1Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. 3A Receiver-Forwarding Decision Scheme Based on Bayesian for NDN-VANET显示文摘Named Data Network(NDN) has caused wide concerns in VANET community because NDN uses a content-centric mechanism that naming content rather than the host. However, integrating NDN into VANET(NDN-VANET) also faces several challenges including consumer/provider mobile, broadcast storm problem and so on. In this paper, we propose a Bayesian-based Receiver Forwarding Decision(BRFD) scheme to mitigate the broadcast storm problem incurred by interest packets in NDN-VANET. In the BRFD, vehicles received an interest packet are required to make forwarding decisions based on Bayesian decision theory according to current network conditions obtained by neighbor interaction. However, the receiver-forwarding decision in BRFD can also cause a conflict issue because multiple vehicles forward copies of the same packet at the same time. So a back-off mechanism is introduced in BRFD. 4Role of the blood-brain barrier in rabies virus infection and protection显示文摘Rabies is an acute,progressive encephalitis caused by infection with rabies virus(RABV).It is one of the most important zoonotic infections and causes more than 70,000 human deaths annually(http://gffzz896b857710fb43b4hxufwvbnp0b6n6pn9.ffgz.tsg.suse.edu.cn).It has long been held that a rabies infection is lethal in humans once the causative RABV reaches the central nervous system(CNS);however,this concept was challenged by the recent recovery of a small number of rabies patients.An analysis of these patients revealed that the blood-brain barrier(BBB)played a major role in protection against the virus.The main reason for the survival of these patients was enhanced BBB permeability after infection with the causative agent(usually bat-originated RABV showing reduced pathogenicity),which allowed immune cells to enter the tissues of the CNS and clear the infection(Willoughby et al.,2005).These fi ndings have been confi rmed in animal infection experiments(Wang et al.,2005;Roy and Hooper,2007,2008;Faber et al.,2009).Thus,the BBB has attracted the attention of scientists interested in the pathogenesis of,and therapeutic approaches,for rabies.This paper introduces the role of the BBB in rabies infections and protection of the CNS and provides insight into future treatments for patients with clinical rabies.Lihua Wang Yuxi Cao Qing Tang Guodong Liang 2013Protein & Cell2013,4,12:2
5Organising a juvenile ratio monitoring programme for 10 key waterbird species in the Yangtze River floodplain:analysis and proposals显示文摘Background:In the face of continued degradation and loss of wetlands in the Yangtze River floodplain(YRF),there is an urgent need to monitor the abundance and distribution of wintering waterbirds.To understand fully observed annual changes,we need to monitor demographic rates to understand factors affecting global population size.Annual reproduction success contributes to dynamic changes in population size and age structure,so an assessment of the juvenile ratio(i.e.first winter birds as a proportion of total number aged)of overwintering waterbirds can be an important indicator of the reproductive success in the preceding breeding season.Methods:During 2016-2019,we sampled juvenile ratios among 10 key waterbird species from the wetlands in the YRF.Based on these data,we here attempt to establish a simple,efficient,focused and reliable juvenile ratio monitoring scheme,to assess consistently and accurately relative annual breeding success and its contribution to the age structure among these waterbird species.Results:We compared juvenile ratio data collected throughout the winter and found that the optimal time for undertaking these samples was in the early stages of arrival for migratory waterbirds reaching their wintering area(early to mid-December).We recommend counting consistently at key points(i.e.those where>1%biogeographical flyway population were counted)at sites of major flyway importance(Poyang Lake,East Dongting Lake,Shengjin Lake,Caizi Lake,Longgan Lake and Chen Lake).Based on this,the error rate of the programme(155 planned points,the count of 10 waterbird species is 826-8955)is less than 5%.Conclusions:We established a juvenile ratio monitoring programme for 10 key waterbird species in the wetlands of the YRF,and discuss the feasibility and necessity of implementing such a future programme,and how to use these data in our monitoring and understanding of the population dynamics of these waterbird populations.Yuxi Wang Iderbat Damba Qingshan Zhao Yanbo Xie Xueqing Deng Rdi Ga Guanhua Liu Zhiwen Xu Yue Li Dali Gao Wenbin Xu Guoxun Chen Lei Cao 2021Avian Research2021,12,4:1

