1Antibody drug conjugate:the“biological missile”for targeted cancer therapy显示文摘Antibody–drug conjugate(ADC)is typically composed of a monoclonal antibody(mAbs)covalently attached to a cytotoxic drug via a chemical linker.It combines both the advantages of highly specific targeting ability and highly potent killing effect to achieve accurate and efficient elimination of cancer cells,which has become one of the hotspots for the research and development of anticancer drugs.Since the first ADC,Mylotarg®(gemtuzumab ozogamicin),was approved in 2000 by the US Food and Drug Administration(FDA),there have been 14 ADCs received market approval so far worldwide.Moreover,over 100 ADC candidates have been investigated in clinical stages at present.This kind of new anti-cancer drugs,known as“biological missiles”,is leading a new era of targeted cancer therapy.Herein,we conducted a review of the history and general mechanism of action of ADCs,and then briefly discussed the molecular aspects of key components of ADCs and the mechanisms by which these key factors influence the activities of ADCs.Moreover,we also reviewed the approved ADCs and other promising candidates in phase-3 clinical trials and discuss the current challenges and future perspectives for the development of next generations,which provide insights for the research and development of novel cancer therapeutics using ADCs.Zhiwen Fu Shijun Li Sifei Han Chen Shi Yu Zhang 2022Signal Transduction and Targeted Therapy2022,7,4:5
2Formal analysis of TPM2.0 key management APIs显示文摘The trusted platform module(TPM),a system component implemented on physical resources,is designed to enable computers to achieve a higher level of security than the security level that it is possible to achieve by software alone.For this reason,the TPM provides a way to store cryptographic keys and other sensitive data in its memory,which is shielded from access by any entity other than the TPM.Users who want to use those keys and data to achieve some security goals are restricted to interact with the TPM through its APIs defined in the TPM specification.Therefore,whether the TPM can provide Protected Capabilities it claimed depends to a large extent on the security of its APIs.In this paper,we devise a formal model,which is accessible to a fully mechanized analysis,for the key management APIs in the TPM2.0 specification.We identify and formalize security properties of these APIs in our model and then successfully use the automated prover Tamarin to obtain the first mechanized analysis of them.The analysis shows that the key management subset of TPM APIs preserves the secrecy of non-duplicable keys for unbounded numbers of fresh keys and handles.The analysis also reports that the key duplication mechanism,used to duplicate a key between two hierarchies,is vulnerable to impersonation attacks,which enable an adversary to recover the duplicated key of the originating hierarchy or import his own key into the destination hierarchy.Aiming at avoiding these vulnerabilities,we proposean approach,which restricts the originating and destination TPMs to authenticate each other’s identity during duplication.Then we formally demonstrate that our approach maintains the secrecy of duplicable keys when they are duplicated.Qianying Zhang Shijun Zhao Yu Qin Dengguo Feng 2014Chinese Science Bulletin2014,59,32:5
3Distinct immune signatures discriminate between asymptomatic and presymptomatic SARS-CoV-2^(pos)subjects显示文摘Increasing numbers of SARS-CoV-2-positive(SARS-CoV-2^(pos))subjects are detected at silent SARS-CoV-2 infection stage(SSIS).Yet,SSIS represents a poorly examined time-window wherein unknown immunity patterns may contribute to the fate determination towards persistently asymptomatic or overt disease.Here,we retrieved blood samples from 19 asymptomatic and 12 presymptomatic SARS-COV-2^(pos)subjects,47 age/gender-matched patients with mild or moderate COVID-19 and 27 normal subjects,and interrogated them with combined assays of 44-plex CyTOF,RNA-seq and Olink.Notably,both asymptomatic and presymptomatic subjects exhibited numerous readily detectable immunological alterations,while certain parameters including more severely decreased frequencies of CD107a^(low)classical monocytes,intermediate monocytes,non-classical monocytes and CD62L^(hi)CD8^(+)Tnaive cells,reduced plasma STC1 level but an increased frequency of CD4^(+)NKT cells combined to distinguish the latter.Intercorrelation analyses revealed a particular presymptomatic immunotype mainly manifesting as monocytic overactivation and differentiation blockage,a likely lymphocyte exhaustion and immunosuppression,yielding mechanistic insights into SSIS fate determination,which could potentially improve 5ARS-CoV-2 management.Shanhe Yu Caixia Di Shijun Chen Mingquan Guo Jiayang Yan Zhaoqin Zhu Li Liu Ruixue Feng Yinyin Xie Ruihong Zhang Juan Chen Mengxi Wang Dong Wei Hai Fang Tong Yin Jinyan Huang Saijuan Chen Hongzhou Lu Jiang Zhu Jieming Qu 2021Cell Research2021,31,11:4
