2HOXC10 up-regulation promotes gastric cancer cell proliferation and metastasis through MAPK pathway显示文摘Objective: As an important regulator of embryonic morphogenesis,homeodomain-containing gene 10(HOXC10) has been found to promote progression of human cancers and its expression indicates poor survival outcome.However,very few studies are available on the role of HOXC10 in gastric carcinoma.Therefore,the aim of this study was to determine the role of HOXC10 in gastric cancer and the potential mechanism underlying its function for cancer biology.Methods: A primary gastric cancer mouse model was obtained via intra-gastric wall injection of gastric cancer cells and was used to evaluate the function of HOXC10 during gastric cancer progression in vivo.Immunohistochemistry was performed to visualize and measure HOXC10 protein expression in gastric cancer tissue.Cells were transfected with plasmids to increase the expression of HOXC10,and si RNA transfection was performed to suppress HOXC10 expression.Reverse transcription polymerase chain reaction(RT-PCR) and western blotting were utilized to measure m RNA and protein expression,respectively.Proliferation,migration,and invasion were investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay,wound healing assay,and matrigel invasion assay in vitro,respectively.Results: HOXC10 expression was significantly increased in gastric cancer tissues compared to matched normal tissues.HOXC10 up-regulation significantly increased tumor volumes in nude mice.Plasmid transfection significantly increased HOXC10 protein and m RNA expressions and effectively promoted cell proliferation.Moreover,HOXC10 up-regulation significantly promoted migration and invasion of gastric cancer cells.Mechanistic investigation showed that HOXC10 up-regulation significantly increased m RNA and protein expression of mitogen-activated protein kinase(MAPK) signaling related genes,including c-myc,c-jun and p53,while also modulating the phosphorylation of c-Jun N-terminal kinase(JNK),extracellular signal-regulated kinase(ERK) and P38 but not their total protein levels.Conclusions: This study demonstrated the tight link between HOXC10 and gastric cancer cell proliferation and metastasis via involvement of the MAPK pathway.Chen Guo Jianing Hou Sheng Ao Xingming Deng Guoqing Lyu 2017Chinese Journal of Cancer Research2017,29,6:10
3Joint contractures in severe burn patients with early rehabilitation intervention in one of the largest burn intensive care unit in China:a descriptive analysis显示文摘Background:Joint contracture is the major clinical complication in burn patients,especially,the severe burn patients.This study aimed to investigate the number and severity of joint contractures in patients with burns affecting greater than or equal to 50%of the total body surface area(TBSA)undergoing early rehabilitation in a burn intensive care unit(BICU).Methods:We analyzed burn patients with burns affecting greater than or equal to 50%of the TBSA admitted to a BICU who received early rehabilitation within 7 days post-injury from January 2011 to December 2015.Demographic and medical information was collected.The range of motion(ROM)of different joints was measured 1 month post-admission.Spearman’s correlation coefficient and logistic regression analysis was used to determine predictors of the presence and severity of contractures.Result:The average affected TBSA of the included burn patients was 67.4%,and the average length of stay in the BICU was 46.2±28.8 days.One hundred and one of 108 burn patients(93.5%)developed at least one joint contracture.The ROM in 67.9%of the affected joints was mildly limited.The majority of contractures in severe burn patients were mild(37.7%)or moderate(33.2%).The wrist was the most commonly affected joint(18.2%),followed by the shoulder,ankle,hip,knee,and elbow.A predictor of the presence of contractures was the length of hospital stay(p=0.049).The severe contracture was related to the area of full-thickness burns,the strict bed rest time,and the duration of rehabilitation in BICU.The length of rehabilitation stay(days)in patients with moderate contracture is 54.5%longer than that in severe contracture(p=0.024)Conclusion:During the long stay in BICU,the length of rehabilitation stay in a BICU could decrease the severity of contractures from severe to moderate in the patients with equal to 50%of the TBSA.Hence,this research reveals the important role of early rehabilitation interventions in severe burn patients.Jianglin Tan Jian Chen Junyi Zhou Huapei Song Huan Deng Ming Ao Gaoxing Luo Jun Wu 2019Burns & Trauma2019,7,1:9
4Tourism sectorization opportunity spectrum model and space partition of tourism urbanization area:a case of the Mayangxi ecotourism area,Fujian province,China显示文摘Nowadays, tourism has become an increasingly significant driving force of modern urbanization in China. However, it has caused varieties of problems, such as resource unreasonable utilization, society unfair play, and environment pollution, etc. Thus, the establishment of an efficient and sustainable space partition model for tourism urbanization, from the geographical perspective, is highly necessary and urgent. Herein, a new evaluation model of tourist town space, referred to as Tourism Sectorization Opportunity Spectrum(TSOS), is established on the relationships between productionliving-ecology spaces. TSOS is a kind of quantitative evaluation model involving such factors as urban living area, ecological conservation area and the tourism industry. Furthermore, the analysis of quantitative production-living-ecology spatial overlap has been utilized for space division in a tourism urbanization area. This paper, using both the TSOS model and GIS technology, analyzes the spatial restructure of the Mayangxi ecotourism area. Theresult demonstrates that tourism industrial space takes the Shanchong Village as a core which shows a pattern of local regional agglomeration and a fuzzy edge extending along the entire Mayangxi River. The urban living area should be consolidated in the Shili village with the supporting facilities and living space for tourism. In addition, Tianzhu Mountain forest park should be the key environmental protection area. The case study on the Mayangxi ecotourism area verifies that the TSOS model has strong operability. 