1Genetic analysis and gene mapping of a narrow leaf mutant in rice (Oryza sativa L.)显示文摘A narrow leaf mutant was obtained after T-DNA transformation conducted on a rice variety Zhonghua 11. Several abnormal morphological characteristics, including semi-dwarf, delayed flowering time, narrow and inward rolling leaves, and lower seed-setting, were observed. The rate of net photosynthesis (un-der saturate light) of flag leaves in the mutant was significantly lower than that of the wild type. More-over, the leaf transpiration rate and stomatal conductance in the mutant flag leaf were lower than those of the wild type at the grain filling stage. It was found that the mutant phenotype was not caused by the T-DNA insertion. Genetic analysis showed that the mutant was controlled by a single recessive gene, designated as nal3(t). A genetic linkage map was constructed using a large F2 mapping population de-rived from a cross between nal3(t) and an indica variety Longtefu B with 6 polymorphic markers on chromosome 12 identified from 366 SSR markers by the BAS method. Gene nal3(t) was mapped be-tween the markers RM7018 and RM3331. Fine mapping of nal3(t) locus was conducted with 22 newly developed STS markers based on the sequence diversity around the region harboring nal3(t) between Nipponbare and 93-11, and nal3(t) was finally mapped to a 136-kb region between the STS markers NS10 and RH12-8.WANG DeKai LIU HeQin LI KeLei LI SuJuan TAO YueZhi 2009Chinese Science Bulletin2009,54,5:19

3Recent advances in cell sheet technology for bone and cartilage regeneration: from preparation to application显示文摘Bone defects caused by trauma,tumour resection,infection and congenital deformities,together with articular cartilage defects and cartilage–subchondral bone complex defects caused by trauma and degenerative diseases,remain great challenges for clinicians.Novel strategies utilising cell sheet technology to enhance bone and cartilage regeneration are being developed.The cell sheet technology has shown great clinical potential in regenerative medicine due to its effective preservation of cell–cell connections and extracellular matrix and its scaffold-free nature.This review will first introduce several widely used cell sheet preparation systems,including traditional approaches and recent improvements,as well as their advantages and shortcomings.Recent advances in utilising cell sheet technology to regenerate bone or cartilage defects and bone–cartilage complex defects will be reviewed.The key challenges and future research directions for the application of cell sheet technology in bone and cartilage regeneration will also be discussed.Yuezhi Lu Wenjie Zhang Jie Wang Guangzheng Yang Shi Yin Tingting Tang Chunhua Yu Xinquan Jiang 2019International Journal of Oral Science2019,11,2:14
4OspTAC2 encodes a pentatricopeptide repeat protein and regulates rice chloroplast development显示文摘Functional chloroplast generation depends on the precise coordination of gene expression between the plastid and the nucleus and is essential for plant growth and development. In this study, a rice(Oryza sativa) mutant that exhibited albino and seedling-lethal phenotypes was isolated from a60Co-irradiated rice population. The mutant gene was identified as an ortholog of the Arabidopsis plastid transcriptionally active chromosome protein 2(p TAC2) gene, and the mutant strain was designated osptac2. Sequence and transcription analyses showed that Osp TAC2 encodes a putative chloroplast protein consisting of 10 pentratricopeptide repeat(PPR) domains and a C-terminal small Mut S-related(SMR) domain. Cytological observations via microscopy showed that the Osp TAC2-green fluorescent fusion protein is localized in the chloroplasts. Transmission electron microscopy revealed that the chloroplast of the osptac2 mutant lacks an organized thylakoid membrane. The transcript levels of all investigated PEP(plastid-encoded RNA polymerase)-dependent genes were dramatically reduced in the osptac2 mutant, whereas the transcript levels of NEP(nuclear-encoded polymerase)-dependent genes were increased. 