| 1 | Clinical importance of aspirin and clopidogrel resistance显示文摘Aspirin and clopidogrel are important components of medical therapy for patients with acute coronary syndromes, for those who received coronary artery stents and in the secondary prevention of ischaemic stroke. Despite their use, a significant number of patients experience recurrent adverse ischaemic events. Interindividual variability of platelet aggregation in response to these antiplatelet agents may be an explanation for some of these recurrent events, and small trials have linked 'aspirin and/or clopidogrel resistance', as measured by platelet function tests, to adverse events. We systematically reviewed all available evidence on the prevalence of aspirin/clopidogrel resistance, their possible risk factors and their association with clinical outcomes. We also identified articles showing possible treatments. After analyzing the data on different laboratory methods, we found that aspirin/clopidogrel resistance seems to be associated with poor clinical outcomes and there is currently no standardized or widely accepted definition of clopidogrel resistance. | 4 | Amyloid-beta peptide and tau protein crosstalk in Alzheimer’s disease显示文摘Alzheimer’s disease is a neurodegenerative disease that accounts for most of the 50-million dementia cases worldwide in 2018.A large amount of evidence supports the amyloid cascade hypothesis,which states that amyloid-beta accumulation triggers tau hyperphosphorylation and aggregation in form of neurofibrillary tangles,and these aggregates lead to inflammation,synaptic impairment,neuronal loss,and thus to cognitive decline and behavioral abnormalities.The poor correlation found between cognitive decline and amyloid plaques,have led the scientific community to question whether amyloid-beta accumulation is actually triggering neurodegeneration in Alzheimer’s disease.The occurrence of tau neurofibrillary tangles better correlates to neuronal loss and clinical symptoms and,although amyloid-beta may initiate the cascade of events,tau impairment is likely the effector molecule of neurodegeneration.Recently,it has been shown that amyloid-beta and tau cooperatively work to impair transcription of genes involved in synaptic function and,more importantly,that downregulation of tau partially reverses transcriptional perturbations.Despite mounting evidence points to an interplay between amyloid-beta and tau,some factors could independently affect both pathologies.Thus,the dual pathway hypothesis,which states that there are common upstream triggers causing both amyloid-beta and tau abnormalities has been proposed.Among others,the immune system seems to be strongly involved in amyloid-beta and tau pathologies.Other factors,as the apolipoprotein Eε4 isoform has been suggested to act as a link between amyloid-beta and tau hyperphosphorylation.Interestingly,amyloid-beta-immunotherapy reduces not only amyloid-beta but also tau levels in animal models and in clinical trials.Likewise,it has been shown that tau-immunotherapy also reduces amyloid-beta levels.Thus,even though amyloid-beta immunotherapy is more advanced than tau-immunotherapy,combined amyloid-beta and tau-directed therapies at early stages of the disease have recently been proposed as a strategy to stop the progression of Alzheimer’s disease. | Alejandro R.Roda Gabriel Serra-Mir Laia Montoliu-Gaya Lidia Tiessler Sandra Villegas | 2022 | Neural Regeneration Research2022,17,8: | 13 |
| 5 | Feature-Based Aggregation and Deep Reinforcement Learning:A Survey and Some New Implementations显示文摘In this paper we discuss policy iteration methods for approximate solution of a finite-state discounted Markov decision problem, with a focus on feature-based aggregation methods and their connection with deep reinforcement learning schemes. We introduce features of the states of the original problem, and we formulate a smaller 'aggregate' Markov decision problem, whose states relate to the features. We discuss properties and possible implementations of this type of aggregation, including a new approach to approximate policy iteration. In this approach the policy improvement operation combines feature-based aggregation with feature construction using deep neural networks or other calculations. It was difficult to clarify the molecular structures and chemical properties of asphaltene due to its structural similarity, composition complexity and source dependences. Many techniques, like Mass spectrometry, Chromatography, Nuclear magnetic resonance spectroscopy, Infrared spectroscopy, Roman spectroscopy, Fluorescence spectroscopy, X-ray diffraction analysis and Small-angle scattering techniques and so on, have revealed some valuable descriptions of asphaltenes chemical compositions and fundamental structures. Moreover, advanced Mass spectrometry,Atomic force microscopy and Scanning tunneling microscopy could provide more clear and essential molecular compounds and structures in asphaltenes. In