1Genome wide abnormal DNA methylome of human blastocyst in assisted reproductive technology显示文摘Proper reprogramming of parental DNA methylomes is essential for mammalian embryonic development.However,it is unknown whether abnormal methylome reprogramming occurs and is associated with the failure of embryonic development.Here we analyzed the DNA methylomes of 57 blastocysts and 29 trophectoderm samples with different morphological grades during assisted reproductive technology(ART) practices.Our data reveal that the global methylation levels of high-quality blastocysts are similar(0.30 ± 0.02,mean ± SD).while the methylation levels of low-quality blastocysts are divergent and away from those of high-quality blastocysts.The proportion of blastocysts with a methylation level falling within the range of 0.30 ± 0.02 in different grades correlates with the live birth rate for that grade.Moreover,abnormal methylated regions are associated with the failure of embryonic development.Furthermore,we can use the methylation data of cells biopsied from trophectoderm to predict the blastocyst methylation level as well as to detect the aneuploidy of the blastocysts.Our data indicate that global abnormal methylome reprogramming often occurs in human embryos,and suggest that DNA methylome is a potential biomarker in blastocyst selection in ART.Guoqiang Li Yang Yu Yong Fan Congru Li Xiaocui Xu Jialei Duan Rong Li Xiangjin Kang Xin Ma Xuepeng Chen Yuwen Ke Jie Yan Ying Lian Ping Liu Yue Zhao Hongcui Zhao Yaoyong Chen Xiaofang Sun Jianqiao Liu Jie Qiao Jiang Liu 2017Journal of Genetics and Genomics2017,44,10:5

7Understanding the interfacial behaviors of benzene alkylation with butene using chloroaluminate ionic liquid catalyst: A molecular dynamics simulation显示文摘To better understand the benzene alkylation with chloroaluminate ionic liquids(ILs) as catalyst, the interfacial properties between the benzene/butene binary reactants and chloroaluminate ILs with varying cation alkyl chain length and different anions were investigated using molecular dynamics(MD) simulations. The results indicate that ILs can obviously improve the interfacial width, solubility and diffusion of reactants compared to H_(2)SO_(4). The longer alkyl chains of cations present a density enrichment at the interface and protrude into the binary reactants phase. Furthermore, the ILs consisting of 1-octyl-3-methylimidazolium cations([Omim]^(+)) and the stronger acidity heptachlorodialuminate anions([Al_(2)Cl_(7)]^(-)) are more beneficial to promote the interfacial width and facilitate the dissolution and diffusion of benzene in both the IL bulk and the interfacial region in comparison to the ones with shorter alkyl chains cations and weaker acidity anions. 9Understanding the interfacial properties of benzene alkylation with 1-dodecene catalyzed by immobilized chloroaluminate ionic liquids using molecular dynamics simulation显示文摘Benzene alkylation catalyzed by immobilized ionic liquids(ILs)on solid carriers is considered as a heterogeneous reaction,in which the interfacial properties play an important role.Hence,the interfacial characteristics between benzene/1-dodecene mixture and immobilized chloroaluminate ILs with different alkyl chain length on the silica substrate were investigated by molecular dynamics simulation.The grafted ILs can obviously promote the enrichment of benzene near the interface,leading to a higher ratio of benzene to dodecene,and the interfacial width increases slightly with increased alkyl chain of grafted cations.At the same time,the grafted cations can also enhance the benzene diffusion and suppress the dodecene diffusion at the interface,which probably helps to inhibit the inactivation of catalysts.This work provides deeply insights into the rational design of novel immo-bilized ILs catalysts for the benzene alkylation.Weizhong Zheng Jialei Sha Weizhen Sun Ling Zhao 2023Green Chemical Engineering2023,4,3:0
