4N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. 6Nanogenerator-Based Self-Powered Sensors for Wearable and Implantable Electronics显示文摘Wearable and implantable electronics(WIEs)are more and more important and attractive to the public,and they have had positive influences on all aspects of our lives.As a bridge between wearable electronics and their surrounding environment and users,sensors are core components of WIEs and determine the implementation of their many functions.Although the existing sensor technology has evolved to a very advanced level with the rapid progress of advanced materials and nanotechnology,most of them still need external power supply,like batteries,which could cause problems that are difficult to track,recycle,and miniaturize,as well as possible environmental pollution and health hazards.In the past decades,based upon piezoelectric,pyroelectric,and triboelectric effect,various kinds of nanogenerators(NGs)were proposed which are capable of responding to a variety of mechanical movements,such as breeze,body drive,muscle stretch,sound/ultrasound,noise,mechanical vibration,and blood flow,and they had been widely used as self-powered sensors and micro-nanoenergy and blue energy harvesters.This review focuses on the applications of self-powered generators as implantable and wearable sensors in health monitoring,biosensor,human-computer interaction,and other fields.The existing problems and future prospects are also discussed.Zhe Li Qiang Zheng Zhong Lin Wang Zhou Li 2020Research2020,,1:17

8Heterotopic pancreas in the gastrointestinal tract显示文摘Heterotopic pancreas is defined as pancreatic tissue found outside the usual anatomical location of the pancreas. It is often an incidental finding and can be found at different sites in the gastrointestinal tract. It may become clinically evident when complicated by pathological changes such as inflammation, bleeding, obstruction, and malignant transformation. In this report, a 60-year-old man with carcinoid syndrome caused by heterotopic pancreatic tissue in the duodenum is described, along with a 62-year-old man with abdominal pain caused by heterotopic pancreatic tissue in the gastric antrum. The difficulty of making an accurate diagnosis is highlighted. The patients remain healthy and symptom-free after follow-up of 1 year. Frozen sections may help in deciding the extent of resection intraoperatively. 11PCA3 and TMPRSS2-ERG gene fusions as diagnostic biomarkers for prostate cancer显示文摘The incidence of prostate cancer(PCa) is rising steadily among males in many countries. Serum prostate-specific antigen(PSA) is widely applied to clinical diagnosis and screening of PCa. However, the so-called grey area of PSA levels 4.0–10.0 ng/m L has a low specificity of 25–40% resulting in a high rate of negative biopsy and overtreatment. So in order to treat PCa patients in early stage, there is an urgent need for new biomarkers in PCa diagnosis. The PCA3 gene, a non-coding RNA(nc RNA) that is highly expressed in prostate cancer(PCa) cells, has been identified as a molecular biomarkers to detect PCa, of which PCA3 has already under clinical application. PCA3 is strongly overexpressed in malignant prostate tissue compared to benign or normal adjacent one. Newly, PCA3 is considered to be a promising biomarker in clinical diagnosis and targeted therapy. The diagnostic significance of PCA3, however, is awaiting further researches. Moreover, it has been demonstrated recently that TMPRSS2-ERG gene fusion is identified as the predominant genetic change in patients diagnosed with PCa. Recent study revealed that combination of the PCA3 and TMPRSS2-ERG gene fusion test optimizes PCa detection compared with that of single biomarker, which would lead to a considerable reduction of the number of prostate biopsies. 12m^6A-dependent biogenesis of circular RNAs in male germ cells显示文摘The majority of circular RNAs(circRNAs)spliced from coding genes contain open reading frames(ORFs)and thus,have protein coding potential.However,it remains unknown what regulates the biogenesis of these ORF-containing circRNAs,whether they are actually translated into proteins and what functions they play in specific physiological contexts.Here,we report that a large number of circRNAs are synthesized with increasing abundance when late pachytene spermatocytes develop into round and then elongating spermatids during murine spermatogenesis.For a subset of circRNAs,the back splicing appears to occur mostly at m^6A-enriched sites,which are usually located around the start and stop codons in linear mRNAs.Consequently,approximately a half of these male germ cell circRNAs contain large ORFs with m^6A-modified start codons in their junctions,features that have been recently shown to be associated with protein-coding potential.Hundreds of peptides encoded by the junction sequences of these circRNAs were detected using liquid chromatography coupled with mass spectrometry,suggesting that these circRNAs can indeed be translated into proteins in both developing(spermatocytes and spermatids)and mature(spermatozoa)male germ cells.The present study discovered not only a novel role of m^6A in the biogenesis of coding circRNAs,but also a potential mechanism to ensure stable and long-lasting protein production in the absence of linear mRNAs,i.e.,through production of circRNAs containing large ORFs and m^6A-modified start codons in junction sequences.Chong Tang Yeming Xie Tian Yu Na Liu Zhuqing Wang Rebekah J.Woolsey Yunge Tang Xinzong Zhang Weibing Qin Ying Zhang Ge Song Weiwei Zheng Juan Wang Weitian Chen Xiongyi Wei Zhe Xie Rachel Klukovich Huili Zheng David R.Quilici Wei Yan 2020Cell Research2020,30,3:12
