1The MDM2-p53 pathway revisited显示文摘The p53 tumor suppressor is a key transcription factor regulating cellular pathways such as DNA repair, cell cycle, apoptosis, angiogenesis, and senescence. It acts as an important defense mechanism against cancer onset and progression, and is negatively regulated by interaction with the oncoprotein MDM2. In human cancers, the TP53 gene is frequently mutated or deleted, or the wild-type p53 function is inhibited by high levels of MDM2, leading to downregulation of tumor suppressive p53 pathways. Thus, the inhibition of MDM2-p53 interaction presents an appealing therapeutic strategy for the treatment of cancer. However, recent studies have revealed the MDM2-p53 interaction to be more complex involving multiple levels of regulation by numerous cellular proteins and epigenetic mechanisms, making it imperative to reexamine this intricate interplay from a holistic viewpoint. 2Population and genetic diversity of Tibetan red deer based on fecal DNA显示文摘The Tibet Shannan Red Deer Nature Reserve is the center of the geographic distribution of Tibetan red deer. This paper presents the results of DNA analysis of 199 red deer fecal pellet groups collected during 2013 and 2014 during the green-plant period. We successfully extracted DNA from 87 DNA pellet groups and determined individual identification with 12 microsatellite loci. We evaluated the genetic diversity of the population and various population estimates with Capwire in R software. The87 successfully extracted pellet groups were from 50 individuals. In the population, the average number of alleles was 7.58 ± 0.18, the average effective number of alleles was 4.58 ± 0.15, and average polymorphism information content was 0.67 ± 0.01. Among the 12 loci,only T123 was moderately polymorphic; the other 11 loci were highly polymorphic. Expected heterozygosity ranged from 0.45 to 0.91, with an average of 0.72 ± 0.01, and average observed heterozygosity was 0.52 ± 0.11.Although Tibetan red deer remains endangered, the high genetic diversity indicates that this population has a good chance of recovery. 3Thermal Performance and Mechanics Characteristic for Double Layer Microchannel Heat Sink显示文摘Double layer micro-channel heat sink(DLMCHS) has been widely used in various electronic devices; however, the existence of the nonuniform thermal strain distribution in actual operation has adverse effect on the overall stability. In this paper, two optimized designs of DLMCHS with cutting baffles on top and bottom layers are presented based on the traditional DLMCHS. The heat transfer and thermal stress performance are numerically analyzed and compared with the traditional DLMCHS. The results indicate that cutting baffles of micro-channels remarkably improves heat transfer and thermal stress performance. The optimized design with cutting baffles on the bottom layer decreases thermal strain but deteriorates heat transfer performance. The model with cutting baffles on the top layer has better combined thermal strain and heat transfer performance, which reduces thermal strain by about 1.5 times and enhances heat transfer by about 26.5%. For the design with cutting baffles on the top board, adding metal foam in the inlet collector can decrease the total minimum thermal strain by 51.4% and maximum temperature by 1.4 K, and increase the Nusselt number by 15%. 4β-Elemene-loaded polymeric micelles intensify anti-carcinoma efficacy and alleviate side effects显示文摘β-Elemene is a volatile oil used for the treatment of cancer,but poor solubility,low bioavailability,and various adverse reactions limit its application.For amelio rating risks of the venous toxicity ofβ-elemene,intravenously injectable micelle ofβ-elemene was prepared using the thin-film hydration method.The results pointed out the micelles were uniformly spherical with about 20.96±0.1966 nm in average diameter and exhibited high entrapment efficiency(99.02%±0.88%).As revealed by drug release studies in vitro,β-elemene micelles had sustained drug release.Compared with freeβ-elemene,the micelles increased the drug cellular uptake and enhanced the