bHLH121 Functions as a Direct Link that Facilitates the Activation of FIT by bHLH IVc Transcription Factors for Maintaining Fe Homeostasis in Arabidopsis显示文摘Iron(Fe)deficiency is prevalent in plants grown in neutral or alkaline soil.Plants have evolved sophisticated mechanisms that regulate Fe homeostasis, ensuring survival. In Arabidopsis,FER-LIKE IRON DEFICIENCY-INDUCED TRANSCRIPTION FACTOR(FIT)is a crucial regulator of Fe-deficiency response.FIT is activated indirectly by basic helix-loop-helix(bHLH)IVc transcription factors(TFs)under Fed eficiency;how ever,it remains unclear which protein(s)act as the linker to mediate the activation of FIT by bHLH IVc TFs.In this study,we characterize the functions of bHLH121 and demonstrate that it directly associates with the FIT promoter. We found that loss-of-function mutations of bHLH121 cause severe Fedeficiency symptoms, reduced Feaccum ulation, and disrupted expression of genes associated with Fehomeostasis.Genetic analysis showed that FIT is epistatic to bHLH121 and FIT overexpression partially rescues the bhlh121 mutant.Further investigations revealed that bHLH IVc TFs interact with and promote nuclear accumulation of bHLH121.We demonstrated that bHLH121 has DNA-binding activity and can bind the prom oters of the FIT and bHLHlb genes, but we did not find that it has either direct transcriptional activation or repression activity tow ard these genes.Meanw hile,we found that bHLH121 functions downstream of and is a direct target of bHLH IVc TFs,and its expression is induced by Fe deficiency in a bHLH IV c-dependent manner.Taken together, these results establish that bHLH121 functions together with bHLH IVc TFs to positively regulate the expression of FIT and thus plays a pivotal role in maintaining Fe homeostasis in Arabidopsis. | Rihua Lei Yang Li Yuerong Cai Chenyang Li Mengna Pu Chengkai Lu Yujie Yang Gang Liang | 2020 | Molecular Plant2020,13,4: | 14
Generation of a Hutchinson-Gilford progeria syndrome monkey model by base editing显示文摘Many human genetic diseases, including Hutchinson-Gilford progeria syndrome(HGPS),are caused by single point mutations.HGPS is a rare disorder that causes premature aging and is usually caused by a de novo point mutation in the LMNA gene.Base editors(BEs)composed of a cytidine deaminase fused to CRISPR/Cas9 nickase are highly efficient at inducing C to T base conversions in a programmable manner and can be used to generate animal disease models with single amino-acid substitutions. Here,we generated the first HGPS monkey model by delivering a BE mRNA and guide RNA(gRNA)targeting the LMNA gene via microinjection into monkey zygotes.Five out of six newborn monkeys carried the mutation specifically at the target site.HGPS monkeys expressed the toxic form of lamin A,progerin, and recapitulated the typical HGPS phenotypes including growth retardation, bone alterations, and vascular abnormalities. Thus, this monkey model genetically and clinically mimics HGPS in humans, demonstrating that the BE system can efficiently and accurately generate patient-specific disease models in non-human primates. | Fang Wang Weiqi Zhang Qiaoyan Yang Yu Kang Yanling Fan Jingkuan Wei Zunpeng Liu Shaoxing Dai Hao Li Zifan Li Lizhu Xu Chu Chu Jing Qu Chenyang Si Weizhi Ji Guang-Hui Liu Chengzu Long Yuyu Niu | 2020 | Protein & Cell2020,11,11: | 11
Wireless whispering-gallery-mode sensor for thermal sensing and aerial mapping显示文摘The Internet of Things(IoT)1,2 employs a large number of spatially distributed wireless sensors to monitor physical environments,e.g.,temperature,humidity,and air pressure, and has many applications, including environmental monitoring3,health care monitoring4,smart cities5,and precision agriculture.A wireless sensor can collect, analyze, and transmit measurements of its environment1,2.Currently,wireless sensors used in the IoT are predominately based on electronic devices that may suffer from electromagnetic interference in many circumstances.Being immune to the electromagnetic interference, optical sensors provide a significant advantage in harsh environments6.Furthermore,by introducing optical resonance to enhance light–matter interactions, optical sensors based on resonators exhibit small footprints, extreme sensitivity, and versatile functionalities7,8,which can significantly enhance the capability and flexibility of wireless