4Enhanced production of seed oil with improved fatty acid composition by overexpressing NAD^(+)-dependent glycerol-3-phosphate dehydrogenase in soybean显示文摘There is growing interest in expanding the production of soybean oils(mainly triacylglycerol, or TAG) to meet rising feed demand and address global energy concerns. We report that a plastidlocalized glycerol-3-phosphate dehydrogenase(GPDH), encoded by GmGPDHp1 gene, catalyzes the formation of glycerol-3-phosphate(G3 P), an obligate substrate required for TAG biosynthesis.Overexpression of GmGPDHp1 increases soybean seed oil content with high levels of unsaturated fatty acids(FAs), especially oleic acid(C18:1), without detectably affecting growth or seed protein content or seed weight. Based on the lipidomic analyses, we found that the increase in G3 P content led to an elevated diacylglycerol(DAG) pool, in which the Kennedy pathwayderived DAG was mostly increased, followed by PC-derived DAG, thereby promoting the synthesis of TAG containing relatively high proportion of C18:1. The increased G3 P levels induced several transcriptional alterations of genes involved in the glycerolipid pathways. In particular, genes encoding the enzymes responsible for de novo glycerolipid synthesis were largely upregulated in the transgenic lines, in-line with the identified biochemical phenotype. 5An open conformation determined by a structural switch for 2A protease from coxsackievirus A16显示文摘Coxsackievirus A16 belongs to the family Picornaviridae,and is a major agent of hand-foot-and-mouth disease that infects mostly children,and to date no vaccines or antivi-ral therapies are available.2A protease of enterovirus is a nonstructural protein and possesses both self-cleavage activity and the ability to cleave the eukaryotic translation initiation factor 4G.Here we present the crystal structure of coxsackievirus A162A protease,which interestingly forms hexamers in crystal as well as in solution.This structure shows an open conformation,with its active site accessible,ready for substrate binding and cleav-age activity.In conjunction with a previously reported“closed”state structure of human rhinovirus 2,we were able to develop a detailed hypothesis for the conforma-tional conversion triggered by two“switcher”residues Glu88 and Tyr89 located within the bll2-cII loop.Substrate recognition assays revealed that amino acid residues P1′,P2 and P4 are essential for substrate specificity,which was verifi ed by our substrate binding model.In addition,we compared the in vitro cleavage effi ciency of 2A pro-teases from coxsackievirus A16 and enterovirus 71 upon the same substrates by fl uorescence resonance energy transfer(FRET),and observed higher protease activity of enterovirus 71 compared to that of coxsackievirus A16.In conclusion,our study shows an open conformation of coxsackievirus A162A protease and the underlying mechanisms for conformational conversion and substrate specifi city.These new insights should facilitate the future rational design of effi cient 2A protease inhibitors.Yao Sun Xiangxi Wang Shuai Yuan Minghao Dang Xuemei Li Xuejun CZhang Zihe Rao 2013Protein & Cell2013,4,10:3
6Experimental Investigation for Co-Combustion Characteristics of Semi-Coke and Bituminous Coal in a 3 MWth Tangential Combustion Facility显示文摘An experimental investigation was conducted in a 3 MW pilot-scale tangential combustion facility to explore the co-combustion characteristics of bituminous coal mixed with semi-coke.The thermal gravimetric analyzer(TGA)was used to obtained fuel thermal analysis.The results presented effects of semi-coke blending ratio(BR)on average furnace temperature,ignition temperature,NO emission and combustion efficiency.The excess air coefficient in main combustion sections and outlet were fixed at 0.85 and 1.2 while BR increased from 0%to 50 wt.%.The temperature profiles of combustion decreases along the height of furnace while average furnace temperature fluctuates slightly with an increasing BR.The concentration of NO has an increasing tendency with the increasing of BR.The ignition temperature obtained from TGA measurement agreed well with experiment result.In addition,combustion efficiency was not sensitive to BR and decreased slightly with the increasing BR.GUAN Jingyu YU Qiang SUN Rui SHEN Tao WANG Minghao YAN Yanfei SONG Xin 2020Journal of Thermal Science2020,29,6:2