5Marginal sealing around integral bilayer scaffolds for repairing osteochondral defects based on photocurable silk hydrogels显示文摘Osteochondral repair remains a major challenge in current clinical practice despite significant advances in tissue engineering.In particular,the lateral integration of neocartilage into surrounding native cartilage is a difficult and inadequately addressed problem that determines the success of tissue repair.Here,a novel design of an integral bilayer scaffold combined with a photocurable silk sealant for osteochondral repair is reported.First,we fabricated a bilayer silk scaffold with a cartilage layer resembling native cartilage in surface morphology and mechanical strength and a BMP-2-loaded porous subchondral bone layer that facilitated the osteogenic differentiation of BMSCs.Second,a TGF-β3-loaded methacrylated silk fibroin sealant(Sil-MA)exhibiting biocompatibility and good adhesive properties was developed and confirmed to promote chondrocyte migration and differentiation.Importantly,this TGF-β3-loaded Sil-MA hydrogel provided a bridge between the cartilage layer of the scaffold and the surrounding cartilage and then guided new cartilage to grow towards and replace the degraded cartilage layer from the surrounding native cartilage in the early stage of knee repair.Thus,osteochondral regeneration and superior lateral integration were achieved in vivo by using this composite.These results demonstrate that the new approach of marginal sealing around the cartilage layer of bilayer scaffolds with Sil-MA hydrogel has tremendous potential for clinical use in osteochondral regeneration.Xiaolin Wu Mingliang Zhou Fei Jiang Shi Yin Sihan Lin Guangzheng Yang Yuezhi Lu Wenjie Zhang Xinquan Jiang 2021Bioactive Materials2021,6,11:7
6Transparent Computing: Spatio-Temporal Extension on von Neumann Architecture for Cloud Services显示文摘The rapid advancements in hardware, software, and computer networks have facilitated the shift of the computing paradigm from mainframe to cloud computing, in which users can get their desired services anytime, anywhere, and by any means. However, cloud computing also presents many challenges, one of which is the difficulty in allowing users to freely obtain desired services, such as heterogeneous OSes and applications, via different light-weight devices. We have proposed a new paradigm by spatio-temporally extending the von Neumann architecture, called transparent computing, to centrally store and manage the commodity programs including OS codes, while streaming them to be run in non-state clients. This leads to a service-centric computing environment, in which users can select the desired services on demand, without concern for these services' administration, such as their installation, maintenance, management, and upgrade. In this paper, we introduce a novel concept, namely Meta OS, to support such program streaming through a distributed 4VP + platform. Based on this platform, a pilot system has been implemented, which supports Windows and Linux environments. We verify the effectiveness of the platform through both real deployments and testbed experiments. 10Characterization and molecular cloning of a serine hydroxymethyltransferase 1(OsSHM_1) in rice显示文摘Serine hydroxymethyltransferase(SHMT) is important for one carbon metabolism and photorespiration in higher plants for its participation in plant growth and development,and resistance to biotic and abiotic stresses. A rice serine hydroxymethyltransferase gene, Os SHM1, an ortholog of Arabidopsis SHM1, was isolated using map-based cloning. The osshm1 mutant had chlorotic lesions and a considerably smaller,lethal phenotype under natural ambient CO2 concentrations,but could be restored to wild type with normal growth under elevated CO2levels(0.5% CO2), showing a typical photorespiratory phenotype. The data from antioxidant enzymes activity measurement suggested that osshm1 was subjected to signi fi cant oxidative stress. Also, Os SHM1 was expressed in allorgans tested(root, culm, leaf, and young panicle) but predominantly in leaves. Os SHM1 protein is localized to the mitochondria. 11Expression and responses to dehydration and salinity stresses of V-PPase gene members in wheat显示文摘Vacuolar H+-translocating pyrophosphatase(V-PPase) is a key enzyme related to plant growth as well as abiotic stress tolerance.In this work,wheat V-PPase genes TaVP1,TaVP2 and TaVP3 were identified.TaVP1 and TaVP2 are more similar to each other than to TaVP3.Their deduced polypeptide sequences preserve the topological structure and essential residues of V-PPases.Phylogenetic studies suggested that monocot plants,at least monocot grasses,have three VP paralogs.TaVP3 transcripts were only detected in developing seeds,and no TaVP2 transcripts were found in germinating seeds.TaVP2 was mainly expressed in shoot tissues and down-regulated in leaves under dehydration.Its expression was up-regulated in roots under high salinity.TaVP1 was relatively more ubiquitously and evenly expressed than TaVP2.Its expression level in roots was highest among the tissues examined,and was inducible by salinity stress.These results indicated that the V-PPase gene paralogs in wheat are differentially regulated spatially and in response to dehydration and salinity stresses.Yuezhi Wang Haibin Xu Guangxiang Zhang Huilan Zhu Lixia Zhang Zhengzhi Zhang Caiqin Zhang Zhengqiang Ma 2009Journal of Genetics and Genomics2009,36,12:3