4Neural stem cell transplantation in the hippocampus of rats with cerebral ischemia/reperfusion injury Activation of the phosphatidylinositol-3 kinase/Akt pathway and increased brain-derived neurotrophic factor expression显示文摘The phosphatidylinositol-3 kinase (PI3K)/Akt pathway and brain-derived neurotrophic factor (BDNF) are involved in neurological functional recovery following cerebral ischemia. Therefore, we hypothesized that mechanisms of neuroprotection by transplantation of neural stem cells (NSCs) on cerebral ischemia contributed to activation of the PI3K/Akt pathway and enhanced BDNF expression. In the present study, Wortmannin (a specific, covalent inhibitor of PI3K) was administered adjacent to ischemic hippocampus by stereotactic transplantation to further confirm the neuroprotective mechanisms of NSC transplantation following cerebral ischemia. Results showed that focal infarct volume was significantly smaller in the NSCs group, but the neurological behavior score in the NSC group was significantly greater than the middle cerebral artery occlusion model group, Wortmannin treatment group, and NSCs + Wortmannin treatment group. Protein expression of BDNF was significantly greater in the NSC group compared with the Wortmannin treatment group and NSCs + Wortmannin treatment group. 7Formal Analysis of Trusted Platform Module Commands for Compromising User Key显示文摘The Trusted Platform Module (TPM) is a dedicated hardware chip designed to provide a higher level of security for computing platform. All TPM functionalities are implemented in TPM commands to achieve specific security goals. We attempt to analyze the security properties of these commands, especially the key management API. Our study utilizes applied pi calculus to formalize the commands and determine how their security properties affect TPM key management. The attacker is assumed to call TPM commands without bounds and without knowing the TPM root key, expecting to obtain or replace the user key. The analysis goal in our study is to guarantee the corresponding property of API execution and the integrity of API data. We analyze the security properties of TPM commands with a process reduction method, identify the key-handle hijack attack on a TPM newly created key, and propose reasonable solutions to solve the problem. Then, we conduct an experiment involving a key-handle attack, which successfully replaces a user key with an attacker's key using malicious TPM software. This paper discloses the weakness of the relationship between the key handle and the key object. 8Spatio-temporal Differentiation in the Incidence of Influenza and Its Relationship with Air Pollution in China from 2004 to 2017显示文摘The Healthy China Initiative is a major health strategy being pursued by the country.To prevent and control different types of diseases as well as their complex variants,research on the spatio-temporal differentiation among and mechanisms of influence of epidemic diseases is growing worldwide.This study analyzed monthly data on the incidence of influenza by using different methods,including Moran’s I,the hotspot analysis model,concentration analysis,and correlation analysis,to determine the characteristics of spatiotemporal differentiation in the incidence of influenza across prefecture-level cities in China from 2004 to 2017,and to examine its relationship with air pollution.According to the results,the overall incidence of influenza in China exhibited a trend of increase from 2004 to 2017,with small peaks in 2009 and 2014.More cases of influenza were recorded in the first and fourth quarters of each year.Regions with higher incidences of influenza were concentrated in northwestern and