1Enhanced production of seed oil with improved fatty acid composition by overexpressing NAD^(+)-dependent glycerol-3-phosphate dehydrogenase in soybean显示文摘There is growing interest in expanding the production of soybean oils(mainly triacylglycerol, or TAG) to meet rising feed demand and address global energy concerns. We report that a plastidlocalized glycerol-3-phosphate dehydrogenase(GPDH), encoded by GmGPDHp1 gene, catalyzes the formation of glycerol-3-phosphate(G3 P), an obligate substrate required for TAG biosynthesis.Overexpression of GmGPDHp1 increases soybean seed oil content with high levels of unsaturated fatty acids(FAs), especially oleic acid(C18:1), without detectably affecting growth or seed protein content or seed weight. Based on the lipidomic analyses, we found that the increase in G3 P content led to an elevated diacylglycerol(DAG) pool, in which the Kennedy pathwayderived DAG was mostly increased, followed by PC-derived DAG, thereby promoting the synthesis of TAG containing relatively high proportion of C18:1. The increased G3 P levels induced several transcriptional alterations of genes involved in the glycerolipid pathways. In particular, genes encoding the enzymes responsible for de novo glycerolipid synthesis were largely upregulated in the transgenic lines, in-line with the identified biochemical phenotype. 2AIEgens in cell-based multiplex fluorescence imaging显示文摘Fluorescence imaging is an important branch of bioimaging.It is non-invasive and provides superior spatial and temporal resolution during the real-time monitoring of biological samples of interest.Although the spatial resolution limit of optical microscopes is about 200 nm,due to the diffraction limit,with the application of super-resolution fluorescence microscopy technologies this limit has been pushed below 30 nm.This makes it feasible to visualize biological structures in subcellular levels and to monitor subcellular biological processes in real time.However,due to the complexity of the biological structure and components within cells,simultaneous staining and monitoring multiple intracellular components with different coloured fluorophores is often needed during multiplex imaging,to better understand biological processes.Aggregation-induced emission luminogens(AIEgen)and AIEgen based nanoparticles(NPs)have presented many advantages in fluorescence imaging,with strong potential for biological science and nano-medicine.Herein this review,we focus on the advantages of AIEgen and AIEgen NP in cell-based fluorescence imaging,and the latest advances of AIEgens in cell-based multiplex imaging are summarized and discussed.The future perspectives are proposed.Yabin Zhou Jin Hua Ben Zhong Tang Youhong Tang 2019Science China Chemistry2019,62,10:3
3Multiplexed imaging detection of live cell intracellular changes in early apoptosis with aggregation-induced emission fluorogens显示文摘Apoptosis is an important process for maintaining tissue homeostasis and eliminating abnormal cells in multicellular organisms.Abnormality in apoptosis often leads to severe diseases such as cancers. Better understanding of its mechanisms and processes is therefore important. Accompanying molecular biology events of apoptosis is a series of cellular morphology changes: nucleus condensation, cell shrinkage and rounding, cell surface blebbing, dynamic blebbing, apoptotic membrane protrusions and nucleus fragmentations and finally, the formation and release of apoptotic bodies. It is difficult to detect cellular changes in the early phase of apoptosis due to the subtle changes at this phase. In the current study, we induced apoptosis in He La cells with H2 O2 and used nuclear dye Hoechst 33258, mitochondria, lysosome and cytoplasmic protein specific aggregation-induced emission fluorogens(AIEgens), TPE-Ph-In, 2 M-DABS and BSPOTPE to successfully perform live cell multiplexed imaging to investigate early apoptosis cellular events. We showed the gradual dissipation of mitochondria membrane potential until it is nondetectable by TPE-Ph-In. 14Establishment and application of national reference panels for SARS-CoV-2 antigen detection kit显示文摘To develop a national reference panel for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)antigen detection kit and establish a quality standard.The cultures of SARS-CoV-2 and other pathogens were collected to establish a national reference panel for SARS-CoV-2 antigen detection.The stability and homogeneity of the reference panel were evaluated.Based on World Health Organization(WHO)guidance and nucleic acid quantitative results,a quality standard reference panel was established.Currently,three generations of SARS-CoV-2 antigen national reference materials with batch numbers 370095–202001,370095–202202,and 370095–202203 have been successfully established.These national reference panels comprised 8 positive samples,20 negative samples,1 repetitive sample,and 1 lower detection limit sample.The stability and homogeneity of the reference panel meet the requirements.The quality standards are as follows:the positive and negative coincidence rates are 8/8 and 20/20,respectively.The 10 test results of the medium and low-concentration repetitive reference materials should be positive,and the color rendering should be uniform(or the coefficient of variance should not be higher than 20.0%).The lower detection limit should be at least 5×105 U/mL(equivalent to copies/mL),and higher concentrations above the lower detection limit must be positive.A national reference panel for the SARS-CoV-2 antigen detection kit has been established.As the standard of SARS-CoV-2 antigen reagents,the reference panel has played a crucial role in the pre-marketing quality evaluation and post-marketing quality supervision in China.Tingting Ma Donglai Liu Keliang Lyu Tingting Gao Dawei Shi Lanqing Zhao Shu Shen Yabin Tian Sihong Xu Haiwei Zhou 2023Biosafety and Health2023,5,6:0
15Approaches to improving critical current density of YBCO superconducting thick films deposited by electrophoresis显示文摘Fabrication of YBCO superconducting thick films by electrophoresis has been widely reported. However, the value of critical current density (Jc) is far from what the practical application requires. The superconducting thick films are obtained using top-seeded melt growth method with different proportional Y2BaCuO5 (Y211) addition. High-pressure oxygen has been used in the annealing process to improve the uniformity of the oxygen content in the superconducting thick films. 16Culture of Eriocheir sinensis Larvae in Ponds in Greenhouse显示文摘A total of 15 pools were selected from a greenhouse, and they were randomly and evenly divided into three groups.In each group, one stocking density of Eriocheir sinensis was arranged. The results showed that after 45-d culture, the crab number per kilogram reached about 300 with survival rate of about 20%; the initial stocking density had significantly effect on the body size of E. sinensis on sell, instead on the survival rate of E. sinensis larvae.
