015,,24:37
3The Reference Genome Sequence of Scutellaria baicalensis Provides Insights into the Evolution of Wogonin Biosynthesis显示文摘Scutellaria baicalensis Georgi is important in Chinese traditional medicine where preparations of dried roots,'Huang Qin,' are used for liver and lung complaints and as complementary cancer treatments. We report a high-quality reference genome sequence for S. baicalensis where 93% of the 408.14-Mb genome has been assembled into nine pseudochromosomes with a super-N50 of 33.2 Mb. Comparison of this sequence with those of closely related species in the order Lamiales, sesamum indicum and Salvia splendens,revealed that a specialized metabolic pathway for the synthesis of 4'-deoxyflavone bioactives evolved in the genus Scu-tellaria. We found that the gene encoding a specific cinnamate coenzyme A ligase likely obtained its newfunc- tion following recent mutations, and that four genes encoding enzymes in the 4'-deoxyflavone pathway are present as tandem repeats in the genome of S. baicalensis. Further analyses revealed that gene duplications, segmental duplication, gene amplification, and point mutations coupled to gene neo- and subfunctionaliza-tions were involved in the evolution of 4'-deoxyflavone synthesis in the genus Scutellaria. Our study not only provides significant insight into the evolution of specific flavone biosynthetic pathways in the mint family, Lamiaceae, but also will facilitate the development of tools for enhancing bioactive productivity by metabolic engineering in microbes or by molecular breeding in plants. 4Efficacy and safety of metformin and sitagliptin based triple antihyperglycemic therapy(STRATEGY):a multicenter,randomized,controlled,non-inferiority clinical trial显示文摘Despite the current guideline's recommendation of a timely stepwise intensification therapy,the 'clinical inertia',termed as the delayed treatment intensification,commonly exists in the real world,which may be partly due to the relatively little substantial evidence and no clear consensus regarding the efficacy and safety of triple oral agents in patients inadequately controlled with dual therapy.In this clinical trial performed in 237 centers in China,5,535 type 2 diabetic patients inadequately controlled by previous therapies were treated with a stable metformin/sitagliptin dual therapy for 20 weeks.The patients who did not reach the glycated hemoglobin A1c(HbA1c) goal were then further randomized into glimepiride,gliclazide,repaglinide,or acarbose group for an additional 24-week triple therapy.A mean HbAlc reduction of 0.85%was observed when sitagliptin was added to the patients inadequately controlled with metformin in 16 weeks.Further HbAlc reductions in the 24-week triple therapy stage were 0.65%in glimepiride group,0.70%in gliclazide group,0.61%in repaglinide group,and 0.45%in acarbose group.The non-inferiority criterion for primary hypotheses was met for gliclazide and repaglinide,but not for acarbose,compared with glimepiride,when added to metformin/sitagliptin dual therapy.The incidences of adverse events(AEs) were 29.2%in the dual therapy stage and30.3%in the triple therapy stage.Metformin/sitagliptin as baseline therapy,with the addition of a third oral antihyperglycemic agent,including glimepiride,gliclazide,repaglinide,or acarbose,was effective,safe and well-tolerated for achieving an HbAlc<7.0%goal in type 2 diabetic patients inadequately controlled with previous therapies.The timely augmentation of up to three oral antihyperglycemic agents is valid and of important clinical benefit to prevent patients from exposure to unnecessarily prolonged hyperglycemia.Wen Xu Yiming Mu Jiajun Zhao Dalong Zhu Qiuhe Ji Zhiguang Zhou Bin Yao Anhua Mao Samuel S.Engel Bin Zhao Yan Bi Longyi Zeng Xingwu Ran Juming Lu Linong Ji Wenying Yang Weiping Jia Jianping Weng 2017Science China(Life Sciences)2017,60,3:19
5Expansion and differentiation of human hepatocyte-derived liver progenitor-like cells and their use for the study of hepatotropic pathogens显示文摘The study of pathophysiological mecha nisms in huma n liver disease has bee n constrained by the in ability to expa nd primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be con verted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD +-dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded sign ifica ntly duri ng in vitro passage. The expa nded cells can readily be con verted back into metabolically functional hepatocytes in vitro and upon tran splantati on in vivo. Under three-dime nsional culture conditions, differentiated cells gen erated from HepLPCs regai ned the ability to support infect io n or reactivatio n of hepatitis B virus (HBV). Our work dem on strates the utility of the con versi on betwee n hepatocyte and liver proge nitor-like cells for studying HBV biology and an tiviral therapies. 6Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 7Tripartite motif.containing 3(TRIM3)inhibits tumor growth and metastasis of liver cancer显示文摘Background:Reduced expression of tripartite motif-containing 3(TRIM3) has been reported to be involved in the pathogenesis of human glioblastoma.In our previous research,we found that TRIM3 expression was markedly reduced in human primary hepatocellular carcinoma(HCC) tissues and that low TRIM3 expression was associated with short survival of HCC patients.However,the role of TRIM3 in liver cancer remains unknown.This study aimed to investigate the function of TRIM3 in liver cancer cells.Methods:The protein levels of TRIM3 in five liver cancer cell lines(SK-Hep1,Hep3 B,Huh7,HepG2,Bel-7402) and one normal liver cell line(L02) were detected with Western blotting.HepG2 and Bel-7402 cells with low TRIM3 expression were infected with recombinant lentiviruses overexpressing TRIM3(LV-TRIM3),whereas Huh7 and Hep3 B cells with high TRIM3 expression were transfected with TRIM3-targeted small interfering RNA(siTRIM3).The functions of TRIM3 in the proliferation,colony formation,cell cycle,migration,invasion,and apoptosis of the above cell lines were examined.The effect ofTRIM3 on tumor growth and metastases in nude mice was also investigated.Results:TRIM3 was overexpressed in HepG2 and Bel-7402 cells with LV-TRIM3 infection,which further reduced proliferation,colony formation,migration,and invasion of both cell lines.Cell cycle analysis showed that TRIM3 overexpression induced G_0/G_1 phase arrest in HepG2 and Bel-7402 cells.Moreover,apoptosis was not increased in HepG2 or Bel-7402 cells overexpressing TRIM3.Contrarily,silencing TRIM3 expression in Huh7 and Hep3 B cells by siTRIM3 led to significantly decreased percentages of both cells in the G_0/G_1 phase and promoted cell proliferation,colony formation,migration,and invasion.In vivo experiment results confirmed that TRIM3 overexpression suppressed tumor growth and metastasis.Conclusions:TRIM3 plays a tumor-suppressing role in the regulation of liver cancer development by reducing cell proliferation through cell cycle arrest at the G_0/G_1 phase.Xu.Qiong Huang Xiao.Fei Zhang Jin.Hua Xia Jie Chao Qiu.Zhong Pan Jing.Jing Zhao Zi.Qi Zhou Chang.Long Chen Yan Tang De.Sheng Weng Jian.Hua Zhang Jian.Chuan Xia 2017Chinese Journal of Cancer2017,36,9:13

9Clinical evaluation following the use of mineralized collagen graft for bone defects in revision total hip arthroplasty显示文摘Revision total hip arthroplasty(THA)with massive bone loss has been a real challenge for orthopaedic surgeons.Here we describe an approach using mineralized collagen(MC)graft to reconstruct acetabulum and femur with massive bone defects.We identified 89 patients suffering acetabular or femoral bone defects after primary THA,who required revision THA for this study.During the surgery,MC was applied to reconstruct both the acetabular and femoral defects.Harris hip score was used to evaluate hip function while radiographs were taken to estimate bone formation in the defect regions.The average follow-up period was 33.662.4 months.None of the components needed re-revised.Mean Harris hip scores were 42.563.5 before operation,75.264.0 at 10th month and 95.063.6 at the final follow-up.There were no instances of deep infection,severe venous thrombosis or nerve palsy.The present study demonstrated that MC graft can serve as a promising option for revision THA with massive bone deficiency.Meanwhile,extended follow-up is needed to further prove its long-term performance.Cheng Huang Liwu Qin Wei Yan Xisheng Weng Xiangjie Huang 2015Regenerative Biomaterials2015,2,4:12

11Renal Protective Activity of Hsian-tsao Extracts in Diabetic Rats显示文摘Objective To investigate the renal protective activity of Hsian-tsao Mesona procumbens Hemsl. water extracts in diabetic rats. Methods Thirty Sprague-dawley female rats were randomly divided into three groups (n=10 each), 'control group' with intraperitoneal saline injection, 'diabetic group' with 60 mg of intraperitoneal streptozotocin injection per kg of body weight and 'Hsian-tsao group' with intragastric administration of Hsian-tsao extraction everyday for 4 weeks after intraperitoneal streptozotocin injection. The body weight and blood sugar were measured before and after model induction in the three groups. Thrombospondin-1 (TSP-1) expressions in the kidney were monitored by immunohistochemistry. Kidney ultrastructural changes were also analyzed by using transmission electron microscopy. Results Before diabetic model induction, there were no significant differences among the three groups in body weight and blood sugar. Four weeks after the induction of diabetes, the differences became statistically significant. Electron microscopy also revealed disruption of the foot processes of the podocytes and other damages in diabetic group. These damages were significantly less severe in Hsian-tsao group when compared with the diabetic group. TSP-1 expressions in the kidney were significantly increased in both the diabetic group and Hsian-tsao group, but it was relatively lower in Hsian-tsao group than in diabetic group. Conclusion Our results showed that Hsian-tsao treatment in the diabetic rats effectively prevented the pathological alterations in the kidney and decreased the TSP-1 expression. 