northern China,and in the coastal areas of southeastern China.Over time,the distribution of regions with a higher incidence of influenza has shifted from the west to the east of the country.A significant relationship was observed between the incidence of influenza and factors related to air pollution.The contents of five air pollutants(PM_(2.5),PM10,SO_(2),NO_(2),and CO)were significantly positively correlated with the incidence of influenza,with a decreasing order of contribution to it of SO_(2)>CO>NO_(2)>PM_(2.5)>PM_(10).The content of O_(3) in the air was negatively correlated with the incidence of influenza.The influence of air pollution-related factors on the incidence of influenza in different regions and seasons showed minor differences.The large-scale empirical results provided here can supply a scientific basis for governmental disease control authorities to formulate strategies for regional prevention and control.SONG Yang ZHANG Yu WANG Tingting QIAN Sitong WANG Shijun 2021Chinese Geographical Science2021,31,5:2
9Effects of Root Zone Temperature on Growth and Photosynthetic Parameters of Grafted Cucumber显示文摘Taking Cucurbita maxima and Cucurbita moschata as root stocks,and‘Jinyou No 3'cucumber as scion,the effects of different root zone temperature conditions optimal temperature( CK)( 18-20 ℃),suboptimal temperature( 13-15 ℃) and low temperature( 8-10 ℃) on the growth and photosynthesis indexes were studied. The results showed that,compared with optimal temperature( CK),suboptimal temperature and low temperature produced a significant inhibition of growth on cucumbers. The plant height,stem diameter,leaf area,number of leaves and dry weight of aboveground part were all reduced,dry weight of underground part and root shoot ratio all increased,while the inhibition was more significant at low temperature. Low and suboptimal temperature conditions also reduced SPAD value,net photosynthetic rate,transpiration rate,intercellular CO_2 concentration and stomatal conductance of the grafted cucumber. 10Problems and countermeasures in construction of transmission line projects in permafrost regions显示文摘Construction of power transmission lines is becoming an important part of permafrost engineering in China.This paper reviews the construction status and problems of transmission lines in different countries,as well as corresponding solutions that would be of practical significance for sustainable engineering practices.Russia has the longest history of transmission line construction in permafrost areas,with transmission lines(mainly 220 kV and 500 kV) spanning approximately 100,000 km.However,all countries suffer from permafrost-related tower foundation stability problems caused by freezing-thawing hazards such as frost heave and thaw settlement,frost lifting,and harmful cryogenic phenomena.As point-line transmission line constructions,the lines,poles and towers should be reasonably selected and installed with a comprehensive consideration of frozen soil characteristics to effectively reduce the occurrence of freezing-thawing disasters.Reinforced concrete pile foundations are widely used in the permafrost regions,and construction in winter is also a universal practice.Moreover,facilitating engineering measures like thermosyphons are an effective way to reduce freezing-thawing hazards and to maintain the stability of tower foundations.GuoShang Wang QiHao Yu YanHui You Ze Zhang Lei Guo ShiJun Wang Yong Yu 2014Research in Cold and Arid Regions2014,6,5:2
11Prediction of solar cycle based on the invariant显示文摘A new method of predicting solar activities has been introduced in this paper. The method can predict both the occurrence time and the maximum number of sunspot at the same time. By studying the variation of sunspot, we find that the combination of the several variables was nearly in-variable during the entire solar cycles, as called invariant. And just only by determining the start time of a cycle, we can predict the occurrence time of cycles peak value accurately. Furthermore, according to observational data of the sunspot cycles, it showed that the sunspot maximum number has correlation not only with the prophase variety of the number in the cycle but also with the anaphase of the previous period. So we can introduce an equivalent regression coefficient, which can dynamically self-adapt to different cycle lengths, and effectively solve the inconsistency between the accuracy and the lead-time of the forecast. It can guarantee the satis-fied accuracy and effectively increases the lead-time of the forecast. This method can predict the maximum sunspot number for solar cycle at the approximate half rise of the period. 