10Photocatalytic degradation of omethoate using NaY zeolite-supported TiO_(2)显示文摘The degradation of omethoate was conducted using H_(2)O_(2) as oxidant,TiO_(2) supported on NaY zeolite as photocatalyst and a 300W lamp as light source.The effect of the calcination temperature of the photocatalyst,the amount of TiO_(2) loaded on NaY zeolite,the photocatalyst amount,the pH value and the radiation time on the degradation ratio of omethoate were investigated.The results show that TiO_(2)/NaY zeolite photocatalyst prepared by sol-gel method had good photocatalysis.The photo-catalytic optimum oxidationconditions of omethoate are as follows:the calcination temperature of the photocatalyst is 550℃,the amount of TiO_(2) loaded on NaY zeolite is 35.2 wt-%,the amount of photocatalyst is 5g/L,pH=8 and the radiation time is 180min.Under these conditions,the removal ratio of omethoate is up to 93%.Dishun ZHAO Jialei WANG Zhigang ZHANG Juan ZHANG 2009Frontiers of Chemical Science and Engineering2009,3,2:0
11A sustained release of BMP2 in urine-derived stemcells enhances the osteogenic differentiation and the potential of bone regeneration显示文摘Cell-based tissue engineering is one of the optimistic approaches to replace current treatments for bone defects.Urine-derived stem cells(USCs)are obtained non-invasively and become one of the promising seed cells for bone regeneration.An injectable BMP2-releasing chitosan microspheres/type I collagen hydrogel(BMP2-CSM/Col I hydrogel)was fabricated.USCs proliferated in a time-dependent fashion,spread with good extension and interconnected with each other in different hydrogels both for 2D and 3D models.BMP2 was released in a sustained mode for more than 28 days.Sustained-released BMP2 increased the ALP activities and mineral depositions of USCs in 2D culture,and enhanced the expression of osteogenic genes and proteins in 3D culture.In vivo,the mixture of USCs and BMP2-CSM/Col I hydrogels effectively enhanced bone regeneration,and the ratio of new bone volume to total bone volume was 38%after 8weeks of implantation.Our results suggested that BMP2-CSM/Col I hydrogels promoted osteogenic differentiation of USCs in 2D and 3D culture in vitro and USCs provided a promising cell source for bone tissue engineering in vivo.As such,USCs-seeded hydrogel scaffolds are regarded as an alternative approach in the repair of bone defects.Shuang Wu Zhao Chen Xi Yu Xin Duan Jialei Chen Guoming Liu Min Gong Fei Xing Jiachen Sun Shishu Huang Zhou Xiang 2022Regenerative Biomaterials2022,9,1:0
12相同剂量长春瑞滨序贯或联合卡培他滨一线治疗转移性乳腺癌的Ⅱ期随机对照临床试验(英文)显示文摘Objective:It remains unclear whether simultaneous use of two chemotherapeutic drugs is better than sequential use.This trial was designed to explore efficacy and safety of sequential vs simultaneous use of vinorelbine and capecitabine at the same dosage as first-line therapy in metastatic breast cancer (MBC).Methods:This was a un-icenter, randomized phase II trial.Patients randomized into the simultaneous group (group A) were simultaneously administered with vinorelbine and capecitabine while those in the sequential group (group B) received vinorelbine followed by capecitabine at the same dosage.Results:Sixty-six patients were screened and 30 patients were randomized into either group.There're significant differences in the clinical benefit rate (CBR) with 80.0% for group A vs 53.3% for group B (P=0.028).With a median follow up time of 13.5 months, there were no significant differences between the two groups in PFS (median PFS:7.70 months for group A vs 7.23 months for group B, P=0.436).Grade III or IV neutropenia (83.3% vs 50.0%, P=0.006), all grades of fatigue (56.7% vs 30.0%, P=0.037) and anorexia (53.3% vs 23.3%, P=0.017) were significantly more frequent in simultaneous group.Conclusion:Simultaneous administration of vinorelbine and capecitabine can bring about improvements in CBR, but cannot translate into long-term benefits, such as progression-free survival (PFS) or overall survival (OS).These findings, combined with a relatively better tolerability in sequential group, showed that both simultaneous and sequential administrations are reasonable options for MBC patients.Shiyang Gu, Ping Zhang, Zhen Jia, Biyun Wang, Jian Zhang, Zhonghua Wang, Haiyi Guo, Jialei Wang, Xinmin Zhao, Xichun Hu Department of Medical Oncology, Fudan University Shanghai Cancer Hospital Department of Oncology, Shanghai Medical College, Fudan University, Shanghai 200032, China 2010The Chinese-German Journal of Clinical Oncology2010,9,9:0
