1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2Air pollution and inhalation exposure to particulate matter of different sizes in rural households using improved stoves in central China显示文摘Household air pollution is considered to be among the top environmental risks in China.To examine the performance of improved stoves for reduction of indoor particulate matter(PM) emission and exposure in rural households, individual inhalation exposure to size-resolved PM was investigated using personal portable samplers carried by residents using wood gasifier stoves or improved coal stoves in a rural county in Central China.Concentrations of PM with different sizes in stationary indoor and outdoor air were also monitored at paired sites. The stationary concentrations of size-resolved PM in indoor air were greater than those in outdoor air, especially finer particles PM0.25. The daily averaged exposure concentrations of PM0.25, PM1.0, PM2.5 and total suspended particle for all the surveyed residents were 74.4 ± 41.1, 159.3 ± 74.3, 176.7 ± 78.1 and 217.9 ± 78.1 μg/m3,respectively. Even using the improved stoves, the individual exposure to indoor PM far exceeded the air quality guideline by WHO at 25 μg/m3. Submicron particles PM1.0 were the dominant PM fraction for personal exposure and indoor and outdoor air. Personal exposure exhibited a closer correlation with indoor PM concentrations than that for outdoor concentrations. Both inhalation exposure and indoor air PM concentrations in the rural households with gasifier firewood stoves were evidently lower than the reported results using traditional firewood stoves. 14Physical mechanism of field modulation effects in ion implanted edge termination of vertical GaN Schottky barrier diodes显示文摘In this study,the physical properties of F ion-implanted GaN were thoroughly studied,and the related electric-field modulation mechanisms in ion-implanted edge termination were revealed.Transmission electron microscopy re.sults indicate that the ion-implanted region maintains a single-crystal structure even with the implantation of high-energy F ions,indicating that the high resistivity of the edge termination region is not induced by amorphization.Alternately,ion implantation-induced deep levels could compensate the electrons and lead to a highly resistive layer In addition to the bulk ffect,the direct bombardment of high-energy F ions resulted in a rough and nitrogen-deficient surface,which was confirmed via atomic force microscopy(AFM)and X-ray photoelectron spectroscopy,The implanted surface with a large density of nitrogen vacancies can accommodate electrons,and it is more conductive than the bulk in the implanted region,which is validated via spreading resistance profiling and conductive AFM measurements.Under reverse bias,the implanted surface can spread the potential in the lateral direction,whereas the acceptor traps capture electrons acting as space charges,shifting the peak electric field into the bulk region in the vertical direction.As a result,the Schottky barrier diode terminated with high-energy F ion-implanted regions exhibits a breakdown voltage of over 1.2 kv.Ruiyuan Yin Chiachia Li Bin Zhang Jinyan Wang Yunyi Fu Cheng P.Wen Yilong Hao Bo Shen Maojun Wang 2022Fundamental Research2022,2,4:1

16Hyperfine Structure of Several Soils from Western Hubei and Inner Mongolia Studied by Mossbauer Spectroscopy显示文摘Soil samples taken from Xilamuren Grassland,Resonant Sand Bay,Inner Mongolia,and Yichang,western Hubei Province were investigated by Mssbauer spectroscopy at room temperature and 20 K.This was supplemented with phase identification and elemental analysis to obtain information about the composition and structure types of Fe-containing compounds.The samples collected from both Xilamuren Grassland and Resonant Sand Bay,Inner Mongolia were found to have small amount of iron 1%-2%.The main phases were determined as silica,albite,and microcline.Two or three dominant doublets were observed in the Mssbauer spectra of these samples,respectively and identified as corresponding to the valence states of Fe2+and Fe3+.A sextet observed in all the spectra is attributed to the hematite phase.Low temperature Mssbauer measurements have revealed Morin transition.Pelenovich V O CHEN Yilong XIAO Renzheng LIU Shouyin FU Dejun 2014Wuhan University Journal of Natural Sciences2014,19,3:0