sensors. Here we provide the first demonstration of a wireless photonic sensor node based on a whisperinggallery-mode(WGM)optical resonator,in which light propagates along the circular rim of such a structure like a sphere,a disk,or a toroid by continuous total internal reflection. The sensor node is controlled via a customized iOS app.Its performance was studied in two practical scenarios:(1)real-time measurement of the air temperature over 12 h and(2)aerial mapping of the temperature distribution using a sensor node mounted on an unmanned drone. Our work demonstrates the capability of WGM optical sensors in practical applications and may pave the way for the large-scale deployment of WGM sensors in the IoT. | Xiangyi Xu Weijian Chen Guangming Zhao Yihang Li Chenyang Lu Lan Yang | 2018 | Light(Science & Applications)2018,7,1: | 8
Zircon SHRIMP U–Pb age of Late Jurassic OIB-type volcanic rocks from the Tethyan Himalaya: constraints on the initial activity time of the Kerguelen mantle plume显示文摘This work presents zircon U–Pb age and wholerock geochemical data for the volcanic rocks from the Lakang Formation in the southeastern Tethyan Himalaya and represents the initial activity of the Kerguelen mantle plume. SHRIMP U–Pb dating of zircons from the volcanic rocks yielded a ^(206) Pb/^(238) U age of 147 ± 2 Ma that reflects the time of Late Jurassic magmatism. Whole rock analyses of major and trace elements show that the volcanic rocks are characterized by high content of Ti O_2(2.62 wt%–4.25 wt%) and P_2O_5(0.38 wt%–0.68 wt%), highly fractionated in LREE/HREE [(La/Yb)N= 5.35–8.31] with no obvious anomaly of Eu, and HFSE enrichment with no obvious anomaly of Nb and Ta, which are similar to those of ocean island basalts and tholeiitic basaltic andesites indicating a mantle plume origin. The Kerguelen mantle plume produced a massive amount of magmatic rocks from Early Cretaceous to the present, which widely dispersed from their original localities of emplacement due to the changing motions of the Antarctic, Australian, and Indian plates. However, our new geochronological and geochemical results indicate that the Kerguelen mantle plume started from the Late Jurassic. Furthermore, we suggest that the Kerguelen mantle plume may played a significant role in the breakup of eastern Gondwanaland according to the available geochronological, geochemical and paleomagnetic data. | Yuruo Shi Chenyang Hou J. | 5 | A haplotype block associated with thousand-kernel weight on chromosome 5DS in common wheat(Triticum aestivum L.)显示文摘Spike number per unit area, number of grains per spike, and thousand-kernel weight(TKW) are important yield components for wheat(Triticum aestivum L.). TKW has the highest heritability among the three components. We validated 27 simple sequence repeat(SSR) loci associated with TKW in an F2:5breeding population grown in four environments. A cfd78265 bpmarker on chromosome 5DS showed the strongest association with TKW and had a significantly positive effect on TKW compared to allele cfd78259 bp, with mean increases of 5.17, 3.63, 4.11, and 5.16 g in the four environments. Markers cfd67 and cfd40 flanking cfd78 also showed significantly positive associations with TKW with increases of 5.11, 3.29, 4.31, and 4.50 g for cfd67205, and4.98, 3.49, 4.06, and 4.84 g for cfd40187 compared with cfd67203 and cfd40190in the four environments, respectively. A major quantitative trait locus for TKW spanning 2.94 c M on chromosome 5DS was detected by association mapping.Strong linkage disequilibrium(LD)(r2> 0.2) was detected Resear among the three linked markers, which formed three haplotype blocks in the F2:5breeding population. Mean TKW differences between Hap B-I and Hap B-II were 5.80, 4.41, 4.02,and 5.06 g in the four environments, respectively. Moreover, significant LD was detected only between cfd78 and cfd67 and between cfd67 and cfd40 in a germplasm collection. | 6 | Oryza sativa FER-LIKE FE DEFICIENCY-INDUCED TRANSCRIPTION FACTOR(OsFIT/OsbHLH156)interacts with OsIRO2 to regulate iron homeostasis显示文摘Iron(Fe)is indispensable for the growth and development of plants. It is well known that FER-LIKE FE DEFICIENCY-INDUCED TRANSCRIPTION FACTOR(FIT)is a key regulator of Fe uptake in Arabidopsis. Here,we identify the Oryza sativa FIT(also known as Osb HLH156)as the interacting partner of IRON-RELATED BHLH TRANSCRIPTION