| 1 | Rho/ROCK pathway and neural regeneration: a potential therapeutic target for central nervous system and optic nerve damage显示文摘· Rho-associated kinase (ROCK) is a serine/threonine kinase and one of the major downstream effectors of the small GTPase RhoA. The Rho/ROCK pathway is closely related to the pathogenesis of several central nervous system (CNS) disorders, and involved in many aspects of neuronal functions including neurite outgrowth and retraction. In the adult CNS, the damaged neuron regeneration is very difficult due to the presence of myelin-associated axon growth inhibitors such as Nogo, myelin-associated glycoprotein (MAG) and oligodendrocyte-myelin glycoprotein (Omgp), etc. The effects of these axon growth inhibitors are reversed by blocking the Rho/ROCK pathway in vitro, and the inhibition of Rho/ROCK pathway can promote axon regeneration and functional recovery in the injured CNS in vivo. In addition, the therapeutic effects of the Rho/ROCK inhibitors have also been demonstrated in some animal models and the Rho/ROCK pathway becomes an attractive target for the development of drugs for treating CNS disorders. | 2 | TOTAL PHOSPHORUS RELEASE FROM BOTTOM SEDIMENTS IN FLOWING WATER显示文摘In this paper, the bottom of the Dianshan Lake was selected as a test sample. The dynamic release of contaminated sediments into the overlying water column was experimentally investigated in an open water channel under different hydrodynamic conditions. The experimental results indicate that the Total Phosphorus (TP) release process can be divided into three stages: rapid release, slow release and equilibration release. In the initial release stage the measured TP concentration changes along the depth. The TP concentration near the sediment-water interface is higher than that near the water surface, but the TP concentration becomes uniform along the depth after 3 h. The dynamic release of re-suspension sediment pollutants is about 6 times higher than the static release of sediment-water interface. | 3 | Laparoscopic total mesorectal excision of low rectal cancer with preservation of anal sphincter:A report of 82 cases显示文摘AIM: To assess the feasibility and efficacy of laparoscopic total mesorectal excision (LTME) of low rectal cancer with preservation of anal sphincter.METHODS: From June 2001 to June 2003, 82 patients with low rectal cancer underwent laparoscopic total mesorectal excision with preservation of anal sphincter. The lowest edge of tumors was below peritoneal reflection and 1.5-7 cm from the dentate line (1.5-5 cm in 48 cases, 5-7 cm in 34 cases).RESULTS: LTME with anal sphincter preservation was performed on 82 randomized patients with low rectal cancer, and 100 % sphincter preservation rate was achieved. There were 30 patients with laparoscopic low anterior resection (LLAR) at the level of the anastomosis below peritoneal reflection and 2 cm above from the dentate line; 27 patients with laparoscopic ultralow anterior resection (LULAR) at the level of anastomoses 2 cm below from the dentate line; and 25 patients with laparoscopic coloanal anastomoses (LC AA) at the level of the anastomoses at or below the dentate line. No defunctioning ileostomy was created in any case. The mean operating time was 120 minutes (ranged from 110-220 min), and the mean operative blood loss was 20 mL (ranged from 5-120 mL).Bowel function was restored and diet was resumed on day 1 or 2 after operation. The mean hospital stay was 8 days (ranged from 5-14). Postoperative analgesics were used in 45 patients. After surgery, 2 patients had urinary retention, one had anastomotic leakage, and another 2patients had local recurrence one year later. | 5 | Effect of Surface Mechanical Attrition Treatment on Corrosion Behavior of 316 Stainless Steel显示文摘The nanocrystalline microstructure of the surface of 316 stainless steel (316SS) induced by surface mechanical attrition treatment (SMAT) was determined by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The technique of hydrogen embrittlement was first used to obtain the information of the brittleness cleavage plane. The effects of SMAT and the following annealing process on the corrosion behavior of 316SS were investigated by potentiodynamic polarization curves and potentiostatic critical pitting temperature measurements. The results demonstrated that the nanocrystalline layer with an average grain size of 19 nm was produced. However, there were