1High glucose causes apoptosis of rabbit corneal epithelial cells involving activation of PERK-eIF2α-CHOP-caspase-12 signaling pathway显示文摘AIM: To investigate the effect of high concentration of glucose(HCG) on double stranded RNA-activated protein kinase-like ER kinase(PERK)-eukaryotic initiation factor-2α(eIF2α)-transcription factor C/EBP homologous protein(CHOP)-cysteine aspartate specific proteinase(caspase-12) signaling pathway activation and apoptosis in rabbit corneal epithelial cells(RCECs). METHODS: RCECs were treated by different concentrations of glucose for 0-48 h. The expressions of PERK, p-PERK, eIF2α, p-eIF2α, 78 k Da glucose-regulated protein 78(GRP78), CHOP, B-cell lymphoma 2(Bcl-2), B-cell lymphoma-2-associated X protein(Bax) and caspase-12 were determined by Western blot. Apoptosis was detected by TUNEL assay. Meanwhile, the function of PERK-eI F2α-CHOP-caspase-12 signaling pathway activation in high glucose-induced apoptosis was evaluated using PERK inhibitor, GSK2606414. RESULTS: HCG significantly promoted the expression of p-PERK, p-eIF2α, GRP78, CHOP, Bax and cleaved caspase-12 in RCECs(P<0.05), while remarkably decreased the expression of Bcl-2 and caspase-12(P<0.05), and the alterations caused by glucose were in concentration-and time-dependent manners. Meanwhile, PERK and eIF2α expressions were not affected in all groups(P>0.05). TUNEL assay showed that the apoptosis rate of RCECs in the HCG group increased significantly in contrast with that in the normal concentration of glucose or osmotic pressure control group(P<0.05), and the apoptosis rate increased with the increase of glucose concentration within limits(P<0.05). GSK2606414 down-regulated the expression of p-PERK and p-eI F2α in the HCG group(P<0.05), while still did not affect the expression of PERK and eIF2α among groups(P>0.05). Correspondingly, GSK2606414 also significantly reduced the apoptosis rate induced by high glucose(P<0.05). 2Dynamic Expression of MicroRNA-127 During Porcine Prenatal and Postnatal Skeletal Muscle Development显示文摘MicroRNAs(miRNAs), evolutionarily conserved non-coding RNAs in length 21-24 bp, play a critical role in skeletal muscle development. In this study, to explore the function of mircoRNA-127 in porcine skeletal muscle development, eight tissue samples from adult pigs and longissimus muscle samples at 26 developmental stages were collected from Tongcheng and Landrace pigs. The spatial-temporal expression profiles of miRNA-127 were carried out using step-loop quantitative real-time PCR(stemloop RT-PCR). To explore the molecular functions of miRNA-127, we predicted its target genes and performed functional annotation using bioinformatics methods. Results suggested that miRNA-127 was abundantly expressed in heart, ovary, uterus and spleen tissues and was weakly expressed in liver, lung, kidney and small intestine in both Tongcheng and Landrace pigs. And miRNA-127 showed significant expression differences in heart, ovary, spleen and uterus tissues between these two breeds. miRNA-127 basically kept at a relatively stable high level in middle and later embryonic stages and a low expression level in early embryonic stages and postnatal stages, but the expression levels of miRNA-127 were higher in Tongcheng pigs than in Landrace at most developmental stages. miRNA-127 potentially regulated 240 candidate genes. Results of Gene Ontology and KEGG pathway analysis indicated that these genes could be involved in many molecular functions and mechanisms, such as regulation of the force of heart contraction, regulation of transcription, regulation of T cell differentiation, MAPK signaling pathway and GnRH signaling pathway. Many significantly enriched GO terms and KEGG pathways were related to skeletal muscle development. 