| 1 | Evolution of Sedimentation and Tectonics of the Youjiang Composite Basin, South China显示文摘Located at the southern margin of the South China plate, the Youjiang basin is a closely related to the NW- and NE-trending syndepositional faults in respect to the configuration and structure of the basin. The evolution of the Youjiang basin progressed through two stages. In the Hercynian period, the opening of the Ailaoshan-Honghe ocean basin gave rise to a number of NW-trending rift belts in the Youjiang area. During this period, deep-water sediments were dominant and the basin was possesed of the characteristics of the rift system of passive continental margins. In the early Indosinian after the Dongwu movement, the circum - Pacific tectonism led to a major change in the configuration and structure of the basin. In the meantime, the Ailaoshan ocean basin began to be subducted towards the northeast, thus causing the basin to be split and expand again, and then the basin developed into the stage of the back -arc basin. | 3 | Microdisk lasers on an erbium-doped lithium-niobite chip显示文摘Lithium niobate on insulator(LNOI) provides a platform for the fundamental physics investigations and practical applications of integrated photonics. However, as an indispensable building block of integrated photonics, lasers are in short supply. In this paper, erbium-doped LNOI laser in the 1550-nm band was demonstrated in microdisk cavities with high quality factors fabricated in batches by UV exposure, inductively coupled plasma reactive ion etching, and chemomechanical polishing. The threshold and conversion efficiency of the erbium-doped LNOI microdisk laser were measured to be lower than 1 m W and 6.5×10^(-5)%, respectively. | 4 | Genetic and Molecular Characterization of H9N2 Avian Influenza Viruses Isolated from Live Poultry Markets in Hubei Province, Central China, 2013–2017显示文摘H9N2 subtype avian influenza virus(AIV)is an influenza A virus that is widely spread throughout Asia,where it jeopardizes the poultry industry and provides genetic material for emerging human pathogens.To better understand the epidemicity and genetics of H9 subtype AIVs,we conducted active surveillance in live poultry markets(LPMs)in Hubei Province from 2013 to 2017.A total of 4798 samples were collected from apparent healthy poultry and environment.Realtime RT-PCR revealed that the positivity rate of influenza A was 26.6%(1275/4798),of which the H9 subtype accounted for 50.3%(641/1275)of the positive samples.Of the 132 H9N2 viral strains isolated,48 representative strains were subjected to evolutionary analysis and genotyping.Phylogenetic analysis revealed that all H9N2 viral genes had 91.1%–100%nucleotide homology,clustered with genotype 57,and had high homology with human H9N2 viruses isolated from2013 to 2017 in China.Using a nucleotide divergence cutoff of 95%,we identified ten distinct H9N2 genotypes that continued to change over time.Molecular analysis demonstrated that six H9N2 isolates had additional potential glycosylation sites at position 218 in the hemagglutinin protein,and all isolates had I155 T and Q226 L mutations.Moreover,44 strains had A558 V mutations in the PB2 protein and four had E627 V mutations,along with H9N2 human infection strains A/Beijing/1/2016 and A/Beijing/1/2017.These results emphasize that the H9N2 influenza virus in Hubei continues to mutate and undergo mammalian adaptation changes,indicating the necessity of strengthening the surveillance of the AIV H9N2 subtype in LPMs. | Zhibin Hu Fuhu Peng Zhenghui Xiong Wanpo Zhang Tingting Li Yuejun Shi Jun Xie Xin Jin Jingjing Huang Hongde Xiao Dingren Bi Nianhua Song Zili Li | 2021 | Virologica Sinica2021,36,2: | 4 |
