| 2 | Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. | 3 | Hf isotopes of zircon megacrysts from the Cenozoic basalts in eastern China显示文摘Cenozoic basalts are widely distributed in east- ern China, and some of them contain zircon megacrysts which are considered to be constituent mineral of the sub- continental lithospheric mantle (SCLM) and petrogenetically related to mantle metasomatism induced by addition of crustal materials. Using the Laser Ablation Multiple Collec- tor Inductively Coupled Plasma Mass Spectrometry (LA-MC-ICPMS), zircon megacrysts from the Cenozoic ba- salts at Changle in Shandong, Mingxi in Fujian, and Penglai in Hainan provinces have been used for Hf isotopic analyses. The data indicate that there is no significant deviation for the different zircon grains in each locale, except those from Penglai. The obtained 176Hf /177Hf ratios are 0.28302 — 0.28308 for Changle, 0.28297—0.28300 for Mingxi, and 0.28288—0.28293 for Penglai, with corresponding εHf values of 8.7—10.8, 7.0—7.9, and 3.9—5.7, respectively. These data display that there existed some regional heterogeneity, but the Hf model ages clustere in the Phanerozoic. Therefore, it is inferred that metasomatism of the lithospheric mantle be- neath eastern China took place in the Phanerozoic, most probably in the Mesozoic-Cenozoic. | 4 | Numerical and experimental study of magnetic fluid seal with large sealing gap and multiple magnetic sources显示文摘To improve the pressure capability of magnetic fluid seal with more than a 0.25 mm single edge gap,a magnetic fluid sealing structure with multiple magnetic sources which has five permanent magnets was designed.Magnetic field distributions under the pole pieces of the magnetic fluid seal with single and multiple magnetic sources were simulated by finite element method and its sealing pressure difference could be calculated according to the theoretical formula of the magnetic fluid seal.The effects of sealing gap height and magnetic source amount on the sealing capability were investigated experimentally.The theoretical and experimental results were compared,analyzed and discussed.The results demonstrated that the magnetic fluid seal with multiple magnetic sources was an effective method to improve the sealing capability for the rotary shaft with large gaps.The theoretical results agreed well with the experimental results when the height of the single edge gap was equal to 0.4 mm.However,it was found that when the height of the single edge gap was larger than 0.4 mm,the difference between the theoretical values and experimental values increased with the gap height because part of magnetic source had less or even no effect on magnetic fluid sealing capability. | YANG XiaoLong ZHANG ZhiLi LI DeCai | 2013 | Science China(Technological Sciences)2013,56,11: | 10 |
| 5 | Age and genesis of the Myanmar jadeite:Constraints from U-Pb ages and Hf isotopes of zircon inclusions显示文摘Myanmar jadeite (jadeitite) is well known for its economical value and distinctive tectonic locality within the collisional belt between India and Eurasian plates. However, it is less studied for its genesis and geodynamic implications due to precipitous topography, adverse weather and local military conflicts in the area. By means of combined ICP-MS and LA-MC-ICPMS techniques, we have carried out in-situ trace elements, U-Pb and Lu-Hf isotopes for zircon inclusions in a piece of jadeite gem sample. CL imaging suggests that the zircons are metasomatic in origin, and contain mineral inclusions of jadeite and omphacite. Seventy-five analyses on 16 grains of the zircons yield a U-Pb age of 158 ± 2 Ma. The Myanmar zircons differ from other types in that they have no significant Eu anomalies despite high HREE concentrations. Measured 176Hf/177Hf ratios range from 0.282976 to 0.283122, with an average value of 0.283066 ± 7; εHf(t) value of 13.8 ± 0.3 (n=75). These results indicate that the Myanmar jadeite was formed in the Late Jurassic, probably by interaction of fluid released from subducted oceanic slab with mantle wedge. | 7 | Host metabolism dysregulation and cell tropism identification in human airway and alveolar organoids upon SARS-CoV-2 infection显示文摘The coronavirus disease 2019(COVID-19)pandemic is caused by infection with the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which is spread primary via respiratory droplets and infects the lungs.Currently widely used cell lines and animals are unable to accurately mimic human physiological conditions because of the abnormal status of cell lines(transformed or cancer cells)and species differences between animals and humans.Organoids are stem cell-derived selforganized three-dimensional culture in vitro and model the physiological conditions of natural organs.Here we showed that SARS-CoV-2 infected and extensively replicated in human embryonic stem cells(hESCs)-derived lung organoids,including airway and alveolar organoids which covered the complete infection and spread route for SARS-CoV-2 within lungs.The infected ceils were ciliated,club,and alveolar type 2(AT2)cells,which were sequentially located from the proximal to the distal airway and terminal alveoli,respectively.Additionally,RNA-seq revealed early cell response to virus infection including an unexpected downregulation of the metabolic processes,especially lipid metabolism,in addition to the well-known upregulation of immune response.Further,Remdesivir and a human neutralizing antibody potently inhibited SARS-CoV-2 replication in lung organoids.Therefore,human lung organoids can serve as a pathophysiological model to investigate the underlying mechanism of SARS-CoV-2 infection and to discover and test therapeutic drugs for COVID-19. | Rongjuan Pei Jianqi Feng Yecheng Zhang Hao Sun Lian Li Xuejie Yang Jiangping He Shuqi Xiao Jin Xiong Ying Lin Kun Wen Hongwei Zhou Jiekai Chen Zhili Rong Xinwen Chen | 2021 | Protein & Cell2021,12,9: | 7 |