FACTOR 2(OsIRO2)that is critical for regulating Fe uptake. The OsIRO2 protein is localized in the cytoplasm and nucleus, but OsFIT facilitates the accumulation of OsIRO2 in the nucleus.Loss-of-function mutations of OsFIT result in decreased Fe accumulation, severe Fe-deficiency symptoms, and disrupted expression of Feuptake genes. In contrast,OsFIT overexpression promotes Fe accumulation and the expression of Fe-uptake genes.Genetic analyses indicate that OsFIT and OsIRO2 function in the same genetic node.Further analyses suggest that OsFIT and OsIRO2 form a functional transcription activation complex to initiate the expression of Fe-uptake genes. Our findings provide a mechanism understanding of how rice maintains Fe homeostasis. | Gang Liang Huimin Zhang Yang Li Mengna Pu Yujie Yang Chenyang Li Chengkai Lu Peng Xu Diqiu Yu | 2020 | Journal of Integrative Plant Biology2020,62,5: | 4
Tailoring the Meso-Structure of Gold Nanoparticles in Keratin-Based Activated Carbon Toward High-Performance Flexible Sensor显示文摘Flexible biosensors with high accuracy and reliable operation in detecting pH and uric acid levels in body fluids are fabricated using well-engineered metaldoped porous carbon as electrode material. The gold nanoparticles@N-doped carbon in situ are prepared using wool keratin as both a novel carbon precursor and a stabilizer. The conducting electrode material is fabricated at 500℃ under customized parameters, which mimics A-B type(two different repeating units) polymeric material and displays excellent deprotonation performance(pH sensitivity). The obtained pH sensor exhibits high pH sensitivity of 57 mV/pH unit and insignificant relative standard deviation of 0.088%.Conversely,the composite carbon material with sp^2 structure prepared at 700℃ is doped with nitrogen and gold nanoparticles, which exhibits good conductivity and electrocatalytic activity for uric acid oxidation. The uric acid sensor has linear response over a range of 1-150 μM and a limit of detection 0.1 μM.These results will provide new avenues where biological material will be the best start, which can be useful to target contradictory applications through molecular engineering at mesoscale. | Aniruddha BPatil Zhaohui Meng Ronghui Wu Liyun Ma Zijie Xu Chenyang Shi Wu Qiu Qiang Liu Yifan Zhang Youhui Lin Naibo Lin Xiang Yang Liu | 2020 | Nano-Micro Letters2020,12,9: | 3
Selective electrocatalytic CO2 reduction enabled by SnO2 nanoclusters显示文摘The development of high-performance electrocatalysts holds the decisive key to the electrochemical CO2 reduction toward value-added products. Formic acid or formate is a desirable reduction product, but its selective production is often challenging. Tin based-materials have attracted great attention for formate production, and yet their performances are far from satisfactory. In this study, we reported the preparation of SnO2 nanoclusters from the controlled self-polymerization of dopamine together with SnO32-, followed by the mild-temperature calcination. The final product consisted of large primary particles that were further made of small secondary SnO2 nanocrystals. When evaluated as the electrocatalyst for CO2 reduction in 0.5 M NaHCO3, our material exhibited impressive activity, selectivity and stability for the selective CO2 reduction to formate. A peak formate Faradaic efficiency of^73% and large partial current density of 16.3 mA/cm2 was achieved at -0.92 V versus reversible hydrogen electrode. | Hui Yang Yang Huang Jun Deng Yunling Wu Na Han Chenyang Zha Leigang Li Yanguang Li | 2019 | Journal of Energy Chemistry2019,28,10: | 3
Two-dimensional multimetallic sulfide nanosheets with multi-active sites to enhance polysulfide redox reactions in liquid Li2S6-based lithium-polysulfide batteries显示文摘The lithium-sulfur battery has attracted enormous attention as being one of the most significant energy storage technologies due to its high energy density and cost-effectiveness.However,the 'shuttle effect' of polysulfide intermediates represents a formidable challenge towards its wide applications.Herein,we have designed and synthesized two-dimensional Cu,Zn and Sn-based multimetallic sulfide nanosheets to construct multi-active sites for the immobilization and entrapment of polysulfides with offering better performance in liquid Li2S6-based lithium-polysulfide batteries. Both experimental