| 1 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 2 | VO2·0.2H2O nanocuboids anchored onto graphene sheets as the cathode material for ultrahigh capacity aqueous zinc ion batteries显示文摘Aqueous Zinc-ion batteries(ZIBs),using zinc negative electrode and aqueous electrolyte,have attracted great attention in energy storage field due to the reliable safety and low-cost.A composite material comprised of VO2·0.2H2O nanocuboids anchored on graphene sheets(VOG)is synthesized through a facile and efficient microwave-assisted solvothermal strategy and is used as aqueous ZIBs cathode material.Owing to the synergistic effects between the high conductivity of graphene sheets and the desirable structural features of VO2·0.2H2O nanocuboids,the VOG electrode has excellent electronic and ionic transport ability,resulting in superior Zn ions storage performance.The Zn/VOG system delivers ultrahigh specific capacity of 423 mAh·g^−1 at 0.25 A·g^−1 and exhibits good cycling stability of up to 1,000 cycles at 8 A·g^−1 with 87%capacity retention.Systematical structural and elemental characterizations confirm that the interlayer space of VO2·0.2H2O nanocuboids can adapt to the reversible Zn ions insertion/extraction.The as-prepared VOG composite is a promising cathode material with remarkable electrochemical performance for low-cost and safe aqueous rechargeable ZIBs. | Dedong Jia Kun Zheng Ming Song Hua Tan Aitang Zhang Lihua Wang Lijun Yue Da Li Chenwei Li Jingquan Liu | 2020 | Nano Research2020,13,1: | 6 |

| 7 | Insulin-like growth factor 1-induced enolase 2 deacetylation by HDAC3 promotes metastasis of pancreatic cancer显示文摘Enolase 2(ENO2)is a key glycolytic enzyme in the metabolic process of glycolysis,but its potential function in pancreatic ductal adenocarcinoma(PDAC)is unclear.In this study,we observed a significant overexpression of ENO2 in PDAC tissues,and its expression was correlated with metastasis and poor prognosis in PDAC patients.K394 was identified as a major acetylation site in ENO2 that regulates its enzymatic activity,cell metabolism and PDAC progression.Knockdown of ENO2 suppressed tumor growth and liver metastasis in PDAC.Re-expression of wild-type(WT)ENO2,but not the K394 acetylation mimetic mutant,could reverse the decreased tumor malignancy.We further characterized histone deacetylase 3(HDAC3)and P300/CBP-associated factor(PCAF)as the potential deacetylase and acetyltransferase for ENO2,respectively.HDAC3-mediated deacetylation was shown to lead to ENO2 activation and enhancement of glycolysis.Importantly,insulin-like growth factor-1(IGF-1)was found to decrease K394 acetylation and stimulate ENO2 activity in a dose-and time-dependent manner.The PI3K/AKT/mTOR pathway facilitated the phosphorylation of HDAC3 on S424,which promoted K394 deacetylation and activation of ENO2.Linsitinib,an oral small-molecule inhibitor of IGF-1R,could inhibit IGF-1-induced ENO2 deacetylation by HDAC3 and the PI3K/AKT/mTOR pathway.Furthermore,linsitinib showed a different effect on the growth and metastasis of PDAC depending on the overexpression of WT versus K394-mutant ENO2.Our results reveal a novel mechanism by which acetylation negatively regulates ENO2 activity in the metastasis of PDAC by modulating glycolysis.Blockade of IGF-1-induced ENO2 deacetylation represents a promising strategy to prevent the development of PDAC. | Yan Zheng Chao Wu Jimeng Yang Yue Zhao Huliang Jia Min Xue Da Xu Feng Yang Deliang Fu Chaoqun Wang Beiyuan Hu Ze Zhang Tianen Li Shican Yan Xuan Wang Peter J.Nelson Christiane Bruns Lunxiu Qin Qiongzhu Dong | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 3 |

| 11 | Environmental and tectonic significance of Late Permian reefs in the Linxi and adjacent areas in Inner Mongolia of China显示文摘Many plateau-edge reefs and reef mounds of the Late Permian have been discovered in Linxi County and adjacent areas of Inner Mongolia, China. These reefs verify that the Hinggan-Inner Mongolia Area was an open and unobstructed sea-trough in Late Permian. Facies and sequences of reef strata in Member 4 and Member 5 of the Linxi Formation were studied for information to reconstruct the closing process of the Linxi-Jiutai chasm in the sea-trough, i.e., to constrain subsidence, uplift, and final closing. Ages of the reef-bearing strata have been determined to be late and end of Late Permian, based on ranges or abundances of diverse fossils. Six reef-building organic assemblages were distinguished, representing different ecologic conditions, according to morphological functions and paleoecological adaptations of organisms including primarily sponges, bryozoans, and calcareous algae. Two types of shelf slopes, the gentle slope and the steep slope, have been interpreted for Member 4 and Member 5 of the Linxi Formation, based on the assemblages. | 17 | Influence of aortic branch arteries on the hemodynamics of patient-specific type B aortic dissection following TEVAR显示文摘Objective:Because of the difficulty in obtaining patient specific flow data,branch arteries are often neglected in the flow simulations of a patient-specific type B aortic dissection(AD)following thoracic endovascular repair(TEVAR).This study aims to investigate the influence of the aortic branch arteries on the hemodynamics and physical flow phenomena commonly associated with aortic modeling of an AD after TEVAR.Methods:The three-dimensional geometry of a type B AD following TEVAR was generated from a high-quality CT dataset.Simulations were performed with all branch arteries,without the four visceral arteries of the abdominal aorta(FVAoAA),and without the upper three branches of the aortic arch(UTBoAA).Results:The removal of the UTBoAA has marginal effect on the flow patterns and hemodynamic parameters in the false lumen(FL);however,regardless of whether there are aorta branches supplied to the FL or not,the omission of FVAoAA will result in a wrong prediction of the flow direction at tears and flow volume to the FL.Conclusions:The omission of aorta branch arteries has great impact on the hemodynamic prediction including flow field and wall shear stress related parameters in a type B AD after TEVAR,which may induce misleading estimates of the potential thrombus in the FL.It is strongly recommended to keep aorta branches in the computational model to predict the post-operative hemodynamic performance in a type B AD following TEVAR. | Yi Jiang Yue Qiu Da Li Ding Yuan Tinghui Zheng Liqing Peng | 2019 | Medicine in Novel Technology and Devices2019,,4: | 1 |
