| 1 | Distribution characteristics of magnetic susceptibility of the surface sediments in the southern Yellow Sea显示文摘The characteristic distributions of magnetic susceptibility (MS) are analyzed on the basis of susceptibility of 172 surface sediment samples in the southern Yellow Sea (SYS). The preliminary results are as follows: first, the distributions clearly correspond to different modern sediment assemblages in the continental sea, which indicates different sediment origins. With the 30 μCGS isoline being taken as demarcation line, the study area can then be divided into section H (high MS value area) and section L (low MS value area). Section H is mainly adjacent to land with two main sources of the Changjiang River and the Huanghe River.Section L is mainly an eddy sediment area, where Yellow Sea Cold Water is entrenched all the year round. The distribution pattern of MS could tell apart strong or weak hydrodynamic conditions and has a close relation to the circulation system in this area. At the areas of the SYS Circumfluent and northern East China Sea (NECS) Circumfluent (weak hydrodynamic), the MS has low values, while in the areas of Coastal Current (strong hydrodynamic), the values are high.At the same time, the oxidizing areas tend to take on higher MS, while the reducing areas have lower one. | 4 | Identification of key genes affecting porcine fat deposition based on coexpression network analysis of weighted genes显示文摘Background:Fat deposition is an important economic consideration in pig production.The amount of fat deposition in pigs seriously affects production efficiency,quality,and reproductive performance,while also affecting consumers’choice of pork.Weighted gene co-expression network analysis(WGCNA)is effective in pig genetic studies.Therefore,this study aimed to identify modules that co-express genes associated with fat deposition in pigs(Songliao black and Landrace breeds)with extreme levels of backfat(high and low)and to identify the core genes in each of these modules.Results:We used RNA sequences generated in different pig tissues to construct a gene expression matrix consisting of 12,862 genes from 36 samples.Eleven co-expression modules were identified using WGCNA and the number of genes in these modules ranged from 39 to 3,363.Four co-expression modules were significantly correlated with backfat thickness.A total of 16 genes(RAD9A,IGF2R,SCAP,TCAP,SMYD1,PFKM,DGAT1,GPS2,IGF1,MAPK8,FABP,FABP5,LEPR,UCP3,APOF,and FASN)were associated with fat deposition.Conclusions:RAD9A,TCAP,SMYD1,PFKM,GPS2,and APOF were the key genes in the four modules based on the degree of gene connectivity.Combining these results with those from differential gene analysis,SMYD1 and PFKM were proposed as strong candidate genes for body size traits.This study explored the key genes that regulate porcine fat deposition and lays the foundation for further research into the molecular regulatory mechanisms underlying porcine fat deposition. | Kai Xing Huatao Liu Fengxia Zhang Yibing Liu Yong Shi Xiangdong Ding Chuduan Wang | 2022 | Journal of Animal Science and Biotechnology2022,13,1: | 3 |
| 5 | Transcriptome sequencing analysis for the identification of stable lncRNAs associated with bovine Staphylococcus aureus mastitis显示文摘Background:Staphylococcus aureus(S.aureus)mastitis is one of the most difficult diseases to treat in lactating dairy cows worldwide.S.aureus with different lineages leads to different host immune responses.Long non-coding RNAs(lncRNAs)are reported to be widely involved in the progress of inflammation.However,no research has identified stable lncRNAs among different S.aureus strain infections.In addition,folic acid(FA)can effectively reduce inflammation,and whether the inflammatory response caused by S.aureus can be reduced by FA remains to be explored.Methods:lncRNA transcripts were identified from Holstein mammary gland tissues infected with different concentrations of S.aureus(in vivo)and mammary alveolar cells(Mac-T cells,in vitro)challenged with different S.aureus strains.Differentially expressed(DE)lncRNAs were evaluated,and stable DE lncRNAs were identified in vivo and in vitro.On the basis of the gene sequence conservation and function conservation across species,key lncRNAs with the function of potentially immune regulation were retained for further analysis.The function of FA on inflammation induced by S.aureus challenge was also investigated.Then,the association analysis between these keys lncRNA transcripts and hematological parameters(HPs)was carried out.Lastly,the knockdown and overexpression of the important lncRNA were performed to validate the gene function on the regulation of cell immune response.Results:Linear regression analysis showed a significant correlation between the expression levels of lncRNA shared by mammary tissue and Mac-T cells(P<0.001,R^(2)=0.3517).lncRNAs PRANCR and TNK2-AS1 could be regarded as stable markers associated with bovine S.aureus mastitis.Several HPs could be influenced by SNPs around lncRNAs PRANCR and TNK2-AS1.The results of gene function validation showed PRANCR regulates the mRNA expression of SELPLG and ITGB2 within the S.aureus infection pathway and the Mac-T cells apoptosis.In addition,FA regulated the expression change of DE lncRNA involved in toxin metabolism and inflammation to fight against S.aureus infection.Conclusions:The remarkable association between SNPs around these two lncRNAs and partial HP indicates the potentially important role of PRANCR and TNK2-AS1 in immune regulation.Stable DE lncRNAs PRANCR and TNK2-AS1 can be regarded as potential targets for the prevention of bovine S.aureus mastitis.FA supplementation can reduce the negative effect of S.aureus challenge by regulating the expression of lncRNAs. | Siyuan Mi Yongjie Tang Gerile Dari Yuanjun Shi Jinning Zhang Hailiang Zhang Xueqin Liu Yibing Liu Usman Tahir Ying Yu | 2022 | Journal of Animal Science and Biotechnology2022,13,2: | 2 |

| 20 | Combination of Click Chemistry and Enzymatic Ligation for Stable and Efficient Protein Immobilization for Single-Molecule Force Spectroscopy显示文摘Protein immobilization is an essential method for both basic and applied research for protein,and covalent,site-specific attachment is the most desirable strategy.Classic methods typically rely on a heterobifunctional cross-linker,such as N-hydroxysuccinimide(NHS)-linker-maleimide,or a similar two-step process.It utilizes the amino-reactive NHS and the thiolreactive maleimide to conjugate protein to the solid support.However,NHS as a chemical is susceptible to hydrolysis during storage and handling,and maleimide reacts nonspecifically with all cysteines available in the protein,leading to an inconsistent result.To solve these problems,we have developed a method by combining a strain-promoted azide–alkyne cycloaddition(SPAAC)click reaction and an OaAEP1(C247A)-based enzymatic ligation.The method was demonstrated by the successful immobilization of enhanced green fluorescent protein(eGFP),which was visualized by fluorescent imaging.Moreover,the correct folding and stability of the immobilized protein were verified by atomic force microscopy-based single-molecule force spectroscopy(AFM-SMFS)measurement with a high success rate(89%).Finally,the strength of the 1,2,3-triazole linkage from the azide-dibenzocyclooctyne(DBCO)-based SPAAC reaction was quantified with an ultrahigh rupture force>1.7 nN.Thus,this stable,efficient,and site-specific immobilization method can be used for many challenging systems,especially SMFS studies. | Shengchao Shi Ziyi Wang Yibing Deng Fang Tian Qingsong Wu Peng Zheng | 2022 | CCS Chemistry2022,4,2: | 0 |