4Preparation of amidoxime-based PE/PP fibers for extraction of uranium from aqueous solution显示文摘A novel amidoxime-based fibrous adsorbent,denoted as PE/PP-g-(PAAc-co-PAO), was prepared by preirradiation grafting of acrylic acid and acrylonitrile onto polyethylene-coated polypropylene skin-core(PE/PP)fibers using 60 Co γ-ray irradiation, followed by amidoximation. The original and modified PE/PP fibers were characterized by a series of characterization methods to demonstrate the attachment of amidoxime groups onto the PE/PP fibers. Breaking strength tests confirmed that the fibrous adsorbent could maintain good mechanical properties. The adsorption capacity of the PE/PP-g-(PAAc-coPAO) fibers was investigated in simulated seawater with an initial uranium concentration of 330 μg/L. The uranium adsorption capacity was 2.27 mg/g-adsorbent after 24 h in simulated seawater, and the equilibrium data were well described by the Freundlich isotherm model. The PE/PP-g-(PAAc-co-PAO) adsorbent exhibited good regeneration and recyclability during five adsorption-desorption cycles.The adsorption test was also performed in simulated radioactive effluents with uranium concentrations of 10 and100 μg/L. The effect of the pH value on the adsorption capacity was also studied. At a very low initial concentration 10 μg/L solution, the PE/PP-g-(PAAc-co-PAO)fiber could remove as much as 93.0% of the uranium, and up to 71.2% of the uranium in the simulated radioactive effluent. These results indicated that the PE/PP-g-(PAAcco-PAO) adsorbent could be used in radioactive effluents over a wide range of pH values. 7Milk fatty acids profiles and milk production from dairy cows fed different forage quality diets显示文摘Thirty lactating Holstein cows were used to investigate the effects of different forages quality on milk fatty acids(FA) profiles and production. The cows were assigned to 3 dietary treatments(n = 10 per treatment) in a randomized block design with 3 repeated measures. They were fed the experimental diets for 90 d with 3 days of collection of samples for analysis at about 27 d intervals(samples were collected on days 28,29,30, 58, 59, 60.88, 89 and 90). The treatments were(DM basis): 1) mixed forages diet(MF) consisting of 3.7% Chinese wild rye, 26.7% corn silage and 23.4% alfalfa hay; 2) corn stalk diet 1(CS1) where corn stalk was used to formulate a similar chemical nutrient level to MF; 3) corn stalk diet 2(CS2) which used corn stalk to formulate a similar forage level to MF for the diet. Dry matter intake and BW were similar between treatments, but daily milk yield, milk fat and protein yield decreased(P < 0.05)in CS1 and CS2 compared with MF, with CS2 being the lowest(P < 0.05). In total FA of milk, the compositions of C18:1 c9, C18:3 and unsaturated FA increased(P < 0.05) in CS1 and CS2 compared with MF,and C18:0 and trans-C18:1 were trended to increase(P < 0.10), but C4:0-C16:0 were decreased(P < 0.05).Compared with cows fed CS2, cows receiving CS1 increased the compositions of C4:0 to C12:0 and C18:2(P < 0.05). 8A specific aggregation-induced emission-conjugated polymer enables visual monitoring of osteogenic differentiation显示文摘Osteogenic differentiation is the basis of bone growth and repair related to many diseases,in which evaluating the degree and ability of osteogenic transformation is quite important and highly desirable.However,fixing or stopping the growth of cells is required for conventional methods to monitor osteogenic differentiation,which cannot realize the full investigation of the dynamic process.Herein,a new anion conjugated polymer featuring aggregation-induced emission(AIE)characteristics is developed with excellent solubility for in-situ monitoring the process of osteogenic differentiation.This novel polymer can bind with osteogenic differentiated cells,and the intracellular fluorescence increases gradually with the enhancement of osteogenic differentiation.Moreover,it possesses good biosafety with negligible effect on cell activity and osteogenic differentiation,which cannot be realized by the typical method of Alizarin Red S staining.Further study shows that the polymer crosses the cell membrane through endocytosis and enriches in lysosomes,whereas no obvious fluorescence is detected with other cells,including non-differentiated osteoblast cells,under the same conditions,demonstrating the high selectivity.This is the first fluorescent probe with excellent specificity to realize real-time observation of the process of osteogenic differentiation.Therefore,PTB-EDTA shows great promise in the study of osteogenic differentiation and related applications.Zhenyu Zheng Taotao Zhou Rong Hu Minjun Huang Xiang Ao Jun Chu Tao Jiang Anjun Qin Zhongmin Zhang 2020Bioactive Materials2020,5,4:3

