| 1 | Fermented soybean meal improves the growth performance, nutrient digestibility, and microbial flora in piglets显示文摘In order to increase nutritive values of soybean meal(SBM), 3 species of microbes were used to ferment SBM. Through a 3 x 3 orthogonal design and parameter measurements of soybean peptide and antinutritional factor contents in the fermented soybean meal(FSBM), it was estimated that the best microbial proportion of Bacillus subtilis, Hansenula anomala and Lactobacillus casei was 2:1:2 for SBM fermentation(P < 0.05). The further piglet feeding experiment showed that 10% FSBM substitute for SBM had no significant effect on growth performance of suckling piglets(d 7-28)(P> 0.05). However, newlyweaned piglets(d 28-38) fed 10% FSBM and different levels of plasma protein obtained higher average daily gain(ADG) and feed conversion ratio(FCR), compared with those without FSBM but with 6% plasma protein(P < 0.05). Piglets(d 38-68) fed diets supplemented with FSBM and soybean protein concentrate(SBPC) at 3.75% and 7.5% respectively increased nutrient digestibility,fecal enzyme activity and lactic acid bacteria counts, and decreased fecal Escherichia coli counts(P < 0.05), compared with the control. | 3 | Synthetic lethal short hairpin RNA screening reveals that ring finger protein 183 confers resistance to trametinib in colorectal cancer cells显示文摘Background: The mitogen-activated extracellular signal-regulated kinase 1/2(MEK1/2) inhibitor trametinib has shown promising therapeutic effects on melanoma, but its efficacy on colorectal cancer(CRC) is limited. Synthetic lethality arises with a combination of two or more separate gene mutations that causes cell death, whereas individual mutations keep cells alive. This study aimed to identify the genes responsible for resistance to trametinib in CRC cells,using a synthetic lethal short hairpin RNA(shRNA) screening approach.Methods: We infected HT29 cells with a pooled lentiviral shRNA library and applied next-generation sequencing to identify shRNAs with reduced abundance after 8-day treatment of 20 nmol/L trametinib. HCT116 and HT29 cells were used in validation studies. Stable ring finger protein 183(RNF183)-overexpressing cell lines were generated by pcDNA4-myc/his-RNF183 transfection. Stable RNF 183-knockdown cell lines were generated by infection of lentiviruses that express RNF183 shRNA, and small interference RNA(siRNA) was used to knock down RNF183 transiently.Quantitative real-time PCR was used to determine the mRNA expression. Western blotting, immunohistochemical analysis, and enzyme-linked immunosorbent assay(ELISA) were used to evaluate the protein abundance. MTT assay,colony formation assay, and subcutaneous xenograft tumor growth model were used to evaluate cell proliferation.Results: In the primary screening, we found that the abundance of RNF183 shRNA was markedly reduced after treatment with trametinib. Trametinib induced the expression of RNF183, which conferred resistance to drug-induced cell growth repression and apoptotic and non-apoptotic cell deaths. Moreover, interleukin-8(IL-8) was a downstream gene of RNF183 and was required for the function of RNF183 in facilitating cell growth. Additionally, elevated RNF183 expression partly reduced the inhibitory effect of trametinib on IL-8 expression. | 13 | Fabrication and Characterization of 3D Graded PDMS Scaffolds Using Vacuum-Assisted Resin Transfer Moulding显示文摘Graded porous scaffold can be applied to study the interactions between cells and scaffold with different pore sizes. Polydimethylsiloxane(PDMS) scaffold with an axial pore size grade was successfully manufactured via vacuum-assisted resin transfer moulding(VARTM) and particle leaching technologies. The properties of graded PDMS scaffolds, including porosity, water absorption, interconnectivity, compression modulus, as well as compression strength were investigated. The results showed that the smaller the size of the NaCl particles is, the higher the porosity and water absorption of graded PDMS scaffolds would be. The graded PDMS scaffold fabricated had a compressive modulus and a compressive strength of 19.69±1.42 kPa and 4.76±0.22 kPa, respectively. Moreover, the graded chitosan(CS)-coated PDMS scaffolds were prepared by using dip-coating technique under low vacuum and their hydrophilicity was examined. | 18 | Orbital and Millennial-Scale Climate Variability over the Past 76 ka in the Western Tarim Basin, Northwest China显示文摘Paleoclimate changes during the last glacial in the arid central Asia are not as well understood as the monsoon-dominated areas of Asia. Here we report a 75-m-long sediment core over past 120 kyr based on astronomical tuning combined with the optically stimulated luminescence(OSL) dating in the Kashi depression of the western Tarim Basin, Northwest China. Analysis of grain size and highresolution gamma ray(GR) logs from the KT11 borehole across the last glacial period yields a climate history for the Tarim Basin which reflects the variations of its temperature and the hydrological cycles. Comparison of these records with north hemisphere summer insolation, the Greenland ice core temperature, stalagmites and the loess from the nearby region indicates that the deposits in the fluvio-lacustrine system of the Kashi depression responded to climate change at the Younger Dryas(YD), six Heinrich cooling events and the Dansgaard-Oeschger cycles. | 20 | Aromatic compounds-mediated synthesis of anatase-free hierarchical TS-1 zeolite:Exploring design strategies via machine learning and enhanced catalytic performance显示文摘Simultaneous achievement of constructing mesopores and eliminating anatase is a long-term pursuit for enhancing the catalytic performance of TS-1.Here,we developed an aromatic compounds-mediated synthesis method to prepare anatase-free and hierarchical TS-1 for olefin epoxidation.A series of hierarchical TS-1 zeolites were prepared by introducing aromatic compounds containing different functional groups via the crystallization process.The formation of intercrystalline mesopores and insertion of titanium into framework were facilitated at different extent.The synergistic coordination of carboxyl and hydroxyl in aromatic compounds with Ti(OH)4 realizes the uniform distribution of titanium species and eliminates the generation of anatase.Noteworthily,eight machine learning models were trained to reveal the mechanism of additive functional groups and preparation conditions on anatase formation and microstructure optimization.The prediction accuracy of most models can reach more than 80%.Benefiting from the larger mesopore volumes(0.37 cm3⋅g−1)and higher content of framework Ti species,TS-DHBDC-48h samples exhibit a higher catalytic performance than other zeolites,giving 1-hexene conversion of 49.3%and 1,2-epoxyhenane selectivity of 99.9%.The paper provides a facile aromatic compounds-mediated synthesis strategy and promotes the application of machine learning toward the design and optimization of new zeolites. | Chang’an Wang Guoqing An Jing Lin Xiaowei Zhang Zhiyuan Liu Yibin Luo Siqi Liu Zhixiang Cheng Tingting Guo Hongyi Gao Ge Wang Xingtian Shu | 2023 | Aggregate2023,4,4: | 0 |