anti-tumor effect in vitro through retarding cell cycle and inducing apoptosis.Meanwhile,the elevated se rum stability o fβ-elemene micelles implied less drug leakage and reduced toxicity.The wound healing and tube formation assay in vitro demonstrated the anti-metastasis and anti-angiogenesis effects ofβ-elemene micelles.Moreover,the pharmacokinetics study showed the AUC and T1/2 ofβ-elemene in micelle group were 1.79 and 1.62 times of that in free fi-elemene group,suggesting the circulation time ofβ-elemene in the blood had been prolonged.In addition,β-elemene micelles showed a favorable antitumor response compared with theβ-elemene solution on C26 colon cance r-bearing mice model.Local irritation study investigated in rabbits indicated that theβ-elemene micelles strikingly mitigated the irritation to the injection sites compared with freeβ-elemene.These results proved that the micelle could be a good candidate as an auspicious drug delivery system ofβ-elemene for the prospective clinical treatment of carcinoma.Qiaoqiao Hu Lan Bai Zejiang Zhu Zhengying Su Peng Bai Minghai Tang Caixia Dou Junfeng Yan Rongsheng Tong Wuyu Zhang Lijuan Chen Lulu Cai 2020Chinese Chemical Letters2020,31,3:2
5In-situ emb e dding zeolitic imidazolate framework derived Co–N–C bifunctional catalysts in carbon nanotube networks for flexible Zn–air batteries显示文摘Recently, the development of high-performance bifunctional oxygen catalysts integrated with flexible conductive scaffolds f or rechargeable metal-air batteries has attracted considerable interest, driving by fastgrowing wearable electronics. Herein, we report a flexible bifunctional oxygen catalyst thin film consisting of Co–N–C bifunctional catalysts embedding in carbon nanotube(CNT) networks. The catalyst is readily prepared by pyrolysis of cobalt-based zeolitic imidazolate frameworks(ZIF-67) that are in-situ synthesized in CNT networks. Such catalyst film demonstrates very high catalytic activities for oxygen reduction(onset potential: 0.91 V, and half-wave potential: 0.87 V vs. RHE) and oxygen evolution(10 m Acm^-2 at 1.58 V) reactions, high methanol tolerance property, and long-term stability(97% current retention). Moreover, our integrated catalyst film shows very good structure flexibility and robustness. Based on the obtained film air electrodes, flexible Zn–air batteries demonstrate low charging and discharging overpotentials(0.82 V at 1 m A cm^-1) and excellent structure stability in the bending tests. 6The nutritional composition and digestion of plants foraged by red deer(Cervus elaphus xanthopygus) in northeast China显示文摘We studied species of plants foraged by red deer in the Muling forest region of northeastern China during January 2014 and 2015. We focused on nutritional composition, energy content, and the digestibility of these plants. Crude protein content varied by plant species.Average crude protein content was 7.5 ± 1.7% of dry matter, ranging from 4.5 to 10.3%. Crude lipid content did not vary by species and averaged 7 ± 1.5%(range5.4–9.8%). Non-fiber carbohydrate(NFC) and neutral detergent fiber(NDF) content varied broadly with average values being 20.1 ± 4.8%(range 13.5–28.8%) and64.7 ± 5.9%(range 53.5–71.8%), respectively. Energy content ranged from 16.7 to 19.7 k J/g. Average dry matter digestibility(DMD) was 46.1 ± 4.8%, with a range from38.7 to 54.6%. Only three plants, Saussurea manshurica,Aralia elata and Equisetum hyemale exhibited DMD [ 50%. NFC content explained most of the variation in energy content(r = 0.451). In contrast, energy content was inversely related to NDF(r =-0.443). There was no correlation between crude protein, crude lipid, and energy content(r_(protein)= 0.004, r_(lipid)=-0.190). S. manshurica,A. elata and E. hyemale had higher NFC and lower NDF content. Thus, these species offered the greatest nutritional value for wintering red deer in the study area. 7Time resource management of OAR based on fuzzy logic priority for multiple target tracking显示文摘For coping with the multiple target tracking in the presence of complex time-varying environments and unknown target information, a time