13Map-based cloning and CRISPR/Cas9-based editing uncover BoNA1 as the causal gene for the no-anthocyanin-accumulation phenotype in curly kale (Brassica oleracea var. sabellica)显示文摘Brassica oleracea comprises several important vegetable and ornamental crops,including curly kale,ornamental kale,cabbage,broccoli,and others.The accumulation of anthocyanins,important secondary metabolites valuable to human health,in these plants varies widely and is responsible for their pink to dark purple colors.Some curly kale varieties lack anthocyanins,making these plants completely green.The genetic basis of this trait is still unknown.We crossed the curly kale inbred line BK2019(without anthocyanins)with the cabbage inbred line YL1(with anthocyanins)and the Chinese kale inbred line TO1000(with anthocyanins)to generate segregating populations.The no-anthocyanin trait was genetically controlled by a recessive gene,bona1.We generated a linkage map and mapped bona1 to a 256-kb interval on C09.We identified one candidate gene,Bo9g058630,in the target genomic region;this gene is homologous to AT5G42800,which encodes a dihydroflavonol-4-reductase-like(DFR-like)protein in Arabidopsis.In BK2019,a 1-bp insertion was observed in the second exon of Bo9g058630 and directly produced a stop codon.To verify the candidate gene function,CRISPR/Cas9 gene editing technology was applied to knock out Bo9g058630.We generated three bona1 mutants,two of which were completely green with no anthocyanins,confirming that Bo9g058630 corresponds to BoNA1.Different insertion/deletion mutations in BoNA1 exons were found in all six of the other no-anthocyanin kale varieties examined,supporting that independent disruption of BoNA1 resulted in no-anthocyanin varieties of B.oleracea.This study improves the understanding of the regulation mechanism of anthocyanin accumulation in B.oleracea subspecies.Kaiwen Yuan Xinyu Zhao Wenru Sun Limei Yang Yangyong Zhang Yong Wang Jialei Ji Fengqing Han Zhiyuan Fang Honghao Lv 2023Horticulture Research2023,10,8:0

15Analysis and Verification of the Conserved MYB Binding Element in the DFR Promoter in Compositae显示文摘Anthocyanins,ubiquitous in the Compositae family,are regulated by MYB(v-myb avian myeloblastosis viral oncogene homolog),playing an important role in anthocyanin synthesis.In this study,we analyzed the regulation pathway in which the MYB protein of subgroup 6 promotes dihydroflavonol reductase(DFR)expression in Compositae,and validated this law in Saussurea medusa through yeast one-hybrid experiments.Our results showed that MYB and DFR underwent purification selection,DFR promoter analysis revealed the presence of MYB binding site(GAGTTGAATGG)and bHLH binding site(CANNTG)at the sense strand of 84–116 nucleotide residues from the start codon.These two motifs were separated by 9–10 nucleotide residues,as existed in the DFR promoters of many Compositae plants.Furthermore,the yeast one-hybrid experiment demonstrated that SmMYB1 can activate the promoter of SmDFR.Our results provide a reference for further functional study of DFR in Compositae.Jialei Guo Fengzhen Li Guomin Shi Weimin Zhao Tao He 2024Phyton-International Journal of Experimental Botany2024,93,2:0
16Prokaryotic Expression and Immunogenicity of Dominant Epitope Region of Goose Parvovirus Structural Protein VP3显示文摘[Objective] The paper was to develop a subunit vaccine candidate for prevention and control of goose parvovirus infection. [Method]Based on the prokaryotic expression system, the antigenic epitopes and locations of the structural protein VP3 were predicted by software analysis,and the region displaying a large portion of antigenic epitopes was amplified by PCR. The target VP3 DNA fragment was inserted into pET-30 a-VP3 vector, was transformed into Escherichia coli BL21 competent cells for protein expression and animal test. The SPF chickens were immunized with the recombinant protein and the antisera were collected for neutralization test by using a goose embryo fibroblast. [Result] The recombinant plasmid was constructed, and the target region of VP3 protein was expressed efficiently in a soluble form. The neutralizing titers of antisera could reach up to-2.608. 17Comparative Transcriptome Analysis Reveals Different Mechanisms of Adaptation to Environment among Three Species of Saussurea DC.