| 8 | Highly Efficient and Stable Hydrogen Production in All pH Range by Two-Dimensional Structured Metal-Doped Tungsten Semicarbides显示文摘Transition-metal-doped tungsten semicarbide nanosheets(M-doped W_(2)C NSs,M=Fe,Co,and Ni)have been synthesized through carburization of the mixture of tungsten trioxide,polyvinylpyrrolidone,and metal dopant.Te nanosheets grow directly on the W mesh and have the lateral dimension of several hundreds of nm to a few�m with a thickness of few tens nm.It is demonstrated that the M-doped W_(2)C NSs exhibit superior electrocatalytic activity for hydrogen evolution reaction(HER).Impressively,the Nidoped W_(2)C NSs(2 at%Ni)with the optimized HER activity show extremely low onset overpotentials of 4,9,and 19 mV and modest Tafel slopes of 39,51,and 87 mV dec−1 in acidic(pH=0),neutral(pH=7.2),and basic(pH=14)solutions,respectively,which is close to the commercial Pt/C catalyst.Density functional theory(DFT)calculations also demonstrate that the Gibbs free energy for H adsorption of Ni-W_(2)C is much closer to the optimal valueG_(H∗)=-0.073 eV as compared to-0.16 eV of W_(2)C.Furthermore,nearly 100%Faradaic efciency and long-term stability are obtained in those environments.Tis realization of highly tolerant metal semicarbide catalyst performing on par with commercial Pt/C in all range of pH ofers a key step towards industrially electrochemical water splitting. | Edison H.Ang Khang N.Dinh Xiaoli Sun Ying Huang Jun Yang Zhili Dong Xiaochen Dong Wei Huang Zhiguo Wang Hua Zhang Qingyu Yan | 2019 | Research2019,,1: | 7 |

| 10 | Synergistic optimization of thermoelectric performance in p-type Ag2Te through Cu substitution显示文摘Superionic conductors,which exhibit liquid-like phonon transport but crystal-like carrier transport,have attracted great attention and broad research interest in the thermoelectric community.Ag_(2)Te is a superionic conductor;however,its small band gap and large Ag vacancy formation energy impede its application as a prominent p-type thermoelectric material.In this work,synergistic optimization of the thermoelectric performance of Ag_(2)Te through Cu substitution is realized through a combination of experimental and theoretical efforts.For the electrical transport,Cu substitution systematically increases the band gap of Ag_(2)Te and reduces the cation vacancy formation energy.These two beneficial effects simultaneously increase the electrical conductivity and suppress the bipolar effect,thereby greatly enhancing the p-type electrical transport properties of Ag_(2)Te.For the thermal transport,alloying Cu_(2)Te with Ag_(2)Te significantly reduces the thermal conductivity through not only point defect scattering but also softening of the interatomic interactions.The latter is attributed to the relatively small Cu atoms vibrating in the oversized 8c sites.This two-fold optimization results in maximum thermoelectric figure of merit zT values of over 1.3 at 773 K for both Ag_(1.2)Cu_(0.8)Te and AgCuTe,demonstrating the great potential of Ag_(2-x)Cu_(x)Te as a promising p-type thermoelectric material system. | Ruonan Wu Zhili Li Yongbo Li Li You Pengfei Luo Jiong Yang Jun Luo | 2019 | Journal of Materiomics2019,5,3: | 4 |
| 11 | Metabolic and Phylogenetic Profile of Bacterial Community in Guishan Coastal Water(Pearl River Estuary), South China Sea显示文摘Characteristics of a microbial community are important as they indicate the status of aquatic ecosystems. In the present study, the metabolic and phylogenetic profile of the bacterioplankton community in Guishan coastal water(Pearl River Estuary), South China Sea, at 12 sites(S1–S12) were explored by community-level physiological profiling(CLPP) with BIOLOG Eco-plate and denaturing gradient gel electrophoresis(DGGE). Our results showed that the core mariculture area(S6, S7 and S8) and the sites associating with human activity and sewage discharge(S11 and S12) had higher microbial metabolic capability and bacterial community diversity than others(S1–5, S9–10). Especially, the diversity index of S11 and S12 calculated from both CLPP and DGGE data(H >3.2) was higher than that of others as sewage discharge may increase water nitrogen and phosphorus nutrient. The bacterial community structure of S6, S8, S11 and S12 was greatly influenced by total phosphorous, salinity and total nitrogen. Based on DGGE fingerprinting, proteobacteria, especially γ- and α-proteobacteria, were found dominant at all sites. In conclusion, the aquaculture area and wharf had high microbial metabolic capability. Many full-text indices require space larger than the original string. In this paper, we introduce the canonical Huffman code to the wavelet tree of a string T[1. . .n]. Compared with Huffman code based wavelet tree, the memory space used to represent the shape of wavelet tree is not needed. In case of large alphabet, this part of memory is not negligible. The operations of wavelet tree are also simpler and more efficient due to the canonical Huffman code. Based on the resulting structure, the multi-key rank and select functions can be performed using at most nH0 + jRj(lglgn + lgn lgjRj)+O(nH0) bits and in O(H0) time for average cases, where H0 is the zeroth order empirical entropy of T.