10Spiral plume structures in turbulent natural convection between two vertical walls显示文摘By means of direct numerical simulation, coherent structures are investigated in turbulent natural convection between two vertical differentially heated walls. It is observed that large-scale spanwise vortices and spiral plume structures exist together in the flow. Spiral plume structures appear at the positions with relatively large helicity, large normal vorticity and high fluctuating temperature. In this note, the shape, the characteristics and formation of the spiral structures are studied and compared with those in Rayleigh-Benard convection. 13Microglia in brain aging:An overview of recent basic science and clinical research developments显示文摘Aging is characterized by progressive degeneration of tissues and organs,and it is positively associated with an increased mortality rate.The brain,as one of the most significantly affected organs,experiences age-related changes,including abnormal neuronal activity,dysfunctional calcium homeostasis,dysregulated mitochondrial function,and increased levels of reactive oxygen species.These changes collectively contribute to cognitive deterioration.Aging is also a key risk factor for neurodegenerative diseases,such as Alzheimer's disease and Parkinson's disease.For many years,neurodegenerative disease investigations have primarily focused on neurons,with less attention given to microglial cells.However,recently,microglial homeostasis has emerged as an important mediator in neurological disease pathogenesis.Here,we provide an overview of brain aging from the perspective of the microglia.In doing so,we present the current knowledge on the correlation between brain aging and the microglia,summarize recent progress of investigations about the microglia in normal aging,Alzheimer's disease,Parkinson's disease,Huntington's disease,and amyotrophic lateral sclerosis,and then discuss the correlation between the senescent microglia and the brain,which will culminate with a presentation of the molecular complexity involved in the microglia in brain aging with suggestions for healthy aging.Haixia Fan Minheng Zhang Jie Wen Shengyuan Wang Minghao Yuan Houchao Sun Liu Shu Xu Yang Yinshuang Pu Zhiyou Cai 2024The Journal of Biomedical Research2024,38,2:0
14Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 15When liver disease diagnosis encounters deep learning:Analysis,challenges,and prospects显示文摘The liver is the second-largest organ in the human body and is essential for digesting food and removing toxic substances.Viruses,obesity,alcohol use,and other factors can damage the liver and cause liver disease.The diagnosis of liver disease used to depend on the clinical experience of doctors,which made it subjective,difficult,and time-consuming.Deep learning has made breakthroughs in various fields;thus,there is a growing interest in using deep learning methods to solve problems in liver research to assist doctors in diagnosis and treatment.In this paper,we provide an overview of deep learning in liver research using 139 papers from the last 5 years.We also show the relationship between data modalities,liver topics,and applications in liver research using Sankey diagrams and summarize the deep learning methods used for each liver topic,in addition to the relations and trends between these methods.Finally,we discuss the challenges of and expectations for deep learning in liver research.Yingjie Tian Minghao Liu Yu Sun Saiji Fu 2023iLIVER2023,2,1:0
16Hyperbranched polymer-cored star polyfluorenes as blue light-emitting materials显示文摘Hyperbranched polymer-cored star polyfluorenes with high molecular weights and narrow molecular weight distribution were prepared by palladium-catalyzed one-pot Suzuki polycondensation of multi- functional cores and an AB-type monomer. The optical, electrochemical and thermal properties of the hyperbranched polymer-cored star polymers were investigated. These polymers exhibited good ther- mal and color stability in solid state, and there was no significant blue-green emission after the poly- mers had been annealed in air for 2.5 h. 18Carboxymethyl chitosan/poly(ethylene oxide)water soluble binder:Challenging application for 