1A Genome-wide Functional Characterization of Arabidopsis Regulatory Calcium Sensors in Pollen Tubes显示文摘Calcium, an ubiquitous second messenger, plays an essential and versatile role in cellular signaling. The diverse function of calcium signals is achieved by an excess of calcium sensors. Plants possess large numbers of calcium sensors, most of which have not been functionally characterized. To identify physiologically relevant calcium sensors in a specific cell type, we conducted a genome-wide functional survey in pollen tubes, for which spatiotemporal calcium signals are well-characterized and required for polarized tip growth. Pollen-specific members of calmodulin (CaM), CaM-like (CML), calcium-dependent protein kinase (CDPK) and calcineurin B-like protein (CBL) families were tagged with green fluorescence protein (GFP) and their localization patterns and overexpression phenotypes were characterized in tobacco pollen tubes. We found that several fusion proteins showed distinct overexpression phenotypes and subcellular localization patterns. CDPK24-GFP was localized to the vegetative nucleus and the generative cell/sperms. CDPK32-GFP caused severe growth depolarization. CBL2-GFP and CBL3-GFP exhibited dynamic patterns of subcellular localization, including several endomembrane compartments, the apical plasma membrane (PM), and cytoskeleton-like structures in pollen tubes. Their overexpression also inhibited pollen tube elongation and induced growth depolarization. 3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 4Secretory/releasing proteome-based identification of plasma biomarkers in HBV-associated hepatocellular carcinoma显示文摘For successful therapy, hepatocellular carcinoma (HCC) must be detected at an early stage. Herein, we used a proteomic approach to analyze the secretory/releasing proteome of HCC tissues to identify plasma biomarkers. Serum-free conditioned media (CM) were collected from primary cultures of cancerous tissues and surrounding noncancerous tissues. Proteomic analysis of the CM proteins permitted the identification of 1365 proteins. The enriched molecular functions and biological processes of the CM proteins, such as hydrolase activity and catabolic processes, were consistent with the liver being the most important metabolic organ. Moreover, 19% of the proteins were characterized as extracellular or membrane-bound. For validation, secretory proteins involved in transforming growth factor-β signaling pathways were validated in plasma samples. Alphafetoprotein (AFP), metalloproteinase (MMP)1, osteopontin (OPN), and pregnancy-specific beta-1-glycoprotein (PSG)9 were significantly increased in HCC patients. The overall performance of MMP1 and OPN in the diagnosis of HCC remained greater than that of AFP. In addition, this study represents the first report of MMP1 as a biomarker with a higher sensitivity and specificity than AFP. Thus, this study provides a valuable resource of the HCC secretome with the potential to investigate serological biomarkers. 7Solute-homogenization model and its experimental verification in Mg-Gd-based alloys显示文摘Composition homogenization in solid solution is important for industrial alloys. In the present work, a solute homogenization model is proposed based on the chemical short-range-order tendency in Mg-Gdbased alloys. After a calculation using the cluster-plus-glue-atom model, the stable Mg-Gd structural unit is derived, [Gd-Mg12]Mg6, where one solute Gd is nearest-neighbored with twelve Mg atoms to form the characteristic hcp cluster [Gd-Mg12] and this cluster is matched with six Mg glue atoms. Such a local unit is then mixed with [Mg-Mg12]Mg3, the stable unit for pure Mg. Assuming that the Gd-containing units are arranged in fcc-or bcc-like lattice points and the Mg units in their octahedral interstices, three proportions between the two units are obtained, 1:1, 2:3, and 1:3, which constitute three solute homogenization modes. The prevailing Mg-Gd-based alloys are consequently classified into three groups, respectively exemplified by GW103 K(Mg-10 Gd-3 Y-0.4 Zr, wt%), GW83 K(Mg-8 Gd-3 Y-0.4 Zr), and GW63 K(Mg-6 Gd-3 Y-0.4 Zr). Mg-Gd-Y-Zr alloys were designed following the model(where Y and Zr were also added in substitution for Gd) and prepared by permanent-mould casting. 