| 2 | Synergistic effect of fluorination on both donor and acceptor materials for high performance non-fullerene polymer solar cells with 13.5% efficiency显示文摘A high performance polymer solar cells(PSCs) based on polymer donor PM6 containing fluorinated thienyl benzodithiophene unit and n-type organic semiconductor acceptor IT-4 F containing fluorinated end-groups were developed. In addition to complementary absorption spectra(300–830 nm) with IT-4 F, the PM6 also has a deep HOMO(the highest occupied molecular) level(-5.50 e V), which will lower the open-circuit voltage(V_(oc)) sacrifice and reduce the E_(loss) of the IT-4 F-based PSCs. Moreover, the strong crystallinity of PM6 is beneficial to form favorable blend morphology and hence to suppress recombination. As a result, in comparison with the PSCs based on a non-fluorinated D/A pair of PBDB-T:ITIC with a medium PCE of 11.2%, the PM6:IT-4 Fbased PSCs yielded an impressive PCE of 13.5% due to the synergistic effect of fluorination on both donor and acceptor, which is among the highest values recorded in the literatures for PSCs to date. Furthermore, a PCE of 12.2% was remained with the active layer thickness of up to 285 nm and a high PCE of 11.4% was also obtained with a large device area of 1 cm^2. In addition, the devices also showed good storage, thermal and illumination stabilities with respect to the efficiency. | 5 | A simple and efficient method for CRISPR/Cas9-induced mutant screening显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9) system provides a technological breakthrough in mutant generation. Several methods such as the polymerase chain reaction(PCR)/restriction enzyme(RE) assay, T7 endonuclease I(T7EI) assay, Surveyor nuclease assay, PAGE-based genotyping assay, and high-resolution melting(HRM) analysis-based assay have been developed for screening CRISPR/Cas9-induced mutants. However, these methods are timeand labour-intensive and may also be sequence-limited or require very expensive equipment. Here, we described a cost-effective and sensitive screening technique based on conventional PCR, annealing at critical temperature PCR(ACT-PCR), for identifying mutants. ACT-PCR requires only a single PCR step followed by agarose gel electrophoresis. We demonstrated that ACT-PCR accurately distinguished CRISPR/Cas9-induced mutants from wild type in both rice and zebrafish. Moreover, the method can be adapted for accurately determining mutation frequency in cultured cells. | 6 | Impact of anthropogenic activities on the sediment microbial communities of Baiyangdian shallow lake显示文摘Microorganisms are recognized as sensors of external disturbance in freshwater ecosystems.In this study,sediment samples from regions with strong human activity interference(four sites),moderate human activity interference(five sites)and minor human activity interference(five sites)in Baiyangdian Lake were collected;and their physiological and chemical properties and heavy metals contents were analyzed.The structures of bacterial and fungal communities were tested using 16S ribosomal Ribonucleic Acid(rRNA)and Internal Transcribed Spacer 2(ITS2)high-throughput sequencing technologies.The results indicated that Proteobacteria was the most abundant bacterial phylum.Ascomycota,Basidiomycota and Ciliophora were the dominant phyla of fungi.Obvious differences of microbial genera among the three regions were observed,especially fungal genera.With intensified disturbance by human activities,the Simpson and Shannon indices of bacteria significantly decreased.The fungal communities were clustered into three groups in accordance with the different human disturbance levels.The bacterial communities of sediment samples under moderate and minor human disturbance appeared similar,but different from those under strong human disturbance.Redundancy analysis indicated that zinc,total phosphorus,copper,and lead were the dominant factors that influenced the bacterial community.Nitrifiers and sulfate-reducing bacteria were influenced by pH,nitrate(NO3 ),and total nitrogen.The fungal community was significantly affected by total phosphorus.This study reveals that sediment microbial community structures among different regions of a shallow lake are mainly impacted by human activities. | Wenjun Wang Yujun Yi Yufeng Yang Yang Zhou Shanghong Zhang Xuan Wang Zhifeng Yang | 2020 | International Journal of Sediment Research2020,35,2: | 14 |

