| 6 | Binding of Divalent H-2K^(d)/IgG2aFc Fusion Protein to Murine Macrophage via Fc-FcR Interaction显示文摘Peptide-MHC class I complex (pMHC) is a specific ligand for TCR recognition, and important for CD8+ T cell activation. Here we described a genetically engineered divalent class I major histocompatibility complex (MHC) molecule, H-2Kd/IgG2aFc, a fusion protein consisting of the extracellular domains of H-2Kd, a murine MHC class I molecule, and the Fc region of IgG2a. This fusion protein is expected to attach the H-2Kd molecule to the surface of murine macrophage (Mφ) through its Fc portion binding to Fc receptor (FcR) of Mφ. cDNAs coding for the extracellular domains of H-2Kd and the Fc region of IgG2a were cloned respectively, and then recombined into plasmid pcDNA3.1(+). The H-2Kd/IgG2aFc protein was expressed by the plasmid-transfected cell line J558L, and purified from its supernatant with a Staphylococcal Protein A (SPA) column. The fusion protein showed a 58.4 kDa band as revealed by SDS-PAGE and Western blotting with murine IgG-specific antibody, which consists with that expected for extracellular domains of H-2Kd heavy chain plus the Fc region of IgG2a. The sandwich ELISA assay with antibodies specific for Fc portion and for H-2Kd indicated the fusion protein consists of both Fc portion and H-2Kd. Peritoneal Mφ of C57BL/6 (H-2Kb) can be stained with H-2Kd specific monoclonal antibody (mAb) after incubated with the H-2Kd/IgG2aFc fusion protein. | 7 | The effect of continuous Jue tone intervention on blood pressure and vasoactive substances in hypertensive rats with a liver-fire hyperactivity pattern显示文摘Objective:Jue tone is a kind of sound,using 3-mi as the main tone,and is melodious,profound,makes people feel comfortable and pleasant.This study aimed to explore the probable mechanism of Jue tone to reduce blood pressure(BP)in hypertensive rats with a liver-fire hyperactivity pattern by observing changes in BP as well as physiochemical indexes in plasma.Methods:Sixteen male spontaneous hypertensive rats(SHR)with a liver-fire hyperactivity pattern were randomly divided into a control group and a Jue tone group.The rats in the Jue tone group were treated with Jue tone(55e65 dB,played by the five elements of music rhythm instrument,a kind of physiotherapy regimen music played by Shen Wu)once a day for 4 weeks.The BP levels in each group were measured twice a week,on Monday and Friday.The levels of angiotensin II(Ang-II),thromboxane B2(TXB2),endothelin-1(ET-1),calcitonin gene-related peptide(cGRP),norepinephrine(NE),cortisol(CORT),and 5-hydroxytryptamine(5-HT).in plasma were detected by enzyme-linked immunosorbent assay after 4 weeks of intervention.Results:BP and the levels of TXB2 and ET-1 in the plasma of the treatment group were significantly lower than those of the control group,while the level of cGRP was significantly higher.Conclusion:Jue tone can reduce the BP of hypertensive rats with a liver-fire hyperactivity pattern.The mechanism may correlate with a reduction in TXB2 and ET-1 and an increase in cGRP with the reduction of reactive oxygen species(ROS). | Qian Zhou Yufeng Chen Sitong Zhang Pei Yang Tian Wang Jianxin Chen Huihui Zhao Wei Wang Xueling Ma | 2022 | Journal of Traditional Chinese Medical Sciences2022,9,1: | 0 |
| 8 | Installation error calibration for MEMS angular measurement system in hypersonic wind tunnel显示文摘The accuracy of model attitude measurement has an important impact on wind tunnel test results. Microelectromechanical System Inertial Measurement Unit(MEMS IMU) provides a feasible way to measure model attitudes with high accuracy. However, the installation error between MEMS IMU coordinate system and the body coordinate system of test models can make the accuracy of the model attitude measurement decrease. In wind tunnel tests, the installation error depends on the relationship between the IMU and the model mechanism before tests. Therefore, infield calibration in wind tunnel