8The Medium Energy X-ray telescope (ME) onboard the Insight-HXMT astronomy satellite显示文摘The Medium Energy X-ray telescope(ME) is one of the three main telescopes on board the Insight hard X-ray modulation telescope(Insight-HXMT) astronomy satellite. ME contains 1728 pixels of Si-PIN detectors sensitive in 5-30 ke V with a total geometrical area of 952 cm^2. The application specific integrated circuit(ASIC) chip, VA32TA6, is used to achieve low power consumption and low readout noise. The collimators define three kinds of field of views(FOVs) for the telescope, 1°×4°, 4°×4°,and blocked ones. Combination of such FOVs can be used to estimate the in-orbit X-ray and particle background components.The energy resolution of ME is ~3 ke V at 17.8 ke V(FWHM) and the time resolution is 255 μs. 10Carbon-Based Metal-Free Electrocatalysts:Past,Present,and Future显示文摘CONSPECTUS:Due to the overuse of fossil fuels,various detrimental effects along with the excess CO_(2)emissions have induced global warming and sea-level rising.To tackle climate change and provide a cleaner environment for the air we breathe and water we consume,the existing energy mix needs to be changed into fossil-free,clean,renewable energy with zero emission(e.g.,fuel cells).While providing a promising and scalable strategy to the energy and environmental challenges,renewable energy processes often involve noble-metal-based catalysts(i.e.,Pt,RuO_(2)).However,the disadvantages of noblemetal-based catalysts,including their high cost and scarcity,have hampered the large-scale application of renewable energy technologies.In 2009,we discovered earth-abundant carbon materials functioning as efficient low-cost,carbon-based metal-free electrocatalysts(C-MFECs)attractive for renewable energy and environmental remediation.Since then,C-MFECs have become an emerging new research field over the world.They are demonstrated to be efficient multifunctional catalysts for various key reactions important to renewable energy and environmental technologies,including oxygen reduction reaction(ORR),hydrogen evolution reaction(HER),oxygen evolution reaction(OER),CO_(2)reduction reaction(CO_(2)RR),and N_(2)reduction reaction(NRR),to name a few.Charge transfer/redistribution induced by heteroatom(e.g.,N)and/or defect doping was recognized as the driving force for the metal-free catalytic activities.This finding has been used as a guidance to design and develop various new and multifunctional C-MFECs for many reactions even beyond the renewable energy and environmental remediation.In this Account,we first summarize our previous work on the development and mechanistic understanding of C-MFECs for ORR,HER,and OER to promote renewable energy conversion and storage.Then,we present recent advances in C-MFECs for new important reactions for environment remediation(e.g.,CO_(2)RR,NRR),seawater splitting,and metal−CO_(2)batteries.However,different dopant locations for C-MFECs even with the same doping element and content can cause variable catalytic properties for heteroatom-doped carbon materials.Therefore,vast opportunities remain for further developing numerous innovative C-MFECs with defined structures to gain a better understanding of their structure-based properties.In this context,we finally conclude with the current challenges and future perspectives in this exciting field.Qingfeng Zhai Ying Pan Liming Dai 2021Accounts of Materials Research2021,2,12:3
11Reversible Al^(3+) storage mechanism in anatase TiO_(2) cathode material for ionic liquid electrolyte-based aluminum-ion batteries显示文摘Rechargeable aluminum ion battery(AIB) with high theoretical specific capacity, abundant elements and low cost engages considerable attention as a promising next generation energy storage and conversion system. Nevertheless, to date, one of the major barriers to pursuit better AIB is the limited applicable cathode materials with the ability to store aluminum highly reversibly. Herein, a highly reversible AIB is proposed using mesoporous TiO2 microparticles(M-TiO2) as the cathode material. The improved performance of Ti O2/Al battery is ascribed to the high ionic conductivity and material stability, which is caused by the stable architecture with a mesoporous microstructure and no random aggregation of secondary particles. In addition, we conducted detailed characterization to gain deeper understanding of the Al^(3+) storage mechanism in anatase Ti O2 for AIB. Our findings demonstrate clearly that Al^(3+)can be reversibly stored in anatase TiO2 by intercalation reactions based on ionic liquid electrolyte. Especially, DFT calculations were used to investigate the accurate insertion sites of aluminum ions in M-Ti O2 and the volume changes of M-TiO2 cells during discharging. As for the controversial side reactions in AIBs, in this work, by normalized calculation, we confirm that M-Ti O2 alone participate in the redox reaction. 