2Labeling of cynomolgus monkey bone marrow-derived mesenchymal stem cells for cell tracking by multimodality imaging显示文摘Recently,transplantation of allogeneic and autologous cells has been used for regenerative medicine.A critical issue is monitoring migration and homing of transplanted cells,as well as engraftment efficiency and functional capability in vivo.Monitoring of superparamagnetic iron oxide(SPIO) particles by magnetic resonance imaging(MRI) has been used in animal models and clinical settings to track labeled cells.A major limitation of MRI is that the signals do not show biological characteristics of transplanted cells in vivo.Bone marrow mesenchymal stem cells(MSCs) have been extensively investigated for their various therapeutic properties,and exhibit the potential to differentiate into cells of diverse lineages.In this study,cynomolgus monkey MSCs(cMSCs) were labeled with Molday ION Rhodamine-BTM(MIRB),a new SPIO agent,to investigate and characterize the biophysical and MRI properties of labeled cMSCs in vitro and in vivo.The results indicate that MIRB is biocompatible and useful for cMSCs labeling and cell tracking by multimodality imaging.Our method is helpful for detection of transplanted stem cells in vivo,which is required for understanding mechanisms of cell therapy.REN ZhenHua WANG JiaYin ZOU ChunLin GUAN YunQian ZHANG Yu Alex 2011Science China(Life Sciences)2011,54,11:9

4Discrimination of False Negative Results in RT-PCR Detection of SARS-CoV-2 RNAs in Clinical Specimens by Using an Internal Reference显示文摘Reverse transcription-polymerase chain reaction(RT-PCR)is an essential method for specific diagnosis of SARS-CoV-2 infection.Unfortunately,false negative test results are often reported.In this study,we attempted to determine the principal causes leading to false negative results of RT-PCR detection of SARS-CoV-2 RNAs in respiratory tract specimens.Multiple sputum and throat swab specimens from 161 confirmed COVID-19 patients were tested with a commercialfluorescent RT-PCR kit targeting the ORF1 ab and N regions of SARS-CoV-2 genome.The RNA level of a cellular housekeeping gene ribonuclease P/MRP subunit p30(RPP30)in these specimens was also assessed by RT-PCR.Data for a total of 1052 samples were retrospectively re-analyzed and a strong association between positive results in SARS-CoV-2 RNA tests and high level of RPP30 RNA in respiratory tract specimens was revealed.By using the ROC-AUC analysis,we identified Ct cutoff values for RPP30 RT-PCR which predicted false negative results for SARS-CoV-2 RT-PCR with high sensitivity(95.03%–95.26%)and specificity(83.72%–98.55%)for respective combination of specimen type and ampli-fication reaction.Using these Ct cutoff values,false negative results could be reliably identified.Therefore,the presence of cellular materials,likely infected host cells,are essential for correct SARS-CoV-2 RNA detection by RT-PCR in patient specimens.RPP30 could serve as an indicator for cellular content,or a surrogate indicator for specimen quality.In addition,our results demonstrated that false negativity accounted for a vast majority of contradicting results in SARS-CoV-2 RNA test by RT-PCR.Yafei Zhang Changtai Wang Mingfeng Han Jun Ye Yong Gao Zhongping Liu Tengfei He Tuantuan Li Mengyuan Xu Luping Zhou Guizhou Zou Mengji Lu Zhenhua Zhang 2020Virologica Sinica2020,35,6:5
5Characterizing the induction of diabetes in juvenile cynomolgus monkeys with different doses of streptozotocin显示文摘Juvenile(2-3 years old) cynomolgus monkeys are frequently used as recipients in non-human primate islet transplantation studies.The aim of this study was to examine the effects of different doses of streptozotocin(STZ),and find the optimal dose for inducing diabetes in these monkeys.Fifteen juvenile(2-3 years old) cynomolgus monkeys were separated into three groups and administered with different doses of STZ(100,68 or 60 mg kg-1).Basal and glucose-stimulated blood glucose,insulin,and C-peptide levels,as well as body weights were monitored.Hepatic and renal function tests and pancreatic immunohistochemistry were performed before and after STZ treatment.Monkeys treated with both 100 and 68 mg kg-1 of STZ exhibited continuous hyperglycemia,which coincided with a nearly complete loss of islet--cells.Two monkeys received 60 mg kg-1 of STZ,but only one became completely diabetic.During the first week following STZ treatment,hepatic and renal function slightly increased in these three groups.However,24 hours post-STZ,serum total bile acid levels were significantly increased in monkeys treated with 100 mg kg-1 than those treated with 68 mg kg-1 of STZ(P<0.05).These data suggest that 100 mg kg-1 and 68 mg kg-1 of STZ can safely induce diabetes in cynomolgus monkeys aged 2-3 years,but 68 mg kg-1 of STZ,rather than 100 mg kg-1 of STZ,may be more appropriate for inducing diabetes in these monkeys.Furthermore,body surface area,rather than body weight,was a more reliable determinant of dosage,where 700 mg m-2 of STZ should be the lower limit for inducing diabetes in juvenile monkeys.ZOU ChunLin WANG JiaYin WANG ShuYan HUANG Fen REN ZhenHua CHEN ZhiGuo ZHANG YU 2012Science China(Life Sciences)2012,55,3:3