tests is necessary to reduce installation errors. To improve attitude measurement accuracy, the least squares quaternion calibration method based on MEMS IMU and six-position calibration procedure are proposed. High-precision three-axis turntable tests are performed. The pitch accuracy after calibration is higher than that before calibration in the angle of attack sweeping tests. The Root-Mean-Square Errors(RMSE) in the roll and yaw are within0.01°, which are smaller than those before calibration. In the roll sweeping tests, RMSE of three attitude angles decrease significantly. In hypersonic wind tunnel tests, the pitch errors before and after calibration are within 0.05° and 0.02° in the angle of attack sweeping tests without wind. In five angle of attack sweeping tests with wind, the deviation between the mean of the pitch and the pitch after the elastic angle correction is within 0.03° and the standard deviation of five tests is within 0.01°. | 9 | Influence of inherent iron and oxygen concentrations on selenite sorption process using bentonite显示文摘The oxygen concentration and inherent Fe in bentonite have a significant influence on the Se(IV) sorption process.In this study,the sorption of selenite on natural bentonite was investigated using a batch experiment method,and the distribution coefficient(K_d)values were obtained in the pH range from 2.0 to 10.0 under oxic/anoxic conditions.The K_d values always reached a maximum value at a pH of 4 under oxic conditions and at a pH of 8 under anoxic conditions;meanwhile,the K_d value under anoxic conditions was larger than the value under oxic conditions,especially in regard to the maximum K_d values.The oxygen conditions have a significant influence on the ratio of redox-sensitive Fe^(2+)/Fe^(3+),which was closely related to the difference in the K_d values under both oxic/anoxic conditions.Ferric selenite and green rust could be responsible for the maximum K_d values under oxic/anoxic conditions.In the leaching experiments,we found that the Fe^(2+) in bentonite could replace Mg^(2+) and Al^(3+) in the octahedral sheet.Spectroscopy methods,such as Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD) and X-ray absorption spectroscopy(XAS) were used to characterize the surface properties of the samples after reaction.Overall,this study shows that the addition of Fe^(2+)-containing materials into backfill/buffer materials under anoxic condition could enhance the sorption of ^(79)Se(IV). | Jiangang He Xueling Qiao Yanlin Shi Yao Li Xiaoyu Yang Wanqiang Zhou Chunli Liu | 2017 | Science China Chemistry2017,60,9: | 0 |
| 10 | Distribution and Functional Characteristics of Voltage-Gated Sodium Channels in Immature Cochlear Hair Cells显示文摘Voltage-gated sodium channels(VGSCs)are transiently expressed in cochlear hair cells before hearing onset and play an indispensable role in shaping spontaneous activity.In this study,we showed that Na^+currents shaped the spontaneous action potentials in developing mouse inner hair cells(IHCs)by decreasing the time required for the membrane potential to reach the action-potential threshold.In immature IHCs,we identified 9 known VGSC subtypes(Navl.la-l.9ot),among which Navl.7a was the most highly expressed subtype and the main contributor to Na+currents in developing hair cells.Electrophysiological recordings of two cochlea-specific Navi.7 variants(CbmNavl.7a and CbmNavl.7b)revealed a novel loss-of-function mutation(C934R)at the extracellular linker between segments 5 and 6 of domain II.In addition,post-transcriptional modification events,such as alternative splicing and RNA editing,amended the gating properties and kinetic features of CbmNavl.7a(C934).These results provide molecular and functional characteristics of VGSCs in mammalian IHCs and their contributions to spontaneous physiological activity during cochlear maturation. | You Zhou Chenchen Xia Manli Yi Xueling Wang Hao Wu Yonghua Ji | 2020 | Neuroscience Bulletin2020,36,1: | 0 |
