| 6 | Effect of blocking Ras signaling pathway with K-Ras siRNA on apoptosis in esophageal squamous carcinoma cells显示文摘OBJECTIVE: To study the effect of RNAi silencing of the K-Ras gene on Ras signal pathway activity in EC9706 esophageal cancer cells. METHODS: EC9706 cells were treated in the following six groups: blank group (no transfection), negative control group (transfection no-carrier), transfection group (transfected with pSilencer-siK-ras), taxol chemotherapy group, taxol chemotherapy plus no-carrier group, taxol chemotherapy plus transfection group. Immunocytochemistry, Reverse transcription-polymerase chain reaction and western blotting were used to analyze the expression of MAPK1 (mitogen-activated protein kinases 1) and cyclin D1 in response to siRNA (small interfering RNA) transfection and taxol treatment. RESULTS: K-Ras (K-Ras gene) siRNA transfection of EC9706 esophageal squamous carcinoma cells decreased the expression of K-Ras, MAPK1 and cyclinD1 at the mRNA and protein level. Reverse transcription-polymerase chain reaction indicated that the expression levels of MAPK1 and cyclin D1 mRNAs were significantly lower in the transfection group than in the blank group (P<0.05). Western blotting showed that 72 h after EC9706 cell transfection, the expression levels of MAPK1 and cyclin D1 proteins had decreased in all groups, and the expression levels in the transfection group were significantly inhibited as compared with the blank group. Apoptosis increased significantly in the transfection group or after addition of taxol as compared with the blank group and the no-carrier group. The degree of apoptosis in the taxol plus transfection group was more severe. CONCLUSION: Apoptosis increased significantly in EC9706 esophageal carcinoma cells after siRNA-mediated inhibition of Ras signaling, with the most obvious increase observed in the transfection plus taxol chemotherapy group. Ras knockdown therefore increased cellular sensitivity to the chemotherapeutic agent, taxol. | 7 | Trajectory Optimization and Power Allocation Scheme Based on DRL in Energy Efficient UAV-Aided Communication Networks显示文摘With flexibility, convenience and mobility, unmanned aerial vehicles(UAVS) can provide wireless communication networks with lower costs, easier deployment, higher network scalability and larger coverage.This paper proposes the deep deterministic policy gradient algorithm to jointly optimize the power allocation and flight trajectory of UAV with constrained effective energy to maximize the downlink throughput to ground users. To validate the proposed algorithm, we compare with the random algorithm, Q-learning algorithm and deep Q network algorithm. The simulation results show that the proposed algorithm can effectively improve the communication quality and significantly extend the service time of UAV. | 11 | Development of a target capture sequencing SNP genotyping platform for genetic analysis and genomic breeding in rapeseed显示文摘Rapeseed(Brassica napus)is an oil crop grown worldwide,making it a key plant species in molecular breeding research.However,the complexity of its polyploid genome increases sequencing costs and reduces sequencing accuracy.Target capture coupled with high-throughput sequencing is an efficient approach for detecting genetic variation at genomic regions or loci of interest.In this study,588 resequenced accessions of rapeseed were used to develop a target capture sequencing SNP genotyping platform named BnaPan50T.The platform comprised 54,765,with 54,058 resequenced markers from the pan-genome,and 855 variant trait-associated markers for 12 agronomic traits.The capture quality of BnaPan50T was demonstrated well in 12 typical accessions.Compared with a conventional genotyping array,BnaPan50T has a high SNP density and a high proportion of SNPs in unique physical positions and in annotated functional genes,promising wide application.Target capture sequencing and wholegenome resequencing in 90 doubled-haploid lines yielded 60%specificity,78%uniformity within tenfold coverage range,and 93%genotyping accuracy for the platform.BnaPan50T was used to construct a genetic map for quantitative trait loci(QTL)mapping,identify 21 unique QTL,and predict several candidate genes for yield-related traits in multiple environments.A set of 132 core SNP loci was selected from BnaPan50T to construct DNA fingerprints and germplasm identification resources.This study provides genomics resources to support target capture sequencing,genetic analysis and genomic breeding of rapeseed. | Xiaodong Li Xumei Liu Yonghai Fan Shengting Li Mengna Yu Mingchao Qian Yuling Chen Hongqiao Chen Xinchun Li Bei Liu Xinfu Xu Cunmin Qu Jiana Li Kun Lu | 2023 | The Crop Journal2023,11,2: | 1 |

| 14 | Changes in the relationship between species richness and belowground biomass among grassland types and along environmental gradients in Xinjiang, Northwest China显示文摘The association between biodiversity and belowground biomass(BGB) remains a central debate in ecology.In this study, we compared the variations in species richness(SR) and BGB as well as their interaction in the top(0–20 cm), middle(20–50 cm) and deep(50–100 cm) soil depths among 8 grassland types(lowland meadow, temperate desert, temperate desert steppe, temperate steppe desert, temperate steppe, temperate meadow steppe, mountain meadow and alpine steppe) and along environmental gradients(elevation, energy condition(annual mean temperature(AMT) and potential evapotranspiration(PET)), and mean annual precipitation(MAP)) based on a 2011–2013 survey of 379 sites in Xinjiang, Northwest China.The SR and BGB varied among the grassland types.The alpine steppe had a medium level of SR but the highest BGB in the top soil depth, whereas the lowland meadow had the lowest SR but the highest BGB in the middle and deep soil depths.The SR and BGB in the different soil depths were tightly associated with elevation, MAP and energy condition;however, the particular forms of trends in SR and BGB depended on environmental factors and soil depths.The relationship between SR and BGB was unimodal in the top soil depth, but SR was positively related with BGB in the middle soil depth.Although elevation, MAP, energy condition and SR had significant effects on BGB, the variations in BGB in the top soil depth were mostly determined by elevation, and those in the middle and deep soil depths were mainly affected by energy condition.These findings highlight the importance of environmental factors in the regulations of SR and BGB as well as their interaction in the grasslands in Xinjiang. | YANG Yuling LI Minfei MA Jingjing CHENG Junhui LIU Yunhua JIA Hongtao LI Ning WU Hongqi SUN Zongjiu FAN Yanmin SHENG Jiandong JIANG Ping'an | 2019 | Journal of Arid Land2019,11,6: | 0 |
| 15 | Energy-Efficient Optimal Guaranteed Cost Intermittent-Switch Control of a Direct Expansion Air Conditioning System显示文摘To improve the energy efficiency of a direct expansion air conditioning(DX A/C) system while guaranteeing occupancy comfort, a hierarchical controller for a DX A/C system with uncertain parameters is proposed. The control strategy consists of an open loop optimization controller and a closed-loop guaranteed cost periodically intermittent-switch controller(GCPISC). The error dynamics system of the closed-loop control is modelled based on the GCPISC principle. The difference,compared to the previous DX A/C system control methods, is that the controller designed in this paper performs control at discrete times. For the ease of designing the controller, a series of matrix inequalities are derived to be the sufficient conditions of the lower-layer closed-loop GCPISC controller. In this way, the DX A/C system output is derived to follow the optimal references obtained through the upper-layer open loop controller in exponential time, and the energy efficiency of the system is improved. Moreover, a static optimization problem is addressed for obtaining an optimal GCPISC law to ensure a minimum upper bound on the DX A/C system performance considering energy efficiency and output tracking error.
