| 1 | Cardioprotective mechanism of SGLT2 inhibitor against myocardial infarction is through reduction of autosis显示文摘Sodium-glucose cotransporter 2(SGLT2)inhibitors reduce cardiovascular mortality in patients with diabetes mellitus but the protective mechanism remains elusive.Here we demonstrated that the SGLT2 inhibitor,Empagliflozin(EMPA),suppresses cardiomyocytes autosis(autophagic cell death)to confer cardioprotective effects.Using myocardial infarction(Ml)mouse models with and without diabetes mellitus,EMPA treatment significantly reduced infarct size,and myocardial fibrosis,thereby leading to improved cardiac function and survival.In the context of ischemia and nutritional glucose deprivation where autosis is already highly stimulated,EMPA directly inhibits the activity of the Na^(+)/H^(+)exchanger 1(NHE1)in the cardiomyocytes to regulate excessive autophagy.Knockdown of NHE1 significantly rescued glucose deprivation-induced autosis.In contrast,overexpression of NHE1 aggravated the cardiomyocytes death in response to starvation,which was effectively rescued by EMPA treatment.Furthermore,in vitro and in vivo analysis of NHE1 and Beclin 1 knockout mice validated that EMPA s cardioprotective effects are at least in part through downregulation of autophagic flux.These findings provide new insights for drug development,specifically targeting NHE1 and autosis for ventricular remodeling and heart failure after Ml in both diabetic and non-diabetic patients. | Kai Jiang Yue Xu Dandan Wang Feng Chen Zizhuo Tu Jie Qian Sheng Xu Yixiang Xu John Hwa Jian Li Hongcai Shang Yaozu Xiang | 2022 | Protein & Cell2022,13,5: | 13 |

| 4 | An experimental and numerical study on the charring rate of timber beams exposed to three-side fire显示文摘To investigate the charring rate of timber beams exposed to three-side fire, a total of fifteen new and used Douglas-Fir timber beams in four groups exposed to three-side fire were experimentally studied, together with the finite element analyses of the temperature distribution in the beam section. The durations of fire exposure were 0 (on a test piece), 10, 15, 20 and 30 min, according to the ISO 834 standard fire curve. The charring depth of each timber beam was calculated by averaging the values at one-third and two-thirds along each cross section to give the charring rate of timber beams. It was found that the timber beam's charring rate reduces as the duration of fire exposure increases and the vertical charring rate is slightly higher than the horizontal one. The areas of beam sections reduce due to charring and the strength and stiffness of the pyrolysis layer near the charring edge decrease owing to the high-temperature. The average horizontal and vertical charring rates are 0.98 and 1.08 mm/min, respectively. To take into account the difference between the test furnace temperature curve and the ISO 834 stand- ard fire curve, some corrections were made for these data to yield the solution for charring rate. With the help of the finite element software ANSYS, the temperature distribution of the wood's cross-section was analyzed. | 5 | Pertuzumab in combination with trastuzumab and chemotherapy for Chinese patients with HER2-positive metastatic gastric or gastroesophageal junction cancer:a subpopulation analysis of the JACOB trial显示文摘Background:The JACOB trial(NCT01774786)was a double-blinded,placebo-controlled,randomized,multicenter,international,phase III trial evaluating the efficacy and safety of adding pertuzumab to trastuzumab and chemo-therapy in first-line treatment of human epidermal growth factor receptor 2(HER2)-positive metastatic gastric cancer/gastroesophageal junction cancer(GEJC).The aim of this analysis was to investigate efficacy and safety outcomes in the Chinese subpopulation from the JACOB trial.Methods:This post hoc subpopulation analysis included all patients recruited in China's Mainland(n=163;20.9%)between June 2013 and January 2016.The patients were randomly assigned in a 1:1 ratio to receive pertuzumab plus trastuzumab and chemotherapy(pertuzumab group;n=82)or placebo plus trastuzumab and chemotherapy(con-trol group;n=81).Intravenous pertuzumab(840 mg)and trastuzumab(8 mg/kg loading and 6 mg/kg maintenance doses)were given every 3 weeks until disease progression or unacceptable toxicity.Chemotherapy was given as per standard regimens/doses of capecitabine or 5-fluorouracil plus cisplatin.The primary endpoint was overall survival(OS);secondary efficacy endpoints included progression-free survival(PFS),and overall objective response rate(ORR).Results:The median OS was 18.7 months in the pertuzumab group and 16.1 months in the control group(hazard ratio[HR]0.75;95%confidence interval[CI]0.49 to 1.14).The median PFS was 10.5 and 8.6 months in the pertuzumab and control groups,respectively(HR 0.85;95%CI 0.60 to 1.21),and the median ORRs were 68.9%and 55.7%,respectively.The treatment effect in