1Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 5Neural Stem Cells Transfected with Leukemia Inhibitory Factor Promote Neuroprotection in a Rat Model of Cerebral Ischemia显示文摘Leukemia inhibitory factor(LIF) contributes to the neuroprotection by neural stem cells(NSCs) after ischemic stroke. Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. 6Breast cancer-specific serum peptide profiles显示文摘Early detection of breast cancer is paramount to successful clinical therapy. Yet, early-stage breast cancer lacks specific symptoms or biomarkers. With the emerging of the mass spectrometric (MS) -based signatures as biomarkers, we investigated breast cancer-related serum profile pattern through class prediction and independent validation, and used Fourier transfer MS to identify breast cancer signature. We now show a distinctive serum peptide pattern that discriminates breast cancer from healthy controls with 93.2% sensitivity and 95.4% specificity. m/z 5901.70 and 4465.74 of ion fragment of FPA and alpha1-antichymotrypsin are found in the signatures that predominantly discriminate breast cancer from healthy individuals. 7Essential sequence of the N-terminal cytoplasmic localization-related domain of huntingtin and its effect on huntingtin aggregates显示文摘Huntington's disease(HD) is caused by abnormal CAG repeat expansion in the 5′-end of the Huntingtin(HTT) gene.In addition to the canonical C-terminal full-length huntingtin(htt) nuclear export signal,a cytoplasmic localization-related domain(CLRD) in the N-terminus of htt has recently been reported.Here,we analyzed this domain by introducing deletion and substitution mutations in a truncated N-terminal htt protein and subsequently monitored htt expression,aggregation and subcellular localization by immunocytochemistry and Western blot analysis.We demonstrated that Htt4-17 was the essential sequence for htt cytoplasmic localization.We also found that the subcellular distribution of htt was altered when Htt1-17 was mutated to contain amino acids of different charges,suggesting a structural requirement of Htt1-17 for the cytoplasmic localization of htt. 9Scale invariant features extraction for stereo vision显示文摘Stable local feature detection is a fundamental component of many stereo vision problems such as 3-D reconstruction, object localization, and object tracking. A robust method for extracting scale-invariant feature points is presented. First, the Harris corners in three-level pyramid are extracted. Then, the points detected at the highest level of the pyramid are correctly propagated to the lower level by pyramid based scale invariant (PBSI) method. The corners detected repeatedly in different levels are chosen as ffnal feature points. Finally, the characteristic scale is obtained based on maximum entropy method. 10Telmisartan prevents high-fat diet-induced hypertension and decreases perirenal fat in rats显示文摘We sought to investigate the effects of telmisartan on high-fat diet-induced hypertension and to explore the possible underlying mechanisms.Rats receiving high-fat diet were randomly divided into two groups,the telmisartan group(n = 9) and the high-fat diet group(n = 10).The control group consisted of age-matched rats on a regular diet(n = 10).At the end of the treatment,the body weight,blood pressure,insulin sensitivity and serum adiponectin levels of all rats were examined,and their visceral fat was extracted and weighed.Our results showed that telmisartan improved insulin resistance and dyslipidemia and increased serum adiponectin levels.Telmisartan also lowered both systolic blood pressure and diastolic blood pressure,and decreased the accumulation of perirenal fat associated with high-fat diet.Furthermore,telmisartan increased adiponectin mRNA expression in the perirenal fat.Correlation analysis showed that both systolic blood pressure and diastolic blood pressure were positively correlated with perirenal fat.These effects of telmisartan may be mediated through decreases in perirenal fat and contributed to the improvement of perirenal fat function.Our findings suggested a strong link between perirenal fat and high-fat diet-induced hypertension,and identified telmisartan as a potential drug for the treatment of obesity-related hypertension.Yaping Wang Yan Song Meng Suo Xin Jin Gang Tian 2012The Journal of Biomedical Research2012,26,3:3