resource management scheme based on chance-constraint programming(CCP) employing fuzzy logic priority is proposed for opportunistic array radar(OAR). In this scheme,the total beam illuminating time is minimized by effective time resource allocation so that the desired tracking performance is achieved. Meanwhile, owing to the randomness of radar cross section(RCS), the CCP is used to balance tracking accuracy and time resource conditioned on the specified confidence level. The adaptive fuzzy logic prioritization, imitating the human decision-making process for ranking radar targets, can realize the full potential of radar. The Bayesian Crame ′r-Rao lower bound(BCRLB) provides us with a low bound of localization estimation root-mean-square error(RMSE), and equally important, it can be calculated predictively. Consequently, it is employed as an optimization criterion for the time resource allocation scheme. The stochastic simulation is integrated into the genetic algorithm(GA) to compose a hybrid intelligent optimization algorithm to solve the CCP optimization problem. 8DNA methylation and alternative splicing modulate FBXW11 gene expression in Holstein bull testis and are correlated with sperm quality显示文摘F-box and WD-40 domain protein 11(FBXW11)is an important component of the E3 ubiquitin-ligase enzyme that plays a key role in the ubiquitin-dependent regulation of spermatogenesis.In our previous research,the mRNA expression of FBXW11 in bull sperm with high motility is significantly higher than that with low motility.In the present study,the protein expression levels of FBXW11 in bull testicular tissues with low-performance sperm quality groups were significantly higher than those in normal performance groups.The immunohistochemistry result demonstrated that FBXW11 protein was located in the periphery of Leydig cells and seminiferous tubules.Three splice variants of the FBXW11 gene,namely,FBXW11-tv1,FBXW11-tv2,and FBXW11-tv3,were identified in testicular tissues.The splicing patterns of the three variants are exon skipping.The transcript FBXW11-tv2 expressions were the highest in each sample.The low-performance groups displayed higher FBXW11-tv1 and FBXW11-tv2 transcript expressions than the normal performance groups.Two CpG islands were located within the 5’UTR and exon 1-2 region of the FBXW11 gene.Bisulfite sequencing PCR results demonstrated that the methylation levels of 11 methylation sites in the CpG island 2 from−99 to−43 in the normal performance groups were significantly lower than those in the low-performance groups.Pearson correlation analysis suggested that the CpG island 2 methylation level was negatively correlated with sperm motility and the transcript FBXW11-tv2 expression level.Our data revealed that alternative splicing and DNA methylation jointly regulated FBXW11 gene expression and were correlated with sperm quality traits during spermatogenesis in Holsteins.YONG LIU ZHIHUA JU QIANG JIANG WENHAO LIU CHUNHONG YANG YARAN ZHANG XIUGE WANG YAPING GAO XIAOCHAO WEI YAN SUN JINPENG WANG MINGHAI HOU LING YANG JINMING HUANG 2021BIOCELL2021,45,1:1

10Update on the status of wolverines in China显示文摘The wolverine, Gulogulo, is an endangered species in China. This is the first report of the overall status of wolverines in China. The geographic distribution and population sizes of wolverines in the Greater Khingan and Altai Mountains were surveyed by line transect from1996–2000 and in 2011, respectively. We identified two geographically distinct groups in the Great Khingan Mountains, with a total population size of about 200 individuals throughout the region(approximately80,000 km^2). Wolverines were also confirmed to be present in the Altai Mountains, where about 100 individuals occupied approximately 28,000 km^2 of mountain forest and tundra. 