this Chinese subpopulation showed consistency with that in the global ITT population with numerically lower HR for OS and PFS compared with the control group.The safety profiles of the pertuzumab and control groups in this Chinese subpopulation analysis were generally comparable.The most common grade 3-5adverse events were neutropenia,anemia,and leukopenia.However,due to the nature of being a post hoc subgroup analysis,the results presented here are descriptive only and need to be interpreted with caution.Conclusions:OS and PFS were numerically improved by adding pertuzumab to trastuzumab and chemotherapy as first-line treatment in Chinese HER2-positive gastric cancer/GEJC patients,and this regimen demonstrated an acceptable safety profile. | Tianshu Liu Yanru Qin Jin Li Ruihua Xu Jianming Xu Shujun Yang Shukui Qin Yuxian Bai Changping Wu Yixiang Mao Haiyan Wu Yilin Ge Lin Shen | 2019 | Cancer Communications2019,39,1: | 6 |
| 6 | Effectiveness of ozonated saline in the treatment of VX2 tumors in rabbits显示文摘Objective To investigate the efficacy, safety, and associated mechanisms of injected ozonated saline in the treatment of VX2 tumors. Methods A total of 90 rabbits bearing VX2 tumors on their left hind legs were randomly divided into three groups. The control group(A) received normal saline, while groups B and C received 20 μg/mL and 40 μg/mL O_3/O_2 ozonated saline, respectively. Rabbits were anesthetized and 2 mL of blood was drawn directly from the heart to measure serum concentrations of interleukin(IL-6) and tumor necrosis factor(TNF-α). The skin covering the VX2 tumor was cut in each rabbit and the maximum and vertical diameters of the tumors were measured under direct visualization. Several milliliters of saline, saline pre-treated with 20 μg/mL O_3/O_2, or saline pre-treated with 40 μg/mL O_3/O_2 were directly injected into the tumors of groups A, B, and C, respectively(injection volume(milliliter) =1/2 volume of the tumor, V = 1/2ab^2). On days 4, 8 and 12 following treatment, 10 rabbits were randomly selected from each group for blood sample collection, and serum IL-6 and TNF-α were measured. The tumor growth rate was calculated by measuring the maximum and vertical diameters of the VX2 tumors under direct visualization. All selected rabbits were euthanized and the tumors, livers, and lungs were removed for pathological examination. The tumor necrosis rate was calculated by cutting the tumors into half along the longitudinal axis and measuring the maximum diameters of the intratumoral necrotic regions. Results The average tumor volume in the three groups increased to different degrees at each time point; however, the average tumor growth rates in groups B and C were substantially lower than that in group A, exhibiting a statistically significant difference. The difference in the tumor growth rate between group B and group C was not statistically significant. The serum concentrations of IL-6 and TNF-α increased in the three groups at each time point, with larger increases occurring in groups B and C; however, the greater increases did not reach statistical significance. Although the diameters of the necrotic areas were larger in both groups B and C than that in group A, significant differences in necrotic area diameters were only found when comparing groups A and C on days 4 and 12 following treatment. Conclusion Direct injection of different concentrations of ozonated saline into VX2 tumors significantly increased intratumoral necrosis and reduced the tumor growth rate. | 7 | Angular-based spatially resolved laser-induced breakdown spectroscopy: a new technique for the effective enhancement of signals without an external time delay system显示文摘In this study, we developed a novel angularbased spatially resolved laser-induced breakdown spectroscopy(ABSR-LIBS) technique. This new method allows the spatially resolved spectroscopic properties of laserinduced plasma to be investigated using different angles and polar distances with a spatially adjustable fiber optics probe. The experimental evaluations suggested that the intensity of the Al signal distributions in the ‘‘side-view''depended greatly on the angle of the collection probe. The location of a collection probe at 50° would facilitate efficient spectral collection and minimize the disturbance of the plasma plume. The spectral signals in the plasma emissions had different distribution at various polar distances using the same collection angle, and the highest signal intensity was observed between 15 and 35 mm polar distances. This ABSR-LIBS technique improved both the LIBS signal and the maximum emission intensity of Al atoms by several times at the best spatial position in our experimental conditions. Furthermore, we successfully obtained well-resolved line emission spectroscopy signals without any external time delay using a nongated spectrometer based on the ABSR-LIBS system.
