| 1 | Xa7, a new executor R gene that confers durable and broad-spectrum resistance to bacterial blight disease in rice显示文摘Bacterial blight(BB)is a globally devastating rice disease caused by Xanthomonas oryzae pv.oryzae(Xoo).The use of disease resistance(R)genes in rice breeding is an effective and economical strategy for the control of this disease.Nevertheless,a majority of R genes lack durable resistance for long-term use under global warming conditions.Here,we report the isolation of a novel executor R gene,Xa7,that confers extremely durable,broad-spectrum,and heat-tolerant resistance to Xoo.The expression of Xa7 was induced by incompatible Xoo strains that secreted the transcription activator-like effector(TALE)AvrXa7 or PthXo3,which recognized effector binding elements(EBEs)in the Xa7 promoter.Furthermore,Xa7 induction was faster and stronger under high temperatures.Overexpression of Xa7 or co-transformation of Xa7 with avrXa7 triggered a hypersensitive response in plants.Constitutive expression of Xa7 activated a defense response in the absence of Xoo but inhibited the growth of transgenic rice plants.In addition,analysis of over 3000 rice varieties showed that the Xa7 locuswas found primarily in the indica and aus subgroups.A variation consisting of an 11-bp insertion and a base substitution(G to T)was found in EBEAvrXa7 in the tested varieties,resulting in a loss of Xa7 BB resistance.Through a decade of effort,we have identified an important BB resistance gene and characterized its distinctive interaction with Xoo strains;these findings will greatly facilitate research on the molecular mechanism of Xa7-mediated resistance and promote the use of this valuable gene in breeding. | Xifeng Chen Pengcheng Liu Le Mei Xiaoling He Long Chen Hui Liu Shurong Shen Zhandong Ji Xixi Zheng Yuchen Zhang Zhenyu Gao Dali Zeng Qian Qian Bojun Ma | 2021 | Plant Communications2021,2,3: | 9 |
| 2 | Hydrothermal growth of hydroxyapatite and ZnO bilayered nanoarrays on magnesium alloy surface with antibacterial activities显示文摘Magnesium alloy(MgA)has been extensively used as orthopedic and cardiovascular scaffolds in virtue of its good biocompatibility,unique biodegradability and excellent mechanical properties.However,poor corrosion resistance and easy infection after implantation seriously limit the potential applications of MgA in the biomedical field.Herein,we fabricated bilayered nanoarrays of hydroxyapatite nanorods(HANRs)and ZnO nanorods(ZnONRs)onto the surface of MgA(MgA-MgO-HANRs-ZnONRs)via micro-arc oxidation(MAO)treatment,microwave-assisted hydrothermal and hydrothermal methods.The morphology and chemical composition of MgA-MgO-HANRs-ZnONRs was characterized by FE-SEM,XRD and EDS,indicating that HANRs-ZnONRs bilayered nanoarrays were fabricated on the surface of MgA-MgO.The surface of MgA-MgO-HANRs-ZnONRs exhibited excellent hydrophilicity as evidenced by the low water contact angle of 3°.Compared with the original MgA,the corrosion resistance of MgA-MgO-HANRs-ZnONRs was obviously improved with decreasing the corrosive current density(icorr)of 2 orders of magnitude.The MgA-MgO-HANRs-ZnONRs performed excellent antibacterial properties with the bactericidal rate of 96.5%against S.aureus and 94.3%against E.coli. | Mengke Peng Fenyan Hu Minting Du Bingjie Mai Shurong Zheng Peng Liu Changhao Wang Yashao Chen | 2020 | Frontiers of Materials Science2020,14,1: | 3 |

| 10 | The functional analysis of transiently upregulated miR-101 suggests a “braking” regulatory mechanism during myogenesis显示文摘Skeletal muscle differentiation is a highly coordinated process that involves many cellular signaling pathways and microRNAs(miRNAs).A group of muscle-specific miRNAs has been reported to promote myogenesis by suppressing key signaling pathways for cell growth.However,the functional role and regulatory mechanism of most non-muscle-specific miRNAs with stage-specific changes during differentiation are largely unclear.Here,we describe the functional characterization of miR-101a/b,a pair of non-muscle-specific miRNAs that show the largest change among a group of transiently upregulated miRNAs during myogenesis in C2C12 cells.The overexpression of miR-101a/b inhibits myoblast differentiation by suppressing the p38/MAPK,Interferon Gamma,and Wnt pathways and enhancing the C/EBP pathway.Mef2a,a key protein in the p38/MAPK pathway,was identified as a direct target of miR-101a/b.Interestingly,we found that the long non-coding RNA(lncRNA)Malat1,which promotes muscle differentiation,interacts with miR-101a/b,and this interaction competes with Mef2a mRNA to relieve the inhibition of the p38/MAPK pathway during myogenesis.These results uncovered a“braking”role in differentiation of transiently upregulated miRNAs and provided new insights into the competing endogenous RNA(ceRNA)regulatory mechanism in myoblast differentiation and myogenesis. | Shurong Liu Shujuan Xie Huafeng Chen Bin Li Zhirong Chen Yeya Tan Jianhua Yang Lingling Zheng Zhendong Xiao Qi Zhang Lianghu Qu | 2021 | Science China(Life Sciences)2021,64,10: | 1 |