addition, several asphaltenes models have succeeded to illustrate aggregation properties asphaltene. In this work, the progress on asphaltene separation, characterization and application was summarized, and the similarities and differences between coal-derived asphaltenes and petroleum asphaltenes were compared. Furthermore, the reactivity of asphaltenes has been discussed in the aspect of hydroprocessing, pyrolysis and gasification. Asphaltene was excellent carbon precursor for functional carbon material due to its high aromaticity and carbon yield;several porous carbon nanosheets from asphaltenes that would be prospective electrode materials after being graphitized were shown. Pitch-based carbon fiber derived from coal-derived asphaltenes displayed a tensile strength of 1.0 GPa and elastic modulus of 350 MPa, respectively. | 10 | Host compatibility interacts with seed dispersal to determine smallscale distribution of a mistletoe in Xishuangbanna,Southwest China显示文摘Aims Mistletoe infection between intra- and interspecific hosts can be restricted by seed dispersal, host–mistletoe compatibility and other factors, yet few studies have linked seed dispersal and seedling establishment together for understanding mistletoe plant distribution and demography together in different anthropogenic disturbance forest types at a local scale. The objectives of this study were to examine how three factors—seed disperser behavior, post-dispersal host compatibility and canopy cover—affect the spatial distribution of a generalist mistletoe Dendrophthoe pentandra (Loranthaceae) in plantation and rainforest within Xishuangbanna, Southwest China.MethodsWe observed mistletoe D. pentandra infection patterns at the scale of individual trees and sixteen 400-m2 forest plots in adjacent plantation and rainforest within Xishuangbanna. To elucidate what determines infection patterns at different scales and in different forest types, we observed the behavior of major avian seed dispersers and carried out a seed inoculation experiment to examine how post-dispersal compatibility and light incidence affect the infection of different hosts.Important FindingsDendrophthoe pentandra displayed an aggregated distribution and infected 10 species in our study site, with a significantly higher infection prevalence and intensity in the plantation than in the tropical forest. Different seed dispersers provided contrasting initial mistletoe templates: the specialist frugivore Dicaeum concolor (plain flowerpecker) preferred to fly between mistletoes in infected trees in the plantation and likely intensified existing infections. In contrast, the dietary generalist Pycnonotus jocosus(red-whiskered bulbul) was more likely to visit uninfected trees, thereby establishing new infections. Thus, seed dispersal appears to be an important determinant of the mistletoes distribution, with deposition patterns providing an initial distribution template and determining small-scale patterns. However, post-dispersal and abiotic factors revealed that different host compatibilities and levels of light incidence in different habitats affected the survival of D. pentandra seedlings. | 17 | Reduced β2-GPI is associated with increased platelet aggregation and activation in patients with prolonged isolated thrombocytopenia after allo-HSCT显示文摘We aimed to measure platelet function and its relationship with β2-GPI in prolonged isolated thrombocytopenia(PT) after allogeneic hematopoietic stem cell transplantation(allo-HSCT).Fifty-six patients with PT and 60 allo-HSCT recipients without PT(non-PT controls) were enrolled.Platelet aggregation and activation,β2-GPI and anti-β2-GPI antibody levels,vWF antigen,and vWF activity were analyzed.The effect of β2-GPI on platelet aggregation was also measured ex vivo.Results showed that ADP-induced platelet aggregation significantly increased(39%±7.5% vs.23%±8.5%,P=0.032),and the platelet expression of both CD62 p(33.6%±11.6% vs.8.5%±3.5%,P<0.001) and PAC-1(42.4%±7.6% vs.6.8%±2.2%,P<0.001) was significantly higher in patients with PT than in those without PT.Significantly lower β2-GPI levels(164.2±12 μg m L^-1 vs.234.2±16 μg mL^-1,P<0.001),higher anti-β2-GPI IgG levels(1.78±0.46 U mL^-1 vs.0.94±0.39 U mL^-1,P<0.001),and increased vWF activity(133.06%±30.50% vs.102.17%±25.90%,P<0.001) were observed in patients with PT than in those without PT.Both ADPinduced platelet aggregation(n=116,r^2=-0.5042,P<0.001) and vWF activity(n=116,r^2=-0.2872,P<0.001) were negatively correlated with β2-GPI levels.In summary,our data suggested that platelet aggregation and activation were significantly higher in patients with PT than in those without PT,which might be associated with reduced β2-GPI levels.The reduced β2-GPI levels might be due to the existence of anti-β2-GPI IgG. | Haixia Fu Jingzhong Zhao Lanping Xu Kaiyan Liu Yu Wang Huan Chen Wei Han Jingzhi Wang Fengrong Wang Xiaojun Huang Xiaohui Zhang | 2019 | Science China(Life Sciences)2019,62,7: | 4 |