11R11 peptides can promote the molecular imaging of spherical nucleic acids for bladder cancer margin identification显示文摘One of the critical problems in bladder cancer(BC)management is the local recurrence of disease.However,achieving the accurate delineation of tumor margins intraoperatively remains extremely difficult due to the lack of effective tumor margin recognition technology.Herein,survivin molecular beacon(MB)and R11 peptide-linked spherical nucleic acids(SNAs)were synthesized as nanoprobes(AuNP-MB@R11)for sensitive detection of BC margins.Physicochemical properties proved that R11 peptides and survivin MB were successfully loaded onto the surface of SNAs.AuNP-MB@R11 had good stability against nuclease activity and high sensitivity and specificity to detect survivin single strand DNA(ssDNA)in vitro.According to cytology,R11 peptides could increase the BC targeting ability and membrane penetrability of SNAs.Notably,R11 peptides significantly promoted the disintegration of lysosomes and the release of SNAs to enhance fluorescence imaging quality.Further RNA sequencing proved that some genes and pathways related to endocytosis and lysosomes were significantly regulated,such as AGPAT5,GPD1L,and GRB2.In orthotopic BC models and a clinical sample from a patient with BC,AuNP-MB@R11 showed a more legible cancerous fluorescence margin and offered remarkably improved detection compared to those achieved by SNAs.R11 peptide-linked SNAs present promising potential to identify BC margin,which may help to improve the R0 resection rate in surgery and improve patients’quality of life.Minghai Ma Pu Zhang Xiao Liang Daxiang Cui Qiuya Shao Haibao Zhang Mengzhao Zhang Tao Yang Lu Wang Nan Zhang Minxuan Jing Lu Zhang Weichao Dan Rundong Song Xi Liu Jiatao Hao Yuhang Chen Lijiang Gu Lei Wang Jinhai Fan 2022Nano Research2022,15,3:1
12Variation in winter daily range area of red deer(Cervus elaphus xanthopygus) based on DNA extracted from fecal samples显示文摘With environmental change and increased human activity,wild red deer(Cervus elaphus xanthopygus)populations and habitat area continue to decrease.Red deer is a main prey species of Amur tiger(Panthera tigris altaica).Factors affecting red deer winter home range in the Wanda montains of northeast China are not well understood.In this study,we aimed to better document red deer daily ranges to support conservation of red deer.A second objective was to develop a practical research method combining macro-and micro-technology for home range research on a variety of rare or endangered wild species.We collected 105 fresh fecal samples of red deer in three different periods during winter from December 2012 to March 2013 in the Wandashan region,Heilongjiang Province,China.Individual and sex identities were analyzed based on fecal DNA using microsatellite DNA.We used samples from deer farms for correction and validation of our identifications from field samples.We used a combination of molecular scatology,3S techniques(GPS),and direct observation to estimate winter daily range areas.Based on eleven microsatellite loci,we identified 18 individuals,10 females and 8 males,with a correction factor of 1.00 and sex ratio of 1:0.8.The mean(±SE)number of alleles was 5.1±0.41,and mean polymorphism information content was 0.63±0.03.Expected heterozygosity ranged from 0.46 to 0.81,with a mean of 0.69±0.03,and mean observed heterozygosity was 0.73±0.07.Sizes of mean daily ranges were 26.1±1.1 ha in December,89.3±1.6 ha in January,and 67.5±1.9 ha in March,indicating the following trend for daily range area:middle winter>late winter>early winter.Mean daily range area was insignificantly larger for males than for females.Estimated winter daily ranges were similar to those reported for Europe.Our combination of macro-and micro-technology proved useful for estimating winter home range areas of red deer.Miao Yang Yue Sun Wei Qi Zhang Hongyi Yuan Minghai Zhang 2019Journal of Forestry Research2019,30,5:1

