2Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 5A new remaining useful life estimation method for equipment subjected to intervention of imperfect maintenance activities显示文摘As the key part of Prognostics and Health Management(PHM), Remaining Useful Life(RUL) estimation has been extensively investigated in recent years. Current RUL estimation studies considering the intervention of imperfect maintenance activities usually assumed that maintenance activities have a single influence on the degradation level or degradation rate, but not on both.Aimed at this problem, this paper proposes a new degradation modeling and RUL estimation method taking the influence of imperfect maintenance activities on both the degradation level and the degradation rate into account. Toward this end, a stochastic degradation model considering imperfect maintenance activities is firstly constructed based on the diffusion process. Then, the Probability Density Function(PDF) of the RUL is derived by the convolution operator under the concept of First Hitting Time(FHT). To implement the proposed RUL estimation method,the Maximum Likelihood Estimation(MLE) is utilized to estimate the degradation related parameters based on the Condition Monitoring(CM) data, while the Bayesian method is utilized to estimate the maintenance related parameters based on the maintenance data. Finally, a numerical example and a practical case study are provided to demonstrate the superiority of the proposed method. 6In vitro effect of iASPP on cell growth of oral tongue squamous cell carcinoma显示文摘iASPP is an inhibitory member of the apoptosis-stimulating proteins of P53(ASPP) family. iASPP is over expressed in several malignant tumors and potentially affects cancer progression. However, the expression and potential role of iASPP in oral tongue squamous cell carcinoma(OTSCC) have not been addressed. In our study, we detected iASPP expression in OTSCC by immunohistochemistry. iASPP expression is up-regulated in OTSCC tissues. Moreover, in clinical pathology specimens, we found that increased iASPP expression correlates with poor differentiation and lymph node metastasis. Using multicellular tumor spheroids(MTS) and flow cytometry, we demonstrated that iASPP down-regulation arrests OTSCC cells at the G0/G1 phase, induces OTSCC cell apoptosis and inhibits OTSCC cell proliferation. 7Alpha-2-macroglobulin as a radioprotective agent: a review显示文摘Radiation is an important modality in cancer treatment, and eighty percent of cancer patients need radiotherapy at some point during their clinical management. However, radiation-induced damage to normal tissues restricts the therapeutic doses of radiation that can be delivered to tumours and thereby limits the effectiveness of the treatment. The use of radioprotectors represents an obvious strategy to obtain better tumour control using a higher dose in radiotherapy. However, most of the synthetic radioprotective compounds studied have shown inadequate clinical efficacy owing to their inherent toxicity and high cost. Hence, the development of radioprotective agents with lower toxicity and an extended window of protection has attracted a great deal of attention, and the identification of alternative agents that are less toxic and highly effective is an absolute necessity. Recent studies have shown that alpha-2-macroglobulin(α2M) possesses radioprotective effects. α2M is a tetrameric, disulfide-rich plasma glycoprotein that functions as a nonselective inhibitor of different types of non-specific proteases and as a carrier of cytokines, growth factors, and hormones. α2M induces protein factors whose interplay underlies radioprotection, which supports the idea that α2M is the central effector of natural radioprotection in the rat. Pretreatment with α2M has also induced a significant reduction of irradiation-induced DNA damage and the complete restoration of liver and body weight. Mihailovi? et al. concluded that the radioprotection provided by α2M was in part mediated through cytoprotection of new blood cells produced in the bone marrow; these authors also indicated that an important aspect of the radioprotective effect of amifostine was the result of the induction of the endogenous cytoprotective capability of α2M. The radioprotective effects of α2M are possibly due to antioxidant, antifibrosis, and anti-inflammatory functions, as well as the maintenance of homeostasis, and enhancement of the DNA repair and cell recovery processes. This review is the first to summarise the observations and elucidate the possible mechanisms responsible for the beneficial effects of α2M. 83D-printed dermis-specific extracellular matrix mitigates scar contraction via inducing early angiogenesis and macrophage M2 polarization显示文摘Scar contraction frequently happens in patients with deep burn injuries.Hitherto,porcine dermal extracellular matrix(dECM)has supplied microenvironments that assist in wound healing but fail to inhibit scar contraction.To overcome this drawback,we integrate dECM into three-dimensional(3D)-printed dermal analogues(PDA)to prevent scar contraction.We have developed thermally gelled,non-rheologically modified dECM powder(dECMp)inks and successfully transformed them into PDA that was endowed with a micron-scale spatial structure.The optimal crosslinked PDA exhibited desired structure,good mechanical properties as well as excellent biocompatibility.Moreover,in vivo experiments demonstrated that PDA could significantly reduced scar contraction and improved cosmetic upshots of split thickness skin grafts(STSG)than the commercially available dermal templates and STSG along.The PDA has also induced an early,intense neovascularization,and evoked a type-2-like immune response.PDA’s superior beneficial effects may attribute to their desired porous structure,the well-balanced physicochemical properties,and the preserved dermis-specific ECM cues,which collectively modulated the expression of genes such as Wnt11,ATF3,and IL1β,and influenced the crucial endogenous signalling pathways.The findings of this study suggest that PDA is a clinical translatable material that possess high potential in reducing scar contraction.Lei Chen Zhiyong Li Yongtai Zheng Fei Zhou Jingling Zhao Qiyi Zhai Zhaoqiang Zhang Tianrun Liu Yongming Chen Shaohai Qi 2022Bioactive Materials2022,7,4:3

11Spectroscopic investigation of the interactions between gold nanoparticles and bovine serum albumin显示文摘The interactions between bovine serum albumin(BSA) and gold nanoparticles(AuNPs) ,and the conformational changes of BSA induced by this interaction,were investigated by UV-visible absorption spectroscopy,fluorescence spectroscopy,and Fourier transform infrared in combination with attenuated total reflection spectroscopy(ATR-FTIR) .The critical adsorption density for preventing AuNP aggregation in 0.1 mol/L phosphate buffered saline(pH 7.2) was 23 BSA molecules per gold particle or 3.8×1012 BSA molecules/cm2.BSA bound to the AuNPs with high affinity(binding constant Ks=7.59×108 L/mol) ,and the intrin-sic fluorescence of BSA was quenched by the AuNPs in accordance with the static quenching mechanism.Both fluorescence spectroscopy and ATR-FTIR showed that AuNPs induced conformational changes in BSA,which resulted in it becoming less compact and increased the polarity of the microenvironment around the tryptophan residue Trp-212.SHI XiuJuan LI Dan XIE Jing WANG Shawn WU ZhaoQiang CHEN Hong 2012Chinese Science Bulletin2012,57,10:2
12One-step preparation of vinyl-functionalized material surfaces:a versatile platform for surface modification显示文摘A simple approach has been developed to functionalize various substrates, such as gold and polyvinylchloride, with dopamine methacrylamide—a molecule with adhesive properties that mimic those of mussels—to produce a versatile and general platform for subsequent surface modification. With active double bonds on the surface, various polymers, such as poly([2-(methacryloyloxy)ethyl]dimethyl-(3-sulfopropyl) ammonium hydroxide(PMEDSAH) and poly(N-vinylpyrrolidone)(PVP), can be grafted by conventional radical polymerization. Double bond surface functionalization and subsequent polymer grafting have been verified by static water contact angle, Fourier transform infrared–attenuated total reflectance(FTIR-ATR) spectroscopy and X-ray photoelectron spectroscopy(XPS) measurements. Protein adsorption assays showed that the polymermodified substrates have good protein-resistant properties.
