1恶性肿瘤骨转移及骨相关疾病临床诊疗中国专家共识(英文)显示文摘Since 1980s, palliative medicine has been an independent educational project. The development of palliative care in China started in the early 1990s. During the last 18 years, we have seen an increasing allocation of resources to palliative care, the establishment of an academic chair in palliative medicine, and more focused interest and awareness of the need for palliative care in the society at large. So the doctors in China have established principles of treatment on clinical diagnosis and treatment of malignant tumor bone metastases and bone related complications. In order to share our experience with the world, after two years’ preparation, the top-level experts in palliative medicine have written the consensus statement on clinical diagnosis and treatment of malignant tumor bone metastases and bone related complications. The overview includes not only the achievements in the world, but the hands-on experience in clinic, especially the experience in combination of TCM and Western medicine. 2Exploratory clinical study of chidamide,an oral subtype-selective histone deacetylase inhibitor,in combination with exemestane in hormone receptor-positive advanced breast cancer显示文摘Objective: The recurrence or progression under endocrine therapy in hormone receptor-positive(HR+)advanced breast cancer(ABC) remained a critical clinical challenge.Chidamide is an oral subtype-selective histone deacetylase(HDAC) inhibitor with multiple functions in tumor growth inhibition and microenvironment modulation via epigenetic reprogramming.The purpose of this study was to evaluate the safety,pharmacokinetics(PK),and preliminary efficacy of chidamide in combination with exemestane in HR+ ABC patients.Methods: Eligible patients were postmenopausal women with HR+ ABC recurrent or progressed to at least one endocrine therapy.Blood samples were obtained in the run-in period and the first day of combination treatment for PK analysis.In combination treatment,patients were given exemestane 25mg daily and chidamide 30mg twice a week(BIW) until progression of disease or intolerable toxicities.A treatment cycle was defined as 4 weeks.Safety,PK parameters,and preliminary efficacy were evaluated.Results: A total of 20 patients were enrolled between July and December,2015.The median number of treatments cycle was 5.2(20.8 weeks) with 2 patients still on treatment at the data cut-off date of October,2017.The treatment-related adverse events(AE) ≥ grade 3 in more than 2 patients were neutropenia(35%),thrombocytopenia(30%),and leucopenia(20%).The plasma exposure of exemestane was consistent in the presence or absence of chidamide.A slight increase in chidamide exposure was noted in the presence of exemestane,probably due to the inter-and intra-patient variations.The best response in 16 evaluable patients was assessed by Response Evaluation Criteria in Solid Tumors(RECIST),including 4 patients with partial response,10 patients with stable disease.The median progression-free survival(PFS) was 7.6 months.Conclusions: The combination of chidamide with exemestane was generally well tolerated with promising preliminary efficacy in HR+ ABC patients.The overall results from this study encourage further pivotal trial in this patient population.Qingyuan Zhang Tao Wang Cuizhi Geng Yue Zhang Jinwen Zhang Zhiqiang Ning Zefei Jiang 2018Chinese Journal of Cancer Research2018,30,6:11

4LSECtin on tumor-associated macrophages enhances breast cancer sternness via interaction with its receptor BTN3A3显示文摘Macrophages have bee n suggested to con tribute to con structing a cancer stem cell(CSC)niche.However,whether and how macrophages regulate the activity of CSCs through juxtacrine signaling are poorly understood.Here we report LSECtin,a transmembrane protein highly expressed on tumor-associated macrophages(TAMs),enhances sternness of breast cancer cells(BCCs).We identified BTN3A3,a B7 family member with previously unknown functions as the receptor for LSECtin on BCCs responsible for stemness-promoting effect of LSECtin.In mice bearing human tumor xenografts,either macrophage-specific ablation of LSECtin or silencing of BTN3A3 in BCCs decreased CSC frequency and tumor growth.Admixture of LSECtin-positive macrophages increased the tumorigenic activity of BCCs dependent on BTN3A3.Disruption of the LSECtin-BTN3A3 axis with BTN3A3-Fc or anti-BTN3A3 mAb has a therapeutic effect on breast cancer.These findings define a juxtacrine signaling mechanism by which TAMs promote cancer stemness.Targeting this axis in the CSC niche may provide potential therapies to breast cancer.Di Liu Qian Lu Xing Wang Jun Wang Ning Lu Zefei Jiang Xiaopeng Hao Jianbin Li Jing Liu Pengbo Cao Guilin Peng Yuandong Tao Dianyuan Zhao Fuchu He Li Tang 2019Cell Research2019,29,5:8

