| 1 | Activation of gibberellin 2-oxidase 6 decreases active gibberellin levels and creates a dominant semi-dwarf phenotype in rice (Oryza sativa L.)显示文摘Gibberellin (GA) 2-oxidase plays a key role in the GA catabolic pathway through 2β-hydroxylation.In the present study,we isolated a CaMV 35S-enhancer activation tagged mutant,H032.This mutant exhibited a dominant dwarf and GA-deficient phenotype,with a final stature that was less than half of its wild-type counterpart.The endogenous bioactive GAs are markedly decreased in the H032 mutant,and application of bioactive GAs (GA3 or GA4) can reverse the dwarf phenotype.The integrated T-DNA was detected 12.8 kb upstream of the OsGA2ox6 in the H032 genome by TAIL-PCR.An increased level of OsGA2ox6 mRNA was detected at a high level in the H032 mutant,which might be due to the enhancer role of the CaMV 35S promoter.RNAi and ectopic expression analysis of OsGA2ox6 indicated that the dwarf trait and the decreased levels of bioactive GAs in the H032 mutant were a result of the up-regulation of the OsGA2ox6 gene.BLASTP analysis revealed that OsGA2ox6 belongs to the class III of GA 2-oxidases,which is a novel type of GA2ox that uses C20-GAs (GA12 and/or GA53) as the substrates.Interestingly,we found that a GA biosynthesis inhibitor,paclobutrazol,positively regulated the OsGA2ox6 gene.Unlike the over-expression of OsGA2ox1,which led to a high rate of seed abortion,the H032 mutant retained normal flowering and seed production.These results indicate that OsGA2ox6 mainly affects plant stature,and the dominant dwarf trait of the H032 mutant can be used as an efficient dwarf resource in rice breeding. | Jian Huang Ding Tang Yi Shen Baoxiang Qin Lilan Hong Aiqing You Ming Li Xin Wang Hengxiu Yu Minghong Gu Zhukuan Cheng | 2010 | Journal of Genetics and Genomics2010,37,1: | 36 |

| 5 | Integral Barrier Lyapunov function-based adaptive control for switched nonlinear systems显示文摘This paper presents an adaptive control method for a class of uncertain strict-feedback switched nonlinear systems. First, we consider the constraint characteristics in the switched nonlinear systems to ensure that all states in switched systems do not violate the constraint ranges. Second, we design the controller based on the backstepping technique, while integral Barrier Lyapunov functions(i BLFs) are adopted to solve the full state constraint problems in each step in order to realize the direct constraints on state variables.Furthermore, we introduce the Lyapunov stability theory to demonstrate that the adaptive controller achieves the desired control goals. | 6 | Molecular analysis of the chloroplast Cu/Zn-SOD gene(AhCSD2) in peanut显示文摘Superoxide dismutase(SOD, EC 1.15.1.1) plays a key role in response to drought stress, and differences in SOD activity changes among cultivars are important under drought conditions. We obtained the full-length DNA of the chloroplast Cu/Zn-SOD gene(Ah CSD2)from 11 allotetraploid cultivars and 5 diploid wild species in peanut. BLAST search against the peanut genome showed that the Ah CSD2 genes g CSD2-1 and g CSD2-2 are located at the tops of chromosome A03(A genome) and B03(B genome), respectively, and both contain 8exons and 7 introns. Nucleotide sequence analyses indicated that g CSD2-2 sequences were identical among all the tested cultivars, while g CSD2-1 sequences showed allelic variations.The amino acid sequences deduced from g CSD2-1 and g CSD2-2 both contain a chloroplast transit peptide and are distinguished by 6 amino acid(aa) residue differences. The other 2aa residue variations in the mature peptide regions give rise to three-dimensional structure changes of the protein deduced from the genes g CSD2-1 and g CSD2-2. Sequences analyses of cultivars and wild species showed that g CSD2-2 of Arachis hypogaea and g Aip CSD2(Arachis ipaensis) are identical, and despite the abundant polymorphic loci between g CSD2-1 of A.hypogaea and sequences from A genome wild species, the deduced amino acid sequence of Ah CSD2-1(A. hypogaea) is identical to that of Adu CSD2(Arachis duranensis), whereas Aco CSD2(Arachis correntina) and Aca CSD2(Arachis cardenasii) both have 2 aa differences in the transit peptide region compared with Ah CSD2-1(A. hypogaea). Based on the Peanut Genome Project, promoter prediction revealed many stress-related cis-acting elements within the potential promoter regions(pp-A and pp-B). pp-A contains more binding sites for drought-associated transcriptional factors than pp-B. | 7 | Engineering rock mechanics practices in the underground powerhouse at JinpingⅠhydropower station显示文摘Based on the analyses of data obtained from the underground powerhouse at Jinping Ⅰ hydropower station,a comprehensive review of engineering rock mechanics practice in the underground powerhouse is first conducted.The distribution of strata,lithology,and initial geo-stress,the excavation process and corresponding rock mass support measures,the deformation and failure characteristics of the surrounding rock mass,the stress characteristics of anchorage structures in the cavern complex,and numerical simulations of