17Effect of the biosynthesis of the volatile compound phenylacetaldehyde on chloroplast modifications in tea (Camellia sinensis) plants显示文摘Plant volatile compounds have important physiological and ecological functions.Phenylacetaldehyde(PAld),a volatile phenylpropanoid/benzenoid,accumulates in the leaves of tea(Camellia sinensis)plants grown under continuous shading.This study was conducted to determine whether PAld production is correlated with light and to elucidate the physiological functions of PAld in tea plants.Specifically,the upstream mechanism modulating PAld biosynthesis in tea plants under different light conditions as well as the effects of PAld on chloroplast/chlorophyll were investigated.The biosynthesis of PAld was inhibited under light,whereas it was induced in darkness.The structural gene encoding aromatic amino acid aminotransferase 1(CsAAAT1)was expressed at a high level in darkness,consistent with its importance for PAld accumulation.Additionally,the results of a transcriptional activation assay and an electrophoretic mobility shift assay indicated CsAAAT1 expression was slightly activated by phytochrome-interacting factor 3-2(CsPIF3-2),which is a light-responsive transcription factor.Furthermore,PAld might promote the excitation of chlorophyll in dark-treated chloroplasts and mediate electron energy transfer in cells.However,the accumulated PAld can degrade chloroplasts and chlorophyll,with potentially detrimental effects on photosynthesis.Moreover,PAld biosynthesis is inhibited in tea leaves by red and blue light,thereby decreasing the adverse effects of PAld on chloroplasts during daytime.In conclusion,the regulated biosynthesis of PAld in tea plants under light and in darkness leads to chloroplast modifications.The results of this study have expanded our understanding of the biosynthesis and functions of volatile phenylpropanoids/benzenoids in tea leaves.Lanting Zeng Xiaochen Zhou Xiumin Fu Yilong Hu Dachuan Gu Xingliang Hou Fang Dong Ziyin Yang 2023Horticulture Research2023,10,3:0

19Anthrax lethal toxin and tumor necrosis factor-a synergize on intestinal epithelia to induce mouse death显示文摘Bacillus anthracis lethal toxin(LT)is a determinant of lethal anthrax.Its function in myeloid cells is required for bacterial dissemination,and LT itself can directly trigger dysfunction of the cardiovascular system.The interplay between LT and the host responses is important in the pathogenesis,but our knowledge on this interplay remains limited.Tumor necrosis factor-a(TNF-a)is a pleiotropic pro-inflammatory cytokine induced by bacterial infections.Since LT accumulates and cytokines,predominantly TNF,amass during B.anthracis infection,co-treatment of TNF+LT in mice was used to mimic in vivo conditions for LT to function in inflamed hosts.Bone marrow transplantation and genetically engineered mice showed unexpectedly that the death of intestinal epithelial cells(IECs)rather than that of hematopoietic cells led to LT+TNF-induced lethality.Inhibition of p38a mitogen-activated protein kinase(MAPK)signaling by LT in IECs promoted TNF-induced apoptosis and necroptosis of IECs,leading to intestinal damage and mouse death.Consistently,p38a inhibition by LT enhanced TNF-mediated cell death in human colon epithelial HT-29 cells.As intestinal damage is one of the leading causes of lethality in anthrax patients,the IEC damage caused by LT+TNF would most likely be a mechanism underneath this clinical manifestation and could be a target for interventions.Xinhe Gao Teng Teng Yifei Liu Tingting Ai Rui Zhao Yilong Fu Peipei Zhang Jiahuai Han Yingying Zhang 2024Protein & Cell2024,15,2:0
20Point-defect-driven flattened polar phonon bands in fluorite ferroelectrics显示文摘The scale-free ferroelectric polarization of fluorite MO_(2)(M=Hf,Zr)due to flat polar phonon bands are promising for nonvolatile memories.Defects are also widely introduced to improve the emergent ferroelectricity.However,their roles are still not fully understood at the atomic-level.Here,we report a significant effect of point-defect-driven flattening of polar phonon bands with more polar modes and polarization contribution in doped MO_(2).The polar phonon bands in La-doped MO_(2)(M=Hf,Zr)can be significantly flattened,compared with pure ones.However,the lower energy barrier with larger polarization of VO-only doped MO_(2) compared with La-doped cases suggest that VO and local lattice distortion should be balanced for high-performance fluorite ferroelectricity.The work is believed to bridge the relation between point defects and the generally enhanced induced ferroelectricity in fluorite ferroelectrics at the atomic-level and inspire their further property optimization via defect-engineering.Pu Ai Fengjun Yan Wen Dong Shi Liu Junlei Zhao Kan-Hao Xue Syed Ul Hasnain Bakhtiar Yilong Liu Qi Ma Ling Miao Mengyuan Hua Guangzu Zhang Shenglin Jiang Wei Luo Qiuyun Fu 2023npj Computational Materials2023,,1:0
