1Transforming growth factor-β in stem cells and tissue homeostasis显示文摘TGF-β 1–3 are unique multi-functional growth factors that are only expressed in mammals, and mainly secreted and stored as a latent complex in the extracellular matrix(ECM). The biological functions of TGF-β in adults can only be delivered after ligand activation, mostly in response to environmental perturbations. Although involved in multiple biological and pathological processes of the human body, the exact roles of TGF-β in maintaining stem cells and tissue homeostasis have not been well-documented until recent advances, which delineate their functions in a given context. Our recent findings, along with data reported by others, have clearly shown that temporal and spatial activation of TGF-β is involved in the recruitment of stem/progenitor cell participation in tissue regeneration/remodeling process, whereas sustained abnormalities in TGF-β ligand activation, regardless of genetic or environmental origin, will inevitably disrupt the normal physiology and lead to pathobiology of major diseases. Modulation of TGF-β signaling with different approaches has proven effective pre-clinically in the treatment of multiple pathologies such as sclerosis/fibrosis, tumor metastasis, osteoarthritis, and immune disorders. 2Intestinal microbiota: a potential target for the treatment of postmenopausal osteoporosis显示文摘Postmenopausal osteoporosis(PMO) is a prevalent metabolic bone disease characterized by bone loss and structural destruction, which increases the risk of fracture in postmenopausal women. Owing to the high morbidity and serious complications of PMO, many efforts have been devoted to its prophylaxis and treatment. The intestinal microbiota is the complex community of microorganisms colonizing the gastrointestinal tract. Probiotics, which are dietary or medical supplements consisting of beneficial intestinal bacteria, work in concert with endogenous intestinal microorganisms to maintain host health. Recent studies have revealed that bone loss in PMO is closely related to host immunity, which is influenced by the intestinal microbiota. The curative effects of probiotics on metabolic bone diseases have also been demonstrated. 3Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 4Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 5Aberrant activation of latent transforming growth factor-β initiates the onset of temporomandibular joint osteoarthritis显示文摘There is currently no effective medical treatment for temporomandibular joint osteoarthritis(TMJ-OA) due to a limited understanding of its pathogenesis. This study was undertaken to investigate the key role of transforming growth factor-β(TGF-β)signalling in the cartilage and subchondral bone of the TMJ using a temporomandibular joint disorder(TMD) rat model, an ageing mouse model and a Camurati–Engelmann disease(CED) mouse model. In the three animal models, the subchondral bone phenotypes in the mandibular condyles were evaluated by μCT, and changes in TMJ condyles were examined by TRAP staining and immunohistochemical analysis of Osterix and p-Smad2/3. Condyle degradation was confirmed by Safranin O staining, the Mankin and OARSI scoring systems and type X collagen(Col X), p-Smad2/3 a and Osterix immunohistochemical analyses. We found apparent histological phenotypes of TMJ-OA in the TMD, ageing and CED animal models, with abnormal activation of TGF-βsignalling in the condylar cartilage and subchondral bone. Moreover, inhibition of TGF-β receptor I attenuated TMJ-OA progression in the TMD models. 7Asian Summer Monsoon Onset in Simulations and CMIP5 Projections Using Four Chinese Climate Models显示文摘The reproducibility and future changes of the onset of the Asian summer monsoon were analyzed based on the simulations and projections under the Representative Concentration Pathways(RCP) scenario in which anthropogenic emissions continue to rise throughout the 21 st century(i.e. RCP8.5) by all realizations from four Chinese models that participated in the Coupled Model Intercomparison Project Phase 5(CMIP5). Delayed onset of the monsoon over the Arabian Sea was evident in all simulations for present-day climate, which was associated with a too weak simulation of the low-level Somali jet in May.A consistent advanced onset of the monsoon was found only over the Arabian Sea in the projections, where the advanced onset of the monsoon was accompanied by an increase of rainfall and an anomalous anticyclone over the northern Indian Ocean. In all the models except FGOALS-g2, the enhanced low-level Somali jet transported more water vapor to the Arabian Sea, whereas in FGOALS-g2 the enhanced rainfall was determined more by the increased wind convergence. Furthermore,and again in all models except FGOALS-g2, the equatorial SST warming, with maximum increase over the eastern Pacific,enhanced convection in the central West Pacific and reduced convection over the eastern Indian Ocean and Maritime Continent region, which drove the anomalous anticyclonic circulation over the western Indian Ocean. In contrast, in FGOALS-g2, there was minimal(near-zero) warming of projected SST in the central equatorial Pacific, with decreased convection in the central West Pacific and enhanced convection over the Maritime Continent. 