| 1 | A Simple Approach to Bioconjugation at Diverse Levels: Metal-Free Click Reactions of Activated Alkynes with Native Groups of Biotargets without Prefunctionalization显示文摘Te efcient bioconjugation of functional groups/molecules to targeted matrix and bio-related species drives the great development of material science and biomedicine,while the dilemma of metal catalysis,uneasy premodifcation,and limited reaction efciency in traditional bioconjugation has restricted the booming development to some extent.Here,we provide a strategy for metal-free click bioconjugation at diverse levels based on activated alkynes.As a proof-of-concept,the abundant native groups including amine,thiol,and hydroxyl groups can directly react with activated alkynes without any modifcation in the absence of metal catalysis.Trough this strategy,high-efcient modifcation and potential functionalization can be achieved for natural polysaccharide,biocompatible polyethylene glycol(PEG),synthetic polymers,cell penetrating peptide,protein,fast whole-cell mapping,and even quick diferentiation and staining of Gram-positive bacteria,etc.Terefore,current metal-free click bioconjugation strategy based on activated alkynes is promising for the development of quick fuorescence labeling and functional modifcation of many targets and can be widely applied towards the fabrication of complex biomaterials and future in vivo labeling and detection. | Xianglong Hu Xueqian Zhao Benzhao He Zheng Zhao Zheng Zheng Pengfei Zhang Xiujuan Shi Ryan T.K.Kwok Jacky W.Y.Lam Anjun Qin Ben Zhong Tang | 2018 | Research2018,,1: | 7 |
| 2 | The chimeric genes in the hybrid lineage of Carassius auratus cuvieri(♀)×Carassius auratus red var.(♂)显示文摘Hybridization can combine the genomes of different strains or species, which leads to changes of genotype and phenotype in the hybrids. In this study, we aimed to investigate the genetic variations of hybrids(WR-F1 and WR-F2) derived from the intraspecific hybridization of white crucian carp(Carassius auratus cuvieri, WCC, ♀) and red crucian carp(Carassius auratus red var., RCC, ♂). Here, we compared the orthologous genes in the liver transcriptomes of hybrids with those of WCC and RCC, and classified the orthologous genes into eight gene patterns within three categories(chimera, mutant, and biparental origin genes).The results revealed 19.04%, 4.17% chimeric genes and 6.90%, 5.05% mutations of orthologous genes in WR-F1 and WR-F2 respectively. Seventeen of twenty-three characterized genes(77%) were confirmed to be the chimeras at the genomic DNA level.The GO classification discovered that some chimeric and mutant genes were related to metabolic process, immune system and developmental process in WR-F1. Our results provide the new evidence that hybridization can combine the parental genomes,leading to changes in the genotype of the resultant hybrids. | 17 | Evaporation and drying characteristics of the sessile ferrofluid droplet under a horizontal magnetic field显示文摘In this study,the evaporation characteristics and drying patterns of various sessile ferrofluid droplets on certain substrate under horizontal magnetic fields of controlled intensities are reported.The effects of droplet concentration and magnetic field intensity on the duration of each evaporation stage and drying patterns of droplets have been systematically investigated.It turned out that a plateau appears at the initial stage of evaporation in the absence of magnetic field and it was found that the plateau value is positively correlated with the concentration of ferrofluid droplets.Under the external magnetic field,the evaporation time of droplets decreases,the stage of contact line retreat extends,the stage of late pinning mode shortens,and the deposition area of ferrofluid droplet decreases compared to that of without magnetics field.The deposition area increases gradually and becomes more uniform with the increase of magnetic field.The decrease of friction force which is due to the decrease of the number of nanoparticles at the contact line under external magnetic field is the main reason for the observed phenomena.We found that the coffee ring and the uniform deposition inside the droplet will be destroyed when the magnetic field intensity is higher than a critical value.Our work has a significant reference value for the evaporation of sessile magnetic fluid droplets under the applied magnetic field,especially when the drying pattern needs to be precisely controlled,such as in spray or biomedicine. | Zhaonan Liu Jiandong Zhou Yang Li Xin Zhuo Xiujuan Shi Dengwei Jing | 2022 | Fundamental Research2022,2,2: | 0 |
| 18 | HAND2-AS1,PRKAA2 and VLDLR predict the risk of peritoneal metastasis in gastric cancer of different Lauren types based on STEPP analysis显示文摘The peritoneum is the most common site of recurrence of gastric cancer(GC).Early occult peritoneal metastasis is difficult to detect by imaging examination.Stratifying the risk of peritoneal metastasis in patients with different Lauren subtypes is of great clinical value.We performed a univariate Cox regression to identify those genes with prognostic value of overall survival(OS)and peritoneal-specified disease-free survival(psDFS)from the Gene Expression Omnibus database.The candidate genes were screened by the Subpopulation Treatment Effect Pattern Plot(STEPP)method.Propensity score matching(PSM)analysis was used to reduce the interference of confounders on the results.Based on the optimal cut-off values determined by the STEPP method,we found overexpression of three genes(HAND2-AS1,PRKAA2,and VLDLR)was correlated with shorter 1-year psDFS among patients with diffuse-type than that of patients with intestinal-type GC,and it is highly significant.Gene Set Enrichment Analysis(GSEA)potentially suggested that the three genes promote the early occurrence of peritoneal metastasis in patients with diffuse-type GC through glucose metabolism-related pathways.These three genes may be potential biomarkers.They can be used to assess the risk of peritoneal metastases to guide treatment decisions and follow-up strategies. | HUI QU RUICHUAN SHI BOWEN YANG XI CHEN YUJIA SONG LINGZI HE YITING SUN YUJING YANG ZHI LI XIUJUAN QU | 2022 | BIOCELL2022,46,3: | 0 |
