1Climate and litter C/N ratio constrain soil organic carbon accumulation显示文摘Soil organic carbon(SOC) plays critical roles in stabilizing atmospheric CO2 concentration, but the mechanistic controls on the amount and distribution of SOC on global scales are not well understood. In turn, this has hampered the ability to model global C budgets and to find measures to mitigate climate change. Here, based on the data from a large field survey campaign with 2600 plots across China’s forest ecosystems and a global collection of published data from forested land, we find that a low litter carbon-to-nitrogen ratio(C/N) and high wetness index(P/PET, precipitation-to-potentialevapotranspiration ratio) are the two factors that promote SOC accumulation, with only minor contributions of litter quantity and soil texture. The field survey data demonstrated that high plant diversity decreased litter C/N and thus indirectly promoted SOC accumulation by increasing the litter quality. We conclude that any changes in plant-community composition, plant-species richness and environmental factors that can reduce the litter C/N ratio, or climatic changes that increase wetness index, may promote SOC accumulation. 3Application of ~1H NMR-based metabolomics for discrimination of different parts and development of a new processing workflow for Cistanche deserticola显示文摘Cistanche deserticola(CD) is one of the two authoritative source plants of Cistanches Herba, a well-known medicinal plant. Herein,~1H NMR spectroscopy was employed to characterize the chemical profile and to distinguish the different parts, as well as to propose a new processing workflow for CD.Signal assignment was achieved by multiple one and two dimensional NMR spectroscopic techniques in combination with available databases and authentic compounds. The upper parts of the plant were distinguished from the lower parts by combining ~1H NMR spectroscopic dataset with multivariate statistical analysis. A new processing method that hyphenated steaming with freeze-drying, was demonstrated to be superior to either steaming coupled with oven-drying or direct freeze-drying via holistic ~1H NMR-based metabolomic characterization. Phenylethanoid glycosides, mainly echinacoside and acteoside, were screened out and confirmed as the chemical markers responsible for exhibiting the superiority of the new processing workflow, whereas serial primary metabolites, especially carbohydrates and tricarboxylic acid cycle metabolites, were found as the primary molecules governing the discrimination between the upper and lower parts of the plant. 4Authentic compound-free strategy for simultaneous determination of primary coumarins in Peucedani Radix using offline high performance liquid chromatography–nuclear magnetic resonance spectroscopy–tandem mass spectrometry显示文摘Herein, a strategy is proposed for the simultaneous determination of primary coumarins in Peucedani Radix(Chinese name: Qianhu). The methodology consists of three consecutive steps: 1) Semi-preparative LC in combination with a home-made automated fraction collection module to fragment the universal metabolome standard into ten fractions(Frs. I–X); 2) LC–accurate MS/MS and quantitative1 H NMR spectroscopy conducted in parallel to acquire the qualitative and quantitative data of each fraction; 3) Robust identification and quantification of components by use of LC coupled to multiple reaction monitoring. In this final step, the most significant fractions(Frs. III–X) were pooled to serve as the pseudo-mixed standard solution. Meticulous online parameter optimization was performed to obtain the optimal parameters, including ion transitions and collision energies. Concerns were particularly paid onto pursuing the parameters being capable of monitoring regiospecific isomers, notably praeruptorin E vs. 3′-isovaleryl-4′-angeloylkhellactone. The quantitative performance of the method was validated according to diverse assays. Eleven primary coumarins(1–11) were unambiguously identified and absolutely quantified, even though no external reference compound was used. 5Redundant CAMTA Transcription Factors Negatively Regulate the Biosynthesis of Salicylic Acid and N-Hydroxypipecolic Acid by Modulating the Expression of SARD1 and CBP60g显示文摘Two signal molecules, salicylic acid (SA) and N-hydroxypipecolic acid (NHP), play critical roles in plant immunity. The biosynthetic genes of both compounds are positively regulated by master immune-regulating transcription factors SARD1 and CBP60g. However, the relationship between the SA and NHP pathways is unclear. CALMODULIN-BINDING TRANSCRIPTION