lots of cracks on the surface, which led to the degradation in the corrosion resistance of 316SS after SMAT. Nevertheless, after annealing treatment, the corrosion resistance of the nanocrystalline surface had been improved greatly. | 8 | Role of Notch signaling pathway in gastric cancer: A meta-analysis of the literature显示文摘AIM: To perform a meta-analysis to quantitatively summarize the evidence for the association between the Notch signaling pathway and gastric cancer(GC).METHODS: An electronic search of the MEDLINE, EMBASE and Chinese National Knowledge Infrastructure,which contain articles published from 1966 onwards,was conducted to select studies for this meta-analysis. RESULTS: Fifteen studies with a total of 1547 gastric cancer cases and 450 controls were included in this meta-analysis. Overall, the expression of Notch1,Notch2, Delta-like 4 and Hes1 was significantly higher in tumor tissues of GC compared to normal tissues.Specifically, stratified analyses showed that significantly increased expression of Notch1 was associated with non-cardia location, > 5 cm size, diffuse type, positive lymphovascular invasion and distal metastasis. Statistically significant higher expression of Notch3 was found in diffuse type GC. Jagged1 was also significantly over-expressed in diffuse type and poor differentiation type of GC. DLL4 was significantly over-expressed in advanced T stage, N stage and TNM stage in GC patients. However, the stratified analysis showed that there was no statistically significant difference in Hes1 expression between different subgroups. | 9 | Preparation of human single chain Fv antibody against hepatitis C virus E2 protein and its identification in immunohistochemistry显示文摘AIM: To screen human single chain Fv antibody (scFv)against hepatitis C virus E2 antigen and identify its applicationin immunohistochemistry.METHODS: The phage antibody library was panned by HCVE2 antigen, which was coated in microtiter plate. After fiverounds of biopanning,56 phage clones were identified specificto HCV E2 antigen. The selected scFv clones were digestedby SfiI/NotI and DNA was sequenced. Then it was subclonedinto the vector pCANTABSE for expression as E-taggedsoluble scFv. The liver tissue sections from normal personand patients with chronic hepatitis B and chronic hepatitis Cwere immunostained with HCV E2 scFv antibody.RESULTS: The data of scFv-E2 DNA digestion and DNAsequencing showed that the scFv gene is composed of 750bp. | 13 | Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. | 14 | The Prevalence of Nonalcoholic Fatty Liver Disease and its Association with Lifestyle/dietary Habits among University Faculty and Staff in Chengdu显示文摘Objective To investigate the prevalence of nonalcoholic fatty liver disease(NAFLD) in different university categories and its association with lifestyle/dietary habits.Methods A cross-sectional study was carried out on 9 378 faculty members and staff who participated in an annual health checkup at three universities selected by random cluster sampling.Demographic,anthropometric,biochemical indices and abdominal ultrasound measurements were collected.A nested case-control study was conducted with 200 NAFLD cases and 200 controls matched by gender,age(±3 years),and university.Results The overall prevalence of NAFLD was 10.3%(13.7% in males and 6.8% in females).The prevalence was significantly higher in the science and engineering university(22.1%) than in the comprehensive universities with(6.4%) and without(10.9%) medical colleges.Obesity/overweight,hyperlipidemia,diabetes mellitus,and family history of NAFLD were independently associated with higher risk of NAFLD,as were frequent consumption of desserts and salty/spicy foods.Using nutritional supplements was a protective factor against NAFLD.Intake of coarse cereals,potatoes,vegetables,fruits,and milk was significantly lower,and intake of red meat,viscera,candies and pastries,cooking oil,and total energy was significantly higher in participants with NAFLD than in controls.Conclusion Science and engineering university faculty and staff are key targets for NAFLD prevention.NAFLD is closely associated with age,gender,university type,metabolic diseases,and lifestyle/dietary habits. | SHI Lei LIU Zhong Wu LI Yun GONG Cai ZHANG Huan SONG Li Juan HUANG Cheng Yu LI Ming | 2012 | Biomedical and Environmental Sciences2012，25，4： | 16 |

On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection.
