2RBD-Fc-based COVID-19 vaccine candidate induces highly potent SARS-CoV-2 neutralizing antibody response显示文摘The pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has posed serious threats to global health and economy,thus calling for the development of safe and effective vaccines.The receptorbinding domain(RBD)in the spike protein of SARS-CoV-2 is responsible for its binding to angiotensin-converting enzyme 2(ACE2)receptor.It contains multiple dominant neutralizing epitopes and serves as an important antigen for the development of COVID-19 vaccines.Here,we showed that immunization of mice with a candidate subunit vaccine consisting of SARS-CoV-2 RBD and Fc fragment of human IgG,as an immunopotentiator,elicited high titer of RBD-specific antibodies with robust neutralizing activity against both pseudotyped and live SARS-CoV-2 infections.The mouse antisera could also effectively neutralize infection by pseudotyped SARS-CoV-2 with several natural mutations in RBD and the IgG extracted from the mouse antisera could also show neutralization against pseudotyped SARS-CoV and SARS-related coronavirus(SARSr-CoV).Vaccination of human ACE2 transgenic mice with RBD-Fc could effectively protect mice from the SARS-CoV-2 challenge.These results suggest that SARS-CoV-2 RBD-Fc has good potential to be further developed as an effective and broad-spectrum vaccine to prevent infection of the current SARS-CoV-2 and its mutants,as well as future emerging SARSr-CoVs and re-emerging SARS-CoV.Zezhong Liu Wei Xu Shuai Xia Chenjian Gu Xinling Wang Qian Wang Jie Zhou Yanling Wu Xia Cai Di Qu Tianlei Ying Youhua Xie Lu Lu Zhenghong Yuan Shibo Jiang 2020Signal Transduction and Targeted Therapy2020,5,1:7

4Altered Motor-Striatal Plasticity and Cortical Functioning in Patients with Schizophrenia显示文摘Patients with schizophrenia undergo changes in brain plasticity. In the present study, we characterized motor cortical-striatal plasticity in such patients. Compared with the potentiation following high-frequency repetitive transcranial magnetic stimulation in the control group, the patients demonstrated impaired plasticity of corticostriatal motor-evoked potentials recorded from hand muscles.Notably, the loss of cortical plasticity was correlated with impaired motor learning in a rotary pursuit task. Moreover,the loss of plasticity was correlated with the symptoms of schizophrenia. 5A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice,rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines.Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge,even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases.Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu 2022Cell Research2022,32,3:5
6A highly potent and stable pan-coronavirus fusion inhibitor as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases显示文摘The development of broad-spectrum antivirals against human coronaviruses(HCoVs)is critical to combat the current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and its variants,as well as future outbreaks of emerging CoVs.We have previously identified a polyethylene glycol-conjugated(PEGylated)lipopeptide,EK1C4,with potent pan-CoV fusion inhibitory activity.However,PEG linkers in peptide or protein drugs may reduce stability or induce anti-PEG antibodies in vivo.Therefore,we herein report the design and synthesis of a series of dePEGylated lipopeptide-based pan-CoV fusion inhibitors featuring the replacement of the PEG linker with amino acids in the heptad repeat 2 C-terminal fragment(HR2-CF)of HCoV-OC43.Among these lipopeptides,EKL1C showed the most potent inhibitory activity against infection by SARS-CoV-2 and its spike(S)mutants,as well as other HCoVs and some bat SARS-related coronaviruses(SARSr-CoVs)tested.The dePEGylated lipopeptide EKL1C exhibited significantly stronger resistance to proteolytic enzymes,better metabolic stability in mouse serum,higher thermostability than the PEGylated lipopeptide EK1C4,suggesting that EKL1C could be further developed as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases.Jie Zhou Wei Xu Zezhong Liu Chao Wang Shuai Xia Qiaoshuai Lan Yanxing Cai Shan Su Jing Pu Lixiao Xing Youhua Xie Lu Lu Shibo Jiang Qian Wang 2022Acta Pharmaceutica Sinica B2022,12,4:5

9Nitrogen partitioning and microbial protein synthesis in lactating dairy cows with different phenotypic residual feed intake显示文摘Background: Residual feed intake(RFI) is an inheritable measure of feed efficiency that is independent on level of production. However, physiological and metabolic mechanisms underlying divergent RFI are not fully elucidated.This study was conducted to investigate dietary nitrogen(N) partitioning and microbial protein synthesis in lactating dairy cows divergent in phenotypic RFI.Results: Thirty Holstein dairy cows(milk yield = 35.3 ± 4.71 kg/d;milk protein yield = 1.18 ± 0.13 kg/d;mean ± standard deviation) were selected for the experiment to derive RFI. After the RFI measurement period of 50 d, the 10 lowest RFI cows and 8 highest RFI cows were selected. The low RFI cows had lower dry matter intake(DMI, P < 0.05) than the high RFI cows, but they produced similar energy-corrected milk. The ratios of milk to DMI(1.41 vs. 1.24, P < 0.01) and energy-corrected milk to DMI(1.48 vs. 1.36, P < 0.01) were greater in low RFI cows than those in the high RFI cows. The low RFI cows had lower milk urea nitrogen than that in the high RFI cows(P = 0.05). Apparent digestibility of nutrients did not differ between two groups(P > 0.10). Compared with high RFI animals, the low RFI cows had a lower retention of N(5.72 vs. 51.4 g/d, P < 0.05) and a higher partition of feed N to milk N(29.7% vs. 26.5%, P < 0.05).Conclusions: The results suggest that differences in N partition, synthesis of microbial protein, and utilization of metabolizable protein could be part of the mechanisms associated with variance in the RFI.Yunyi Xie Zezhong Wu Diming Wang Jianxin Liu 2019Journal of Animal Science and Biotechnology2019,10,4:4