6Treatment patterns for adjuvant docetaxel-based chemotherapy in early-stage breast cancer in China:A pooled retrospective analysis of four observational studies显示文摘Objective:Adjuvant docetaxel-based chemotherapy is frequently used for operable early breast cancer(EBC).This study investigated patterns of use of docetaxel(T)in real-life clinical practice in China.Methods:This was a retrospective pooled analysis of the Asia-Pacific Breast Initiatives(APBI)Ⅰ(2006–2008)and Ⅱ(2009–2011)registries,and two Chinese observational studies;BC STATE(2011–2014)and BC Local Registry(2007–2010).Female Chinese adults(≥18 years)with operable breast cancer treated with docetaxel-based adjuvant chemotherapy were included in the analysis.Patients with metastatic disease were excluded.The primary endpoint was assessment of treatment patterns and patient profiles.A logistic regression analysis was conducted to identify factors associated with choice of adjuvant chemotherapy regimen.Results:Data from 3,020 patients were included.The most frequently used adjuvant regimen was docetaxel/anthracycline combination[n=1,421(47.1%);of whom 52.0%received T/epirubicin(E)/cyclophosphamide(C)],followed by docetaxel/other[n=705(23.3%);of whom 72.8%received TC],docetaxel/anthracycline sequential[n=447(14.8%);of whom 40.9%and 39.6%received 5-Fu/EC-T and EC-T,respectively],and'other'[n=447(14.8%);of whom 91.5%received T].A significant association was found between adjuvant therapy with docetaxel/anthracycline combination and patient weight,menopausal status and estrogen receptor status.Conclusions:Real-world data revealed that docetaxel/anthracycline combination is the most commonly used category of docetaxel-based adjuvant therapy for patients with operable breast cancer in China;of which TEC is the most frequently used regimen.Binghe Xu Zhimin Shao Shui Wang Zefei Jiang Xichun Hu Xiaohua Zhang Xiru Li Jinping Liu Mengquan Li Shu Wang 2018Chinese Journal of Cancer Research2018,30,3:5

8Adsorption of BSA on Carbon-coated Fe_3O_4 Microspheres Activated with 1-ethyl-3-(3-dimethylaminopropyl)-Carbodiimide Hydrochloride显示文摘Carbon-coated Fe_3O_4( Fe_3O_4/C) microspheres activated with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride(EDC) were prepared, characterized and applied to adsorb bovine serum albumin(BSA). The prepared magnetic microspheres had spherical core-shell structure with a uniform and continuous carbon coating coupled with activation by EDC, and possessed superparamagnetic characteristics. The experimental results showed that the adsorption amount of BSA on the EDC-activated Fe_3O_4/C(Fe_3O_4/C-EDC) microspheres was higher than that on the Fe_3O_4/C microspheres. The maximum adsorption of BSA on Fe_3O_4/C-EDC microspheres occurred at pH 4.7, which was the isoelectric point of BSA. At low concentrations(below 1.0 M), salt had no noticeable effect on BSA adsorption. The BSA adsorption of Fe_3O_4/C-EDC microspheres had a better fit to the Langmuir model than the Freundlich isotherm and Temkin isotherm model, and the kinetic data were well described by the pseudo-second-order model. The adsorption equilibrium could be reached within 20 min. 9Effectiveness of second-line anti-HER2 treatment in HER2-positive metastatic breast cancer patients previously treated with trastuzumab:A real-world study显示文摘Objective:Several studies have demonstrated different benefits for patients whose disease progressed despite previous trastuzumab treatment.Due to limited real-world data,we evaluate the effectiveness of anti-human epidermal growth factor receptor 2(HER2)therapy(lapatinib or trastuzumab)plus chemotherapy or chemotherapy alone in patients who were previously treated with trastuzumab-containing regimens and investigate factors associated with effectiveness.And we further show the effectiveness of the two anti-HER2 therapy groups.Methods:A total of 342 HER2-positive metastatic breast cancer(MBC)patients whose disease progressed during prior anti-HER2(trastuzumab)and standard chemotherapy therapy from Department of Breast Oncology,the Fifth Medical Center of Chinese PLA General Hospital,from August 2010 to December 2016 were included.Seventy-eight patients received standard chemotherapy only,148 patients continued to receive trastuzumab and switched to other chemotherapy drugs,and 116 patients received tyrosine-kinase inhibitors(TKIs;lapatinib)and chemotherapy.The main outcome measures were progression-free survival(PFS),overall response rate(ORR),and clinical benefit rate(CBR).Subgroup analyses were conducted to identify patient characteristics associated with the greatest