12Metabolic reprogramming and redox adaptation in sorafenib-resistant leukemia cells:detected by untargeted metabolomics and stable isotope tracing analysis显示文摘Background:Internal tandem duplications(ITD)within the juxtamembrane domain of FMS-like tyrosine kinase 3(FLT3)represent a poor prognostic indicator in acute myeloid leukemia(AML).Therapeutic benefits of tyrosine kinase inhibitors,such as sorafenib,are limited due to the emergence of drug resistance.While investigations have been con-ducted to improve the understanding of the molecular mechanisms underlying the resistance to this FLT3 inhibitor,a profile of cell functioning at the metabolite level and crosstalk between metabolic pathways has yet to be created.This study aimed to elucidate the alteration of metabolomic profile of leukemia cells resistant to the FLT3 inhibitor.Methods:We established two sorafenib-resistant cell lines carrying FLT3/ITD mutations,namely the murine BaF3/ITD-R and the human MV4-11-R cell lines.We performed a global untargeted metabolomics and stable isotope-labe-ling mass spectrometry analysis to identify the metabolic alterations relevant to the therapeutic resistance.Results:The resistant cells displayed fundamentally rewired metabolic profiles,characterized by a higher demand for glucose,accompanied by a reduction in glucose flux into the pentose phosphate pathway(PPP);and by an increase in oxidative stress,accompanied by an enhanced glutathione synthesis.We demonstrated that the highest scoring network of altered metabolites in resistant cells was related to nucleotide degradation.A stable isotope tracing experiment was performed and the results indicated a decrease in the quantity of glucose entering the PPP in resistant cells.Further experiment suggested that the inhibition of major enzymes in the PPP consist of glucose-6-phosphate dehydrogenase deficiency(G6PD)in the oxidative arm and transketolase(TKT)in the non-oxidative arm.In addition,we observed that chronic treatment with sorafenib resulted in an increased oxidative stress in FLT3/ITD-positive leu-kemia cells,which was accompanied by decreased cell proliferation and an enhanced antioxidant response.Conclusions:Our data regarding comparative metabolomics characterized a distinct metabolic and redox adaptation that may contribute to sorafenib resistance in FLT3/ITD-mutated leukemia cells.Xin You Weiye Jiang Wenhua Lu Hui Zhang Tiantian Yu Jingyu Tian Shijun Wen Guillermo Garcia-Manero Peng Huang Yumin Hu 2019Cancer Communications2019,39,1:1
13Nitrogen and atomic Fe dual-doped porous carbon nanocubes as superior electrocatalysts for acidic H2-O_(2) PEMFC and alkaline Zn-air battery显示文摘Air cathodes with high electrocatalytic activity are vital for developing H2/O_(2) proton exchange membrane fuel cells(PEMFC)and Zn-air batteries.However,the state-of-the-art air cathodes suffer from either limited catalytic activity or high cost,which thus hinder their applications.Herein,we designed ZIF-8 derived nitrogen and atomic iron dual-doped porous carbon nanocubes as high-quality catalysts for ORR,through a novel gas-doping approach.The porous carbon nanocubic architecture and abundant Fe-Nxactive species endow ZIF-8 derived single atomic iron catalyst(PCN-A@Fe SA)with superior catalytic activity,and surpass Pt/C and a majority of the reported catalysts.Both XAS and DFT calculations suggest that Fe2+N4 moieties are the main active centers that are favorable for oxygen affinity and OH*intermediate desorption,which can result in promising catalytic performance.Most importantly,PCNA@Fe SA can achieve power density of 514 m W cm^(-2) as cathodic catalyst in a PEMFC and discharge peak power density of 185 m W cm^(-2) in an alkaline Zn-air battery.The outstanding performance is derived from both the high specific surface area and high-density of iron single atom in nitrogen doped nanocubic carbon matrix.Xiudong Shi Zonghua Pu Bin Chi Mingrui Liu Siyan Yu Long Zheng Lijun Yang Ting Shu Shijun Liao 2021Journal of Energy Chemistry2021,30,8:1