13Effect of amlodipine combined with telmisartan or compound amiloride on plasma ET-1 concentration and left ventricular diastolic function of essential hypertension patients with left ventricular hypertrophy显示文摘Objective: To observe the effect of different combination antihypertensive therapies on plasma endothelin-1(ET-1) concentration and left ventricular diastolic function of essential hypertension patients with left ventricular hypertrophy(LVH). Methods: A total of 111 patients with essential hypertension were selected, including 60 cases(34 males, 26 females) aged(60.7±5.6) years with simple hypertension, and 51 cases(28 males, 23 females) aged(61.8±7.0) years with essential hypertension complicated with LVH. Essential hypertension patients with LVH were randomly divided into the group of amlodipine(2.5 mg/d) combined with telmisartan(40 mg/d, n=26) and the group of amlodipine(2.5 mg/d) combined with amiloride(half tablet/d, n=25),and the treatment lasted for 1 year. Echocardiography was performed before and after the treatment. Left ventricular mass index(LVMI), left ventricular isovolumic relaxation time(IVRT) and other indicators were detected, and plasma ET-1 concentrations were measured for comparative analysis. And 56 patients(31 males, 25 females) aged(59.3±6.7) years with normal blood pressure in the same period in our hospital were selected as the normal control group. Results: The general clinical characteristics were similar between hypertensive LVH group, simple hypertensive group and normal healthy control group. Plasma ET-1 concentrations, LVMI and IVRT of hypertensive LVH group were significantly higher than those of normal control group and simple hypertension group, and the difference was statistically significant. The patients' baseline blood pressure, ET-1, LVMI, interventricular septal thickness(IVST), left ventricular posterior wall thickness(LVPWT), left ventricular end-diastolic diameter(LVEDd) and other clinical parameters showed no significant difference(P>0.05) between the group of amlodipine combined with telmisartan and the group of amlodipine combined with amiloride. It was found that compared with that before treatment, blood pressure could be effectively controlled(P<0.05), and LVMI, IVST, LVPWT and IVRT(P<0.05) were all lowered, and ET-1(P<0.01) was significantly reduced after 1 year of antihypertensive therapy in both the group of amlodipine combined with telmisartan and the group of amlodipine combined with amiloride. The group of amlodipine combined with telmisartan was better in lowering blood pressure and reducing LVMI, IVST, LVPWT, IVRT and ET-1 than the group of amlodipine combined with amiloride(P<0.05). 14All in one theranostic nanoplatform enables efficient anti-tumor peptide I delivery for triple-modal imaging guided cancer therapy显示文摘Developing a reliable system to efficiently and safely deliver peptide drugs into tumor tissues still remains a great challenge since the instability of peptide drugs and low ability to traverse the cell membrane. Herein, we constructed a multifunctional nanoplatform based on porous europium/gadolinium (Eu/Gd)-doped NaLa(MoO4)2 nanoparticles (NLM NPs) to deliver antitumor peptide of B-cell lymphoma/leukemia-2-like protein 11 (BIM) for cancer therapy. The porous NLM NPs exhibited inherent photoluminescent, magnetic and X-ray absorbable properties, which enable them for triple-modal bioimaging, including fluorescence, magnetic resonance imaging (MRI) and computed tomography (CT). This triple-modal bioimaging can contribute to monitoring NLM NPs biodistribution and guiding therapy in vitro and in vivo. Furthermore, the NLM NPs showed negligible cytotoxicity in vitro and tissue toxicity in vivo. Importantly, NLM NPs could load the antitumor peptide of BIM and efficiently improve the resistance of peptide drugs to proteolysis. The BIM peptide was efficiently delivered into the tumor cells by NLM NPs, which can inhibit the growth and promote the apoptosis of cancer cells in vitro, significantly inhibit the tumor growth in vivo. Notably, NLM-BIM theranostic nanoplatform exhibits low systemic toxicity and fewer side effects in vivo. 15Impact of Lithium-Ion Coordination on Lithium Electrodeposition显示文摘The lithium