5V LiNi0.5Mn1.5O4 cathode显示文摘Carboxymethyl chitosan/poly(ethylene oxide)(CCTS/PEO) composite is firstly reported as a water soluble binder for the application of 5V LiNi_(0.5)Mn_(1.5)O_4 cathode in Li-ion batteries. Both CCTS and PEO show a high electrochemical oxidation potential of above 5.0 V(vs. Li/Li+). The electrochemical performances of LiNi_(0.5)Mn_(1.5)O_4(LNMO) cathodes with CCTS/PEO composite binders of different mass rates are investigated, it is found that LiNi_(0.5)Mn_(1.5)O_4 cathode with an optimized CCTS/PEO(85/15, w/w) composite exhibits a slightly better cycling performance than that of polyvinylidene fluoride(PVDF), retaining 81.4% capacity as compared with 79.8% for PVDF at 0.5C rate after 200 cycles. LNMO with PEO/CCTS(85/15,w/w) exhibited the better rate capability than that of PVDF. 19Single-cell profiling reveals Müller glia coordinate retinal intercellular communication during light/dark adaptation via thyroid hormone signaling显示文摘Light adaptation enables the vertebrate visual system to operate over a wide range of ambient illumination.Regulation of phototransduction in photoreceptors is considered a major mechanism underlying light adaptation.However,various types of neurons and glial cells exist in the retina,and whether and how all retinal cells interact to adapt to light/dark conditions at the cellular and molecular levels requires systematic investigation.Therefore,we utilized single-cell RNA sequencing to dissect retinal cell-type-specific transcriptomes during light/dark adaptation in mice.The results demonstrated that,in addition to photoreceptors,other retinal cell types also showed dynamic molecular changes and specifically enriched signaling pathways under light/dark adaptation.Importantly,Müller glial cells(MGs)were identified as hub cells for intercellular interactions,displaying complex cell‒cell communication with other retinal cells.Furthermore,light increased the transcription of the deiodinase Dio2 in MGs,which converted thyroxine(T4)to active triiodothyronine(T3).Subsequently,light increased T3 levels and regulated mitochondrial respiration in retinal cells in response to light conditions.As cones specifically express the thyroid hormone receptor Thrb,they responded to the increase in T3 by adjusting light responsiveness.Loss of the expression of Dio2 specifically in MGs decreased the light responsive ability of cones.These results suggest that retinal cells display global transcriptional changes under light/dark adaptation and that MGs coordinate intercellular communication during light/dark adaptation via thyroid hormone signaling.Min Wei Yanping Sun Shouzhen Li Yunuo Chen Longfei Li Minghao Fang Ronghua Shi Dali Tong Jutao Chen Yuqian Ma Kun Qu Mei Zhang Tian Xue 2023Protein & Cell2023,14,8:0
20CHARACTERISTIC FEATURES OF MUELLER MATRIX PATTERNS FOR POLARIZATION SCATTERING MODEL OF BIOLOGICAL TISSUES显示文摘We developed a model to describe polarized photon scattering in biological tissues.In this model,tissues are simplified to a mixture of scatterers and surrounding mpdium.There are t wo types ofscatterers in the model:solid spheres and infinitely long solid cylinders.Variables related to thescatterers include:the densities and sizes of the spheres and cylinders,the orientation and angulardistribution of cylinders.Variables related to the surrounding medium include:the refractiveindex,absorption coeficient and birefringence.In this paper,as a development we introduce anoptical activity effct to the model.By comparing experiments and Monte Carlo simulations,we analyze the backscater ing Mueller matrix patterms of several tissue-like media,and summarizethe different effects coming from anisotropic scattering and optical properties.In addition,wepropose a possible method to extract the optical activity values for tissues.Both the experimentaland simulated results show that,by analyzing the Mueller matrix patterns,the microstructureand optical properties of the medium can be obtained.The characteristic features of Muellermatrix patterns are potentilly powerful tools for studying the contrast mechanisms of polari-zation imaging for medical diagnosis.E DU HONGHUI HE NAN ZENG CELONG LIU YIHONG GUO RAN LIAO MINGHAO SUN YONGHONG HE HUI MA 2014Journal of Innovative Optical Health Sciences2014,7,1:0