13Epidemiological and etiological investigation of dengue fever in the Fujian province of China during 2004–2014显示文摘Dengue fever(DF) is a vector-borne disease and a tremendous socioeconomic burden on tropical and subtropical countries worldwide. To explore the characteristics of DF epidemic in the Fujian province, information of DF cases in Fujian during 2004–2014 was collected and analyzed. The complete E genes of 48 viral isolates were amplified and sequenced for phylogenetic analysis. A total of 733 cases was reported, of which 612(83.5%) occurred during the peak period from August to October. Additionally, 76%(190/250) of imported cases originated from Southeast Asia countries, by the epidemiological investigation. Phylogenetic analysis of the 48 viral isolates revealed that three genotypes(I, IV, V) of DENV1, and one genotype each of DENV2(cosmopolitan) and DENV3(I) circulated in Fujian during 2004–2014. Similar to the results of the epidemiological investigations, the source of most of the viral isolates, including imported and indigenous cases, may be Southeast Asia countries; however, importation from adjacent provinces was also observed in recent years. Overall, DF is considered an imported epidemic disease in Fujian. 14Petrogenetic contrastive studies on the Mesozoic early stage ore-bearing and late stage ore-barren granites from the southern Anhui Province显示文摘Yanshanian magmatisms are intensive in the southern Anhui Province and can be divided into early(152–137 Ma)and late(136–122 Ma) stages. A Yanshanian granitic zone was found to crop out along Qingshan to Changgai areas in the Tunxi district in Field investigation which has a genetic link with molybdenum multiple metal mineralization. To be a representative syenitic granite in the southern Anhui Province, the Huangshan pluton has not been found so far to have any genetic link with mineralization. Zircon LA-ICP-MS dating indicate that the four granitic bodies from the Qingshan-Changgai zone have concurrent formed ages from 140±4 to 141±2 Ma, belonging to the Yanshanian early stage magmatism. However, the Huangshan granite is dated to be 129±2 Ma, belonging to the Yanshanian late stage magmatism. The Qingshan-Changgai granites show high SiO2 and K2 O contents, low P2O5 contents and middle Al2O3 contents and are high-K calc-alkaline series metaluminum I-type granite. These rocks are characterized by enrichments in the large ion lithophile elements and light rare earth elements(REE), depletions in the high field-strength elements, and middle degree negative anomalies of Eu, geochemical features of arc or continent crustal derived magma affinities. These rocks have 87Sr/86Sr(t) ratios from 0.7120 to 0.7125, εNd(t) values from.7.24 to.4.38 and zircon εHf(t) values of.4.4 to 6.7, similar to that of the coeval ore-bearing granodiorites in the southern Anhui Province. Integrated geochemical studies indicate that the Yanshanian ore-bearing granodiorites were formed by partial melting of the Meso-Neoproterozoic accreted thickened low crust. Meanwhile, the Qingshan-Changgai granites were formed through a AFC process of plagioclase+amphibole+Shangxi Group of magmas that formed the ore-bearing granodiorites. The Huangshan granites are characterized by high SiO2 and K2 O contents, moderate Al2O3 contents, seagull shape REE distributed pattern and distinct Eu negative abnormities. Comparing with the Qingshan-Changgai granites, the Huangshan granites show more Ba, Sr, P,and Ti negative abnormities with no Nb and Ta depletions and are high-K calc-alkaline series metaluminum A-type granite. εHf(t)values of the Huangshan granites are from.6.6 to.1.2, similar to that of the early stage ore-bearing granodiorites, indicating that they were also formed by anatexis of the Meso-Neoproterozoic accreted crust, but their magma sources might be residual granulitic crust which ever underwent Yanshanian early stage I-type intermediate-acid magma extraction. Comparing studies on the two stages granites indicate that the early stage granites derived from a relative thickened low crust under a lower temperature condition. Their magma sources were Meso-Neoproterozoic accreted crust which enriched in ore-forming materials and further became more enriched through processes of magma AFC evolution. However, the late stage A-type granites originated from relative shallow crust under a higher temperature condition. Their magma source was depleted in ore-forming materials due to the early stage magma extraction and thus had weak ore-forming capacity. 