| 5 | The impact of genomic relatedness between populations on the genomic estimated breeding values显示文摘In genomic selection, prediction accuracy is highly driven by the size of animals in the reference population(RP).Combining related populations from different countries and regions or using a related population with large size of RP has been considered to be viable strategies in cattle breeding. The genetic relationship between related populations is important for improving the genomic predictive ability. In this study, we used 122 French bulls as test individuals. The genomic estimated breeding values(GEBVs) evaluated using French RP, America RP and Chinese RP were compared.The results showed that the GEBVs were in higher concordance using French RP and American RP compared with using Chinese population. The persistence analysis, kinship analysis and the principal component analysis(PCA) were performed for 270 French bulls, 270 American bulls and 270 Chinese bulls to interpret the results. All the analyses illustrated that the genetic relationship between French bulls and American bulls was closer compared with Chinese bulls. Another reason could be the size of RP in China was smaller than the other two RPs. | 12 | Analysis of nucleosome positioning in promoters of miRNA genes and protein-coding genes显示文摘Nucleosome positioning in promoters is important for gene transcription regulation. In this paper, with a nucleosome prediction model, curvature profile, the characteristics of nucleosome positioning in promoters are analyzed for miRNA genes and protein-coding genes. In the vicinity of transcription start site (TSS), there is a nucleosome-free region (NFR) followed by a positioned nucleosome at ~200 bp downstream of TSS. A similar characteristic is observed in independent intronic promoters and intergenic promoters, namely, both types of promoters have a longer NFR in 0—-400 bp upstream of TSS. Moreover, transcription factor binding sites (TFBSs) locate in the NFR with a high concentration. However, nucleosome pattern in dependent intronic promoters are like that in protein-coding promoters, with two nucleosomes positioned at -200—-400 bp and -400—-600 bp upstream of TSS. | 14 | Enhanced protective immunity against SARS-CoV-2 elicited by a VSV vector expressing a chimeric spike protein显示文摘SARS-CoV-2 and SARS-CoV are genetically related coronavirus and share the same cellular receptor ACE2.By replacing the VSV glycoprotein with the spikes(S)of SARS-CoV-2 and SARS-CoV,we generated two replication-competent recombinant viruses,rVSVSARS-CoV-2 and rVSV-SARS-CoV.Using wild-type and human ACE2(hACE2)knock-in mouse models,we found a single dose of rVSV-SARS-CoV could elicit strong humoral immune response via both intranasal(i.n.)and intramuscular(i.m.)routes.Despite the high genetic similarity between SARS-CoV-2 and SARS-CoV,no obvious cross-neutralizing activity was observed in the immunized mice sera.In macaques,neutralizing antibody(NAb)titers induced by one i.n.dose of rVSV-SARS-CoV-2 were eight-fold higher than those by a single i.m.dose.Thus,our data indicates that rVSV-SARS-CoV-2 might be suitable for i.n.administration instead of the traditional i.m.immunization in human.Because rVSV-SARS-CoV elicited significantly stronger NAb responses than rVSV-SARS-CoV2 in a route-independent manner,we generated a chimeric antigen by replacing the receptor binding domain(RBD)of SARS-CoV S with that from the SARS-CoV-2.rVSV expressing the chimera(rVSV-SARS-CoV/2-RBD)induced significantly increased NAbs against SARS-CoV-2 in mice and macaques than rVSV-SARS-CoV-2,with a safe Th1-biased response.Serum immunized with rVSV-SARS-CoV/2-RBD showed no cross-reactivity with SARS-CoV.hACE2 mice receiving a single i.m.dose of either rVSV-SARS-CoV-2 or rVSV-SARSCoV/2-RBD were fully protected against SARS-CoV-2 challenge without obvious lesions in the lungs.Our results suggest that transplantation of SARS-CoV-2 RBD into the S protein of SARS-CoV might be a promising antigen design for COVID-19 vaccines. | Hongyue Li Yuhang Zhang Dong Li Yong-Qiang Deng Hongde Xu Chaoyue Zhao Jiandong Liu Dan Wen Jianguo Zhao Yongchun Li Yong Wu Shujun Liu Jiankai Liu Junfeng Hao Fei Yuan Shuguang Duo Cheng-Feng Qin Aihua Zheng | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 1 |