8Development and evaluation of a regional ocean-atmosphere coupled model with focus on the western North Pacific summer monsoon simulation:Impacts of different atmospheric components显示文摘A regional ocean atmosphere coupled model (ROAM) is developed through coupler OASIS3,and is composed of regional climate model RegCM3 and CREM (Climate version of Regional Eta Model) as its atmospheric component and of a revised Princeton ocean model (POM2000) as its oceanic component.The performance of the ROAM over the western North Pacific summer monsoon region is assessed by the case simulation of warm season in 1998.Impacts of different atmospheric model components on the performance of ROAM are investigated.Compared with stand-alone simulation,CREM (RegCM3) produces more (or less) rainfall over ocean area with inclusion of the air-sea coupling.Different biases of rainfall are caused by the different biases of SST derived from the coupled simulation.Warm (or cold) SST bias simulated by CREM_CPL (RegCM3_CPL) increases (or decreases) the evaporation at sea surface,then increases (or decreases) the rainfall over ocean.The analyses suggest that the biases of vertical profile of temperature and specific humidity in stand-alone simulations may be responsible for the SST biases in regional coupled simulations.Compared with reanalysis data,the warmer (or colder) and moister (or dryer) lower troposphere simulated in CREM (RegCM3) produces less (or more) sea surface latent heat flux.Meanwhile,the more unstable (or stable) lower troposphere produces less (or more) cloudiness at low-level,which increases (or decreases) the solar radiation reaching on the sea surface.CREM (RegCM3) forced by observed SST overestimates (or underestimates) the sea surface net heat flux,implying a potential warm (or cold) heat source.After coupling with POM2000,the warm (or cold) heat source would further increase (or decrease) the SST.The biases of vertical profile of temperature and specific humidity may be ascribed to the different representation of cumulus convection in atmospheric models.ZOU LiWei ZHOU TianJun 2012Science China Earth Sciences2012,55,5:8

10The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. 12Berberine mediates root remodeling in an immature tooth with apical periodontitis by regulating stem cells from apical papilla differentiation显示文摘Once pulp necrosis or apical periodontitis occurs on immature teeth,the weak root and open root apex are challenging to clinicians.Berberine(BBR)is a potential medicine for bone disorders,therefore,we proposed to apply BBR in root canals to enhance root repair in immature teeth.An in vivo model of immature teeth with apical periodontitis was established in rats,and root canals were filled with BBR,calcium hydroxide or sterilized saline for 3 weeks.The shape of the roots was analyzed by micro-computed tomography and histological staining.In vitro,BBR was introduced into stem cells from apical papilla(SCAPs).Osteogenic differentiation of stem cells from apical papilla was investigated by alkaline phosphatase activity,mineralization ability,and gene expression of osteogenic makers.The signaling pathway,which regulated the osteogenesis of SCAPs was evaluated by quantitative real time PCR,Western blot analysis,and immunofluorescence.In rats treated with BBR,more tissue was formed,with longer roots,thicker root walls,and smaller apex diameters.In addition,we found that BBR promoted SCAPs osteogenesis in a time-dependent and concentration-dependent manner.BBR induced the expression ofβ-catenin and enhancedβ-catenin entering into the nucleus,to up-regulate more runt-related nuclear factor 2 downstream.BBR enhanced root repair in immature teeth with apical periodontitis by activating the canonical Wnt/β-catenin pathway in SCAPs.Yujia Cui Jing Xie Yujie Fu Chuwen Li Liwei Zheng Dingming Huang Changchun Zhou Jianxun Sun Xuedong Zhou 2020International Journal of Oral Science2020,12,2:5
