1Emerging biotechnology applications in natural product and synthetic pharmaceutical analyses显示文摘Pharmaceutical analysis is a discipline based on chemical, physical, biological, and information technologies. At present, biotechnological analysis is a short branch in pharmaceutical analysis;however, bioanalysis is the basis and an important part of medicine. Biotechnological approaches can provide information on biological activity and even clinical efficacy and safety, which are important characteristics of drug quality. Because of their advantages in reflecting the overall biological effects or functions of drugs and providing visual and intuitive results, some biotechnological analysis methods have been gradually applied to pharmaceutical analysis from raw material to manufacturing and final product analysis,including DNA super-barcoding, DNA-based rapid detection, multiplex ligation-dependent probe amplification, hyperspectral imaging combined with artificial intelligence, 3D biologically printed organoids,omics-based artificial intelligence, microfluidic chips, organ-on-a-chip, signal transduction pathwayrelated reporter gene assays, and the zebrafish thrombosis model. 2Induction of avirulence by AVR-Pita1 in virulent U.S. field isolates of Magnaporthe oryzae’显示文摘The AVR-Pita1 gene,from the Chinese isolate O-137 of Magnaporthe oryzae,is an effector that determines the efficacy of the Pi-ta rice blast resistance gene.In the present study,the avirulence function of AVR-Pita1 was induced by transformation of field isolates(TM2,ZN19,B2 and B8)that originally were collected from the U.S.and are virulent on Pi-ta-carrying rice cultivars.The presence of AVR-Pita1 from O-137 in independent transformants was detected by PCR using AVR-Pita1 specific primers and verified by DNA sequencing and Southern blot analysis using the AVR-Pita1 coding region as a probe.The results of pathogenicity assays showed that the AVR-Pita1-transformed isolates were not able to infect rice cultivars Katy and Drew carrying Pi-ta.Control isolates that were transformed with inserts lacking the AVR-Pita1gene remained virulent.Our findings demonstrate that AVR-Pita1 can be used to induce novel gene-specific blast resistance in nature.Yuntao Dai Eugenia Winston James C.Correll Yulin Jia 2014The Crop Journal2014,2,1:2

9Stall flutter prediction based on multi-layer GRU neural network显示文摘The modeling of dynamic stall aerodynamics is essential to stall flutter, due to the flow separation in a large-amplitude pitching oscillation process. A newly neural network based Reduced Order Model(ROM) framework for predicting the aerodynamic forces of an airfoil undergoing large-amplitude pitching oscillation at various velocities is presented in this work. First, the dynamic stall aerodynamics is calculated by solving RANS equations and the transitional SST-γ model. Afterwards, the stall flutter bifurcation behavior is calculated by the above CFD solver coupled with structural dynamic equation. The critical flutter speed and limit-cycle oscillation amplitudes are consistent with those obtained by experiments. A newly multi-layer Gated Recurrent Unit(GRU) neural network based ROM is constructed to accelerate the calculation of aerodynamic forces. The training and validation process are carried out upon the unsteady aerodynamic data obtained by the proposed CFD method. The well-trained ROM is then coupled with the structure equation at a specific velocity, the Limit-Cycle Oscillation(LCO) of stall flutter under this flow condition is predicted precisely and more quickly. In order to predict both the critical flutter velocity and LCO amplitudes after bifurcation at different velocities, a new ROM with GRU neural network considering the variation of flow velocities is developed. The stall flutter results predicted by ROM agree well with the CFD ones at different velocities. Finally, a brief sensitivity analysis of two structural parameters of ROM is carried out. 10Progress and prediction of multicomponent quantification in complex systems with practical LC-UV methods显示文摘Complex systems exist widely,including medicines from natural products,functional foods,and biological samples.The biological activity of complex systems is often the result of the synergistic effect of multiple components.In the quality evaluation of complex samples,multicomponent quantitative analysis(MCQA)is usually needed.To overcome the difficulty in obtaining standard products,scholars have proposed achieving MCQA through the“single standard to determine multiple components(SSDMC)”approach.This method has been used in the determination of multicomponent content in natural source drugs and the analysis of impurities in chemical drugs and has been included in the Chinese Pharmacopoeia.Depending on a convenient(ultra)high-performance liquid chromatography method,how can the repeatability and robustness of the MCQA method be improved?How can the chromatography conditions be optimized to improve the number of quantitative components?How can computer software technology be introduced to improve the efficiency of multicomponent analysis(MCA)?These are the key problems that remain to be solved in practical MCQA.First,this review article summarizes the calculation methods of relative correction factors in the SSDMC approach in the past five years,as well as the method robustness and accuracy evaluation.Second,it also summarizes methods to improve peak capacity and quantitative accuracy in MCA,including column selection and twodimensional chromatographic analysis technology.Finally,computer software technologies for predicting chromatographic conditions and analytical parameters are introduced,which provides an idea for intelligent method development in MCA.This paper aims to provide methodological ideas for the improvement of complex system analysis,especially MCQA.Xi Chen Zhao Yang Yang Xu Zhe Liu Yanfang Liu Yuntao Dai Shilin Chen 2023Journal of Pharmaceutical Analysis2023,13,2:0