12Quantitative deconvolution of autocorrelations and cross correlations from two-dimensional lifetime decay maps in fluorescence lifetime correlation spectroscopy显示文摘Fluorescence correlation spectroscopy(FCS) is a widely used method for measuring molecular diffusion and chemical kinetics. However, when a mixture of fluorescent species is taken into account, the conventional FCS method has limitations in extracting autocorrelations for different species and cross correlations between different species. Recently developed fluorescence lifetime correlation spectroscopy(FLCS) based on time-tagged time-resolved(TTTR) photon recording, which can record the global and micro arrival time for each individual photon, has been used to discriminate different species according to fluorescence lifetime. Here, based on two-dimensional lifetime decay maps constructed from TTTR photon stream, we have developed a quantitative lifetime-deconvolution FCS model(LDFCS) to extract precise chemical rates for chemical conversions in multi-species systems. 13Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 14Effect of multiphase content on temperature-dependent electrical conductivity in silicone rubber composites显示文摘To investigate how the multiphase structures affect the electrical conductivity in semicrystalline polymer composites,herein,an accurate multiphase content calculation method is proposed and verified,which combines amorphous phase information in broadband dielectric spectroscopy and crystalline phase information in differential scanning calorimetry.Taken aluminium hydroxide(ATH)filled silicone rubber as an example,it is found that the rigid amorphous fraction(RAF)corresponding to the chains constrained by crystals(RAF_(cry))is not linearly increased with crystalline fraction(CF).Compared to non-isothermal crystallisation,RAF caused by ATH/silicone rubber interface(RAF_(int))after isothermal crystallisation at 213 K changes little,while mobile amorphous fraction and RAFcry is attenuated and CF is strengthened.Based on the calculated structures of ATH filled silicone rubber,activation energy of conductivities during cooling is dominated by the thermal transition for conductive ions and shortened distance among the conductive ions through shrunk volumes of the amorphous phase.Our findings deepen the understanding of multiphase content in semi-crystalline polymer composites and its relationship with electrical conductivity,which can be applied in manipulating electrical performance of semi-crystalline polymers by fillers.Ying Lin Yuhao Liu Bin Cao Jianyi Xue Liming Wang Jianing Wang Lijian Ding 2023High Voltage2023,8,2:2
15Nogo-A and Nogo-A receptor expression in periventricular white matter of experimental autoimmune encephalomyelitis rats显示文摘BACKGROUND:Previous studies have focused on the correlation between Nogo-A expression and multiple sclerosis or between Nogo-A receptor(NgR) expression and multiple sclerosis in the central nervous system.Expression patterns of Nogo-A and NgR remain poorly understood in rat models of experimental autoimmune encephalomyelitis(EAE).OBJECTIVE:To observe dynamic changes in Nogo-A and NgR protein expression,and to verify the correlation between Nogo-A and NgR protein,as well as expression patterns at various time points,in periventricular tissue of EAE rats.DESIGN,TIME AND SETTING:A neuroimmunological,randomized,controlled experiment was performed at the Clinical Institute of Hunan People's Hospital of China from September to November 2008.MATERIALS:Immunohistochemistry(streptavidin-biotin-peroxidase complex method) kit was purchased from Boster,China.METHODS:A total of 60 female,Wistar rats,aged 6-8 weeks,were randomly assigned to EAE and control groups(n = 30,respectively).Guinea pig spinal cord homogenate,self-made complete Freund's adjuvant(0.2 mL/100 g),and pertussis vaccine(0.2 mL) were subcutaneously injected into the hindlimb foot pad of rats from the EAE group to create rat models of EAE.Complete Freund's adjuvant(0.2 mL) was infused into rats from the control group.MAIN OUTCOME MEASURES:Nogo-A and NgR protein expression was determined in periventricular white matter using immunohistochemical methods.Neurological scores were determined in all rats.RESULTS:Rats from the EAE group developed acute-onset EAE following immunization.The pathogenetic symptoms reached a peak on day 15,and neurological scores were also greatest at this time point.Neurological scores decreased with recovery of the illness.Nogo-A was shown to be expressed in neuronal cells and oligodendrocytes,and expression increased 11 days after immunization(P < 0.01),decreased by day 13(P < 0.01),and then increased again by day 15.Nogo-A expression remained greater in the EAE group compared with the control group at day 30(P < 0.01).In the EAE group,NgR protein was primarily expressed on the surface of neuronal bodies and axons.NgR expression increased 13-18 days after immunization(P < 0.01 or P < 0.05).CONCLUSION:Nogo-A and NgR protein expression altered with disease course in periventricular white matter of EAE rats.Results suggested that Nogo-A and NgR were involved in EAE occurrence.Liming Tan Ying Chi Chunyu Wang1, Jun Wu1, Hainan Zhang1, Xiangmin Shen1, ShujuanDuan1, Bo JiangI Jun Wu Hainan Zhang Xiangmin Shen Shujuan Duan Bo Jiang 2010Neural Regeneration Research2010,5,8:2
