| 3 | Experimental Analysis of the Flow Characteristics of an Adjustable Critical-Flow Venturi Nozzle显示文摘The response of an adjustable critical-flow Venturi nozzle is investigated through a set indoor experiments aimed to determine the related critical flow rate,critical pressure ratio,and discharge coefficient.The effect of a variation in the cone displacement and liquid content on the critical flow characteristics is examined in detail and it is shown that the former can be used to effectively adjust the critical flow rate.The critical pressure ratio of the considered nozzle is above 0.85,and the critical flow control deviation of the gas flow is within±3%.Liquid flow can reduce the gas critical mass flow rate accordingly,especially for the cases with larger liquid volume and lower inlet pressure.The set of results and conclusions provided are intended to support the optimization of steam injection techniques in the context of heavy oil recovery processes. | Chun Ye Jingjing Gao Zhihui Wang Weibiao Zheng Yibei Wang Xingkai Zhang Ming Liu | 2023 | Fluid Dynamics & Materials Processing2023,19,3: | 0 |
| 4 | Two types of coumarins-specific enzymes complete the last missing steps in pyran- and furanocoumarins biosynthesis显示文摘Pyran-and furanocoumarins are key representatives of tetrahydropyrans and tetrahydrofurans,respectively,exhibiting diverse physiological and medical bioactivities.However,the biosynthetic mechanisms for their core structures remain poorly understood.Here we combined multiomics analyses of biosynthetic enzymes in Peucedanum praeruptorum and in vitro functional verification and identified two types of key enzymes critical for pyran and furan ring biosynthesis in plants.These included three distinct P.praeruptorum prenyltransferases(PpPT1e3)responsible for the prenylation of the simple coumarin skeleton 7 into linear or angular precursors,and two novel CYP450 cyclases(PpDC and PpOC)crucial for the cyclization of the linear/angular precursors into either tetrahydropyran or tetrahydrofuran scaffolds.Biochemical analyses of cyclases indicated that acid/base-assisted epoxide ring opening contributed to the enzyme-catalyzed tetrahydropyran and tetrahydrofuran ring refactoring.The possible acid/base-assisted catalytic mechanisms of the identified cyclases were theoretically investigated and assessed using site-specific mutagenesis.We identified two possible acidic amino acids Glu303 in PpDC and Asp301 in PpOC as vital in the catalytic process.This study provides new enzymatic tools in the epoxide formation/epoxide-opening mediated cascade reaction and exemplifies how plants become chemically diverse in terms of enzyme function and catalytic process. | Yucheng Zhao Yuedong He Liangliang Han Libo Zhang Yuanzheng Xia Fucheng Yin Xiaobing Wang Deqing Zhao Sheng Xu Fei Qiao Yibei Xiao Lingyi Kong | 2024 | Acta Pharmaceutica Sinica B2024,14,2: | 0 |
| 5 | Characteristics and expression of the TCP transcription factors family in Allium senescens reveal its potential roles in drought stress responses显示文摘Allium senescens,is an important economic and ecological grassland plant with drought-resistant characteristics.A TCP protein transcription factor is important in the regulation of plant development and adverse responses.However,the mechanism by which TCP transcription functions in drought resistance in Allium senescens is still not clear.Here,we obtained a total of 190,305 transcripts with 115,562 single gene clusters based on RNA-Seq sequencing of Allium senescens under drought stress.The total number of bases was 97,195,096 bp,and the average length was 841.06 bp.Furthermore,we found that there were eight genes of the TCP family that showed an upregulated expression trend under drought stress in Allium senescens.We carried out an investigation to determine the evolution and function of the AsTCP family and how they produce an effect in drought resistance.The 14 AsTCP genes were confirmed and divided into class I and class II containing CIN and CYC/TBI subfamilies,respectively.We also found that the expression of AsTCP17 was remarkably upregulated with drought treatment.Besides,the transformation of AsTCP17 in Arabidopsis revealed that the protective enzymes,namely polyphenol oxidase(POD)and superoxide dismutase(SOD),were increased by 0.4 and 0.8 times,respectively.Chlorophyll content was also increased,while the H2O2 and malondialdehyde(MDA)contents were decreased.Staining assays with 3,3′-diaminobenzidine(DAB)also suggested that the AsTCP17 downregulates reactive oxygen species(ROS)accumulation.In addition,overexpression of the AsTCP17 affected the accumulation of drought-related hormones in plants,and the synthesis of ABA.The expression of AtSVP and AtNCED3,related ABA synthesis pathway genes,indicated that the level of expression of AtSVP and AtNCED3 was obviously enhanced,with the overexpression of line 6 showing a 20.6-fold and 7.0-fold increase,respectively.Taken together,our findings systematically analyze the AsTCPs family at the transcriptome expression level in Allium senescens,and we also demonstrated that AsTCP17 protein,as a positive regulator,was involved in drought resistance of Allium senescens.In addition,our research contributes to the comprehensive understanding of the drought stress defense mechanism in herbaceous plants. | XIAOHONG FU JIE ZHAO DANDAN CAO CHENGXING HE ZIYI WANG YIBEI JIANG JIANFENG LIU GUIXIA LIU | 2023 | BIOCELL2023,47,4: | 0 |
| 6 | Novel phthalimides regulating PD-1/PD-L1 interaction as potential immunotherapy agents显示文摘Programmed cell death 1(PD-1)/programmed cell death ligand 1(PD-L1)have emerged as one of the most promising immune checkpoint targets for cancer immunotherapy.Despite the inherent advantages of small-molecule inhibitors over antibodies,the discovery of small-molecule inhibitors has fallen behind that of antibody drugs.Based on docking studies between small molecule inhibitor and PD-L1 protein,changing the chemical linker of inhibitor from a flexible chain to an aromatic ring may improve its binding capacity to PD-L1 protein,which was not reported before.A series of novel phthalimide derivatives from structure-based rational design was synthesized.P39 was identified as the best inhibitor with promising activity,which not only inhibited PD-1/PD-L1 interaction(IC_(50)=8.9 nmol/L),but also enhanced killing efficacy of immune cells on cancer cells.Co-crystal data demonstrated that P39 induced the dimerization of PD-L1 proteins,thereby blocking the binding of PD-1/PD-L1.Moreover,P39 exhibited a favorable safety profile with a LD_(50)>5000 mg/kg and showed significant in vivo antitumor activity through promoting CD8^(+)T cell activation.All these data suggest that P39 acts as a promising small chemical inhibitor against the PD-1/PD-L1 axis and has the potential to improve the immunotherapy efficacy of T-cells. | Chengliang Sun Yao Cheng Xiaojia Liu Gefei Wang Wenjian Min Xiao Wang Kai Yuan Yi Hou Jiaxing Li Haolin Zhang Haojie Dong Liping Wang Chenguang Lou Yanze Sun Xinmiao Yu Hongbin Deng Yibei Xiao Peng Yang | 2022 | Acta Pharmaceutica Sinica B2022,12,12: | 0 |
| 7 | Photocatalytic degradation of 2,4,6-trinitrotoluene显示文摘A Pt/TiO2 photocatalyst was used in the photocatalytic degradation of 2,4,6-trinitrotoluene (TNT) under the irradiation of the simulated sunlight. The results show that the destruction of TNT is faster and more complete with the photocatalyst. The photocatalytic degradation of TNT follows first order kinet-ics. The transformation of nitryl groups of TNT molecules into nitrite and nitrate ions may be acceler-ated in the presence of the Pt/TiO2 photocatalyst.