13EEG-correlated fMRI of P3b component in P300 waves显示文摘Electroencephalography-correlated functional magnetic resonance imaging (EEG/fMRI) can be used to identify blood oxygen level-dependent (BOLD) signal changes associated with both physiological and pathological EEG events. Here, we implemented continuous and simulta-neous EEG/fMRI to identify BOLD signal changes related to P3b component of P300, and 64 channels of EEG were re-corded in 11 subjects during Landot Ring task inside a 1.5 T functional magnet resonance (MR) scanner using an MR-compatible EEG recording system. Functional scanning by echoplanar imaging covered almost the entire cerebrum every 2 s, leaving gaps of 2 s without scanning. Off-line MRI artifact subtraction software was applied to obtain continu-ous EEG data. Additionally, a P300 wave matched filter was constructed to inspect P300 wave occurrence following every target stimulus, target stimuli inspected to induce P300 were detected and their MRI scan number were then used as input for an event-related fMRI analysis. Finally MRI statistical parametric maps were constructed and corrected for multi-ple comparisons. By random effect group analysis, activa-tions were detected in the right superior parietal lobule and bilaterally in inferior parietal lobule(p<0.001, uncorrected). 17Proteome and transcriptome profile analysis reveals regulatory and stress-responsive networks in the russet fruit skin of sand pear显示文摘The epidermal tissues of the cuticular membrane(CM)and periderm membrane(PM)confer first-line protection from environmental stresses in terrestrial plants.Although PM protection is essentially ubiquitous in plants,the protective mechanism,the function of many transcription factors and enzymes,and the genetic control of metabolic signaling pathways are poorly understood.Different microphenotypes and cellular components in russet(PM-covered)and green(CM-covered)fruit skins of pear were revealed by scanning and transmission electron microscopy.The two types of fruit skins showed distinct phytohormone accumulation,and different transcriptomic and proteomic profiles.The enriched pathways were detected by differentially expressed genes and proteins from the two omics analyses.A detailed analysis of the suberin biosynthesis pathways identified the regulatory signaling network,highlighting the general mechanisms required for periderm formation in russet fruit skin.The regulation of aquaporins at the protein level should play an important role in the specialized functions of russet fruit skin and PM-covered plant tissues.Yuezhi Wang Meisong Dai Danying Cai Zebin Shi 2020Horticulture Research2020,7,1:1

20Maintenance of species boundaries in three sympatric Ligularia(Senecioneae,Asteraceae)species显示文摘The key process in speciation concerns the formation and maintenance of reproductive isolating barriers between diverging lineages.Although species boundaries are frequently investigated between two species across many taxa,reproductive isolating barriers among multiple species(>2)that would represent the most common phenomenon in nature,remain to be clarified.Here,we use double digest restriction-site associated DNA(ddRAD)sequencing to examine patterns of hybridization at a sympatric site where three Ligularia species grow together and verify whether those patterns contribute to the maintenance of boundaries among species.The results based on the RAD SNP datasets indicated hybridization Ligularia cyathiceps?L.duciformis and L.duciformis?L.yunnanensis were both restricted to F1s plus a few first-generation backcrosses and no gene introgression were identified,giving rise to strong reproductive isolation among hybridizing species.Moreover,hybrid swarm simulation,using HYBRIDLAB,indicated the RAD SNP datasets had sufficient discriminatory power for accurate hybrid detection.We conclude that parental species show strong reproductive isolation and they still maintain species boundaries,which may be the key mechanism to maintain species diversity of Ligualria in the eastern Qinghai-Tibetan Plateau and adjacent areas.Moreover,this study highlights the effectiveness of RAD sequencing in hybridization studies.Ningning Zhang Yongpeng Ma Ryan A.Folk Jiaojun Yu Yuezhi Pan Xun Gong 2018Journal of Integrative Plant Biology2018,60,10:1
