| 1 | Self-adjusted elastic action and its CBM pool-forming effect of the high rank coal reservoir显示文摘The coal reservoir is a kind of geologic body with considerable elasticity, and its response to stress is more sensitive than the routine ones. In the pool-forming process of the coalbed methane, (CBM) the action of many external geodynamic factors in the coal reservoir is represented by the ability of CBM’s diffusion and migration in the coal reservoir. Therefore, the occurrence of the natural fissures in the coal reservoir becomes a key that determines whether the CBM pool can be formed or not. Based on the principle, the authors have designed a comprehensive method of physical modeling to study the elasticity of the coal matrix block with different coal ranks in the light of the solid to fluid coupling, have established the model of the volume deformation (self-adjusted elastic effect) of the coal reservoir under the condition of the effective stress-adsorption/desorption, and have obtained a correct understanding of the law of the elastic deformation. | 2 | Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. | 3 | Quercetin promotes human epidermal stem cell proliferation through the estrogen receptor/β-catenin/c-Myc/cyclin A2 signaling pathway显示文摘Skin epidermal stem cells(EpSCs)play an important role in wound healing.Quercetin is a phytoestrogen reported to accelerate skin wound healing,but its effect on EpSCs is unknown.In this study,we investigated the effect of quercetin on human EpSC proliferation and explored the underlying mechanisms.We found that quercetin at 0.1~1μM significantly promoted EpSC proliferation and increased the number of cells in S phase.The pro-proliferative effect of quercetin on EpSCs was confirmed in cultured human skin tissue.Mechanistic studies showed that quercetin significantly upregulated the expressions ofβ-catenin,c-Myc,and cyclins A2 and E1.Inhibitor forβ-catenin or c-Myc significantly inhibited quercetin-induced EpSC proliferation.Theβ-catenin inhibitor XAV-939 suppressed quercetin-induced expressions ofβ-catenin,c-Myc,and cyclins A2 and E1.The c-Myc inhibitor 10058-F4 inhibited the upregulation of c-Myc and cyclin A2 by quercetin.Pretreatment of EpSCs with estrogen receptor(ER)antagonist ICI182780,but not the G protein-coupled ER1 antagonist G15,reversed quercetin-induced cell proliferation and upregulation ofβ-catenin,c-Myc,and cyclin A2.Collectively,these results indicate that quercetin promotes EpSC proliferation through ER-mediated activation ofβ-catenin/c-Myc/cyclinA2 signaling pathway and ER-independent upregulation of cyclin E1 and that quercetin may accelerate skin wound healing through promoting EpSC proliferation.As EpSCs are used not only in clinic to treat skin wounds but also as seed cells in skin tissue engineering,quercetin is a useful reagent to expand EpSCs for basic research,skin wound treatment,and skin tissue engineering. | Zhaodong Wang Guangliang Zhang Yingying Le Jihui Ju Ping Zhang Dapeng Wan Qiang Zhao Guangzhe Jin Hao Su Jinwei Liu Jiaxuan Feng Yi Fu Ruixing Hou | 2020 | Acta Biochimica et Biophysica Sinica2020,52,10: | 3 |

| 6 | VPS4B mutation impairs the osteogenic differentiation of dental follicle cells derived from a patient with dentin dysplasia type I显示文摘A splicing mutation in VPS4B can cause dentin dysplasia type I(DD-I),a hereditary autosomal-dominant disorder characterized by rootless teeth,the etiology of which is genetically heterogeneous.In our study,dental follicle cells(DFCs)were isolated and cultured from a patient with DD-I and compared with those from an age-matched,healthy control.In a previous study,this DD-I patient was confirmed to have a loss-of-function splicing mutation in VPS4B(IVS7+46C>G).The results from this study showed that the isolated DFCs were vimentin-positive and CK14-negative,indicating that the isolated cells were derived from the mesenchyme.DFCs harboring the VPS4B mutation had a significantly higher proliferation rate from day 3 to day 8 than control DFCs,indicating that VPS4B is involved in cell proliferation.The cells were then replenished with osteogenic medium to investigate how the VPS4B mutation affected osteogenic differentiation.Induction of osteogenesis,detected by alizarin red and alkaline phosphatase staining in vitro,was decreased in the DFCs from the DD-I patient compared to the