13Simple synthesis of sandwich-like SnSe2/rGO as high initial coulombic efficiency and high stability anode for sodium-ion batteries显示文摘Metal selenides owing to their high theoretical capacity and good conductivity are considered as one of the potential candidates for the anode materials of sodium-ion batteries(SIBs).However,their practical applications are greatly restricted by the poor cycling performances and complicated synthesis methods.In this work,a sandwich-like Sn Se2/reduced graphene oxide(r GO)composite with a small amount of r GO(7.3%)is synthesized by a simple one-pot solvothermal technique.The as-synthesized Sn Se2/r GO shows improved initial coulombic efficiency(ICE)of 73.7%,high capacity of 402.0 m Ah g-1 after 150 cycles at 0.1 A g-1 with a retention of 86.2%and outstanding rate performances.The abundant Sn-O-C bonds of synthesized material not only accelerate the charge transfer at the interface but also enhance the mechanical strength to accommodate volume variation and prevent active material loss during cycling.Moreover,the compact structure leads to thin solid electrolyte interface(SEI)so that high initial coulombic efficiency was obtained.Furthermore,full cells are assembled to test its potential application.This work offers a simple method to synthesize Sn Se2/r GO as a candidate anode for SIBs.Tingting Wang Kaiwei Yang Juan Shi Shangrui Zhou Liwei Mi Hongping Li Weihua Chen 2020Journal of Energy Chemistry2020,29,7:4

15Introduction to the Regional Coupled Model WRF4-LICOM: Performance and Model Intercomparison over the Western North Pacific显示文摘Regional coupled modeling is one of the frontiers of regional climate modeling,but intercomparison has not been well coordinated.In this study,a community regional climate model,WRF4,with a resolution of 15 km,was coupled with a high-resolution(0.1°)North Pacific Ocean model(LICOM_np).The performance of the regional coupled model,WRF4_LICOM,was compared to that of another regional coupled model,RegCM4_LICOM,which was a coupling of version 4 of the Regional Climate Model(RegCM4)with LICOM_np.The analysis focused on the 2005 western North Pacific summer monsoon rainfall.The results showed that the regional coupled models with either RegCM4 or WRF4 as their atmospheric model component simulated the broad features over the WNP reasonably well.Quantitative intercomparison of the regional coupled simulations exhibited different biases for different climate variables.RegCM4_LICOM exhibited smaller biases in its simulation of the averaged June–July–August SST and rainfall,while WRF4_LICOM better captured the tropical cyclone(TC)intensity,the percentage contributions of rainfall induced by TCs to the total rainfall,and the diurnal cycle of rainfall and stratiform percentages,especially over land areas.The different behaviors in rainfall simulated by the two models were partly ascribed to the behaviors in the simulated western North Pacific subtropical high(WNPSH).The stronger(weaker)WNPSH in WRF4_LICOM(RegCM4_LICOM)was driven by overestimated(underestimated)diabatic heating,which peaked at approximately 450 hPa over the region around the Philippines in association with different condensation–radiation processes.Coupling of WRF4 with LIOCM is a crucial step towards the development of the next generation of regional earth system models at the Chinese Academy of Sciences.Liwei ZOU Tianjun ZHOU Jianping TANG Hailong LIU 2020Advances in Atmospheric Sciences2020,37,8:4

17Progress report on the study of wood-decaying fungi in China显示文摘This study addressed three important aims:(1) undermining the previously obtained raw data about wood-decaying fungi(WDF) distribution and continuously investigating permanent plots to address certain scientific questions in ecology,(2) resolving the higher-level phylogeny of WDF with the help of multiple loci,and(3) testing and estimating the medicinal values of species that are closely related to well-known medicinal species.More than 1200 species and 2469 strains of WDF in China were identified from 28908 specimens collected from a series of field investigations.Using these materials,studies in multiple disciplines,such as ecology,taxonomy and phylogeny,and medicine,have been performed.With respect to ecology,the diversity of wood-decaying polypores significantly differed among a boreal forest zone,a temperate and warm temperate forest zone,and a tropical and subtropical forest zone.For instance,from north to south,the number and proportion of brown-rot species and the proportion of species found on fallen trunks were both decreased.The ecological patterns of wood-decaying polypores on gymnosperm and angiosperm trees were also explored by a case study in Northeast China.Although the total species richness was similar between the two tree groups,several other characteristics were significantly different,such as community structure and richness in certain substrates.The taxonomy and phylogeny of wide samples were referred to and their phylogenetic positions were resolved or at least partially established.In particular,phylogenetic knowledge about four genera,Fomitiporia,Ganoderma,Inonotus and Perenniporia,which include medicinal species,was essential for further research to determine the medicinal values of these types of fungi.Among these medicinal species,we mainly focused on Inonotus obliquus for its medicinal purposes.Polyphenols,polysaccharides and lanostane-type triterpenoids,extracted from the sterile conk of this species,could dramatically decrease levels of free radicals,DPPH and hydroxyl radicals,respectively.The metabolic profiles(both production and composition) of cultured I.obliquus mycelia could be altered by co-culture with other medicinal species or by induction of S-nitrosylation and denitrosylation,which may enhance the antioxidant capacity of I.obliquus.ZHOU LiWei DAI YuCheng 2012Chinese Science Bulletin2012,57,33:3