FACTOR 1 (CAMTA1), CAMTA2, and CAMTA3 are known redundant negative regulators of plant immunity, but the underlying mechanism also remains largely unknown. In this study, through chromatin immunoprecipitation and electrophoretic mobility shift assays, we uncovered that CBP60g is a direct target of CAMTA3, which also negatively regulates the expression of SARD1, presumably via an indirect effect. The autoimmunity of camta3-1 is suppressed by sard1 cbp60g double mutant as well as ald1 and fmo1, two single mutants defective in NHP biosynthesis. Interestingly, a suppressor screen conducted in the camta1/ 2/ 3 triple mutant background yielded various mutants blocking biosynthesis or signaling of either SA or NHP, leading to nearly complete suppression of the extreme autoimmunity of camta1/ 2/ 3, suggesting that the SA and NHP pathways can mutually amplify each other. 7A MEMS Device for in-situ TEM Test of SCS Nanobeam显示文摘In-situ tensile testing in TEM (transmission electron microscopy) is a useful tool for studying mechanical properties of nano-structures because it can provide quanti-tative information on sample deformation at atomic scale. To facilitate in-situ TEM tensile tests of SCS (single crystal silicon) nanobeam, a MEMS tensile-testing chip was designed and fabricated. The chip was fabricated by means of bulk micro-machining and wafer bonding techniques. An SCS nanobeam, a comb drive ac-tuator, a force sensor beam and an electron beam window were integrated into the chip. With the on-chip comb-drive-actuator stretching the nanobeam and in-situ TEM observation, tensile test on a 90 nm-thick nanobeam was performed and the strain-stress relationship was obtained. 9Separation and Manipulation of Rare-earth Oxide Particles by Dielectrophoresis显示文摘A challenge in chemical engineering is the separation and purification of rare-earth elements and their compounds. We report the design and manufacture of a dielectrophoresis(DEP) microchip of microelectrode arrays. This microchip device is constructed in order to use DEP to capture micro-particles of rare-earth oxides in petro-leum. Dielectrophoretic behavior of micro-particles of rare-earth oxides in oil media is explored. The dielectropho-retic effects of particles under different conditions are investigated. It is showed that the prepared microchip is suit-able for use in the investigation of dielectrophoretic responses of the rare-earth oxides in oil media. The factors such as frequency,particle size and valence of rare-earth metal are discussed. When the frequency is fixed,the transla-tion voltage decreases as particle size increases. Lower frequencies are more effective for manipulation of inorganic particles in oil media. Particles of the same rare-earth oxide with different size,as well as particles of different rare-earth oxides,are captured in different regions of the field by regulating DEP conditions. 10Analysis of the efficacy of transcatheter arterial infusion chemotherapy in the treatment of pancreatic carcinoma显示文摘Objective:To evaluate the clinical efficacy of infusion of gemcitabine(GEM) and fluorouracil(5-FU)through the celiac artery and superior mesenteric artery in the treatment of pancreatic carcinoma(PC).Methods:We analyzed 20 patients diagnosed clinically or pathologically with PC,without metastases,who had an estimated survival duration of>3 months in our department from May 2009 to December 2014.Nine patients were treated directly without surgical resection of the tumor,while the other 11 patients were treated after surgery.In all patients,the femoral artery was punctured using the Seldinger technique,and a catheter was placed in the opening of the celiac artery or the superior mesenteric artery.We administered 500 mg/m2 GEM and 500 mg/m2 5-FU.Observational data included data on clinical efficacy and survival rates during the follow-up period of 3-72 months.Results:Twenty patients were treated 85 times with transcatheter arterial infusion chemotherapy(TAI).The survival rates were 80%,40%,35%,20%,10%,and 5% at 3,6,12,24,and 72 months,respectively.Conclusion: TAI chemotherapy with GEM and 5-FU may be a therapeutic option for the treatment of PC.Chunhui Nie Yuelin Zhang Guanhui Zhou Tanyang Zhou Tongyin Zhu Baoquan Wang Shengqun Chen Hongliang Wang Ziniu Yu Li Jing Junhui Sun 2021Journal of Interventional Medicine2021,4,1:4

