| 3 | Arsenic trioxide alleviates acute graft-versus-host disease by modulating macrophage polarization显示文摘This study aimed to explore macrophage polarization in acute graft-versus-host disease after hematopoietic stem cell transplantation, and investigated if arsenic trioxide(ATO) could correct this imbalance. In the colon of GVHD mice, we found that the number of F4/80+iNOS+ cells as well as the expression intensity of TNF-α and IL-1β was greater in the GVHD group than in the BM group, whereas the number of F4/80+CD206+ cells and the expression intensity of IL-10 and TGF-β was greater in the BM group than in the GVHD group. We investigated the effect of ATO on GVHD mice, and found that ATO treatment clearly improved the survival of the mice and reduced the severity of GVHD. In addition, ATO reduced the number of F4/80+iNOS+ cells, and increased the number of F4/80+CD206+ cells in the colon of GVHD mice. Furthermore, ATO sharply decreased CD86 and CD80 expression, and increased CD163 and CD206 expression in macrophages induced from aGVHD patients. Therefore,ATO can modulate the M1 and M2 phenotype in GVHD mice or in macrophages from aGVHD patients. | 5 | Large-Scale Discovery of Non-conventional Peptides in Maize and Arabidopsis through an Integrated Peptidogenomic Pipeline显示文摘Non-conventional peptides(NCPs),which include small open reading frame-encoded peptides,play critical roles in fundamental biological processes.In this study,we developed an integrated peptidogenomic pipeline using high-throughput mass spectra to probe a customized six-frame translation database and applied it to large-scale identification of NCPs in plants.A total of 1993 and 1860 NCPs were unambiguously identified in maize and Arabidopsis,respectively.These NCPs showed distinct characteristics compared with conventional peptides and were derived from introns,3′UTRs,5′UTRs,junctions,and intergenic regions.Furthermore,our results showed that translation events in unannotated transcripts occur more broadly than previously thought.In addition,we found that dozens of maize NCPs are enriched within regions associated with phenotypic variations and domestication selection,indicating that they potentially are involved in genetic regulation of complex traits and domestication in maize.Taken together,our study developed an integrated peptidogenomic pipeline for large-scale identification of NCPs in plants,which would facilitate global characterization of NCPs from other plants.The identification of large-scale NCPs in both monocot(maize)and dicot(Arabidopsis)plants indicates that a large portion of plant genome can be translated into biologically functional molecules,which has important implications for functional genomic studies. | Shunxi Wang Lei Tian Haijun Liu Xiang Li Jinghua Zhang Xueyan Chen Xingmeng Jia Xu Zheng Shubiao Wu Yanhui Chen Jianbing Yan Liuji Wu | 2020 | Molecular Plant2020,13,7: | 4 |

| 7 | Climate Characteristics of Abnormal Double-Blocking Activities over the Ural Mountains and Sea of Okhotsk显示文摘By using NCEP–NCAR daily reanalysis data for June–August of 1948–2009, the synoptic situation of the doubleblocking high pressure process over the Ural Mountains–Sea of Okhotsk region was identified objectively, and the climatic characteristics and dynamic mechanism during the double blockings were also investigated. The results suggest that the Urals–Okhotsk double-blocking high experienced obviously an abrupt change around 1977. After the change, the occurrence frequency and number of sustained days of the blocking high reduced significantly.Moreover, the zonal spacing between the Urals blocking and the Okhotsk blocking also decreased, while their meridional locations did not change much. The double blockings became strengthened in general, and the blocking high over the Sea of Okhotsk extended northward evidently. However, during the process without double-blocking activities, the intensity and location of the double-blocking high showed no noticeable variation before and after the abrupt change. In addition, dynamic diagnosis shows that after the abrupt change, the energy