| 1 | Sequencing of Cultivated Peanut, Arachis hypogaea, Yields Insights into Genome Evolution and Oil Improvement显示文摘Cultivated peanut (Arachis hypogaea) is an allotetraploid crop planted in Asia, Africa, and America for edible oil and protein. To explore the origins and consequences of tetraploidy, we sequenced the allotetraploid A. hypogaea genome and compared it with the related diploid Arachis duranensis and Arachis ipaensis genomes. We annotated 39 888 A-subgenome genes and 41 526 B-subgenome genes in allotetraploid peanut. The A. hypogaea subgenomes have evolved asymmetrically, with the B subgenome resembling the ancestral state and the A subgenome undergoing more gene disruption, loss, conversion, and transposable element proliferation, and having reduced gene expression during seed development despite lacking genome-wide expression dominance. Genomic and transcriptomic analyses identified more than 2 500 oil metabolism-related genes and revealed that most of them show altered expression early in seed development while their expression ceases during desiccation, presenting a comprehensive map of peanut lipid biosynthesis. | 4 | Tribological behaviour diagnostic and fault detection of mechanical seals based on acoustic emission measurements显示文摘Acoustic emission(AE) has been studied for monitoring the condition of mechanical seals by many researchers, however to the best knowledge of the authors, typical fault cases and their effects on tribological behaviour of mechanical seals have not yet been successfully investigated. In this paper, AE signatures from common faults of mechanical seals are studied in association with tribological behaviour of sealing gap to develop more reliable condition monitoring approaches. A purpose-built test rig was employed for recording AE signals from the mechanical seals under healthy and faulty conditions. The collected data was then processed using time domain and frequency domain analysis methods. The study has shown that AE signal parameters: root mean squared(RMS) along with AE spectrum, allows fault conditions including dry running, spring out and defective seal faces to be diagnosed under a wide range of operating conditions. However, when mechanical seals operate around their transition point, conventional signal processing methods may not allow a clear separation of the fault conditions from the healthy baseline. | 5 | White Matter Abnormalities in Major Depression Biotypes Identified by Diffusion Tensor Imaging显示文摘Identifying data-driven biotypes of major depressive disorder(MDD) has promise for the clarification of diagnostic heterogeneity. However, few studies have focused on white-matter abnormalities for MDD subtyping. This study included 116 patients with MDD and118 demographically-matched healthy controls assessed by diffusion tensor imaging and neurocognitive evaluation.Hierarchical clustering was applied to the major fiber tracts, in conjunction with tract-based spatial statistics, to reveal white-matter alterations associated with MDD.Clinical and neurocognitive differences were compared between identified subgroups and healthy controls. With fractional anisotropy extracted from 20 fiber tracts, cluster analysis revealed 3 subgroups based on the patterns of abnormalities. Patients in each subgroup versus healthy controls showed a stepwise pattern of white-matter alterations as follows: subgroup 1(25.9% of patient sample),widespread white-matter disruption;subgroup 2(43.1% of patient sample), intermediate and more localized abnormalities in aspects of the corpus callosum and left cingulate;and subgroup 3(31.0% of patient sample),possible mild alterations, but no statistically significant tract disruption after controlling for family-wise error. The neurocognitive impairment in each subgroup accompanied the white-matter alterations: subgroup 1, deficits in sustained attention and delayed memory;subgroup 2, dysfunction in delayed memory;and subgroup 3, no significant deficits. | 6 | Neurocognitive Graphs of First-Episode Schizophrenia and Major Depression Based on Cognitive Features显示文摘Neurocognitive deficits are frequently observed in patients with schizophrenia and major depressive disorder(MDD). The relations between cognitive features may be represented by neurocognitive graphs based on cognitive features, modeled as Gaussian Markov random fields. However, it is unclear whether it is possible to differentiate between phenotypic patterns associated with the differential diagnosis of schizophrenia and depression using this neurocognitive graph approach. In this study, we enrolled 215 first-episode patients with schizophrenia(FES), 125 with MDD, and 237 demographically-matched healthy controls(HCs). The cognitive performance of all participants was evaluated using a battery of neurocognitive tests. The graphical LASSO model was trained with aone-vs-one scenario to learn the conditional independent structure of neurocognitive features of each group. Participants in the holdout dataset were classified into different groups with the highest likelihood. A partial correlation matrix was transformed from the graphical model to further explore the neurocognitive graph for each group. The classification approach identified the diagnostic class for individuals with an average accuracy of 73.41% for FES vs HC, 67.07% for MDD vs HC, and 59.48% for FES vs MDD. Both of the neurocognitive graphs for FES and MDD had more connections and higher node centrality than those for HC. Mutagenesis is an important tool in crop improvement and is free of the regulatory restrictions imposed on genetically modified