Racemic and scalemic mixtures of these enantiomers were separated by HPLC on a chiral semi-preparative column.Their structures including absolute configurations were determined by extensive spectroscopic analysis in conjunction with the calculation of electronic circular dichroism(ECD) spectra. The enantiomer pairs possess parent structures of 2-oxo-1,2,3,4-tetrahydroquinoline-4-carboxylic acid, indolo[2,1-b]quinazolinone, and 3-thioxohexahydro-1H-pyrrolo[1,2-c]imidazol-1-one, respectively. | 10 | Clinical characteristics and effectiveness of antiviral agents in hospitalized children with infectious mononucleosis in China:A multicenter retrospective study显示文摘Importance:The clinical characteristics of infectious mononucleosis(IM)in Chinese children have not been evaluated in multicenter studies,and the effectiveness of antiviral treatment are controversial.Objective:To investigate the clinical characteristics of Chinese children with IM and current status of antiviral therapy for affected patients.Methods:Hospitalized patients with IM were enrolled between 2018 and 2020 in five children’s hospitals in China.The clinical characteristics were compared among four age groups:<3 years,3-<6 years,6-<10 years,and≥10 years.The clinical characteristics of IM and effectiveness of antiviral therapy were compared among patients receiving acyclovir(ACV),ganciclovir(GCV),and no antiviral therapy(i.e.,non-antiviral group).Results:In total,499 patients were analyzed;most patients were 3-<6 years of age.The most common symptoms and signs included fever(100%),lymphadenopathy(98.6%),pharyngitis(86.4%),eyelid edema(76.8%),and snoring(72.9%).There were significant differences in rash,hepatomegaly,and liver dysfunction among the four age groups.Patients aged<3 years had a lower incidence of liver dysfunction and a higher incidence of rash.Among the 499 patients,50.1%were treated with GCV,26.3%were treated with ACV,and 23.6%received no antiviral therapy.Compared with the non-antiviral group,patients in the ACV and GCV groups had longer durations of fever(P<0.001).There were no significant differences in the incidences of complications among the three treatment groups.Interpretation:The incidence of IM in Chinese children peaked at 3-<6 years of age.Clinical features of IM varied according to age.Patients receiving antiviral therapy exhibited more serious clinical manifestations than did patients without antiviral therapy.The effectiveness of antiviral therapy for IM requires further analysis. | Huili Hu Huiling Deng Jing Bi Yi Xu Shuangjie Li Yue Xie Xinrong Sun Dongmeng Wang Xufang Li Wenxian Ouyang Bing Hu Yufeng Zhang He Tang Chunxiao Fang Hui Zhang Lingyun Guo Chen Wang Tianyi Wang Fengxia Yang Tao Jiang Zhengde Xie Gang Liu | 2021 | Pediatric Investigation2021,5,3: | 12 |

Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. | 13 | Personalized Real-Time Movie Recommendation System:Practical Prototype and Evaluation显示文摘With the eruption of big data,practical recommendation schemes are now very important in various fields,including e-commerce,social networks,and a number of web-based services.Nowadays,there exist many personalized movie recommendation schemes utilizing publicly available movie datasets(e.g.,MovieLens and Netflix),and returning improved performance metrics(e.g.,Root-Mean-Square Error(RMSE)).However,two fundamental issues faced by movie recommendation systems are still neglected:first,scalability,and second,practical usage feedback and verification based on real implementation.In particular,Collaborative Filtering(CF)is one of the major prevailing techniques for implementing recommendation systems.However,traditional CF schemes suffer from a time complexity problem,which makes them bad candidates for real-world recommendation systems.In this paper,we address these two issues.Firstly,a simple but high-efficient recommendation algorithm is proposed,which exploits users1 profile attributes to partition them into several clusters.For each cluster,a virtual opinion leader is conceived to represent the whole cluster,such that the dimension of the original useritem matrix can be significantly reduced,then a Weighted Slope One-VU method is designed and applied to the virtual opinion leader-item matrix to obtain the recommendation results.Compared to traditional clusteringbased CF recommendation schemes,our method can significantly reduce the time complexity,while achieving comparable recommendation performance.Furthermore,we have constructed a real personalized web-based movie recommendation system,MovieWatch,opened it to the public,collected user feedback on recommendations,and evaluated the feasibility and accuracy of our system based on this real-world data. | Jiang Zhang Yufeng Wang Zhiyuan Yuan Qun Jin | 2020 | Tsinghua Science and Technology2020,25,2: | 10 |