of the double-blocking high mainly propagated vertically from low to high levels, and transferred horizontally from low to high latitudes; whereas, previously, before the abrupt change, this occurred from high to low latitudes. Meanwhile, the development of a low pressure system was fueled between the double blockings, which meant that the double-blocking activity was beneficial to the maintenance and development of the Northeast China cold vortex. | 9 | Chemical composition of walnuts from three regions in China显示文摘To reveal the characteristic chemicals of walnuts from different origins, we analyzed fatty acid composition,tocopherols, phytosterols and total phenolic content (TPC) of walnuts from three main producing regions inChina. The results showed that walnuts were rich in polyunsaturated fatty acids, and the ratio of ω-6 to ω-3 fattyacids was close to the recommendation of Chinese Nutrition Society. Moreover, walnuts contain high contents oftocopherols (331.20–414.71 mg/kg), phytosterols (97.17–110.35 mg/100 g) and phenols (38.51–48.08 mg GAE/kg). Significant chemical differences exist among walnuts from three production regions. The highest content ofpolyunsaturated fatty acids was found in walnuts from the northern China, the highest content of tocopherols inwalnuts from southwest China, and the highest contents of phytosterol and TPC in walnuts from northwest China.However, there was no significant difference in the tocopherol, phytosterol and TPC content of walnuts betweenthe Northern China and Northwest China. | 13 | Inhibition of the maize salt overly sensitive pathway by ZmSK3 and ZmSK4显示文摘Soil salinity is a worldwide problem that adversely affects plant growth and crop productivity. The salt overly sensitive (SOS) pathway is evolutionarily conserved and essential for plant salt tolerance. In this study, we reveal how the maize shaggy/glycogen synthase kinase 3-like kinases ZmSK3 and ZmSK4, orthologs of brassinosteroid insensitive 2 in Arabidopsis thaliana, regulate the maize SOS pathway. ZmSK3 and ZmSK4 interact with and phosphorylate ZmSOS2, a core member of the maize SOS pathway. The mutants defective in ZmSK3 or ZmSK4 are hyposensitive to salt stress, with higher salt-induced activity of ZmSOS2 than that in the wild type. Furthermore, the Ca^(2+) sensors ZmSOS3 and ZmSOS3-like calcium binding protein 8 (ZmSCaBP8) activate ZmSOS2 to maintain Na^(+)/K^(+) homeostasis under salt stress and may participate in the regulation of ZmSOS2 by ZmSK3 and ZmSK4. | 20 | Three Novel Mutations of APC Gene Found in a Chinese Family with Familial Adenomatous Polyposis显示文摘Objective:To identify the causative adenomatous polyposis coli(APC)gene defects associated with a pedigree of familial adenomatous polyposis(FAP).Methods:FAP was diagnosed based on clinical manifestations,family history,as well as endoscopic and pathological examinations.The blood samples of the FAP pedigree members,colonic polyp patients,and normal individuals were collected.Genomic DNA was then extracted from those samples.APC mutation analysis was conducted via direct polymerase chain reaction(PCR)sequencing.Results:Three synonymous mutations and a missense mutation were found:c.5034G>A(p.Glyl678Gly),c.5465T>A(p.Vall 822Asp),c.5880G>A(p.Prol960Pro),and c.5274T>G(p.Serl758Ser)・Among them,the homozygous mutation on APC gene c.5034G>A has been reported,while the other three mutations have not been reported in the Chinese Han population.Individuals with c.5465T>A(p.Vall822ASP)missense mutation eventually suffer from colon cancer and have poor prognosis.We found no mutation in patients with simple intestinal polyp and in normal individuals.In addition,there were homozygous and heterozygous mutations in different patients from the same family.Conclusion:Three new mutations of APC gene were firstly reported in Han population.The missense mutation of c.5465T>A(p.Vall 822Asp)may be the cause of carcinogenesis in this FAP pedigree with poor prognosis. | Shujuan Gao Min Lin Yan Jin Zhuona Wang Yunqing Zhu Guisheng Liu Xueyan Guo | 2022 | Journal of Clinical and Nursing Research2022,6,3: | 0 |