organisms. Targeting Induced Local Lesions in Genomes(TILLING), which combines traditional chemical mutagenesis with high‐throughput genome‐wide screening for point mutations in desired genes, offers a powerful way to create novel mutant alleles for both functional genomics and improvement of crops. TILLING is generally applicable to genomes whether small or large, diploid or evenallohexaploid, and shows great potential to address the major challenge of linking sequence information to the function of genes and to modulate key traits for plant breeding. TILLING has been successfully applied in many crop species and recent progress in TILLING is summarized below, especially on the developments in mutation detection technology, application of TILLING in gene functional studies and crop breeding. The potential of TILLING/EcoTILLING for functional genetics and crop improvement is also discussed. | 12 | Quantification of occlusions influencing the tree stem curve retrieving from single-scan terrestrial laser scanning data显示文摘Background: The stem curve of standing trees is an essential parameter for accurate estimation of stem volume.This study aims to directly quantify the occlusions within the single-scan terrestrial laser scanning(TLS) data,evaluate its correlation with the accuracy of the retrieved stem curves, and subsequently, to assess the capacity of single-scan TLS to estimate stem curves.Methods: We proposed an index, occlusion rate, to quantify the occlusion level in TLS data. We then analyzed three influencing factors for the occlusion rate: the percentage of basal area near the scanning center, the scanning distance and the source of occlusions. Finally, we evaluated the effects of occlusions on stem curve estimates from single-scan TLS data.Results: The results showed that the correlations between the occlusion rate and the stem curve estimation accuracies were strong(r = 0.60–0.83), so was the correlations between the occlusion rate and its influencing factors(r = 0.84–0.99). It also showed that the occlusions from tree stems were the main factor of the low detection rate of stems, while the non-stem components mainly influenced the completeness of the retrieved stem curves.Conclusions: Our study demonstrates that the occlusions significantly affect the accuracy of stem curve retrieval from the single-scan TLS data in a typical-size(32 m × 32 m) forest plot. However, the single-scan mode has the capacity to accurately estimate the stem curve in a small forest plot(< 10 m × 10 m) or a plot with a lower occlusion rate, such as less than 35% in our tested datasets. | 13 | Desiccation tolerance in bryophytes: the rehydration proteomes of Bryum argenteum provide insights into the resuscitation mechanism显示文摘Bryum argenteum Hedw. is a desiccation tolerant bryophyte and belongs to one of the most important components of the biological soil crusts(BSCs) found in the deserts of Central Asia. Limited information is available on rehydration-responsive proteins in desiccation tolerant plants. As a complement to our previous research analyzing the rehydration transcriptome, we present a parallel quantitative proteomic effort to study rehydration-responsive proteins. Bryophyte gametophores were desiccated(Dry) and rehydrated for 2 h(R2) and 24 h(R24). Proteins from Dry, R2 and R24 gametophores were labeled by isobaric tags for relative and absolute quantitation(iTRAQ) to determine the relative abundance of rehydration-responsive proteins. A total of 5503 non-redundant protein sequences were identified and 4772(86.7%) protein sequences were annotated using Gene Ontology(GO) terms and Pfam classifications. Upon rehydration 239 proteins were elevated and 461 proteins were reduced as compared to the desiccated protein sample. Differentially up-regulated proteins were classified into a number of categories including reactive oxygen species scavenging enzymes, detoxifying enzymes, Late Embryogenesis Abundant(LEA) proteins, heat shock proteins, proteasome components and proteases, and photosynthesis and translation related proteins. | 15 | Comparative outcomes of subcutaneous and transvenous cardioverter-defibrillators显示文摘Background: The comparative outcomes of subcutaneous implantable cardioverter-defibrillator (S-ICD) and transvenous (T-ICD) have not been well studied. The aim of this study was to evaluate the safety and efficacy of currently available S-ICD and T-ICD. Methods: The study included 86 patients who received an S-ICD and 1:1 matched to those who received single-chamber T-ICD by gender, age, diagnosis, left ventricular ejection fraction (LVEF), and implant year. The clinical outcomes and implant complications were compared between the two groups. Results: The mean age of the 172 patients was 45 years, and 129 (75%) were male. The most common cardiac condition was hypertrophic cardiomyopathy (HCM, 37.8%). The mean LVEF was 50%. At a mean follow-up of 23 months, the appropriate and inappropriate ICD therapy rate were 1.2% vs. 4.7%(X^2= 1.854, P=0.368) and 9.3% vs. 3.5%(X^2 = 2.428, P = 0.211) in S-ICD and T-ICD groups respectively. There were no significant differences in device-related major and minor complications between the two groups (7.0% vs. 3.5%, X^2 = 1.055, P = 0.496). The S-ICD group had higher T-wave oversensing than T-ICD group (9.3% vs. 0%, X^2 = 8.390, P=0.007). Sixty-five patients had HCM (32 in S-ICD and 33 in T-ICD). The incidence of major complications was not significantly different between the two groups. Conclusions: The efficacy of an S-ICD is comparable to that of T-ICD, especially in a dominantly HCM patient population.