7Sinorhizobium meliloti nifA gene exerts a pleiotropic effect on nodulation through the enhanced plant defense response显示文摘Sinorhizobium meliloti nifA gene is required for the expression of a bunch of nif and fix genes. Here, we report its pleiotropic effects on the nodule formation. Compared with wild type strain, nifA mutant sig- nificantly reduced nodule suppression rate in split-root system. The plants inoculated with mutant strain produced lower amount of daidzein and less necrotic cells on their roots. In addition, the defense genes failed to be evoked by nifA mutant at the early nodulation stage. These findings indicated that host defense response was one of the mechanisms mediated by nifA gene to regulate nodule formation during symbiosis. Even though nifA mutant could increase the number of nodules in host plant, it synthesized lower Nod factors than wild type. 14Identification of sweet spots in shale-type and siltstone-type“shale oil systems”:A case study of the Chang 7 Member in Ordos Basin显示文摘Mid-high maturity shale oil is the most realistic field for the scale breakthrough of terrestrial shale oil production in China.Generally,three deficiencies hinder shale oil development in China:heavy oil density,small sweet spot areas,and poor distribution continuity.Thus,identifying the“sweet spots”in shale oil reservoirs is critical for the efficient exploration and development of terrestrial shale oil.This study targets the siltstone type(Class-Ⅱshale oil)and pure shale type(Class-Ⅲshale oil)of the Chang 7 Member in the Ordos Basin,and identifies three stratigraphic units,namely the hydrocarbon accumulation unit,hydrocarbon generation unit,and hydrocarbon retention unit,which together constitute the in-source“shale oil system”.The hydrocarbon accumulation unit is mainly siltstone,where the hydrocarbons are migrated from shales.It has favorable porethroat network connectivity with a pore connectivity ratio of 32–57%,being the siltstone-type sweet spots.The hydrocarbon generation unit is mainly composed of high-TOC mudstone/shale and is the main contributor to in-source hydrocarbon generation and expulsion.This unit has high three-dimensional connectivity(28–30%),as shown by the pore-throat network model,associated with vertical paths for hydrocarbon expulsion.The hydrocarbon retention unit is mainly composed of low-TOC mudstone/shale retaining self-generated and migrated hydrocarbons.The pore connectivity rate is 17–42%,and the pore-throat network connectivity direction is uneven.Light and low-carbon-number hydrocarbons are preferentially trapped or even sealed in small pores of the retention unit,forming the typical mudstone/shale-type sweet spots.In the process called shale oil intrasource migration,the oil migrates in source rocks causing component fractionation,which allows more shale oil to enrich in the hydrocarbon accumulation and retention units to form sweet spots,compared with the hydrocarbon generation unit.The migration paths include the one from mudstone/shale to siltstone interlayers and that from the high-TOC mudstone/shale intervals to the low-TOC intervals.The in-source accumulation of shale oil shows the differentiated enrichment model featuring“high-TOC mudstone/shale generating hydrocarbons,low-TOC mudstone/shale retaining hydrocarbons,siltstone accumulating hydrocarbons and multiple intra-source migration paths”.In the Ordos Basin,the organic-lean(TOC 1–3%)mudstone/shale intervals appear to be the sweet spots of shale oil,where there are abundant medium-short-chain hydrocarbons retained with high flowability.After fracturing stimulation,their production conditions may be even superior to those of siltstones.This proposed idea changes the previous strategy to look for sweet spots in high-TOC intervals derived from the shale gas industry.Songqi PAN Qiulei GUO Caineng ZOU Zhenhua JING Ming YUAN Ying HE He ZHENG Ying MU Zhi YANG Shixiang LI Xinping ZHOU Songtao WU Hanlin LIU Feng YANG Yuanjia HAN 2023Science China Earth Sciences2023,66,7:1

16Optofluidic microring dye laser embedded in polydimenthylsiloxane with reduced threshold显示文摘We demonstrate an integrated optofluidic microring dye laser with a relatively low threshold on a polydimenthylsiloxane(PDMS) chip.The chip was fabricated through conventional soft lithography.It consists of a liquid waveguide with microring structure providing the feedback.A reduced threshold is realized due to the unique design of the bus waveguide across the center of the microring structure,which results in a great reduction in the cavity losses.Laser dye rhodamine 6G(R6G) dissolved in benzyl alcohol was injected into the microfluidic channel as the gain medium.When the dye laser was pumped with a pulsed laser at 532 nm,the dye laser oscillation was achieved with a threshold of only 4?5 μJ mm-2.The convenience in fabrication and operation makes the optofluidic microring dye laser a promising underlying photonic component in the integrated optofluidic systems.SHEN ZhenHua ZOU Yun CHEN XianFeng 2013Chinese Science Bulletin2013,58,7:1