5Nutritional Quality of Mixed Silage of Lablab purpureus and Sweet Sorghum显示文摘[Objective] The paper was to evaluate the fermentation quality of mixed silage of Lablab purpureus and sweet sorghum,and to find out the appropriate mixing ratio.[Method]L.purpureus were mixed with sweet sorghum at different proportions,to identify the sensory character and quality of silage.[Result]The nutrient content of mixed silage of L.purpureus and sweet sorghum at different proportions decreased significantly at 30-60 d,while no significant changes were observed after 60 d.Mixed silage of L.purpureus and sweet sorghum had the best effort at the proportion of 3∶7; followed by the proportion of 5∶5.These two proportions significantly improved dry matter( DM) content and effectively alleviated the crude protein( CP) loss of raw materials; significantly improved the contents of crude fiber and crude ash; and significantly reduce ammonia nitrogen/total nitrogen( AT/TN).[Conclusion]From the perspective of silage quality,the appropriate mixing ratio of L.purpureus and sweet sorghum is 3∶7 or 5∶5.Liu Hailin Deng Huifen Xu Huaqin Sun Ao 2017Animal Husbandry and Feed Science2017,9,6:4
6Flight dynamics characteristics of canard rotor/wing aircraft in helicopter flight mode显示文摘The aerodynamic layout of the Canard Rotor/Wing(CRW) aircraft in helicopter flight mode differs significantly from that of conventional helicopters. In order to study the flight dynamics characteristics of CRW aircraft in helicopter mode, first, the aerodynamic model of the main rotor system is established based on the blade element theory and wind tunnel test results. The aerodynamic forces and moments of the canard wing, horizontal tail, vertical tail and fuselage are obtained via theoretical analysis and empirical formula. The flight dynamics model of the CRW aircraft in helicopter mode is developed and validated by flight test data. Next, a method of model trimming using an optimization algorithm is proposed. The flight dynamics characteristics of the CRW are investigated by the method of linearized small perturbations via Simulink. The trim results are consistent with the conventional helicopter characteristics, and the results show that with increasing forward flight speed, the canard wing and horizontal tail can provide considerable lift,which reflects the unique characteristics of the CRW aircraft. Finally, mode analysis is implemented for the linearized CRW in helicopter mode. The results demonstrate that the stability of majority modes increases with increasing flight speed. However, one mode that diverges monotonously,and the reason is that the CRW helicopter mode has a large vertical tail compared to the conventional helicopter. 7An Inverse Power Generation Mechanism Based Fruit Fly Algorithm for Function Optimization显示文摘As a novel population-based optimization algorithm, fruit fly optimization(FFO) algorithm is inspired by the foraging behavior of fruit flies and possesses the advantages of simple search operations and easy implementation. Just like most population-based evolutionary algorithms, the basic FFO also suffers from being trapped in local optima for function optimization due to premature convergence.In this paper, an improved FFO, named IPGS-FFO, is proposed in which two novel strategies are incorporated into the conventional FFO. Specifically, a smell sensitivity parameter together with an inverse power generation mechanism(IPGS) is introduced to enhance local exploitation. Moreover,a dynamic shrinking search radius strategy is incorporated so as to enhance the global exploration over search space by adaptively adjusting the searching area in the problem domain. The statistical performance of FFO, the proposed IPGS-FFO, three state-of-the-art FFO variants, and six metaheuristics are tested on twenty-six well-known unimodal and multimodal benchmark functions with dimension 30, respectively. 8Cholesterylation of Smoothened is a calcium-accelerated autoreaction involving an intramolecular ester intermediate显示文摘Hedgehog(Hh)is a morphogen that binds to its receptor Patched 1 and activates Smoothened(SMO),thereby governing embryonic development and postnatal tissue homeostasis.Cholesterol can bind and covalently conjugate to the luminal cysteine-rich domain(CRD)of human SMO at the D95 residue(D99 in mouse).The reaction mechanism and biological function of SMO cholesterylation have not been elucidated.Here,we show that the SMO-CRD undergoes auto cholesterylation which is boosted by calcium and involves an intramolecular ester intermediate.In cells,Hh stimulation elevates local calcium concentration in the SMO-localized endosomes through store-operated calcium entry.In addition,we identify the signaling-incompetent SMO D95E mutation,and the D95E mutant SMO can bind cholesterol but cannot be modifed or activated by cholesterol.The homozygous Smo^(O99E/D99E) knockin mice are embryonic lethal with severe developmental delay,demonstrating that cholesterylation of CRD is required for full-length SMO activation.Our work reveals the unique autocatalytic mechanism of SMO cholesterylation and an unprecedented role of calcium in Hh signaling.Ao Hu Jing-Zan Zhang Jie Wang Chen-Chen Li Meng Yuan Gang Deng Zi-Cun Lin Zhi-Ping Qiu Hu-Yue Liu Xian-Wei Wang Peng-Cheng Wei Xiao He Xiaolu Zhao Wen-Wei Qiu Bao-Liang Song 2022Cell Research2022,32,3:3