显示文摘Saussurea medusa,Saussurea hypsipeta and Saussurea obvallata are typical alpine snowline plants growing in the Qinghai-Tibet plateau.They are characterized by a specialized morphology.S.medusa and S.hypsipeta have very dense trichomes on whole plant,whereas S.obvallata has transparent bracts covered inflorescences.The different forms reflect their adaptation to cold environments.To investigate the different mechanisms of adaptation of these species to cold temperatures,transcriptome sequencing was performed in three species of Saussurea DC.A total of 116394137237 and 113879 Unigenes were identified from S.medusa,S.hypsipeta and S.obvallata,respectively.Of these,55909(48.03%),65519(47.74%)and 51889(45.56%)Unigenes were matched in public databases.GO analysis identified that most of annotated Unigenes in the three species of plants were related to cellular,metabolic,and single−organism processes,and binding and catalytic activities.The differential expression of 37 genes related to environmental adaptation were discovered by pairwise comparisons.Of these,two candidate genes(Interaptin-like and CSLB3)related to trichome development were identified only in S.medusa and S.hypsipeta,which was consistent with their distinct morphology.Our data can provide a valuable resource for the further studies on the adaptive mechanisms of molecular and functional ecology in Saussurea DC.Weimin Zhao Fengzhen Li Guomin Shi Jialei Guo Guifang He Tao He 2022Phyton-International Journal of Experimental Botany2022,91,7:0
18Overbooking-Enabled Task Scheduling and Resource Allocation in Mobile Edge Computing Environments显示文摘Mobile Edge Computing(MEC)is proposed to solve the needs of Inter-net of Things(IoT)users for high resource utilization,high reliability and low latency of service requests.However,the backup virtual machine is idle when its primary virtual machine is running normally,which will waste resources.Overbooking the backup virtual machine under the above circumstances can effectively improve resource utilization.First,these virtual machines are deployed into slots randomly,and then some tasks with cooperative relationship are off-loaded to virtual machines for processing.Different deployment locations have different resource utilization and average service response time.We want tofind a balanced solution that minimizes the average service response time of the IoT application while maximizing resource utilization.In this paper,we propose a task scheduler and exploit a Task Deployment Algorithm(TDA)to obtain an optimal virtual machine deployment scheme.Finally,the simulation results show that the TDA can significantly increase the resource utilization of the system,while redu-cing the average service response time of the application by comparing TDA with the other two classical methods.The experimental results confirm that the perfor-mance of TDA is better than that of other two methods.Jixun Gao Bingyi Hu Jialei Liu Huaichen Wang Quanzhen Huang Yuanyuan Zhao 2023Intelligent Automation & Soft Computing2023,,7:0
19Effect of catgut embedding at acupoints versus non-acupoints in abdominal obesity:a randomized clinical trial显示文摘OBJECTIVE:To explore the difference of catgut embedding effect between acupoints and non-acupoints in patients with abdominal obesity(AO).METHODS:In this multicenter,double-blind,randomized controlled trial,all subjects were randomly assigned into the acupoint catgut embedding(ACE)group and control group(catgut embedding at non-acupoints).With a 12-week actual intervention period and a 4-week period of follow-up.Waist circumference(WC),body weight,body mass index(BMI),hip circumference(HC)and appetite were applied and assessed at baseline and after 6,12 and 16 weeks.RESULTS:After the total intervention phase(12 weeks),the WC,body weight,BMI,HC and visual analogue scale scores of appetite,decreased significantly in the two groups as compared to the baseline(P<0.001).Meanwhile,after the 4-week follow-up,the indicators still decreased significantly in the ACE group(P<0.001).At 12 and 16 weeks,catgut embedding at acupoints showed significantly advantages to non-acupoints in WC and appetite(P<0.05).No serious adverse events were observed in ACE group and control group.CONCLUSIONS:Catgut embedding at acupoints and non-acupoints are all effective and safe for AO.ACE can effectively treat AO as expected and deliver lasting results.ZHANG Xinghe LI Qifu YI Rong XING Chonghui JIN Yuhao MENG Jiangqiong FENG Jialei ZHAO Siwen LIANG Fanrong GUO Taipin 2023Journal of Traditional Chinese Medicine2023,43,4:0