measurements and theoretical computations demonstrate that the interfacial multi-active sites of multimetallic sulfides not only accelerate the multi-chained redox reactions of highly diffusible polysulfides, but also strengthen affinities toward polysulfides.By adopting multimetallic sulfide nanosheets as the sulfur host, the liquid Li2 S6-based cell exhibits an impressive rate capability with 1200 mAh/g and retains 580 mAh/g at 0.5 mA/cm^(2) after 1000 cycles. With high sulfur mass loading conditions, the cell with 2.0 mg/cm^(2) sulfur loading delivers a cell capacity of 1068 mAh/g and maintains 480 mAh/g with 0.8 mA/cm^(2) and 500 cycles. This study provides new insights into the multifunctional material design with multi-active sites for elevated lithium-polysulfide batteries. | Chenyang Zha Donghai Wu Yuwei Zhao Jun Deng Jinghua Wu Rong Wu Meng Yang Lin Wang Houyang Chen | 2021 | Journal of Energy Chemistry2021,30,1: | 3

Targeted therapeutics and novel signaling pathways in nonalcohol-associated fatty liver/steatohepatitis(NAFL/NASH)显示文摘Non-alcohol-associated fatty liver/steatohepatitis(NAFL/NASH)has become the leading cause of liver disease worldwide.NASH,an advanced form of NAFL,can be progressive and more susceptible to developing cirrhosis and hepatocellular carcinoma.Currently, lifestyle interventions are the most essential and effective strategies for preventing and controlling NAFL without the development of fibrosis. While there are stll limited appropriate drugs specifically to treat NAFL/NASH,growing progress is being seen in elucidating the pathogenesis and identifying therapeutic targets. In this review,we discussed recent developments in etiology and prospective therapeutic targets,as well as pharmacological candidates in pre/clinical trials and patents, with a focus on diabetes, hepatic lipid metabolism, inflammation, and fibrosis.Importantly, growing evidence elucidates that the disruption of the gut-liver axis and microbederived metabolites drive the pathogenesis of NAFL/NASH.Extracellular vesicles(EVs)act as a signaling mediator, resulting in lipid accumulation, macrophage and hepatic stellate cell activation, further promoting inflammation and liver fibrosis progression during the development of NAFL/NASH.Targeting gut microbiota or EVs may serve as new strategies for the treatment of NAFL/NASH.Finally,other mechanisms, such as cell therapy and genetic approaches, also have enormous therapeutic potential.Incorporating drugs with different mechanisms and personalized medicine may improve the efficacy to better benefit patients with NAFL/NASH. | Xiaohan Xu Kyle L.Poulsen Ljuan Wu Shan Liu Tatsunori Miyata Qiaoling Song Qingda Wei Chenyang Zhao Chunhua Lin Jinbo Yang | 2022 | Signal Transduction and Targeted Therapy2022,7,9: | 2
A Sub-100 mg Electromagnetically Driven Insect-inspired Flapping-wing Micro Robot Capable of Liftoff and Control Torques Modulation显示文摘Inspired by the unique, agile and efficient flapping flight of insects,we present a novel sub-100 mg,electromagnetically driven,tailless,flapping-wing micro robot. This robot utilizes two optimized electromagnetic actuators placed back to back to drive two wings separately, then kinematics of each wing can be independently controlled, which gives the robot the ability to generate all three control torques of pitch, roll and yaw for steering. To quantify the performance of the robot,a simplified aerodynamic model is used to estimate the generated lift and torques, and two customized test platforms for lift and torque measurement are built for this robot. The mean lift generated by the robot is measured to be proportional to the square of the input voltage amplitude. The three control torques are measured to be respectively proportional to three decoupled parameters of the control voltages, therefore the modulation of three control torques for the robot is independent, which is helpful for the further controlled flight. All these measured results fit well with the calculated results of the aerodynamic model.Furthermore, with a total weight of 96 mg and a wingspan of 3.5 cm,this robot can generate sufficient lift to take off. | Chenyang Wang Weiping Zhang Yang Zou Ran Meng Jiaxin Zhao Mingchen Wei | 2020 | Journal of Bionic Engineering2020,17,6: | 2
