| 1 | Development of a Haploid-Inducer Mediated Genome Editing System for Accelerating Maize Breeding显示文摘Crop breeding aims to generate pure in bred lines with multiple desired traits. Doubled haploid (DH) and genome editing using CRISPR/Cas9 are two powerful game-changing technologies in crop breeding. However, both of them still fall short for rapid generation of pure elite lines with integrated favorable traits. Here, we report the development of a Haploid-Inducer Mediated Genome Editing (IMGE) approach, which utilizes a maize haploid inducer line carrying a CRISPR/Cas9 cassette targeting for a desired agronomic trait to pollinate an elite maize in bred line and to generate genome-edited haploids in the elite maize background. | 4 | FHY3 and FAR1 Integrate Light Signals with the miR156-SPL Module-Mediated Aging Pathway to Regulate Arabidopsis Flowering显示文摘In response to competition for light from their neighbors,shade-intolerant plants flower precociously to ensure reproductive success and survival.However,the molecular mechanisms underlying this key developmental switch are not well understood.Here,we show that a pair of Arabidopsis transcription factors essential for phytochrome A signaling,FAR-RED ELONGATED HYPOCOTYL3(FHY3)and FAR-RED IMPAIRED RESPONSE1(FAR1),regulate flowering time by integrating environmental light signals with the miR156-SPL module-mediated aging pathway.We found that FHY3 and FAR1 directly interact with three flowering-promoting SQUAMOSA-PROMOTER BINDING PROTEIN-LIKE(SPL)transcription factors,SPL3,SPL4,and SPL5,and inhibit their binding to the promoters of several key flowering regulatory genes,including FRUITFUL(FUL),LEAFY(LFY),APETALA1(AP1),and MIR172C,thus downregulating their transcript levels and delaying flowering.Under simulated shade conditions,levels of SPL3/4/5 proteins increase,whereas levels of FHY3 and FAR1 proteins decline,thus releasing SPL3/4/5 from FHY3/FAR1 inhibition to allow activation of FUL,LFY,AP1,and MIR172C and,consequently,early flowering.Taken together,these results unravel a novel mechanism whereby plants regulate flowering time by integrating environmental cues(such as light conditions)and an internal developmental program(the miR156-SPL module-mediated aging pathway). | Yurong Xie Qin Zhou Yongping Zhao Quanquan Li Yang Liu Mengdi Ma Baobao Wang Rongxin Shen Zhigang Zheng Haiyang Wang | 2020 | Molecular Plant2020，13，3： | 8 |

| 6 | Effect of vernalization on tuberization and flowering in the Tibetan turnip is associated with changes in the expression of FLC homologues显示文摘The turnip(Brassica rapa var. rapa) is a biennial crop that is planted in late summer/early fall and forms fleshy tubers for food in temperate regions. The harvested tubers then overwinter and are planted again the next spring for flowering and seeds. FLOWERING LOCUS C(FLC) is a MADS-box transcription factor that acts as a major repressor of floral transition by suppressing the flowering promoters FT and SOC1. Here we show that vernalization effectively represses tuber formation and promotes flowering in Tibetan turnip. We functionally characterized four FLC homologues(BrrFLC1,FLC2, FLC3, and FLC5), and found that BrrFLC2 and BrrFLC1 play a major role in repressing flowering in turnip and in transgenic Arabidopsis. In contrast, tuber formation was correlated with BrrFLC1 expression in the hypocotyl and was repressed under cold treatment following the quantitative downregulation of BrrFLC1. Grafting experiments of non-vernalized and vernalized turnips revealed that vernalization independently suppressed tuberization in the tuber or hypocotyl of the rootstock or scion, which occurred in parallel with the reduction in BrrFLC1 activity. | 7 | Reduning injection for fever, rash, and ulcers in children with mild hand, foot, and mouth disease: a randomized controlled clinical study显示文摘OBJECTIVE: To assess the efficacy and safety of Reduning injection for fever, rash, and ulcers in children with mild hand, foot, and mouth disease(HFMD). METHODS: A stratified-block randomized, double-blind, parallel-controlled, and multicenter clinical trial was conducted with 360 patients in five hospitals across China: Quanzhou Children's Hospital, Shijiazhuang No. 5 Hospital, Shanghai Public Health Centre, Hunan Provincial Children's Hospital, and Kaifeng Children's Hospital. Patients were randomized into three groups with 120 in each. Group A was treated with Western Medicine, group B with Reduning injection, a Chinese herbal medicine, and group C with both Reduning injection and Western Medicine. Results were compared for treatment efficacy and safety on HFMD. The clinical outcomes