15Single-cell analysis of angiotensin-converting enzyme II expression in human kidneys and bladders reveals a potential route of 2019 novel coronavirus infection显示文摘Background::Since 2019,a novel coronavirus named 2019 novel coronavirus(2019-nCoV)has emerged worldwide.Apart from fever and respiratory complications,acute kidney injury has been observed in a few patients with coronavirus disease 2019.Furthermore,according to recent findings,the virus has been detected in urine.Angiotensin-converting enzyme II(ACE2)has been proposed to serve as the receptor for the entry of 2019-nCoV,which is the same as that for the severe acute respiratory syndrome.This study aimed to investigate the possible cause of kidney damage and the potential route of 2019-nCoV infection in the urinary system.Methods::We used both published kidney and bladder cell atlas data and new independent kidney single-cell RNA sequencing data generated in-house to evaluate ACE2 gene expression in all cell types in healthy kidneys and bladders.The Pearson correlation coefficients between ACE2 and all other genes were first generated.Then,genes with r values larger than 0.1 and P values smaller than 0.01 were deemed significant co-expression genes with ACE2.Results::Our results showed the enriched expression of ACE2 in all subtypes of proximal tubule(PT)cells of the kidney.ACE2 expression was found in 5.12%,5.80%,and 14.38%of the proximal convoluted tubule cells,PT cells,and proximal straight tubule cells,respectively,in three published kidney cell atlas datasets.In addition,ACE2 expression was also confirmed in 12.05%,6.80%,and 10.20%of cells of the proximal convoluted tubule,PT,and proximal straight tubule,respectively,in our own two healthy kidney samples.For the analysis of public data from three bladder samples,ACE2 expression was low but detectable in bladder epithelial cells.Only 0.25%and 1.28%of intermediate cells and umbrella cells,respectively,had ACE2 expression.Conclusion::This study has provided bioinformatics evidence of the potential route of 2019-nCoV infection in the urinary system.Wei Lin Jue Fan Long-Fei Hu Yan Zhang Joshua DOoi Ting Meng Peng Jin Xiang Ding Long-Kai Peng Lei Song Rong Tang Zhou Xiao Xiang Ao Xiang-Cheng Xiao Qiao-Ling Zhou Ping Xiao Yong Zhong 2021Chinese Medical Journal2021,,8:1
16Prediction of pandemic risk for animal-origin coronavirus using a deep learning method显示文摘Background:Coronaviruses can be isolated from bats,civets,pangolins,birds and other wild animals.As an animalorigin pathogen,coronavirus can cross species barrier and cause pandemic in humans.In this study,a deep learning model for early prediction of pandemic risk was proposed based on the sequences of viral genomes.Methods:A total of 3257 genomes were downloaded from the Coronavirus Genome Resource Library.We present a deep learning model of cross-species coronavirus infection that combines a bidirectional gated recurrent unit network with a one-dimensional convolution.The genome sequence of animal-origin coronavirus was directly input to extract features and predict pandemic risk.The best performances were explored with the use of pre-trained DNA vector and attention mechanism.The area under the receiver operating characteristic curve(AUROC)and the area under precision-recall curve(AUPR)were used to evaluate the predictive models.Results:The six specifc models achieved good performances for the corresponding virus groups(1 for AUROC and 1 for AUPR).The general model with pre-training vector and attention mechanism provided excellent predictions for all virus groups(1 for AUROC and 1 for AUPR)while those without pre-training vector or attention mechanism had obvi‑ously reduction of performance(about 5–25%).Re-training experiments showed that the general model has good capabilities of transfer learning(average for six groups:0.968 for AUROC and 0.942 for AUPR)and should give reason‑able prediction for potential pathogen of next pandemic.The artifcial negative data with the replacement of the coding region of the spike protein were also predicted correctly(100%accuracy).With the application of the Python programming language,an easy-to-use tool was created to implements our predictor.Conclusions:Robust deep learning model with pre-training vector and attention mechanism mastered the features from the whole genomes of animal-origin coronaviruses and could predict the risk of cross-species infection for early warning of next pandemic.Zheng Kou Yi‑Fan Huang Ao Shen Saeed Kosari Xiang‑Rong Liu Xiao‑Li Qiang 2021Infectious Diseases of Poverty2021,10,5:0