10A hydrothermal investigation system for the Qianlong-Ⅱ autonomous underwater vehicle显示文摘Qianlong-Ⅱ is a fully autonomous underwater vehicle designed for the investigation of submarine resources,particularly polymetallic sulfides. It was used to successfully explore hydrothermal fields on the Southwest Indian Ridge. Here, we summarized the exploration of hydrothermal systems using Qianlong-Ⅱ, including detailed descriptions of its implementation along with the systems used for data management and fast mapping. We also introduced a method to remove platform magnetic interference using magnetic data while Qianlong-Ⅱ is spinning. Based on hydrothermal anomalies collected by Qianlong-Ⅱ, we developed a rapid method for locating hydrothermal vents. 11Comparative Study of Three Warm-season Forage Varieties in Southern China显示文摘The plant height,tiller number,yield of three warm-season forage varieties in Southern China were studied for consecutive two years.The result showed that the three varieties all adapted to the environmental conditions.The annual fresh grass yield of Pennisetum purpureum Schum ev.Taiwan,Guimu-1 hybrid P.purpureum and P.sinese Roxb were 220.85,163.18,186.20 t/hm^2;the crude protein content were 17.39%,19.02%and 20.91%;the crude fat content were 6.37%,5.37%and 4.69%;and the crude fiber content were 33.49%,32.22%and 33.58%,respectively.P.purpureum Schum ev.Taiwan had stronger cold resistance,fast growth after overwintering and strong resistance,and no plant diseases and insect pests were found during the test.P.purpureum Schum ev.Taiwan is a high yield and high quality forage variety,which is worth popularizing and applying.Sun Ao Deng Huifen Li Caijun Jiang Guitao Zhang Xu Li Jianbo 2019Animal Husbandry and Feed Science2019,11,3:2
12High birth weight and its interaction with physical activity influence the risk of obesity in early school-aged children显示文摘Background High birth weight(HBW)is associated with childhood obesity,but with inconsistent results.This study investigated the relationship between HBW and childhood obesity,and further explored the interaction of HBW with behavioral and socio-economic determinants of obesity.Methods This cross-sectional study enrolled 1906 grade-two children of Guangzhou,China,from June to November,2016.Overweight/obesity corresponded to a body mass index higher than the sex–age-specific criteria.Abdominal obesity was assessed using the sex-specific waist–height ratio cutoffs.The association of HBW with obesity was evaluated in multivariable logistic regression model.The relative excess risk due to interaction(RERI)and the attributable proportion of interaction(AP)indices were used to measure additive interaction,while applying the interaction of OR index for multiplicative interaction assessment.Results Children with HBW had an increased risk of overweight/obesity[odds ratio(OR)=2.42,95%confidence interval(CI)=1.56–3.76]compared with those without HBW.Significant additive interaction of HBW with physical activity was found for overweight/obesity[relative excess risk due to interaction(RERI)=2.69,95%CI=0.62–4.75;attributable proportion of interaction(AP)=0.72,95%CI=0.42–1.02].The HBW children with insufficient activity had higher odds of overweight/obesity compared to the non-HBW children with sufficient activity(OR=3.75,95%CI=2.06–6.83).In addition,we identified a significant interaction of HBW with household income for abdominal obesity(RERI=1.20,95%CI=0.02–2.37;AP=0.76,95%CI=0.16–1.36).Conclusions HBW confers an increased risk for childhood overweight/obesity.Physical activity attenuates the effect of HBW on overweight/obesity,and HBW possibly synergistically interacts with high household income to promote abdominal obesity in childhood.Jia-Ru Deng Wei-Qing Tan Shu-Yi Yang Li-Ping Ao Jian-Ping Liang Li-Xia Li Yan-Hui Gao Yi Yang Li Liu 2020World Journal of Pediatrics2020,16,4:2

16DOA estimation of incoherently distributed sources using importance sampling maximum likelihood显示文摘In this paper, an importance sampling maximum likelihood(ISML) estimator for direction-of-arrival(DOA) of incoherently distributed(ID) sources is proposed. Starting from the maximum likelihood estimation description of the uniform linear array(ULA), a decoupled concentrated likelihood function(CLF) is presented. A new objective function based on CLF which can obtain a closed-form solution of global maximum is constructed according to Pincus theorem. To obtain the optimal value of the objective function which is a complex high-dimensional integral,we propose an importance sampling approach based on Monte Carlo random calculation. Next, an importance function is derived, which can simplify the problem of generating random vector from a high-dimensional probability density function(PDF) to generate random variable from a one-dimensional PDF. Compared with the existing maximum likelihood(ML) algorithms for DOA estimation of ID sources, the proposed algorithm does not require initial estimates, and its performance is closer to CramerRao lower bound(CRLB).