| 14 | Overexpression of BnKCS1-1,BnKCS1-2,and BnCER1-2 promotes cuticular wax production and increases drought tolerance in Brassica napus显示文摘Higher amounts of cuticular wax in plants have been associated with improved plant stress tolerance and increased potential for industrial use.In this study,orthologs of KCS1 and CER1 in Arabidopsis,designated BnKCS1-1,BnKCS1-2,and BnCER1-2,were isolated from Brassica napus.Transcription of BnKCS1-1 and BnKCS1-2 in B.napus were induced by abscisic acid(ABA)and drought treatment,while transcription of BnCER1-2 was induced only by drought treatment.All three gene transcripts decreased significantly when plants were treated with methyl jasmonate(MeJA)or subjected to cold stress.Overexpression of BnKCS1-1,BnKCS1-2,and BnCER1-2 under the control of the CaMV35S promoter led to a significant increase in cuticular wax on transgenic B.napus leaves.BnKCS1-1 and BnKCS1-2 overexpression led to similar differences from non-transformed plants,with significantly higher levels of aldehydes(C29 and C30),alkanes(C28,C29,and C31)and secondary alcohols(C28 and C29),and a significantly lower level of C29 ketone.Overexpression of BnCER1-2 led to an increase in alkanes(C27,C28,C29,and C31),a decrease in secondary alcohols(C28 and C29),and insignificant changes in other wax components.Scanning electron microscopy revealed that overexpression of BnKCS1-1,BnKCS1-2,and BnCER1-2 in B.napus resulted in a higher density of wax crystals on the leaf surface than observed in non-transformed plants.Transgenic plants showed a reduced rate of water loss and increased drought tolerance compared to non-transformed plants.These results suggest that BnKCS1-1,BnKCS1-2,and BnCER1-2 gene products can modify the cuticular wax of B.napus.Changing cuticular waxes using transgenic approaches is a new strategy for genetic improvement of plant drought tolerance and provides an opportunity for development of B.napus as a surface-wax crop. | Yanmei Wang Shurong Jin Yi Xu Shuai Li Shuangjuan Zhang Zheng Yuan Jiana Li Yu Ni | 2020 | The Crop Journal2020,8,1: | 1 |

| 20 | CREB1 contributes colorectal cancer cell plasticity by regulating lnc RNA CCAT1 and NF-κB pathways显示文摘The CREB1 gene encodes an exceptionally pleiotropic transcription factor that frequently dysregulated in human cancers.CREB1 can regulate tumor cell status of proliferation and/or migration;however,the molecular basis for this switch involvement in cell plasticity has not fully been understood yet.Here,we first show that knocking out CREB1 triggers a remarkable effect of epithelial-mesenchymal transition(EMT)and leads to the occurrence of inhibited proliferation and enhanced motility in HCT116colorectal cancer cells.By monitoring 45 cellular signaling pathway activities,we find that multiple growth-related pathways decline significantly while inflammatory pathways including NF-κB are largely upregulated in comparing between the CREB1wild-type and knocked out cells.Mechanistically,cells with CREB1 knocked out show downregulation of MYC as a result of impaired CREB1-dependent transcription of the oncogenic lnc RNA CCAT1.Interestingly,the unbalanced competition between the coactivator CBP/p300 for CREB1 and p65 leads to the activation of the NF-κB pathway in cells with CREB1 disrupted,which induces an obvious EMT phenotype of the cancer cells.Taken together,these studies identify previously unknown mechanisms of CREB1 in CRC cell plasticity via regulating lnc RNA CCAT1 and NF-κB pathways,providing a critical insight into a combined strategy for CREB1-targeted tumor therapies. | Bin Li Lisi Zheng Jiayi Ye Chenmin Zhang Jie Zhou Qiaojuan Huang Yanhua Guo Luqin Wang Peng Yu Shurong Liu Qiao Lin Yuxia Luo Hui Zhou Jianhua Yang Lianghu Qu | 2022 | Science China(Life Sciences)2022,65,8: | 0 |