clinical benefit.Results:After a median follow-up of 26.2(range,2.0-56.0)months,PFS significantly improved with anti-HER2 therapy compared with chemotherapy alone:median 6.0 months with lapatinib[95%confidence interval(95%CI),4.53-7.47],4.5 months with trastuzumab(95%CI,3.99-5.01)vs.3.0 months with chemotherapy alone(95%CI,2.42-3.58);stratified hazard ratio(HR)=0.70,95%CI,0.60-0.81;P<0.0001.The ORR values were 33.6%,25.0%and 12.8%,respectively,the CBR values were 60.3%,48.6%and 26.9%,respectively.The effectiveness of lapatinib group and trastuzumab group were further analyzed.In multivariate analysis,lapatinib group was associated with a longer PFS,after controlling other potential confounders(HR=0.68,95%CI,0.52-0.90;P=0.006).Conclusions:The combination of TKIs and chemotherapy was effective in this cohort previously treated with trastuzumab treatment.Therefore,TKIs combined with chemotherapy is an option for Chinese HER2-positive MBC patients previously treated with trastuzumab treatment.Wei Zhao Li Bian Tao Wang Shaohua Zhang Jianbin Li Fengrui Xu Zefei Jiang 2020Chinese Journal of Cancer Research2020,32,3:1

13Improved Fatigue and Damage Tolerant Material Design for Aerospace Industry显示文摘Various micro-mechanical and micro-structural influences on fatigue crack growth resistance of the material havebeen investigated over the years. It is widely recognized that resistance to fatigue crack growth can be differentiatedinto ’intrinsic’ and ’extrinsic’. The separation of intrinsic and extrinsic crack growth resistance has constituted amajor theme of fatigue research in the last 30 years, with the concept of crack closure or crack tip shielding beingused to rationalize a wide range of micro-structural and mechanical influences on fatigue crack growth behavior. Anaccurately quantitative understanding of intrinsic and extrinsic effects on crack growth is essential to directed alloydesign for improved fatigue resistance, and/or improved structural service life. 14Underwater Image Classification Based on EfficientnetB0 and Two-Hidden-Layer Random Vector Functional Link显示文摘The ocean plays an important role in maintaining the equilibrium of Earth’s ecology and providing humans access to a wealth of resources.To obtain a high-precision underwater image classification model,we propose a classification model that combines an EfficientnetB0 neural network and a two-hidden-layer random vector functional link network(EfficientnetB0-TRVFL).The features of underwater images were extracted using the EfficientnetB0 neural network pretrained via ImageNet,and a new fully connected layer was trained on the underwater image dataset using the transfer learning method.Transfer learning ensures the initial performance of the network and helps in the development of a high-precision classification model.Subsequently,a TRVFL was proposed to improve the classification property of the model.Net construction of the two hidden layers exhibited a high accuracy when the same hidden layer nodes were used.The parameters of the second hidden layer were obtained using a novel calculation method,which reduced the outcome error to improve the performance instability caused by the random generation of parameters of RVFL.Finally,the TRVFL classifier was used to classify features and obtain classification results.The proposed EfficientnetB0-TRVFL classification model achieved 87.28%,74.06%,and 99.59%accuracy on the MLC2008,MLC2009,and Fish-gres datasets,respectively.The best convolutional neural networks and existing methods were stacked up through box plots and Kolmogorov-Smirnov tests,respectively.The increases imply improved systematization properties in underwater image classification tasks.The image classification model offers important performance advantages and better stability compared with existing methods.ZHOU Zhiyu LIU Mingxuan JI Haodong WANG Yaming ZHU Zefei 2024Journal of Ocean University of China2024,23,2:0
15Giant nonlinear Hall effect in twisted bilayer WSe_(2)显示文摘The recently discovered nonlinear Hall effect(NHE)in a few non-interacting systems provides a novel mechanism for generating second-harmonic electrical Hall signals under time-reversal-symmetric conditions.Here,we introduce a new approach to engineering an NHE by using twisted moiréstructures.We found that the twisted WSe_(2)bilayer exhibited an NHE when the Fermi level was tuned to the moiréflat bands.When the first moiréband was half-filled,the nonlinear Hall signal exhibited a sharp peak with a generation efficiency that was at least two orders of magnitude greater than those obtained in previous experiments.We discuss the possible origins of the diverging generation efficiency in twisted WSe_(2)based on resistivity measurements,such as moiré-interface-induced correlation effects and mass-diverging-type continuous Mot transition.This study demonstrates not only how interaction effects can combine with Berry curvature dipoles to produce novel quantum phenomena,but also the potential of NHE measurements as a new tool for studying quantum criticality.Meizhen Huang Zefei Wu Jinxin Hu Xiangbin Cai En Li Liheng An Xuemeng Feng Ziqing Ye Nian Lin Kam Tuen Law Ning Wang 2023National Science Review2023,10,4:0