dendrite and parasitic reactions are two major challenges for lithium(Li)metal anode—the most promising anode materials for high-energy-density batteries.In this work,both the dendrite and parasitic reactions that occurred between the liquid electrolyte and Li-metal anode could be largely inhibited by regulating the Li+-solvation structure.The saturated Li+-solvation species exist in commonly used LiPF 6 liquid electrolyte that needs extra energy to desolvation during Li-electrodeposition.Partial solvation induced high-energy state Li-ions would be more energy favorable during the electron-reduction process,dominating the competition with solvent reduction reactions.The Li-symmetric cells that are cycling at higher temperatures show better performance;the cycled lithium metal anode with metallic lustre and the dendrite-free surface is observed.Theoretical calculation and experimental measurements reveal the existence of high-energy state Li+-solvates species,and their concentration increases with temperature.This study provides insight into the Li+-solvation structure and its electrodeposition characteristics.Li Sheng Yanzhou Wu Jiekang Tian Li Wang Jianlong Wang Yaping Tang Hong Xu Xiangming He 2023Energy & Environmental Materials2023,6,1:2

17Amorphization activated FeB_(2) porous nanosheets enable efficient electrocatalytic N_(2) fixation显示文摘Designing active,robust and cost-effective catalysts for the nitrogen reduction reaction(NRR) is of paramount significance for sustainable electrochemical NH3 synthesis.Transition-metal diborides(TMB_2)have been recently theoretically predicted to be a new class of potential NRR catalysts,but direct experimental evidence is still lacking.Herein,we present the first experimental demonstration that amorphous FeB_2 porous nanosheets(a-FeB_2 PNSs) could be a highly efficient NRR catalyst,which exhibited an NH3 yield of 39.8 μg h^(-1) mg^(-1)(-0.3 V) and a Faradaic efficiency of 16.7%(-0.2 V),significantly outperforming their crystalline counterpart and most of existing NRR catalysts.First-principle calculations unveiled that the amorphization could induce the upraised d-band center of a-FeB_2 to boost d-2π~* coupling between the active Fe site and ~*N_2 H intermediate,resulting in enhanced ~*N_2 H stabilization and reduced reaction barrier.Out study may facilitate the development and understanding of earth-abundant TMB_2-based catalysts for electrocatalytic N_2 fixation.Ke Chu Weicong Gu Qingqing Li Yaping Liu Ye Tian Wuming Liu 2021Journal of Energy Chemistry2021,30,2:2

19Ribonuclease H-like gene SMALL GRAIN2 regulates grain size in rice through brassinosteroid signaling pathway显示文摘Grain size is a key agronomic trait that determines the yield in plants.Regulation of grain size by brassinosteroids(BRs)in rice has been widely reported.However,the relationship between the BR signaling pathway and grain size still requires further study.Here,we isolated a rice mutant,named small grain2(sg2),which displayed smaller grain and a semi-dwarf phenotype.The decreased grain size was caused by repressed cell expansion in spikelet hulls of the sg2 mutant.Using map-based cloning combined with a Mut Map approach,we cloned SG2,which encodes a plant-specific protein with a ribonuclease H-like domain.SG2 is a positive regulator downstream of GLYCOGEN SYNTHASE KINASE2(GSK2)in response to BR signaling,and its mutation causes insensitivity to exogenous BR treatment.Genetical and biochemical analysis showed that GSK2 interacts with and phosphorylates SG2.We further found that BRs enhance the accumulation of SG2 in the nucleus,and subcellular distribution of SG2 is regulated by GSK2 kinase activity.In addition,Oryza sativa OVATE family protein 19(OsOFP19),a negative regulator of grain shape,interacts with SG2 and plays an antagonistic role with SG2 in controlling gene expression and grain size.Our results indicated that SG2 is a new component of GSK2-related BR signaling response and regulates grain size by interacting with Os OFP19.Yunshuai Huang Hui Dong Changling Mou Ping Wang Qixian Hao Min Zhang Hongmin Wu Fulin Zhang Tengfei Ma Rong Miao Kai Fu Yaping Chen Ziyan Zhu Cheng Chen Qikai Tong Zhuoran Wang Shirong Zhou Xi Liu Shijia Liu Yunlu Tian Ling Jiang Jianmin Wan 2022Journal of Integrative Plant Biology2022,64,10:1
