| 3 | Ginsenoside Rg1 protects against transient focal cerebral ischemic injury and suppresses its systemic metabolic changes in cerabral injury rats显示文摘Ginsenoside Rg1(GR),a major bioactive compound of traditional Chinese medicine,such as Panax ginseng or Radix Notoginseng,has been shown to exert neuroprotective effects against ischemic stroke.However,pharmacokinetic studies have suggested that GR could not be efficiently transported through the blood–brain barrier.The mechanism by which GR attenuates cerebral ischemic injury in vivo remains largely unknown.Therefore,this study explored potential neuro-protective effects of GR through its systemic metabolic regulating mechanism by using mass spectrometry–based metabolomic profiling.Rats with middle cerebral artery occlusion(MCAO) were treated with GR intravenously.Their metabolic profiles in serum were measured by gas chromatography coupled with mass spectrometry on 1 and 3 days after MCAO.GR exhibited a potent neuro-protective effect by significantly decreasing the neurological scores and infarct volume in the MCAO rats.Moreover,18 differential metabolites were tentatively identified,all of which appeared to correlate well with these disease indices.Our findings suggested that GR carries a therapeutic potential in stroke possibly through a feed-back mechanism by regulating systematic metabolic mediation. | Mingbao Lin Wei Sun Wan Gong Yasi Ding Yuanyan Zhuang Qi Hou | 2015 | Acta Pharmaceutica Sinica B2015,5,3: | 7 |

| 6 | Profiling kidney microRNAs from juvenile grass carp(Ctenopharyngodon idella) after 56 days of oral exposure to decabromodiphenyl ethane显示文摘Grass carp(Ctenopharyngodon idella) is one of the most important species in China.Decabromodiphenyl ethane(DBDPE) is a brominated flame retardant that has been used widely in industry, and has been observed to accumulate in the tissues of fish from South China. Evidence has shown that DBDPE is toxic to aquatic animals, but the molecular response has been unclear. MicroRNAs(miRNAs) are small noncoding and negative regulatory RNAs that are 20–24 nucleotides in length, which are involved in a wide range of biological processes. We took advantage of deep-sequencing techniques to accurately and comprehensively profile the kidney mi RNA expression of grass carp after 8 weeks of oral exposure to DBDPE. After mapping sequencing data to the genome and Expressed Sequence Tags(ESTs) of grass carp, we identified 493 miRNAs in the sequenced grass carp samples, which included 51 new miRNAs. The results indicated that 5 miRNAs were significantly down-regulated and 36 miRNAs were significantly up-regulated(FDR < 0.001,1.5-fold change) after DBDPE exposure. Real-time quantitative PCR(RT-q PCR) was performed on 4 miRNAs from the two samples, and the sequencing and RT-q PCR data were consistent. | 14 | Rice FLOURY ENDOSPERM22, encoding a pentatricopeptide repeat protein, is involved in both mitochondrial RNA splicing and editing and is crucial for endosperm development显示文摘Most of the reported P-type pentatricopeptide repeat(PPR) proteins play roles in organelle RNA stabilization and splicing. However, P-type PPRs involved in both RNA splicing and editing have rarely been reported, and their underlying mechanism remains largely unknown. Here, we report a rice floury endosperm22(flo22) mutant with delayed amyloplast development in endosperm cells. Map-based cloning and complementation tests demonstrated that FLO22 encodes a mitochondrion-localized P-type PPR protein.Mutation of FLO22 resulting in defective transsplicing of mitochondrial nad1 intron 1 and perhaps causing instability of mature transcripts affected assembly and activity of complex Ⅰ, and mitochondrial morphology and function. RNA-seq analysis showed that expression levels of many genes involved in starch and sucrose metabolism were significantly down-regulated in the flo22mutant compared with the wild type, whereas genes related to oxidative phosphorylation and the tricarboxylic acid cycle were significantly upregulated. In addition to involvement in splicing as a P-type PPR protein, we found that FLO22 interacted with DYW3, a DYW-type PPR protein, and they may function synergistically in mitochondrial RNA editing.