16原癌基因Fra-1在乳腺恶性肿瘤组织中的表达(英文)显示文摘Objective:Invasion and metastasis are the most significant and intrinsic biological characteristics of cancers,also which are main factors of malignant tumor causing treatment failure and death.Recent studies have found that Fra-1 plays an important role on cell migration,invasion,and maintaining malignant phenotype of transformed cells.But there are few studies about the expression and location of Fra-1 in breast tissues and cells being reported.This study just aims to discuss the expression and location of transcription factor Fra-1 in benign and malignant human breast tissues.Methods:The expression of Fra-1 was investigated by immunohistochemistry in neoplastic breast diseases ranging from benign fibroadenoma to very aggressive undifferentiated carcinoma.The correlations of Fra-1 expression with other indicators of breast carcinoma prognosis (ER,PR and ErbB2 receptors) were analyzed.Results:All neoplastic breast tissues,either benign or malignant breast tissues,were nuclear immunoreactive for Fra-1-recognizing antibody.In 85% of benign tumors (17/20),the immunoreactive for Fra-1-recognizing antibody as exclusively restricted to the nuclei.In three cases (3/20,15%),focal unequivocal cytoplasmic staining was also exhibited.Strong positive nuclear staining for Fra-1 was easily seen in all types of breast carcinomas.However the nuclear/cytoplasmic concomitant immunoreactivity was observed in all types of breast carcinomas.A clear shift in Fra-1 immunoreactivity,from an exclusively nuclear to a simultaneous nuclear and cytoplasmic localization was noticed in 90.2% (37/41) of breast carcinomas.No inverse relationship between Fra-1 and ER and PR protein levels was noticed in malignant tumors.The relative expression level of Fra-1 was not correlated with the expression of ErbB2.Conclusion:The overall expression,pattern and intensity of Fra-1 proteins were correlated with breast oncogenesis.Overexpression of Fra-1,leading to a persistent high cytoplasmic accumulation,may play a role in the process of breast carcinogenesis.Yuhua Song Jing Wang Xiaoyun Yu Santai Song Zefei Jiang 2012The Chinese-German Journal of Clinical Oncology2012,11,6:0
17Large Eddy Simulation of Micro-Particles Transport with Different Mass Flow Rate in Turbulent Planar Jet Flow显示文摘The motion of micro-particles with different mass flow rate in the planer turbulent jet flow has been simulated,using LES method to obtain the flow vorticity evolution and Lagrangian method to track micro-particles.The results showed that when the flow rate is small,the particles more likely to present in the vortex periphery,the distribution pattern is similar to the flow pattern.When the flow rate is high,some particles will escape from the motion region to the original static region,so that in the jet region,particles are relatively evenly distributed.When the flow field is full developed,the particles average concentration in the y direction affected by the mass flow rate relative slightly,the normalized mean particles concentrations at different flow rate were similar to Gaussian shape.LIN Peifeng WU Dichong ZHU Zefei 2012Journal of Thermal Science2012,21,6:0

20Asymptotic analysis of nonlinear micro-film buckling显示文摘The buckling design of micro-films has various potential applications to engineering.The substrate prestrain,interconnector buckling amplitude and critical strain are important parameters for the buckling design.In the presented analysis,the buckled film shape was described approximately by a trigonometric function and the buckled film amplitude was obtained by minimizing the total strain energy.However,this method only generates the first-order approximate solution for the nonlinear buckling.In the present paper,an asymptotic analysis based on the rigorous nonlinear differential equation for the buckled micro-film deformations is proposed to obtain more accurate relationship of the buckling amplitude and critical strain to prestrain.The obtained results reveal the nonlinear relation and are significant to accurate buckling design of micro-films.YING ZuGuang WANG Yong ZHU ZeFei 2012Science China(Technological Sciences)2012,55,7:0