12C:N:P stoichiometry of Ericaceae species in shrubland biomes across Southern China:influences of climate,soil and species identity显示文摘Aims Carbon(C),nitrogen(N)and phosphorus(P)stoichiometry strongly affect functions and nutrient cycling within ecosystems.However,the related researches in shrubs were very limited.In this study,we aimed to inves-tigate leaf stoichiometry and its driving factors in shrubs,and whether stoichiometry significantly differs among closely related species.Methods We analyzed leaf C,N and P concentrations and their ratios in 32 species of Ericaceae from 161 sites across southern China.We examined the relationships of leaf stoichiometry with environmen-tal variables using linear regressions,and quantified the interactive and independent effects of climate,soil and species on foliar stoi-chiometry using general linear models(GLM).Important Findings The foliar C,N and P contents of Ericaceae were 484.66,14.44 and 1.06 mg g−1,respectively.Leaf C,N and P concentrations and their ratios in Ericaceae were significantly related with latitude and altitude,except the N:P insignificantly correlated with latitude.Climate(mean annual temperature and precipitation)and soil properties(soil C,N and P and bulk density)were significantly influenced element stoichiom-etry.The GLM analysis showed that soil exerted a greater direct effect on leaf stoichiometry than climate did,and climate affected leaf traits mainly via indirect ways.Further,soil properties had stronger influ-ences on leaf P than on leaf C and N.Among all independent factors examined,we found species accounted for the largest proportion of the variation in foliar stoichiometry.These results suggest that species can largely influence foliar stoichiometry,even at a lower taxonomic level.Qiang Zhang Qing Liu Huajun Yin Chunzhang Zhao Lin Zhang Guoying Zhou Chunying Yin Zhijun Lu Gaoming Xiong Yuelin Li Jiaxiang Li Wenting Xu Zhiyao Tang Zongqiang Xie 2019Journal of Plant Ecology2019,12,2:4
13Investigating the relationships between facets of work task and selection and query-related behavior显示文摘Purpose: This study aims to explore the relationships between different facets of work task and selection and query-related behavior.Design/methodology/approach:An experiment was conducted to explore the issue. The researcher recruited 24 participants and assigned six simulated work task situations to each of them. Each experiment lasted around 2 hours and was recorded by the software tool Morae.Findings: Time(frequency) and time(length) are more closely related to user's selection and query-related behavior compared to the facet ‘process' of work task. Knowledge level of work task topic, degree of work task difficulty, and subjective work task complexity are significantly correlated with selection and query-related behavior. Work task difficulty and work task complexity are different concepts. Subjective work task complexity, work task difficulty, and knowledge of work task topic are significantly correlated with user's selection and query-related behavior.Research limitations/implications: The limitations of this study include a small sample size,limited work task situations, and possible spurious relationships. This study has implications in informing task-based information seeking/search/retrieval research and interactive information retrieval(IIR) systems design.Originality/values: Previous studies usually did not touch upon how different facets of work tasks affected interactive activities. Some studies examining task complexity and information behavior were concerned with how work tasks affect users' behavior at information-seeking level, rather than at information search level. 16Negative regulation of resistance protein-mediated immunity by master transcription factors SARD_1 and CBP60g显示文摘Salicylic acid(SA) is an essential defence hormone in plants. Upon pathogen infection, induced biosynthesis of SA is mediated by Isochorismate synthase 1(ICS1), whose gene transcription is controlled mainly through two redundant transcription factors, SAR Deficient 1(SARD_1) and Calmodulinbinding protein 60-like g(CBP60g). Although these master transcription factors regulate not only positive,but also negative regulators of immunity, how they control signaling events downstream of different immune receptors is unclear. Using autoimmune mutants activating immunity mediated by different receptors we show that, although the sard1 cbp60 g double mutant almost fully suppresses the activation of defence mediated by suppressor of npr1-1, constitutive 2(snc2), it strikingly enhances snc1, which carries a gain-of-function mutation in an intracellular nucleotidebinding leucine-rich repeat(NLR) immune receptor.This negative regulation of immunity is achieved through the transcriptional regulation of negative regulators, such as Nudix hydrolase homolog 6(NUDT6).