13Effect of Magnetic Field on Tribological Properties of Lubricating Oils with and without Tricresyl Phosphate显示文摘Tribological properties of neat 150 SN mineral base oil and the oils doped with different contents of tricresyl phosphate(TCP) under magnetic field or non-magnetic field were evaluated on a four-ball tribotester, on which an external magnetic field was applied. Furthermore, the morphology and the tribochemical characteristics of the worn surfaces were examined with a scanning electron microscope(SEM) and an X-ray photoelectron spectrograph(XPS). The tribological test results showed that the magnetic field improved anti-wear properties but impaired the friction-reducing properties of neat base oil and the TCP-doped oils. The worn surfaces tested in magnetic field were characterized by a slighter wear than those tested in normal condition, especially for the surfaces lubricated with the TCP-doped oils. Furthermore, the results of XPS analysis indicated that tribochemical films on the surfaces tested with TCP-doped oils were mainly composed of ferriccontaining compounds such as Fe_2O_3, Fe_3O_4 and FePO_4. Under the influence of a magnetic field, the atomic concentrations of oxygen and phosphorous in the frictional sub-surfaces were higher than those without magnetic impact. 15Improvement of Magnetic Field on Tribological Properties of Lubricating Oils with Zinc Butyloctyldithiophosphate显示文摘Tribological properties of 150 SN mineral oil and the oils doped with different contents of zinc butyloctyldithiophosphate(T202) under magnetic field or non-magnetic field were evaluated on a four-ball tribotester by applying an external magnetic field around the friction region. Moreover, the morphology and the tribochemical characteristics of worn surfaces were examined by a scanning electron microscope(SEM) and an X-ray photoelectron spectrometer(XPS). Then the lubrication mechanisms were discussed. The tribological test results indicated that the wear scar diameters(WSDs) of steel balls lubricated by the T202-containing lubricating oils and the friction coefficients of the corresponding oil under magnetic field were smaller than those without magnetic affection. The worn surface lubricated with the T202-formulated oils in a magnetic field was smoother than that obtained under the normal condition. Furthermore, the results of XPS analysis indicated that tribochemical films on the surfaces lubricated with T202-doped oils were mainly composed of compounds such as FeSO_4, FeS and ZnS. The atomic concentrations of oxygen, sulfur, iron, zinc and phosphorus species identified in T202 under magnetic field were higher than those without magnetic impact. 17Endovascular treatment with or without intravenous alteplase for acute ischaemic stroke due to basilar artery occlusion显示文摘Background and purpose It remains controversial if endovascular treatment(EVT)can improve the outcome of patients with acute basilar artery occlusion(BAO).This study aims to compare the functional outcomes between EVT with and without intravenous thrombolysis(IVT)first in patients who had acute ischaemic stroke(AIS)due to BAO.Methods Patients who had AIS with BAO who underwent EVT within 24 hours of onset were enrolled in this multicentre cohort study,and the efficacy and safety were compared between IVT+EVT and direct EVT.The primary outcome was 90-day functional independence.All outcomes were assessed with adjusted OR(aOR)from the multivariable logistic regression.In addition,a meta-analysis was performed on all recently published pivotal studies on functional independence after EVT in patients with BAO.Results Of 310 enrolled patients with BAO,241(78%)were treated with direct EVT and 69(22%)with IVT+EVT.Direct EVT was associated with a worse functional outcome(aOR,0.46(95%CI 0.24 to 0.85),p=0.01).IVT+EVT was associated with a lower percentage of patients who needed≥3 passes of stent retriever(10.14%vs 20.75%).The meta-analysis regression revealed a potential positive correlation between bridging with IVT first and functional independence(r=0.14(95%CI 0.05 to 0.24),p<0.01).Conclusions This study showed that compared with direct EVT,EVT with IVT first was associated with better functional outcomes in patients with BAO treated within 24 hours of onset.The meta-analysis demonstrated similar favourable efficacy of IVT first followed by EVT in patients with BAO.Ximing Nie David Wang Yuehua Pu Yufei Wei Qixuan Lu Hongyi Yan Xin Liu Lina Zheng Jingyi Liu Xinxuan Yang Yarong Ding Dacheng Liu Wanying Duan Zhe Zhang Zhonghua Yang Miao Wen Weibin Gu Xinyi Hou Xinyi Leng Yuesong Pan Zhongrong Miao Liping Liu 2022Stroke & Vascular Neurology2022,7,3:10

20Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups.
