| 1 | Akt Inhibitor Perifosine Prevents Epileptogenesis in a Rat Model of Temporal Lobe Epilepsy显示文摘Accumulating data have revealed that abnormal activity of the mTOR(mammalian target of rapamycin)pathway plays an important role in epileptogenesis triggered by various factors. We previously reported that pretreatment with perifosine, an inhibitor of Akt(also called protein kinase B), abolishes the rapamycin-induced paradoxical increase of S6 phosphorylation in a rat model induced by kainic acid(KA). Since Akt is an upstream target in the mTOR signaling pathway, we set out to determine whether perifosine has a preventive effect on epileptogenesis. Here, we explored the effect of perifosine on the model of temporal epilepsy induced by KA in rats and found that pretreatment with perifosine had no effect on the severity or duration of the KA-induced status epilepticus. However, perifosine almost completely inhibited the activation of p-Akt and p-S6 both acutely and chronically following the KA-induced status epilepticus.Perifosine pretreatment suppressed the KA-induced neuronal death and mossy fiber sprouting. The frequency of spontaneous seizures was markedly decreased in rats pretreated with perifosine. Accordingly, rats pretreated with perifosine showed mild impairment in cognitive functions. | 3 | 紫杉醇脂质体与紫杉醇联合铂类同步放化疗治疗宫颈癌的随机对照研究(英文)显示文摘Objective:The aim of the study was to compare the efficacy,side effect and influence on the survival rate of two chemotherapy regimens,paclitaxel liposome combined with platinum and paclitaxel combined with platinum,in concurrent chemoradiotherapy for cervical carcinoma.Methods:The 162 cases with primary cervical carcinoma diagnosed between January 2008 and November 2009 in Jiangxi Maternal and Child Health Hospital(China) were enrolled in this randomized controlled trial.Seventy-one cases were included in paclitaxel group and 91 in paclitaxel liposome group.And the chemotherapy doses were as follows:paclitaxel liposome and paclitaxel 135 mg/m2;cisplatin 80 mg/m2 or carboplatin AUC 4-6;then repeated every 21 days for two or three times.Radical radiotherapy was given to both groups at the same time.Efficacy was evaluated according to the tumor regression six months later and follow-up was done consistently.Results:The overall response rates of paclitaxel group and paclitaxel liposome group were 90.1% and 89 % respectively(P > 0.05).The one year cumulative survival was 91.4% for paclitaxel group and 89.2% for paclitaxel liposome group(P > 0.05).The incidence rates of adverse effects such as rash,gastrointestinal toxicity,bone marrow suppression and muscle/joint pain in paclitaxel liposome group were much lower than those in paclitaxel group(P < 0.05),while there was no difference regarding hair loss,hepatic function damage,peripheral neuritis and other aspects(P > 0.05).Conclusion:Paclitaxel liposome plus platinum is a safe and effective method for staging IIa-IV cervical carcinomas.While the long-term efficacy should be further observed. | Siyuan Zeng Ling Li Meiling Zhong Wei Jiang Yun Xiao | 2012 | The Chinese-German Journal of Clinical Oncology2012,11,2: | 3 |
| 4 | Suppression of PFKFB3-driven glycolysis restrains endothelial-to-mesenchymal transition and fibrotic response显示文摘Endothelial-to-mesenchymal transition(EndoMT),the process wherein endothelial cells lose endothelial identity and adopt mesenchymal-like phenotypes,constitutes a critical contributor to cardiac fibrosis.The phenotypic plasticity of endothelial cells can be intricately shaped by alteration of metabolic pathways,but how endothelial cells adjust cellular metabolism to drive EndoMT is incompletely understood.Here,we identified 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(PFKFB3)as a critical driver of EndoMT via triggering abnormal glycolysis and compromising mitochondrial respiration.Pharmacological suppression of PFKFB3 with salvianolic acid C(SAC),a phenolic compound derived from Salvia miltiorrhiza,attenuates EndoMT and fibrotic response.PFKFB3-haplodeficiency recapitulates the anti-EndoMT effect of SAC while PFKFB3-overexpression augments the magnitude of EndoMT and exacerbates cardiac fibrosis.Mechanistically,PFKFB3-driven glycolysis compromises cytoplasmic nicotinamide adenine dinucleotide phosphate(reduced form,NADPH)production via hijacking glucose flux from pentose phosphate pathway.Efflux of mitochondrial NADPH through isocitrate/α-ketoglutarate shuttle replenishes cytoplasmic NADPH pool but meanwhile impairs mitochondrial respiration by hampering mitochondrial iron-sulfur cluster biosynthesis.SAC disrupts PFKFB3 stability by accelerating its degradation and thus maintains metabolic homeostasis in endothelial cells,underlying its anti-EndoMT effects.These findings for the first time identify the critical role of PFKFB3 in triggering EndoMT by driving abnormal glycolysis in endothelial cells,and also highlight the therapeutic potential for pharmacological intervention of PFKFB3(with SAC or other PFKFB3 inhibitors)to combat EndoMT-associated fibrotic responses via metabolic regulation. | Hao Zeng Ting Pan Meiling Zhan Renaguli Hailiwu Baolin Liu Hua Yang Ping Li | 2022 | Signal Transduction and Targeted Therapy2022,7,10: | 1 |

| 7 | Decision Difficulty, Work-Family Conflict, and Social Support for Chinese Nurses in Guangzhou after the Two-Child Policy显示文摘Background:AlthoughChina's Mainland implemented a two-child policy (every legally married may have two children) on January 1, 2016, women in China are becoming increasingly cautious in their decision-making about family planning, due to work-family conflicts and a lack of social support. Objective:The aim of this study was to investigate Chinese nurses' intents to have a second child, and to explore the relationship between decision making difficulties in terms of childbearing intents, work-family conflict, and social support for Chinese nurses. Methods:Data were collected at three different hospital levels in four districts of Guangzhou, from 711 nurses of childbearing age (18-50 years) who had had at least one child. A structured questionnaire was used, composed of the Decisional Conflict Scale (DCS), Work-Family Conflict Scale (WFCS), and Scale of Perceived Social Support (SPSS). Results:Participants’ mean age was 33.6 years (18-50). The mean scores of decisional conflict by DCS, work-family conflict by WFCS and perceived social support by SPSS were 40.9 (SD = 14.8), 55.83 (SD = 15.18) and 57.85 (SD = 15.77), respectively. Work-family conflict was positively correlated with decisional conflict (P < 0.01), and social support was negatively related to decisional conflict (P < 0.01). By logistic regression analysis, family interference with work (FIW), especially time-based FIW, was a significant predictor of nurses having decisional conflicts in their childbearing intents (OR=1.083, 95% CI: 1.045 to 1.123). Perceived social support was another significant predictor of decisional difficulty (OR=0.981, 95% CI: 0.970 to 0.992). Discussion:This study found that female nurses, working night shifts and with temporary job contract had higher decisional conflicts in intents of having a second child. The results showed that for Chinese nurses, work-family conflict influences decisional conflicts about having a second child.