19Accurate Assessment and Tracking the Process of Liver-Specific Injury by the Residual Tissue Activity of Carboxylesterase 1 and Dipeptidyl Peptidase 4显示文摘Accurately assessing and tracking the progression of liver-specific injury remains a major challenge in the field of biomarker research.Here,we took a retrospective validation approach built on the mutuality between serum and tissue biomarkers to characterize the liver-specific damage of bile duct cells caused by a-naphthyl isothiocyanate(ANIT).We found that carboxylesterase 1(CES1),as an intrahepatic marker,and dipeptidyl peptidase 4(DPP-IV),as an extrahepatic marker,can reflect the different pathophysiologies of liver injury.Levels of CES1 and DPP-IV can be used to identify liver damage itself and the inflammatory state,respectively.While the levels of the conventional serological biomarkers alkaline phosphatase(ALP),alanine aminotransferase(ALT),and aspartate aminotransferase(AST)were all concomitantly elevated in serum and tissues after ANIT-induced injury,the levels of bile acids decreased in bile,increased in serum,and ascended in intrahepatic tissue.Although the level of γ-glutamyl transpeptidase(γ-GT)changed in an opposite direction,the duration was much shorter than that of CES1 and was quickly restored to normal levels.Therefore,among the abovementioned biomarkers,only CES1 made it possible to specifically determine whether the liver cells were destroyed or damaged without interference from inflammation.CES1 also enabled accurate assessment of the anti-cholestasis effects of ursodeoxycholic acid(UDCA;single component)and Qing Fei Pai Du Decoction(QFPDD;multicomponent).We found that both QFPDD and UDCA attenuated ANIT-induced liver damage.UDCA was more potent in promoting bile excretion but showed relatively weaker anti-injury and antiinflammatory effects than QFPDD,whereas QFPDD was more effective in blocking liver inflammation and repairing liver damage.Our data highlights the potential of the combined use of CES1(as an intrahepatic marker of liver damage)and DPP-IV(as an extrahepatic marker of inflammation)for the accurate evaluation and tracking of liver-specific injury—an application that allows for the differentiation of liver damage and inflammatory liver injury.Qiusha Pan Peifang Song Zhenhua Ni Xingkai Qian Anqi Wang Liwei Zou Yong Liu Ping Wang Weidong Zhang Hong Ma Ling Yang 2022Engineering2022,,12:0
20Major biological events and fossil energy formation: On the development of energy science under the earth system framework显示文摘In geological history, one major life explosion and five mass extinction events occurred. These major biological and environmental events affected the evolution of the Earth ecosystem and controlled the formation of organic-rich strata. The life explosion occurred in Cambrian and the five mass extinction events happened at the end of Ordovician, Late Devonian, end of Permian, end of Triassic, and end of Cretaceous, respectively. They are corresponded to the formation of multiple suites of organic-rich strata globally, which are crucial to the formation, evolution and distribution of the fossil energy on Earth. China’s energy is characterized by 'unlimited new energy, rich in coal but insufficient in oil and gas ', and it is determined to rely on new energy sources to achieve ' energy independence ' and carbon neutrality. From the perspective of the Earth system evolution, we investigate the multiple relationships between energy and Earth, energy and environment, as well as energy and human beings, and carry out comprehensive research on energy. Energy science refers to the science of studying the various energy sources formation and distribution, evaluation and selection, production and utilization, orderly replacement, development prospects, etc. in temporal and spatial scales based on the evolution of the Earth system. The connotation of energy science includes three core contents:(1) The relationship between the Earth and energy, including the formation of energy in the Earth system and the feedback of energy consumption to the Earth’s climate and environment;(2) The relationship between the Earth environment and the human beings, including the Earth environment breeding human beings and human activities transforming the earth environment;(3) The relationship between the energy and the human beings, including the development of energy technology by human beings and the progress of human society driven by energy utilization. The energy science focuses on the formation and development of fossil energy, development and orderly replacement of new energy, exploration and utilization of energy in deep earth and deep space, and energy development strategy and planning.