18Two interannual variability modes of the Northwestern Pacific Subtropical Anticyclone in boreal summer显示文摘Using the reanalysis data and 20th century simulation of coupled model FGOALS_gl developed by LASG/IAP, we identified two distinct interannual modes of Northwestern Pacific Subtropical Anticyclone (NWPAC) by performing Empirical Orthogonal Function (EOF) analysis on 850 hPa wind field over the northwestern Pacific in summer. Based on the associated anomalous equatorial zonal wind, these two modes are termed as 'Equatorial Easterly related Mode' (EEM) and 'Equatorial Westerly related Mode' (EWM), respectively. The formation mechanisms of these two modes are similar, whereas the maintenance mechanisms, dominant periods, and the relationships with ENSO are different. The EEM is associated with El Ni o decaying phase, with the anomalous anticyclone established in the preceding winter and persisted into summer through local positive air-sea feedback. By enhancing equatorial upwelling of subsurface cold water, EEM favors the transition of ENSO from El Ni o to La Ni a. The EWM is accompanied by the El Ni o events with long persistence, with the anomalous anticyclone formed in spring and strengthened in summer due to the warm Sea Surface Temperature anomalies (SSTA) forcing from the equatorial central-eastern Pacific. The model well reproduces the spatial patterns of these two modes, but fails to simulate the percentage variance accounted for by the two modes. 19Simulation of the western North Pacific summer monsoon by regional ocean–atmosphere coupled model:impacts of oceanic components显示文摘A successful simulation of the western North Pacific summer monsoon needs a regional ocean–atmosphere coupled model(ROAM). How the performance of ROAM relies on the oceanic component model remains unknown. In this study, the authors investigated the effects of different oceanic components on the simulation of western North Pacific(WNP) summer monsoon in a ROAM. Three cases of simulations were performed, viz. the summer of 1998(El Nin o decaying phase), 2004(El Nin o developing phase), and 1993(the non-ENSO phase). Results show that the coupled simulations for different ENSO phases exhibit improvements in the simulation of location of Meiyu rainband and spatial distribution of monsoon low-level flow over WNP, whereas the systemic cold biases of sea surface air temperature are further increased. The coupled simulations with different oceanic components show similar performance, which is not ENSO phase dependent. For the case of the summer of 1998, a slightly stronger western Pacific subtropical high and colder sea surface air temperature are found in the simulation with colder sea surface temperature(SST) biases. The colder SST biases are partly contributed by the ocean dynamics processes because the sea surface net flux favors a warmer SST. 20Injectable kartogenin and apocynin loaded micelle enhances the alleviation of intervertebral disc degeneration by adipose-derived stem cell显示文摘Cell transplantation has been proved the promising therapeutic effects on intervertebral disc degeneration(IVDD).However,the increased levels of reactive oxygen species(ROS)in the degenerated region will impede the efficiency of human adipose-derived stem cells(human ADSCs)transplantation therapy.It inhibits human ADSCs proliferation,and increases human ADSCs apoptosis.Herein,we firstly devised a novel amphiphilic copolymer PEG-PAPO,which could self-assemble into a nanosized micelle and load lipophilic kartogenin(KGN),as a single complex(PAKM).It was an injectable esterase-responsive micelle,and showed controlled release ability of KGN and apocynin(APO).Oxidative stimulation promoted the esterase activity in human ADSCs,which accelerate degradation of esterase-responsive micelle.Compared its monomer,the PAKM micelle possessed better bioactivities,which were attributed to their synergistic effect.It enhanced the viability,autophagic activation(P62,LC3 II),ECM-related transcription factor(SOX9),and ECM(Collagen II,Aggrecan)maintenance in human ADSCs.Furthermore,it is demonstrated that the injection of PAKM with human ADSCs yielded higher disc height and water content in rats.Therefore,PAKM micelles perform promoting cell survival and differentiation effects,and may be a potential therapeutic agent for IVDD.Chao Yu Dongdong Li Chenggui Wang Kaishun Xia Jingkai Wang Xiaopeng Zhou Liwei Ying Jiawei Shu Xianpeng Huang Haibin Xu Bin Han Qixin Chen Fangcai Li Jianbin Tang Chengzhen Liang Nigel Slater 2021Bioactive Materials2021,6,10:3