8Soxllb regulates the migration and fate determination of Müller glia-derived progenitors during retina regeneration in zebrafish显示文摘The transcription factor Sox11 plays important roles in retinal neurogenesis during vertebrate eye development.However,its function in retina regeneration remains elusive.Here we report that Sox11 b,a zebrafish Sox11 homolog,regulates the migration and fate determination of Müller glia-derived progenitors(MGPCs)in an adult zebrafish model of mechanical retinal injury.Following a stab injury,the expression of Sox11 b was induced in proliferating MGPCs in the retina.Sox11 b knockdown did not affect MGPC formation at 4 days post-injury,although the nuclear morphology and subsequent radial migration of MGPCs were alte red.At 7 days post-injury,Sox11 b knockdown res ulted in an increased proportion of MGPCs in the inner retina and a decreased propo rtion of MGPCs in the outer nuclear layer,compared with controls.Furthermore,Sox11 b knockdown led to reduced photoreceptor regeneration,while it increased the numbe rs of newborn amacrines and retinal ganglion cells.Finally,quantitative polymerase chain reaction analysis revealed that Sox11 b regulated the expression of Notch signaling components in the retina,and Notch inhibition partially recapitulated the Sox11 b knockdown phenotype,indicating that Notch signaling functions downstream of Sox11 b.Our findings imply that Sox11 b plays key roles in MGPC migration and fate determination during retina regeneration in zebrafish,which may have critical im plications for future explorations of retinal repair in mammals.Kaida Song Zihao Lin Lining Cao Bowen Lu Yuxi Chen Shuqiang Zhang Jianfeng Lu Hui Xu 2023Neural Regeneration Research2023,18,2:0
9Inhibition of fibroblast activation protein ameliorates cartilage matrix degradation and osteoarthritis progression显示文摘Fibroblast activation protein(Fap)is a serine protease that degrades denatured type I collagen,α2-antiplasmin and FGF21.Fap is highly expressed in bone marrow stromal cells and functions as an osteogenic suppressor and can be inhibited by the bone growth factor Osteolectin(Oln).Fap is also expressed in synovial fibroblasts and positively correlated with the severity of rheumatoid arthritis(RA).However,whether Fap plays a critical role in osteoarthritis(OA)remains poorly understood.Here,we found that Fap is significantly elevated in osteoarthritic synovium,while the genetic deletion or pharmacological inhibition of Fap significantly ameliorated posttraumatic OA in mice.Mechanistically,we found that Fap degrades denatured type II collagen(Col II)and Mmp13-cleaved native Col II.Intra-articular injection of r Fap significantly accelerated Col II degradation and OA progression.In contrast,Oln is expressed in the superficial layer of articular cartilage and is significantly downregulated in OA.Genetic deletion of Oln significantly exacerbated OA progression,which was partially rescued by Fap deletion or inhibition.Intra-articular injection of r Oln significantly ameliorated OA progression.Taken together,these findings identify Fap as a critical pathogenic factor in OA that could be targeted by both synthetic and endogenous inhibitors to ameliorate articular cartilage degradation.Aoyuan Fan Genbin Wu Jianfang Wang Laiya Lu Jingyi Wang Hanjing Wei Yuxi Sun Yanhua Xu Chunyang Mo Xiaoying Zhang Zhiying Pang Zhangyi Pan Yiming Wang Liangyu Lu Guojian Fu Mengqiu Ma Qiaoling Zhu Dandan Cao Jiachen Qin Feng Yin Rui Yue 2023Bone Research2023,11,1:0

11Synthesis of atomically dispersed cationic nickel-confined mesoporous ZSM-48(ANMZ-48)directed by metal complexes in amphiphilic molecules显示文摘The synthesis of mesoporous zeolite-anchored atomically dispersed metal catalysts(ADCs)is a considerable challenge in chemistry and materials science.Here we report the synthesis of atomically dispersed cationic nickel-confined mesoporous ZSM-48(ANMZ-48)by in situ hydrothermal reaction employing a designed tri-functional metal complex template,by which the triquaternary ammonium groups in the hydrophilic region direct the formation of ZSM-48 zeolite;the aromatic groups in the hydrophobic tail generate the mesopores through π-π stacking;and the complexes formed by nickel ions coordinated with terpyridyl groups generate atomically dispersed Ni2+confined in zeolite frameworks due to the strong sintering resistance generated by the strong coordination interaction.The ANMZ-48 is consisting of stacking of sheet-like ZSM-48 domains connected by multiply crystal twinning sharing the common(011)plane,generating abundant of imbedded mesopores with the uniform thickness of~2.4 nm and with the width of 10-50 nm.The excellent catalytic activity and stability of ANMZ-48 were also reflected in the dry reforming of methane(DRM)reaction.Yu Chen Quanzheng Deng Yiru Mao Yuxi Fang Zhihua Cao Honggen Peng Shunai Che Lu Han 2024Science China Chemistry2024,67,1:0