control DFCs.Furthermore,we also found that the VPS4B mutation in the DD-I patient downregulated the expression of osteoblast-related genes,such as ALP,BSP,OCN,RUNX2,and their encoded proteins.These outcomes confirmed that the DD-I-associated VPS4B mutation could decrease the capacity of DFCs to differentiate during the mineralization process and may also impair physiological root formation and bone remodeling.This might provide valuable insights and implications for exploring the pathological mechanisms underlying DD-I root development. | Qiang Li Fangli Lu Tianxuan Chen Ke Zhang Yuping Lu Xiaocong Li Yingying Wang Ling Liu Qing Tian Fu Xiong Dong Chen | 2020 | International Journal of Oral Science2020,12,3: | 2 |
| 7 | Integrated Fertilization Regimes Boost Heavy Metals Accumulation and Biomass of Sedum alfredii Hance显示文摘The hyperaccumulator Sedum alfredii Hance(S.alfredii)may be employed for zinc(Zn)and cadmium(Cd)-polluted soil remediation.However,the low phytoremediation efficiency,related to the low biomass production,limits its use with that purpose.In this experiment,nitrogen(N),phosphorus(P),and potassium(K)fertilizers,and organic manure were applied to investigate the phytoremediation ability of S.alfredii.Hydroponic and pot experiments were conducted using Zn-Cd polluted soil.The hydroponic experiment indicated that appropriate fertilizer application could increase(p<0.05)the amount of accumulated Zn and Cd in S.alfredii.When N supply ranged from 0.5 to 2.5 mmol L^(−1),it could improve growth and accumulation of Zn and Cd in whole plants of S.alfredii.The 1 mmol L^(−1) N was an optimal N dosage for shoot biomass production and Cd accumulation in shoots,while the 2.5 mmol L^(−1) was an optimal N dosage for Zn accumulation in shoots.Both low(<0.05 mmol L^(−1))and high(>0.8 mmol L^(−1))P supply decreased growth,and Zn/Cd accumulation in whole plants of the studied species.The 0.1 mmol L^(−1) P was an optimal dosage for S.alfredii biomass production and Zn/Cd accumulation in shoots.The supply levels within the range from 0.3 to 1 mmol L^(−1) K could significantly improve the biomass production of S.alfredii and its capability to accumulate Zn and Cd in the biomass.The 0.5 mmol L^(−1) K was an optimal dosage for the whole biomass production and Zn accumulation in shoots,while the 1 mmol L^(−1) was an optimal K dosage for Zn accumulation in shoots,which was 17.2%higher than the control.Moreover,the soil pot experiment showed that the combination of organic(fermented manure)and inorganic fertilizers made significant effects on the Zn and Cd-polluted soil remediation by S.alfredii.These effects varied,however,with the application of different proportions of N,P,K and organic matter.The Zn accumulation by S.alfredii reached the highest efficiency ability under the highest fertilizer mixing rate(N:50 mg kg^(−1),P:40 mg kg^(−1),K:100 mg kg^(−1),organic matter:1%).Even more,S.alfredii showed the strongest ability to accumulate Cd with a lower fertilizer mixing rate(N:25mg kg^(−1),P:20mg kg^(−1),K:50 mg kg^(−1),organic matter:0.5%). | Kangning Zhou Yingying Zhang Jiasen Wu Chunying Dou Zihao Ye Zhengqian Ye Weijun Fu | 2021 | Phyton-International Journal of Experimental Botany2021,90,4: | 2 |

| 17 | Spectroscopic Measurements of Impurity Spectra on the EAST Tokamak显示文摘Ultraviolet (UV) and visible impurity spectra (200~750 nm) are commonly used to study plasma and wall interactions in magnetic fusion plasmas. Two optical multi-channel analysis (OMA) systems have been installed for the UV-visible spectrum measurement on EAST. These two OMA systems are both equipped with the Czerny-Turner (C-T) type spectrometer. The upper vacuum vessel and inner divertor baffle can be viewed simultaneously through two optical lenses. The OMA1 system is mainly used for multi-impurity lines radiation measurement. A 280 nm wavelength range can be covered by a 300 mm focal length spectrometer equipped with a 300 grooves/mm grating. The Dα/Hα line shapes can be resolved by the OMA2 system. The focal length is 750 mm. The spectral resolution can be up to 0.01 nm using a 1800 grooves/mm grating. The impurity behaviour and hydrogen ratio evolution after boroniztion, lithium coating, and siliconization are compared. Lithium coating has shown beneficial effects on the reduction of edge recycling and low Z impurity (C,O) influx.