15A fast algorithm for calculating surface electric field of converter valve shield system by multipole boundary element method显示文摘The surface electric field analysis of the converter valve shield system is a large-scale electrostatic field problem, which is difficult to analyse. The fast multipole boundary element method(FMBEM), which is suitable for solving large-scale problems,can accelerate the computation speed and conserve memory. However, the coefficient matrix implicitly formed by using the FMBEM is sometimes ill-conditioned, especially for large-scale problems; thus, the convergence of iteration is poor. In this paper, a fast solver is proposed to improve efficiency. First, an adaptive GMRES(m) with variant restart parameter is adjusted for the Galerkin FMBEM. In addition, the sparse approximate inverse preconditioner is improved, and a new sparsity pattern is proposed for the multiscale problem derived from the converter valve shield system. The numerical results show that the accuracy can meet the engineering requirements compared with the finite element method. Compared with other solvers and preconditioners, the algorithm can achieve a satisfactory convergence rate and reduce the computation time. 16Classical swine fever virus NS5A protein antagonizes innate immune response by inhibiting the NF-κB signaling显示文摘The NS5A non-structural protein of classical swine fever virus(CSFV)is a multifunctional protein involved in viral genomic replication,protein translation,assembly of infectious virus particles,and regulation of cellular signaling pathways.Previous report showed that NS5A inhibited nuclear factor kappa B(NF-κB)signaling induced by poly(I:C);however,the mechanism involved has not been elucidated.Here,we reported that NS5A directly interacted with NF-κB essential modulator(NEMO),a regulatory subunit of the IκB kinase(IKK)complex,to inhibit the NF-κB signaling pathway.Further investigations showed that the zinc finger domain of NEMO and the aa 126–250 segment of NS5A are essential for the interaction between NEMO and NS5A.Mechanistic analysis revealed that NS5A mediated the proteasomal degradation of NEMO.Ubiquitination assay showed that NS5A induced the K27-linked but not the K48-linked polyubiquitination of NEMO for proteasomal degradation.In addition,NS5A blocked the K63-linked polyubiquitination of NEMO,thus inhibiting IKK phosphorylation,IκBαdegradation,and NF-κB activation.These findings revealed a novel mechanism by which CSFV inhibits host innate immunity,which might guide the drug design against CSFV in the future.Jinfu Sun Jiaying Li Liming Li Haixiao Yu Ping Ma Yingnan Wang Jinqi Zhu Zezhong Feng Changchun Tu 2023Virologica Sinica2023,38,6:0
17Regulating Li transport in Li-magnesium alloy for dendrite free Li metal anode显示文摘Li metal has become a strong candidate for anode due to its high theoretical specific capacity and lowest electrochemical potential.However,the poor reversibility caused by continuous chemical and electrochemical degradation hinders the practical application of Li metal.Solid-solution-based metal alloy phases have been proposed as hosts for regulating the non-dendrite electrodeposition,but the fundamental understanding remains unclear due to the drastically different deposition behaviors of Li on them.Here we found the difference in the diffusion coefficient of Li atoms on solid-solution-based metal alloy phases(Li-Mg and Li-Ag alloys)was a major contributor to the different deposition behaviors.The low Li atom diffusion coefficient of Li-Mg alloy showed a preferential Li accumulation on the upper surface rather than the inward-growth plating of Li atoms into alloy foil in Li-Ag alloy.By the process of secondary recrystallization,we improved the diffusion coefficient of Li atoms in Li-Mg alloy that facilitates the inward transfer rather than surface plating of Li atoms.In this case,the recrystallized Li-Mg alloy underwent a solidsolution phase change in the delithiation-lithiation cycles which yielded a high Coulombic efficiency of 99.3%with a reversible gravimetric capacity of 2,874 mAh·g−1 and superior cycling stability over 5,000 h without dendrite growth.Jinxi Wang Yadong Ye Hongmin Zhou Wei Zhang Zhaowei Sun Jiawen Zhu Hongchang Jin Huanyu Xie Haoliang Huang Yi Cui Rong Huang Zezhong Li Song Jin Hengxing Ji 2023Nano Research2023,16,6:0
18Simplified parasitic capacitance extraction of shield in HVDC converter system with boundary element method显示文摘It is critical to build a wide-band circuit model to conduct research on the characteristics of the electromagnetic disturbance source during the localization of high voltage direct current(HVDC)technology.Parasitic capacitance is most essential for modeling the equivalent circuit,so a fast and accurate computation of capacitance parameters plays a vital role.Because of the large size and complex structure of the converter equipment,it is impossible to obtain capacitance parameters by means of measurement or simulating calculation with finite element software.In this paper,a simplified method of capacitance extraction based on boundary element method is proposed,which can provide an efficient means of establishing simulation models.In the method presented,simulation model of the shield may not be chamfered.Consequently,the edge and corner of the shield do not need to be handled with a sphere,cylinder and other curved surface model.The availability of this method is demonstrated by comparing the capacitance parameters of chamfered shield with that of non-chamfered shield.Shili LIU Zezhong WANG Jing SUN 2010Frontiers of Electrical and Electronic Engineering in China2010,5,4:0