12A novel undifferentiated spermatogonia-speci?c surface protein 1(USSP1) in neonatal mice显示文摘Mammalian spermatogenesis is maintained by a rare population of spermatogonial stem cells(SSCs),which are important for male fertility. SSCs remain a subset of undifferentiated spermatogonia, which can be isolated by a combination of surface markers. Specific markers to identify and isolate undifferentiated spermatogonia are lacking. Ussp1, a transcript previously annotated as long noncoding RNA(RIKEN cDNA 4933427D06, Gene ID: 232217), virtually encodes a membrane protein, USSP1, in a highly testisspecific manner in mouse. We demonstrate its expression on the membrane of undifferentiated spermatogonia by a homemade polyclonal rabbit antibody against the protein. In vivo, USSP1^+ clusters consist mainly of As, Apr(GFRa1^+) and Aal(PLZF^+) cells. USSP1^+ cells exhibit enrichment of undifferentiated spermatogonia, as shown by increased expression of SSC self-renewal molecular markers and the potential to form SSC clones in vitro and in vivo. However, Ussp1 knockout did not affect the number of SSCs or spermatogenesis in mice. Thy1^+ cells from Ussp1 null mice did not show any defect in the SSC colony formation capacity, indicating that USSP1 is not essential for SSC self-renewal. Our data demonstrate that Ussp1 is specifically expressed in undifferentiated murine spermatogonia, indicating the potential to sort undifferentiated spermatogonia with USSP1 antibodies. 13Action Mechanism of Antibacterial Hydrolysate from Ruditapes philippinarum显示文摘[Objectives] The antibacterial mechanism of protein hydrolysate from Ruditapes philippinarum( named RPPH) was studied in this article. [Methods]The integrity of bacteria's wall and membrane was determined by some traditional ways. [Results]The growth of Staphylococcus aureus and Bacillus subtilis were inhibited by RPPH in the logarithmic phase. The activity of alkaline phosphatase could be detected in the culture solution. The results showed that the protein content and the conductivity of two kinds of bacteria increased with the extension of incubation time. The results of scanning electron microscope revealed that it emerged the phenomenon of agglomeration with the extension of response time in the culture solution of S. aureus,and the cell shape became irregular,a large number of cells stuck together,afterwards intracellular material was released from bacteria,and the boundaries among cells completely disappeared. The cell surface of B. subtilis became rough,the cells began to adhere,intercellular boundaries became blurred,subsequently cells broke,and then intracellular material leaked out after treating for 9 h. The form of B. subtilis could be roughly identified from the remnants of the bacterial debris,and most of the bacteria had been completely cracked and died. Transmission electron microscope results showed that the surface of S. aureus became rough,with uneven distribution of cytoplasm,and darker substances appeared in the middle,followed by leakage of large numbers of intracellular material. Massive cells became dead when treating for 9 h. B. subtilis cells began to shrink,and cytoplasm was distributed unevenly. The significant phenomenon of plasmolysis and rupture of cell wall could be observed. Afterwards,intracellular material spilled out,only the residual and ambiguity nucleoplasm area could be seen,and cells were killed. 14Brazilein intervention for sciatic nerve injury in BALB/c mice显示文摘Brazilein has been shown to exert an immune regulation effect on regeneration in the central nervous system,but there is currently no consensus regarding its effect on sciatic nerve injury.In the current study,western blot and real-time PCR detection revealed that,after 16 and 8 g/kg of brazilein intervention,the amount of S100 protein and mRNA at L4-6 spinal segments of sciatic nerve injured mice was significantly greater than after 4 g/kg brazilein administration,and in an untreated model group.Luxol Fast Blue myelin staining revealed that neural regeneration after 16 and 8 g/kg of brazilein intervention was significantly better than after 4 g/kg brazilein administration,and in the model group.In addition,electrophysiological and immunohistochemical examinations further confirmed the effect of brazilein in repairing sciatic nerve injury in BALB/c mice.These results indicated that brazilein was able to activate the S100 in anterior horn cells of the spinal cord,promoting sciatic nerve regeneration and repair.Both moderate and high doses were found to exert significant effects.Zhan Zhang Jian Cao Weitian Yin Lisen Li Yuxi Jia Haoyu Liu Mingming Zhao Rihua Jiang 2011Neural Regeneration Research2011,6,12:0
