| 1 | Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual. | Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang | 2020 | Protein & Cell2020,11,4: | 44 |
| 2 | Effect of Boschniakia rossica on expression of GSTP,p53 and p21^(ras)proteins in early stage of chemical hepatocarcinogenesis and its anti-inflammatory activities in rats显示文摘AIM To investigate the effect of Boschniakiarossica(BR)extract on expression of GST-P,p53 and p21rasproteins in early stage of chemicalhepatocarcinogenesis in rats and its anti-inflammatory activities.METHODS The expression of tumor marker-placental form glutathione S-transferase(GST-P),p53 and p21rasproteins were investigated byimmunohistochemical techniques and ABCmethod.Anti-inflammatory activities of BR werestudied by xylene and croton oil-induced mouseear edema,carrageenin,histamine and hotscald-induced rat pow edema,adjuvant-inducedrat arthritis and cotton pellet-induced mousegranuloma formation methods.RESULTS The 500 mg/kg of BR-H2O extractfractionated from BR-Methanol extract hadinhibitory effect on the formation of DEN-inducedGST-P-positive foci in rat liver(GST-P stainingwas 78% positive in DEN+AAF group vs 20%positive in DEN+AAF+BR group,P<0.05)andthe expression of mutant p53 and p21rasproteinwas lower than that of hepatic preneoplasticlesions(33% and 22% positive respectively inDEN+AAF group vs negative in DEN+AAF+BRgroup).Both CH2Cl2 and H2O extracts from BRhad anti-inflamatory effect in xylene and crotonoil-induced mouse ear edema(inhibitory rateswere 26%-29% and 35%-59%,respectively). BR-H2O extract exhibited inhibitory effect incarrageenin,histamine and hot scald-inducedhind paw edema and adjuvant-induced arthritis inrats and cotton pellet-induced granulomaformation in mice.CONCLUSION BR extract exhibited inhibitory effect on formation of preneoplastic hepatic foci in early stage of rat chemical hepato-carcinogenesis. | 5 | Integrating morphological spatial pattern analysis and the minimal cumulative resistance model to optimize urban ecological networks: a case study in Shenzhen City, China显示文摘Background:With the increasing fragmentation of landscape induced by rapid urbanization,the construction of ecological networks is of great signifcance to alleviate the degradation of urban habitats and protect natural envi‑ronments.However,there is considerable uncertainty when constructing ecological networks,especially the difer‑ent approaches to selecting ecological sources.We used the southern Chinese city of Shenzhen as a study area to construct and optimize ecological networks using a coupling approach.Ecological source areas were extracted using morphological spatial pattern analysis(MSPA)and the landscape index method.Ecological networks were con‑structed using the minimal cumulative resistance(MCR)model and the gravity model.Stepping stones and ecological fault points were added in corridors to optimize the ecological network.Results:Ten core areas with maximum importance patch values were extracted by the landscape index method as ecological source areas according to MSPA,after which corridors between ecological sources were constructed based on the MCR model.The constructed ecological networks were optimized using 35 stepping stones and 17 ecologi‑cal fault points.The optimized ecological networks included 11 important corridors,34 general corridors,and seven potential corridors.The results of corridor landscape-type analysis showed that a suitable ecological corridor is 60 to 200 m wide.Conclusions:Overall,our results imply that ecological source areas can be identifed virtually,and that ecological networks can be signifcantly optimized by combining MSPA and MCR models.These results provide a methodologi‑cal reference for constructing ecological networks,and they will be useful for urban planning and biodiversity protec‑tion in Shenzhen and other similar regions around the world. | Yang‑Yang Li Yu‑Zhe Zhang Zhi‑Yun Jiang Cheng‑Xuan Guo Ming‑Yue Zhao Zhi‑Guang Yang Ming‑Yan Guo Bing‑Yue Wu Quan‑Ling Chen | 2021 | Ecological Processes2021,10,1: | 3 |

| 9 | Ultra-small nanodots coated with oligopeptides providing highly negative charges to enhance osteogenic differentiation of hBMSCs better than osteogenic induction medium显示文摘For bone regenerative engineering,it is a promising method to form skeletal tissues differentiating from human bone morrow mesenchyme stem cells(hBMSCs).However,it is still a critical challenge to efficiently control ostogenesis and clearly reveal the influence factor.To this end,the fluorescent gold nanodots(Au NDs) with highly negative charges as osteogenic induction reagent are successfully synthesized,which display better than commercial osteogenic induction medium through the investigations of ALP activity(2.5 folds) and cytoskeleton staining(1.5 folds).Two kinds of oligopeptides with different bio-structures(cysteine,Cys and glutathione,GSH) are selected for providing surficial charges on Au NDs.It is revealed that Au-Cys with more negative charges(-51 mV) play better role than Au-GSH(-19 mV) in osteogenic differentiation,when both of them have same size(~2 nm),sphere shape and show similar cell uptake amount.To explore deeply,osteogenesis related signaling pathways are monitored,revealing that the enhancement of osteogenic differentiation was through autophagy signaling pathway triggered by Au-Cys.And the promotion of highly negative charges in osteogenic diffe rentiation was further proved via sliver nanodots(Ag NDs,Ag-Cys and Ag-GSH) and carbon nanodots(CDs,Cys-CDs and GSH-CDs).This work indicates part of insights during hBMSCs differentiation and provides a novel strategy in osteogenic differentiation process. | Yueqi Zhao Lin Meng Kai Zhang Yuanqing Sun Yue Zhao Zhe Yang Yangliu Lin Xinrui Liu Hongchen Sun Bai Yang Quan Lin | 2021 | Chinese Chemical Letters2021,32,1: | 1 |
| 10 | Analysis of clinical characteristics and treatment of pancreatic cystic tumors显示文摘Objective: To summarize experience in the diagnosis and treatment of pancreatic cystic neoplasms.Methods: This is a retrospective study of 207 patients who were diagnosed with pancreatic cystic tumors at Peking Union Medical College Hospital between Jan 2009 and Mar 2014.Clinical data, such as clinical manifestations, radiological and pathological images and surgical recordings, were collected.Results: Of the 207 included patients, females accounted for 76.81%, and the mean patient age was 52.04 years.Malignancy was more common in older patients who presented with marasmus and jaundice.Other risk factors included solid components in the tumor, a large tumor size, and elevated levels of tumor markers.Surgical treatment was required when a malignant tumor was suspected.The operation approach was selected based on the location, size and characteristics of the tumor.The position of the tumor relative to the pancreatic duct also played a significant role.Conclusions: No specific symptoms were observed for the patients with pancreatic cystic tumors.Imaging played an important role in making a differential diagnosis.Furthermore, surgical treatment should be proposed for patients with significant symptoms and potentially malignant tumors.The tumor resection rate is high, suggestive of good prognosis. | Lei You Jianchun Xiao Zhe Cao Wanying Zhang Quan Liao Menghua Dai Taiping Zhang Yupei Zhao | 2016 | Chinese Journal of Cancer Research2016,28,5: | 1 |