| 15 | Regulatory effects of antitumor agent matrine on FOXO and PI3K-AKT pathway in castration-resistant prostate cancer cells显示文摘We previously demonstrated that matrine could inhibit the proliferating, migrating, as well as invading processes of both PC-3 and DU145 cells. However, the underlying molecular mechanisms have not yet been clearly defined. In this study, using various techniques such as high throughput sequencing technology, bioinformatics, quantitative real-time PCR, and immunoblot analysis,we aimed to understand whether matrine serves as a novel regulator of FOXO and PI3K-AKT signaling pathway. DU145 and PC-3 cell lines were cultured for 24 h in vitro. Cells were treated with either matrine or control serum for 48 h, followed by extraction of total RNA. The RNA was sequenced using HiSeq 2500 high-throughput sequencing platform (Illumina). A gene library was established and quality analysis of read data carried out. Integrated database from the website DAVID was used to analyze Gene Ontology (GO), and Kyoto encyclopedia of genes and genomes (KEGG) pathway of differential genes was used for pathway analysis, screening for fold differences of more than two times. The FOXO and PI3K-AKT signaling pathways were screened, and expression levels of mRNA and core protein detected by real-time PCR and immunoblotting, respectively. High throughput sequencing and GO analysis revealed that differentially expressed genes before and after treatment played an important role in cell metabolic process, growth process, anatomical structure formation, cellular component organization, and biological regulation. KEGG signal pathway analysis revealed that FOXO and PI3K-AKT signal pathways had a significant difference between before and after matrine-treated androgen-independent prostate cancer cells PC-3 and DU145. Real-time PCR showed that matrine treatment led to a significant increase in the expression levels of FOXO1A, FOXO3A, FOXO4, and FOXO6 in DU145 and PC-3 cells (P<0.01 or P<0.05), whereas the PI3K expression levels decreased (P<0.01). Similarly, immunoblotting revealed a significant increase (P<0.05) in the expression levels of FOXO1A FOXO3A, FOXO4, and FOXO6 in both PC-3 and DU145 cells, whereas PI3K expression levels decreased (P<0.05). Matrine had a broad regulating effect on the mRNA expression profiles of both PC-3 and DU145 cells. Matrine may inhibit cell proliferation, migration, as well as invasion, and induce apoptosis in both PC-3 and DU145 cells through FOXO and PI3K-AKT signaling pathways. | 18 | Optimizing mechanical property and cytocompatibility of the biodegradable Mg-Zn-Y-Nd alloy by hot extrusion and heat treatment显示文摘The Mg-Zn-Y-Nd alloy is a new type of degradable material for biomedical application. In the present study, Mg-6Zn-1.2Y-0.8Nd alloy was fabricated, and then extrusion and heat treatment were conducted to optimize its mechanical properties and cytocompatibility. The microstructure observation, mechanical property, degradation behavior and cytocompatibility tests were conducted on the Mg-Zn-Y-Nd alloy with three different states: as-cast(alloy C), as-extruded(alloy E) and extruded + heat treated(alloy EH).The results show that alloy C consists of coarse grains and continuous secondary phases. The extrusion process has caused incomplete recrystallization, and results in a mixed grain structure of elongated grains and small equiaxed grains(alloy E). The heat treatment process has promoted the recrystallization and homogenized the grain structure(alloy EH). Both the strength and ductility of the alloy has been improved by extrusion, but the following heat treatment has decreased the strength and increased the ductility.The degradation behavior of the alloy C and E alloys does not show much difference, but improves slightly in alloy EH, because the heat treatment has homogenized the microstructure and released the residual stress in the alloy. The directly and indirectly cell viability tests indicate that alloy EH exhibits the best cytocompatibility, which should be ascribed to its relative uniform degradation and low ion releasing rate.