surrounding rock mass stability and anchor support performance are presented.The results indicate that the underground powerhouse of Jinping Ⅰ hydropower station is characterized by high to extremely high geo-stresses during rock excavation.Excessive surrounding rock mass deformation and high stress of anchorage structures,surrounding rock mass unloading damage,and local cracking failure of surrounding rock masses,etc.,are mainly caused by rock mass excavation.Deformations of surrounding rock masses and stresses in anchorage structures here are larger than those found elsewhere:20%of extensometers in the main powerhouse record more than 50 mm with the maximum at around 250 mm observed in the downstream sidewall of the transformer hall.There are about 25%of the anchor bolts having recorded stresses of more than 200 MPa.Jinping Ⅰ hydropower plant is the first to have an underground powerhouse construction conducted in host rocks under extremely high geo-stress conditions,with the ratio of rock mass strength to geo-stress of less than 2.0.The results can provide a reference to underground powerhouse construction in similar geological conditions. | Aiqing Wu Jimin Wang Zhong Zhou Shuling Huang Xiuli Ding Zhihong Dong Yuting Zhang | 2016 | Journal of Rock Mechanics and Geotechnical Engineering2016,8,5: | 4 |
| 8 | A linearly-independent higher-order extended numerical manifold method and its application to multiple crack growth simulation显示文摘The numerical manifold method(NMM)can be viewed as an inherent continuous-discontinuous numerical method,which is based on two cover systems including mathematical and physical covers.Higher-order NMM that adopts higher-order polynomials as its local approximations generally shows higher precision than zero-order NMM whose local approximations are constants.Therefore,higherorder NMM will be an excellent choice for crack propagation problem which requires higher stress accuracy.In addition,it is crucial to improve the stress accuracy around the crack tip for determining the direction of crack growth according to the maximum circumferential stress criterion in fracture mechanics.Thus,some other enriched local approximations are introduced to model the stress singularity at the crack tip.Generally,higher-order NMM,especially first-order NMM wherein local approximations are first-order polynomials,has the linear dependence problems as other partition of unit(PUM)based numerical methods does.To overcome this problem,an extended NMM is developed based on a new local approximation derived from the triangular plate element in the finite element method(FEM),which has no linear dependence issue.Meanwhile,the stresses at the nodes of mathematical mesh(the nodal stresses in FEM)are continuous and the degrees of freedom defined on the physical patches are physically meaningful.Next,the extended NMM is employed to solve multiple crack propagation problems.It shows that the fracture mechanics requirement and mechanical equilibrium can be satisfied by the trial-and-error method and the adjustment of the load multiplier in the process of crack propagation.Four numerical examples are illustrated to verify the feasibility of the proposed extended NMM.The numerical examples indicate that the crack growths simulated by the extended NMM are in good accordance with the reference solutions.Thus the effectiveness and correctness of the developed NMM have been validated. | Dongdong Xu Aiqing Wu Cong Li | 2019 | Journal of Rock Mechanics and Geotechnical Engineering2019,11,6: | 4 |
| 9 | Cloning and characterization of rice RH3 gene induced by brown planthopper显示文摘Experiments have showed that the histone H3 gene is correlated with development, cell speciality and stress response. The RH3 full-length cDNA was isolated from the cDNA library of rice infested by brown planthopper (BPH) with EST (Accession no. BU572343) screened from rice SSH library as probe. This gene encodes histone H3 protein in-cluding 136 amino acids, with one amino acid different from a kind of disease resistance-related protein in rice (AF467728). At the position 126, the aspartic acid is replaced by lysine. The time course results showed that the expression of the RH3 began to increase at 8 h after BPH-feeding, and got to its peak at 96 h. Regulations of the gene expression in treatments with stress/defense signal molecules were ana-lyzed by Northern blot. Water deficit and Pyricularia grisea increased the expression of RH3 while ABA down-regulated the gene. The enhanced accumulation of RH3 transcripts in the vascular bundle and short cell of stem after BPH feeding was revealed by RNA in situ hybridization. Based on the analysis of these data, the variations of atmospheric compositions in solar and geomagnetic quiet conditions are reported first, then a detailed discussion on the atmospheric composition variations under the so lar and geomagnetic disturbed conditions is given. The results show that near the altitude of 400 km the variations of main atmospheric compositions corre sponding to solar disturbances are more remarkable in the sunlit area than in the shade area.
