1Critical function of DNA methyltransferase 1 in tomato development and regulation of the DNA methylome and transcriptome显示文摘DNA methylation confers epigenetic regulation on gene expression and thereby on various biological processes.Tomato has emerged as an excellent system to study the function of DNA methylation in plant development.To date,regulation and function of DNA methylation maintenance remains unclear in tomato plants.Here,we report the critical function of tomato(Solanum lycopersicum)Methyltransferase 1(Sl MET1)in plant development and DNA methylome and transcriptome regulation.Using CRISPR-Cas9 gene editing,we generated slmet1 mutants and observed severe developmental defects with a frame-shift mutation,including small and curly leaves,defective inflorescence,and parthenocarpy.In leaf tissues,mutations in Sl MET1 caused CG hypomethylation and CHH hypermethylationon a whole-genome scale,leading to a disturbed transcriptome including ectopic expression of many RIN target genes such as ACC2 in leaf tissues,which are normally expressed in fruits.Neither the CG hypomethylation nor CHH hypermethylation in the slmet1 mutants is related to tissue culture.Meanwhile,tissue culture induces non-CG hypomethylation,which occurs more frequently at gene regions than at TE regions.Our results depict Sl MET1-and tissue culture-dependent tomato DNA methylomes,and that Sl MET1 is required for maintaining a normal transcriptome and normal development of tomato.Yu Yang Kai Tang Tatsiana U Datsenka Wenshan Liu Suhui Lv Zhaobo Lang Xingang Wang Jinghui Gao Wei Wang Wenfeng Nie Zhaoqing Chu Heng Zhang Avtar K HANDa Jian-Kang Zhu Huiming Zhang 2019Journal of Integrative Plant Biology2019,61,12:3

4Multimaterial additive manufacturing manipulator for fabricating magnetoelectric pressure sensors显示文摘The simultaneous placement of different types of materials including polymers,ceramics,and metals,in their desired positions could be adopted to manufacture end-use devices/apparatuses with diverse functionalities and significantly reduce the fabrication cost and time.However,existing additive manufacturing(AM)approaches can only treat one material species at a time due to their intrinsic working mechanisms.Here we develop an AM manipulator for manufacturing a wide variety of material species,including polymers,ceramics,and metals,through a multifilament transport strategy assisted by laser power.The six-jaw manipulator contains three pairs of filament delivering/cutting systems for transporting diverse materials and a beam of tunable laser as the thermal source.The whole apparatus is integrated into a robotic manipulator to create a multifreedom manufacturing platform.With this innovation,products with multiple material species and desired complex geometries can be fabricated on demand.Furthermore,we synthesize a multimaterial(polymer/ceramic/metal)printed magnetoelectric pressure sensor that can convert applied mechanical forces to electricity and maintain efficiency even after undergoing 10000 cycles of pressure/recovery.With this multimaterial filament transport and laser manufacturing strategy,our AM manipulator exhibits promising application in the advanced manufacturing of embedded electronics,sensors,soft robotics,and customizable medical devices.LIU ZhuFeng LI ZhaoQing WANG Qi WANG Yan YANG Lei NIE Xiang GENG Peng WU ZhenHua TANG SiHan YANG Lei WU HongZhi SHI YunSong CHEN Peng SU Bin ZHANG LiChao YAN ChunZe SHI YuSheng 2022Science China(Technological Sciences)2022,65,11:1

7Predictors of mortality in patients with acute smallbowel perforation transferred to ICU after emergency surgery:a single-centre retrospective cohort study显示文摘Background Although small-bowel perforation is a life-threatening emergency even after immediate surgical intervention,studies have rarely investigated surgical outcomes due to its relatively low incidence.This study aimed to investigate the outcomes of emergency surgery for patients with small-bowel perforation transferred to the intensive care unit(ICU)and the risk factors for mortality.Methods Consecutive patients with small-bowel perforation who were confirmed via emergency surgery and transferred to the ICU in Zhongshan Hospital,Fudan University(Shanghai,China)between February 2011 and May 2020 were retrospectively analysed.Medical records were reviewed to determine clinical features,laboratory indicators,surgical findings,and pathology.Results A total of 104 patients were included in this study,among whom 18(17.3%),59(56.7%),and 27(26.0%)underwent perforation repair,segmental resection with primary anastomosis,and small-bowel ostomy,respectively.Malignant tumours were the leading cause of perforation in these patients(40.4%,42/104).The overall post-operative complication rate and mortality rates were 74.0%(77/104)and 19.2%(20/104),respectively.Malignant tumour-related perforation(odds ratio[OR],4.659;95%confidence interval[CI],1.269–17.105;P=0.020)and high post-operative arterial blood-lactate level(OR,1.479;95%CI,1.027–2.131;P=0.036)were identified as independent risk factors for post-operative mortality in patients with small-bowel perforation transferred to the ICU.Conclusions Patients with small-bowel perforation who are transferred to the ICU after emergency surgery face a high risk of post-operative complications and mortality.Moreover,those patients with malignant tumour-related perforation and higher post-operative blood-lactate levels have poor prognosis.Jianzhang Wu Ping Shu Hongyong He Haojie Li Zhaoqing Tang Yihong Sun Fenglin Liu 2022Gastroenterology Report2022,10,1:0
8Bovine Placental Bioactive Proteins Preparation and Efficacy Evaluation显示文摘Taking the qualified bovine placenta as the research object,a series of pure physical separation and extraction processes such as tissue fragmentation,ultrasound,freeze-thawed,centrifugation and filtration were used to prepare the bovine placental bioactive proteins.The results of protein concentration showed that bovine placental bioactive proteins prepared in this study could reach(11.33±2.09)mg/mL under optimal experimental condition.The determination of specific components showed that there were abundant exosome like vesicles in the product with particle size ranging from 30 to 150 nm,pie structure under transmission electron microscope and expressing CD9,CD63 and TSG101 by western blot detection.The results of cell efficacy evaluation showed that bovine placental bioactive proteins could significantly promote the proliferation of human skin fibroblasts and had anti-inflammatory effect via down-regulating IL-6 and TNF-αexpressionin RAW264.7 cells.Li Xuejia Jiang Jiaohua Tang Huijuan Liang Jiafu Zhang Zhaoqing Qi Kangyi Yue Kun Ma yanyan Chen donghuang Chen Haijia Pang keliang 2023China Detergent & Cosmetics2023,8,2:0
