| 8 | Generation of Recombinant Equine Influenza Vaccine Candidate RgH3N1 Virus by Reverse Genetics显示文摘The antigenic variation of influenza A virus hemagglutinin (HA) glycoproteins requires frequent changes in vaccine formulation. The new strategy of creating influenza seed strains for vaccine production is to generate 7 + 1 reassortants that contain seven genes from a high-yield virus A/Puerto Rico/8/34[A/PR/8/34](H1N1) and the HA gene from the circulating strains. By using this DNA-based cotransfection technique, we generated 7 + 1 reassortants rgH3N1 which had the antigenic determinants of influenza virus A/Songbird/HongKong/102/00[SB/HK/01](H3N8) and 7 other genes from A/PR/ 8/34. The hemagglutinin of A/Songbird/HongKong/102/00 is 96.3% homologous to that of A/Equine/Jilin/98[Eq/Jl/89] (H3N8). The resulting virus rgH3N1 grows to high HA titers in chicken embryonated eggs, allowing vaccine preparation in unconcentrated allantoic fluid. The rgH3N1 is stable after multiple passages in embryonated eggs. | 15 | Effects of Soil Types and Moisture Contents on Emergence Rate of Contarinia morulae Jiang显示文摘In order to explore the relationship between soil moisture content and the severity of occurrence of Contarinia morulae Jiang,the effects of different soil types and moisture contents on the emergence rate of C. morulae were investigated. The results showed that soil moisture contents had a significant effect on the emergence rate of C. morulae. When the soil moisture content was 10%,the emergence rate of C. morulae was the highest in both soil types. When the soil moisture content was too high or too low,the emergence rate of C. morulae was significantly reduced. Within the suitable water content range,the emergence rate of C. morulae in sandy soil was significantly higher than that in sandy loam soil. The trend of emergence of C. morulae in the two soil types was basically the same. | 18 | Osteogenesis Capacity of a Novel BMP/a-TCP Bioactive Composite Bone Cement显示文摘To improve the osteogenesis ability of α-trkalcium phosphate (a-TCP ) bone cement, a novel BMP/ a- TCP composite bone cement was prepared. By measuring the setting time and compressive strength, the hydration characteristic of bone cemtwnt icts eralnated. Animal experiments including histological observation, radiographic investigation as well as digital image analyses reveal the difference of osteogenesis ability among BMP, a TCP bone cetnent and BMPI a-TCP composite bone cement, Results show that a-TCP bone cement possesses excellent hydration and setting properties as well as high mechanical property, Comparison experiments show that BMPI a-TCP composite bonc cement has a stronger osteogenesis obility. The gross observation of the implant site does not exhibit any inflammation or necrosis. Histohwcal analyses reveal that the material has good osteointegration with host bone, and new bone formntion is detected within the materials, which are degrading. Strong osteogenesis ability of the composite is due to not only the excellent osteoconductive potential but also the osteoinductive potential contributed by attire BMP releasing and the material degradation . Large skull defect could be well-healed ty filling BMPl a-TCP composite bone cement.