Photoacoustic detection of follicular thyroid carcinoma using targeted Nano-Au-Tripods显示文摘Follicular thyroid carcinoma(FTC)is the second most common form of thyroid malignancy, and it is associated with more aggressive growth and worse long-term survival outcomes relative to papillary thyroid carcinoma(PTC).Reliable approaches to preoperative FTC detection, however, remain to be established.Herein,a targeted Affibody-Au-Tripod nanoprobe was developed and successfully utilized to facilitate the targeted photoacoustic imaging(PAI)of epidermal growth factor receptor(EGFR)-positive cells and tumors. These Affibody-Au-Tripods were found to be highly sensitive and specific for cells expressing EGFR when used as a PA contrast agent in vitro, and studies conducted in an FTC-133 subcutaneous tumor model system in mice further revealed that these Affibody-Au-Tripods were able to specifically target these EGFR-expressing tumors while providing a strong photoacoustic signal in vivo.Importantly, these nanoprobes exhibited negligible cytotoxicity and robust chemical and physical stability, making Affibody-Au-Tripods promising candidates for targeted PAI-based FTC diagnosis. In addition, these nanoprobes have the potential to facilitate the individualized treatment of patients harboring EGFRpositive tumors. | Yang Gui Kai Cheng Ruojiao Wang Sirui Liu Chenyang Zhao Rui Zhang Ming Wang Zhen Cheng Meng Yang | 2022 | Chinese Journal of Chemical Engineering2022,35,4: | 2
Enhanced ionic diffusion interface in hierarchical metal-organic framework@layered double hydroxide for high-performance hybrid supercapacitors显示文摘Layered double hydroxides(LDHs)with abundant accessible active sites are promising electrode materials for hybrid supercapacitor(HSC)due to their ultrahigh theoretical capacitances. However, the structural agglomeration of LDH leads to poor rate capability and durability.Herein,we construct a diffusion-controlled interface in hierarchical architecture of metal-organic framework(MOF)HKUST-1@cobalt-nickel LDH(denoted as HKUST-1@CoNiLDH)through an in situ etching/electro-deposition strategy. The rapid charge transfer and ionic diffusion in HKUST-1@CoNiLDH deliver a remarkable specific capacity of 297.23 mAh·g^(−1) at 1 A·g^(−1),superior to mostly reported LDH-based electrodes.More importantly, the as-prepared HKUST-1@CoNiLDH//activated carbon HSC exhibit a high energy density of 39.8 Wh·kg^(−1) at a power density of 799.9 W·kg^(−1) with an outstanding capacitance retention of 90%after 5,000 charge–discharge cycles. The in-depth understanding of the ionic diffusion among the MOF/LDH interfaces will greatly promote the further development of designing and synthesizing high performance energy conversion and storage devices. | Yanan Zhang Junlei Chen Chenyang Su Keyao Chen Huabin Zhang Yuhao Yang Wenhuan Huang | 2022 | Nano Research2022,15,10: | 2

A rice XANTHINE DEHYDROGENASE gene regulates leaf senescence and response to abiotic stresses显示文摘Xanthine dehydrogenase, a member of the molybdenum enzyme family, participates in purine metabolism and catalyzes the generation of ureides from xanthine and hypoxanthine. However, the mechanisms by which xanthine dehydrogenase affects rice growth and development are poorly understood. In the present study, we identified a mutant with early leaf senescence and reduced tillering that we named early senescence and less-tillering 1(esl1). Map-based cloning revealed that ESL1 encodes a xanthine dehydrogenase, and it was expressed in all tissues. Chlorophyll content was reduced and chloroplast maldevelopment was severe in the esl1 mutant. Mutation of ESL1 led to decreases in allantoin, allantoate, and ABA contents. Further analysis revealed that the accumulation of reactive oxygen species in esl1 resulted in decreased photosynthesis and impaired chloroplast development, along with increased sensitivity to abscisic acid and abiotic stresses. Ttranscriptome analysis showed that the ESL1 mutation altered the expression of genes involved in the photosynthesis process and reactive oxygen species metabolism. Our results suggest that ESL1 is involved in purine metabolism and the induction of leaf senescence. These findings reveal novel molecular mechanisms of ESL1 gene-mediated plant growth and leaf senescence. | Jiangmin Xu Chenyang Pan Han Lin Hanfei Ye Sheng Wang Tao Lu Qianyu Chen Kairu Yang Mei Lu Qian Qian Deyong Ren Yuchun Rao | 2022 | The Crop Journal2022,10,2: | 1