15Time-consistent investment-reinsurance strategies towards joint interests of the insurer and the reinsurer under CEV models显示文摘The present paper studies time-consistent solutions to an investment-reinsurance problem under a mean-variance framework.The paper is distinguished from other literature by taking into account the interests of both an insurer and a reinsurer jointly.The claim process of the insurer is governed by a Brownian motion with a drift.A proportional reinsurance treaty is considered and the premium is calculated according to the expected value principle.Both the insurer and the reinsurer are assumed to invest in a risky asset,which is distinct for each other and driven by a constant elasticity of variance model.The optimal decision is formulated on a weighted sum of the insurer’s and the reinsurer’s surplus processes.Upon a verification theorem,which is established with a formal proof for a more general problem,explicit solutions are obtained for the proposed investment-reinsurance model.Moreover,numerous mathematical analysis and numerical examples are provided to demonstrate those derived results as well as the economic implications behind.ZHAO Hui WENG ChengGuo SHEN Yang ZENG Yan 2017Science China Mathematics2017,60,2:9
16Liuwei Dihuang pill suppresses metastasis by regulating the wnt pathway and disrupting β-catenin/T cell factor interactions in a murine model of triple-negative breast cancer显示文摘OBJECTIVE:To investigate if the Liuwei Dihuang pill(LWDHP)can inhibit metastasis to the liver and lungs in mice bearing triple-negative breast cancer(TNBC),and the molecular mechanism underpinning this action.METHODS:Ninety-nine TNBC bearing-mice were distributed randomly to five groups:control(Con),paclitaxel(PTX),low-dose LWDHP(LLP,2.3 g·kg^-1·d^-1),middle-dose LWDHP(MLP,4.6 g·kg^-1·d^-1)and high-dose LWDHP(HLP,9.2 g·kg^-1·d^-1).The LWDHP were administered(p.o.)to the agonal stage.The morphology of BC cells was observed by hematoxylin&eosin staining.Expression of axin-2,β-catenin,T cell factor(TCF),cyclin-D1 and vascular endothelial growth factor(VEGF)was detected by western blotting or immunofluorescence.β-catenin/TCF-1 interaction was measured using a co-immunoprecipitation assay.RESULTS:After LWDHP treatment,metastasis of BC cells to the lungs and liver was inhibited,expression of axin-2 was increased,expression of TCF-1,β-catenin,cyclin-D1 and VEGF was decreased,andβ-catenin/TCF-1 interaction was disrupted.CONCLUSION:The LWDHP could inhibit metastasis of BC cells to the liver and lungs.The molecular mechanism underlying this action may be regulation of protein expression andβ-catenin/TCF-1 interactions in the Wnt pathway.Zheng Lixiang Zheng Qing Yu Zhipeng Wang Jian Ren Xiaoying Gong Yan Yang Xue Hunag Ping Weng Meizhi Liu Hongning Liu Haizhou 2019Journal of Traditional Chinese Medicine2019,39,6:9
17OsCIPK7 point-mutation leads to conformation and kinase-activity change for sensing cold response显示文摘Calcineurin B-like interacting protein kinases(CIPKs) play important roles via environmental stress.However, less is known how to sense the stress in molecular structure conformation level. Here, an Os CIPK7 mutant via TILLING procedure with a point mutation in the kinase domain showed increased chilling tolerance, which could be potentially used in the molecular breeding. 19Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
20Magneto-thermo-mechanical characterization of giant magnetostrictive materials显示文摘The variations of magnetization and magnetostriction with temperature and stress were investigated through the analysis of the effective field,induced by temperature and stress.A nonlinear magnetostrictive model of giant magnetostrictive materials was proposed.The proposed model can be used to calculate the magnetostrictive characterization of giant magnetostrictive materials in different temperatures and under different stresses.The coupling effects of axial stress,magnetic field,and temperature on the magnetostriction of a Terfenol-D rod were numerically simulated as well as experimentally tested.Comparison between the calculating and experimental results shows that the proposed model can better describe the magneto-thermo-mechanical characteristics of Terfenol-D rod under different temperatures and compressive stress.Therefore,the proposed model possesses an important significance for the design of magnetostrictive devices.Li Wang Bo-Wen Wang Zhi-Hua Wang Ling Weng Wen-Mei Huang Yan Zhou 2013Rare Metals2013,32,5:7