were observed as follows: fever and onset time of antifebrile effect(time to bring the body temperature down ≥0.5℃ after medication); cumulative time for fever recovery(body temperature recovering to normal and lasting more than 24 h without medication); cumulative time for rash disappearance(without new rashes or ulcers appearing and the original ones fading away); and cumulative time for mouth ulcer disappearance. RESULTS: For the onset time of the antifebrile effect, there was no statistical difference between groups A and B(P>0.05) and groups B and C(P> 0.05). However, there was a statistical difference between groups A and C(P<0.05), and the effect in group C was the best. For the cumulative time for rash disappearance, there was no statistical difference between groups A and B(P>0.05). There were statistical differences between groups A and C, and groups B and C(P<0.05), and the effect in group C was the best. For the cumulative time for mouth ulcers disappearance, there were no statistical differences among the three groups(P>0.05). CONCLUSION: Reduning injection with Western Medicine for symptomatic treatment is most effective for mild HFMD. | 10 | Cow, yak, and camel milk diets differentially modulated the systemic immunity and fecal microbiota of rats显示文摘Cow milk is most widely consumed; however,non-cattle milk has gained increasing interest because of added nutritive values. We compared the health effects of yak,cow,and camel milk in rats. By measuring several plasma immune factors,significantly more interferon-c was detected in the camel than the yak(P = 0.0020) or cow(P = 0.0062) milk group. Significantly more Ig M was detected in the yak milk than the control group(P = 0.0071). The control group had significantly less interleukin 6 than the yak(P = 0.0499)and cow(P = 0.0248) milk groups. The fecal microbiota of the 144 samples comprised mainly of the Firmicutes(76.70 ± 11.03%),Bacteroidetes(15.27 ± 7.79%),Proteobacteria(3.61 ± 4.34%),and Tenericutes(2.61 ± 2.53%) phyla. Multivariate analyses revealed a mild shift in the fecal microbiota along the milk treatment. We further identified the differential microbes across the four groups. At day 14,22 and 28 differential genera and species were identified(P = 0.0000–0.0462),while 8 and 11 differential genera and species(P = 0.0000–0.0013) were found at day 28. Some short-chain fatty acid and succinate producers increased,while certain health-concerned bacteria(Prevotella copri,Phascolarctobacterium faecium,and Bacteroides uniformis) decreased after 14 days of yak or camel milk treatment. | 12 | Protective effect of Tanreqing injection on axon myelin damage in the brain of mouse model for experimental autoimmune encephalomyelitis显示文摘OBJECTIVE: To evaluate the effect of Tanreqing injection on axon myelin in the mouse brain of experimental autoimmune encephalomyelitis(EAE).METHODS: An EAE model was established by myelin oligodendrocyte glycoprotein(MOG)35-55 immunization in C57BL/6 mice. Mice were randomly divided into the following groups: normal, model,prednisone acetate(PA)(6 mg/kg), Tanreqing high dose(5.14 m L/kg), Tanreqing low dose(2.57 m L/kg). On the day of immunization, both Tanreqing groups were treated by intraperitoneal injection,with the PA group treated by intragastrical perfusion after T cell response, and the other groups treated with saline. Changes in body weight, neurological deficit score, incidence rate, mortality rate,and course of disease were observed for all mice.Brain tissue was isolated and stained with hematoxylin-eosin, and pathological investigations performed to evaluate axon myelin damage by transmission electron microscopy(TEM). Myelin basic protein and microtubule associated protein-2 were analyzed by immunohistochemistry.RESULTS: Tanreqing injection significantly prolonged EAE latency and decreased the neurological deficit score, alleviated infiltration of inflammatory cells in the focus area, up-regulated hippocampal MBP expression at the acute stage and the remission stage, and increased microtubule associated protein-2 expression in the EAE brain to varying degrees in the acute stage. By using the electromechanically coupling spring-type interface model and the generalized self-consistent method(GSM), we analytically derived the effective electroelastic moduli. The present solutions include as special cases the three-phase cylindrical piezoelectric composites with perfect interfaces as well as the two-phase(matrix/fiber) case with imperfect or perfect interfaces. Selected calculations are graphically shown to demonstrate dependence of the effective moduli on the interfacial properties.