16Carbon-based bifunctional electrocatalysts for oxygen reduction and oxygen evolution reactions:Optimization strategies and mechanistic analysis显示文摘Electrocatalysts are one of the essential components for the devices of high-efficiency green energy storage and conversion,such as metal-air cells,fuel cells,and water electrolysis systems.While catalysts made from noble metals possess high catalytic performance in both oxygen reduction reaction(ORR)and oxygen evolution reaction(OER),their scarcity and expensiveness significantly limit large-scale applications.In this regard,metal-free/non-noble metal carbon-based catalysts have become competitive alternatives to replace catalysts made of noble metals.Nevertheless,low catalytic ORR/OER performance is the challenge of carbon-based catalysts for the commercial applications of metal-air batteries.To solve the problem of poor catalytic performance,two strategies have been proposed:(1)controlling the microstructure of the catalysts to expose more active sites as the channels of rapid mass and electron transfer;and(2)reducing the reaction energy barrier by optimizing the electronic structures of the catalysts via surface engineering.Here,we review different types of bifunctional ORR/OER electrocatalysts with the activated surface sites.We focus on how the challenge can be overcome with different methods of material synthesis,structural and surface characterization,performance validation/optimization,to outline the principles of surface modifications behind catalyst designs.In particular,we provide critical analysis in the challenges that we are facing in structural design and surface engineering of bifunctional ORR/OER catalysts and indicate the possible solution for these problems,providing the society with clearer ideas on the practical prospects of noble-metal-free electrocatalysts for their future applications.Huidong Xu Jack Yang Riyue Ge Jiujun Zhang Ying Li Mingyuan Zhu Liming Dai Sean Li Wenxian Li 2022Journal of Energy Chemistry2022,31,8:2

19Auxin signaling module OsSK41-OsIAA10-OsARF regulates grain yield traits in rice显示文摘Auxin is an important phytohormone in plants, and auxin signaling pathways in rice play key roles in regulating its growth, development, and productivity. To investigate how rice grain yield traits are regulated by auxin signaling pathways and to facilitate their application in rice improvement, we validated the functional relationships among regulatory genes such as OsIAA10, OsSK41, and OsARF21 that are involved in one of the auxin(OsIAA10) signaling pathways. We assessed the phenotypic effects of these genes on several grain yield traits across two environments using knockout and/or overexpression transgenic lines.Based on the results, we constructed a model that showed how grain yield traits were regulated by OsIAA10 and OsTIR1, OsAFB2, and OsSK41 and OsmiR393 in the OsSK41-OsIAA10-OsARF module and by OsARF21 in the transcriptional regulation of downstream auxin response genes in the OsSK41-OsIAA10-OsARF module. The population genomic analyses revealed rich genetic diversity and the presence of major functional alleles at most of these loci in rice populations. The strong differentiation of many major alleles between Xian/indica and Geng/japonica subspecies and/or among modern varieties and landraces suggested that they contributed to improved productivity during evolution and breeding. 20UV light absorbers executing synergistic effects of passivating defects and improving photostability for efficient perovskite photovoltaics显示文摘Metal halide perovskite-based solar cells(PSCs) have rapidly-increased power conversion efficiency(PCE)exceeding 25% but poor stability especially under ultraviolet(UV) light. Meanwhile, non-radiative recombination caused by diverse defects in perovskite absorbers and related interfaces is one of the major factors confining further development of PSCs. In this study, we systematically investigate the role of 2-(2-hydroxy-5-methylphenyl)benzotriazole(UVP) additive in perovskite layers. By adjusting the amount of doped UVP, the quality of perovskite absorbers is significantly improved with enlarged grains, longer lifetime and diffusion length of charge carriers. Furthermore, UVP not only reduces defects for less nonradiative recombination, but also matches energy level alignment for efficient interfacial charge extraction. X-ray photoelectron spectroscopy confirms that N-donor of UVP molecule coordinates with undercoordinated Pb^(2+) on the surface. Interestingly, UVP incorporated in PbI_(2) protects the perovskite by absorbing UV through the opening and closing of the chelating ring. Eventually, the UVP treated PSCs obtain a champion PCE of 22.46% with remarkably enhanced UV stability, retaining over 90% of initial PCE after 60 m W/cm^(2) strong UV irradiation for 9 h while the control maintaining only 74%.