19Reproducibility of macular perfusion parameters in non-proliferative diabetic retinopathy patients by two different OCTA sweep modes显示文摘·AIM:To assess the reproducibility of macular perfusion parameters in non-proliferative diabetic retinopathy(NPDR)patients measured by different examiners and two different sweep modes of optical coherence tomography angiography(OCTA).·METHODS:Ninety-eight(98 eyes)patients with NPDR were included in this study.All participates were performed three times using Cirrus OCTA with Angiography 3×3 mm^(2)and 6×6 mm^(2)sweep mode by two examiners.The macular foveal avascular zone(FAZ)and vessel density(VD)in the superficial retinal layer(SRL)were measured.The reproducibility of the measurements was evaluated with intraclass correlation coefficients(ICC)and coefficient of variation(CoV).·RESULTS:The intra-mode ICCs of Angiography 3×3 mm^(2)and 6×6 mm^(2)sweep mode were 0.957 to 0.959 and 0.964 to 0.977,respectively;and the inter-mode ICCs were 0.962 to 0.970.The intra-examiner ICCs of macular perfusion parameters were>0.950;and the inter-examiner ICCs were0.928 to 0.969.All CoVs were<1.0%.·CONCLUSION:Cirrus OCTA can measure macular perfusion parameters in NPDR patients with excellent reproducibility.The measurements of FAZ and VD in the SRL determined by Angiography 3×3 mm^(2)and 6×6 mm^(2)sweep mode are highly consistent and both sweep modes are suitable for macular perfusion parameters measurement.Shen Qu Ao Rong Yun-Li Niu Xin Liu Yu-Shan Zhang Chun-Yu Liu Yan-Long Bi 2022International Journal of Ophthalmology(English edition)2022,15,9:0
20Pars plana vitrectomy for retinal detachment using perfluoro-n-octane as intraoperative tamponade:a multicenter,randomized,non-inferiority trial显示文摘AIM:To evaluate the efficacy and safety of perfluoro-n-octane(PFO)for ophthalmic surgery versus F-Octane as an intraoperative tamponade in pars plana vitrectomy(PPV)in management of retinal detachment.METHODS:This multicenter,prospective,randomized,double-masked,parallel-controlled,non-inferiority trial was conducted in three ophthalmology clinical centers in China.Patients with retinal detachment,who were eligible for PPV were consecutively enrolled.Participants were assigned to PFO for ophthalmic surgery or F-Octane for intraocular tamponade in a 1:1 ratio.Best-corrected visual acuity(BCVA),intraocular pressure(IOP)measurement,and dilated fundus examination were performed preoperatively and at 1,7±1,28±3d postoperatively.The primary outcome was complete retinal reattachment rate at postoperative day one.The non-inferiority margin was set at 9.8%.The secondary outcomes included intraoperative retinal reattachment rate,and mean changes in IOP and BCVA from baseline to 1,7±1,28±3d postoperatively,respectively.Safety analyses were presented for all randomly assigned participates in this study.RESULTS:Totally 124 eligible patients completed the study between Mar.14,2016 and Jun.7,2017.Sixty of them were randomly assigned to the PFO for ophthalmic surgery group,and 64 were assigned to the F-Octane group.Baseline characteristics were comparable between the two groups.Both groups achieved 100%retinal reattachment at postoperative day one(difference 0,95%CI:-6.21%to 5.75%,P=1).The pre-defined noninferiority criterion was met.No significant difference was observed in intraoperative retinal reattachment rate(difference 1.77%,P=0.61),mean changes in IOP(difference 0.36,-0.09,2.22 mm Hg at 1,7±1,28±3d postoperatively,with all P>0.05)and BCVA(difference 0.04,-0.02,0.06 logMAR at 1,7±1,28±3d postoperatively,all P>0.05)between the two groups.No apparent adverse events related to the utilization of PFO were reported.CONCLUSION:In patients with retinal detachment undergoing PPV,PFO for ophthalmic surgery is non-inferior to F-Octane as an intraocular tamponade,and both are safe and well-tolerated.Xin Shi Wei-Jun Wang Ying Fan Hai-Yun Liu Hong Wang Yu-Hui Chen Ao Rong Zhi-Feng Wu Xun Xu Kun Liu 2024International Journal of Ophthalmology(English edition)2024,17,1:0