14Multiplex ratiometric gold nanoprobes based on surface-enhanced Raman scattering enable accurate molecular detection and imaging of bladder cancer显示文摘Recently,surface-enhanced Raman scattering(SERS)has been successfully used in the non-invasive detection of bladder tumor(BCa).The internal standard method was considered as an effective ratiometric strategy for calibrating signal fluctuation originated from the interference of measurement conditions and samples.However,it is still difficult to detect the target mRNA quantitatively using the current ratiometric SERS nanosensors.In this study,we developed an internal reference based ratiometric SERS assay.Two kinds of molecular beacons(MB)carrying Raman reporter molecules were anchored on sea-urchinlike Au nanoclusters(AuNCs).Thymidine kinase1(TK1)MBs with hexachlorofluorescein(HEX)were used to capture tumor biomarker TK1 mRNA,and glyceraldehyde 3-phosphate dehydrogenase(GAPDH)MBs with 5(6)-carboxyfluorescein(FAM)were used to offer internal standard signals.The internal reference GAPDH MB can reflect the consistent content of the GAPDH mRNA in single cells.The ratiometric method(I745/I645)can more accurately reflect the content of target mRNA in single cells.The ratiometric nanoprobes had excellent stability(coefficient of variation:0.3%),high sensitivity(detection limit:3.4 pM),high specificity(capable of single-base mismatch recognition)and ribozyme-resistant stability.Notably,the nanoprobes can effectively distinguish BCa cells from normal cells,and it was easy to contour the single BCa cell using the ratiometric method.By combining asymmetric polymerase chain reaction(PCR)and ratiometric nanoprobes,it was easy to distinguish the SERS ratio(I_(745)/I_(645))as low concentration as 10-14 M.Further clinical detection in urine samples from patients with BCa confirmed its potential for early noninvasive diagnosis of BCa with the sensitivity of 80%and specificity of 100%,which is superior to the current urine cytological method.Xiao Liang Pu Zhang Minghai Ma Tao Yang Xiangwei Zhao Rui Zhang Minxuan Jing Rundong Song Lei Wang Jinhai Fan 2022Nano Research2022,15,4:1

19Effects of Soil Types and Moisture Contents on Emergence Rate of Contarinia morulae Jiang显示文摘In order to explore the relationship between soil moisture content and the severity of occurrence of Contarinia morulae Jiang,the effects of different soil types and moisture contents on the emergence rate of C. morulae were investigated. The results showed that soil moisture contents had a significant effect on the emergence rate of C. morulae. When the soil moisture content was 10%,the emergence rate of C. morulae was the highest in both soil types. When the soil moisture content was too high or too low,the emergence rate of C. morulae was significantly reduced. Within the suitable water content range,the emergence rate of C. morulae in sandy soil was significantly higher than that in sandy loam soil. The trend of emergence of C. morulae in the two soil types was basically the same. 20Redefinition of the Odorrana versabilis Group,with a New Species from China(Anura,Ranidae,Odorrana)显示文摘Taxonomic studies of the Odorrana versabilis group have been consistently disorganized,and they have often been incorrectly associated with an abandoned subgenus or genus Bamburana in existing literature.In this study,we conducted an integrative taxonomic analysis using molecular phylogenetic analysis of two mitochondrial gene fragments(16S rRNA and CO1)and morphological examination of 41 specimens collected during herpetological surveys in southern China.Our results confirmed the monophyly of O.versabilis group which exhibited distinct morphological traits.We also identified a cryptic lineage sister with O.exiliversabilis.Additionally,our study revealed that O.trankieni was nested within O.nasuta.Consequently,we redefined the morphological characteristics of the O.versabilis group,described a new species in this group,O.confusa sp.nov.,and proposed that O.trankieni be considered a junior synonym of O.nasuta.One of the most significant findings of this study was the observation of distinct morphological changes in adult males across all species within the O.versabilis group between the non-breeding and breeding seasons.This study contributes to research on various aspects of Asian amphibian fauna,including taxonomy,ecology,evolution,and physiology.Hanming SONG Siyu ZHANG Shuo QI Zhitong LYU Zhaochi ZENG Yongheng ZHU Minghai HUANG FuchenLUAN Zufei SHU Yuening GONG Zhifa LIU Yingyong WANG 2023Asian Herpetological Research2023,14,4:0
