| 1 | Adipose-derived mesenchymal stem cell transplantation promotes adult neurogenesis in the brains of Alzheimer's disease mice显示文摘In the present study,we transplanted adipose-derived mesenchymal stem cells into the hippocampi of APP/PS1 transgenic Alzheimer’s disease model mice.Immunofluorescence staining revealed that the number of newly generated(BrdU+)cells in the subgranular zone of the dentate gyrus in the hippocampus was significantly higher in Alzheimer’s disease mice after adipose-derived mesenchymal stem cell transplantation,and there was also a significant increase in the number of BrdU+/DCX+neuroblasts in these animals.Adipose-derived mesenchymal stem cell transplantation enhanced neurogenic activity in the subventricular zone as well.Furthermore,adipose-derived mesenchymal stem cell transplantation reduced oxidative stress and alleviated cognitive impairment in the mice.Based on these findings,we propose that adipose-derived mesenchymal stem cell transplantation enhances endogenous neurogenesis in both the subgranular and subventricular zones in APP/PS1 transgenic Alzheimer’s disease mice,thereby facilitating functional recovery. | Yufang Yan Tuo Ma Kai Gong Qiang Ao Xiufang Zhang Yao Gong | 2014 | Neural Regeneration Research2014,9,8: | 16 |

| 3 | Effects of multidrug resistance, antisense RNA on the chemosensitivity of hepatocellular carcinoma cells显示文摘BACKGROUND: Multidrug resistance is a major obstacle in cancer chemotherapy. We examined whether the antisense RNA of multidrug resistance gene 1 (mdr1) could reverse multidrug resistance in the human hepatocellular carcinoma (HCC) cell line SMMC7721/ADM. METHODS: The recombinant adenoviruses pAdEasy- GFP-ASmdr1 product was produced by the adenoviral vector AdEasy system, which can express antisense RNA against the mdr1 gene. Following that, the recombinant adenovirus was transfected into the P-glycoprotein- producing multidrug resistance cell line, SMMC7721/ADM human HCC cells resistant to adriamycin (ADM) and daunorubicin (DNR). In order to investigate the reversal of multidrug resistance phenotype, we measured the expression of mdr1 mRNA by RT-PCR and the production of P-glycoprotein by flow cytometry. The sensitivities for ADM and DNR SMMC7721/ADM cells were examined by [3-(4, 5-dimethylthi-azol-2-yl)-2,5 diphenyl-terazolium bromide] (MTT) analysis. RESULTS: The low-level expression of mdr1 mRNA and P-glycoprotein production were observed in parental sensitive cells SMMC/7721 in addition to the overexpressionof mdr1 mRNA and P-glycoprotein in SMMC7721/ADM cells. The transfection of antisense-RNA into SMMC7721/ ADM cells resulted in decreases of mdr1 mRNA and P-glycoprotein, but increase of drug sensitivities. The sensitivities of transfected SMMC7721/ADM cells to ADM and DNR in IC50 reduced by 31.25% and 62.96% respectively. CONCLUSIONS: Mdr1 antisense RNA can increase the sensitivities of SMMC7721/ADM cells to anticancer drug by decreasing the expression of the mdr1 gene and inhibiting P-glycoprotein expression. | 4 | Effects of inorganic arsenic on growth and microcystin production of a Microcystis strain isolated from an algal bloom in Dianchi Lake,China显示文摘Our previous data have shown that inorganic arsenic concentrations were high in Dianchi Lake,China,where Microcystis blooms often occur.To explore the relationship between arsenic and the growth of Microcystis,the effects of arsenite[As(III)]and arsenate[As(V)]on the growth and toxin production of M.aeruginosa strain FACHB 905 were tested.Results showed that M.aeruginosa FACHB 905 was tolerant to inorganic arsenic and its growth was not inhibited when the concentration of As(III)was below 10-5 mol L-1 or that of As(V)below 10-3 mol L-1.Total microcystin production was stimulated in the presence of 10-7 mol L-1 As(III)and the response of this M.aeruginosa strain to As(III)seemed to be a typical inverted U-shaped hormesis.The content increase of microcystin-LR per cell indicated that the toxicity was enhanced as M.aeruginosa FACHB 905 was exposed to As(V).Considering the relatively high concentration of inorganic arsenic in Dianchi Lake(139μg L-1 in epilimnetic water),the origin of the M.aeruginosa strain,inorganic arsenic favors survival of M.aeruginosa FACHB 905 and may stimulate its microcystin production and cellular toxicity. | GONG Yan AO HongYi LIU BiBo WEN Sheng WANG Zhi HU DingJing ZHANG XingZhong SONG LiRong LIU JianTong | 2011 | Chinese Science Bulletin2011,56,22: | 6 |

| 9 | Engineering microparticles based on solidified stem cell secretome with an augmented pro-angiogenic factor portfolio for therapeutic angiogenesis显示文摘Tissue (re)vascularization strategies face various challenges, as therapeutic cells do not survive long enough in situ, while the administration of pro-angiogenic factors is hampered by fast clearance and insufficient ability to emulate complex spatiotemporal signaling. Here, we propose to address these limitations by engineering a functional biomaterial capable of capturing and concentrating the pro-angiogenic activities of mesenchymal stem cells (MSCs). In particular, dextran sulfate, a high molecular weight sulfated glucose polymer, supplemented to MSC cul-tures, interacts with MSC-derived extracellular matrix (ECM) components and facilitates their co-assembly and accumulation in the pericellular space. Upon decellularization, the resulting dextran sulfate-ECM hybrid material can be processed into MIcroparticles of SOlidified Secretome (MIPSOS). The insoluble format of MIPSOS protects protein components from degradation, while facilitating their sustained release. Proteomic analysis demonstrates that MIPSOS are highly enriched in pro-angiogenic factors, resulting in an enhanced pro-angiogenic bioactivity when compared to naïve MSC-derived ECM (cECM). Consequently, intravital microscopy of full-thickness skin wounds treated with MIPSOS demonstrates accelerated revascularization and healing, far superior to the ther-apeutic potential of cECM.
