1RESEARCH PROGRESS IN NONLINEAR ANALYSIS OF HEART ELECTRIC ACTIVITIES显示文摘Nonlinear science research is a hot point in the world. It has deepened our cognition of determinism and randomicity, simplicity and com-plexity, noise and order and it will profoundly influ-ence the progress of the study of natural science, including life science. Life is the most complex nonlinear system and heart is the core of lifecycle system. In the late more than 20 years, nonlinear research on heart electric activities has made much headway. The commonly used parameters are based on chaos and fractal theory, such as correlation dimension, Lyapunov ex-ponent, Kolmogorov entropy and multifractal singu-larity spectrum. This paper summarizes the commonly used methods in the nonlinear study of heart electric signal. Then, considering the shortages of the above tradi-tional nonlinear parameters, we mainly introduce the results on short-term heart rate variability (HRV) signal (500 R-R intervals) and HFECG signal (1-2s). 2Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
3Chimeric antigen receptor T cell targeting EGFRvIII for metastatic lung cancer therapy显示文摘Lung cancer is the most common incident cancer and the leading cause of cancer death. In recent years, the development of tumor immunotherapy especially chimeric antigen receptor T (CAR-T) cell has shown a promising future. Epidermal growth factor receptor variant III (EGFRvlll) is a tumor-specific mutation expressed in various types of tumors and has been detected in non-small cell lung cancer with a mutation rate of 10%. Thus, EGFRvIII is a potential antigen for targeted lung cancer therapy. In this study, CAR vectors were constructed and transfected into virus-packaging cells. Then, activated T cells were infected with retrovirus harvested from stable virus-producing single clone cell lines. CAR expression on the surfaces of the T cells was detected by flow cytometry and Western blot. The function of CAR-T targeting EGFRvIII was then evaluated. The EGFRvIII-CAR vector was successfully constructed and confirmed by DNA sequencing. A stable virus-producing cell line was produced from a single clone by limited dilution. The culture conditions for the cell line, including cell density, temperature, and culture medium were optimized. After infection with retrovirus, CAR was expressed on more than 90% of the T cells. The proliferation of CAR-T cells were induced by cytokine and specific antigen in vitro. More importantly, EGFRvIII-CART specifically and efficiently recognized and killed A549-EGFRvIII cells with an effector/target ratio of 10:1 by expressing and releasing cytokines, including perforin, granzyme B, IFN-γ, and TNF-α. The in vivo study indicated that the metastasis of A549-EGFRvIII cells in mice were inhibited by EGFRvIII-CART cells, and the survival of the mice was significantly prolonged with no serious side effects. EGFRvIII-CART showed significantly efficient antitumor activity against lung cancer cells expressing EGFRvlll in vivo and in vitro. 4The positive function of selenium supplementation on reducing nitrate accumulation in hydroponic lettuce(Lactuca sativa L.)显示文摘High nitrate(NO_3^- ) in vegetables, especially in leaf vegetables poses threaten to human health. Selenium(Se) is an important element for maintaining human health, and exogenous Se application during vegetable and crop production is an effective way to prevent Se deficiency in human bodies. Exogenous Se shows positive function on plant growth and nutrition uptake under abiotic and/or biotic stresses. However, the influence of exogenous Se on NO_3^- accumulation in hydroponic vegetables is still not clear. In the present study, hydroponic lettuce plants were subjected to six different concentrations(0, 0.1, 0.5, 5, 10 and 50 μmol L–1) of Se as Na2 Se O3. The effects of Se on NO_3^- content, plant growth, and photosynthetic capacity of lettuce(Lactuca sativa L.) were investigated. The results showed that exogenous Se positively decreased NO_3^- content and this effect was concentration-dependent. The lowest NO_3^- content was obtained under 0.5 μmol L–1 Se treatment. The application of Se enhanced photosynthetic capacity by increasing the photosynthesis rate(Pn), stomatal conductance(Cs) and the transpiration efficiency(Tr) of lettuce. The transportation and assimilation of NO_3^- and activities of nitrogen metabolism enzymes in lettuce were also analysed. The NO_3^- efflux in the lettuce roots was markedly increased, but the efflux of NO_3^- from the root to the shoot was decreased after treated with exogenous Se. Moreover, Se application stimulated NO_3^- assimilation by enhancing nitrate reductase(NR), nitrite reductase(Ni R), glutamine synthetase(GS) and glutamate synthase enzyme(GOGAT) activities. These results provide direct evidence that exogenous Se shows positive function on decreasing NO_3^- accumulation via regulating the transport and enhancing activities of nitrogen metabolism enzyme in lettuce. 9The nitritation performance of biofilm reactor for treating domestic wastewater under high dissolved oxygen显示文摘The objective of this study was to investigate the nitritation performance in a biofilm reactor for treating domestic wastewater.The reactor was operated in continuous feed mode from phases 1 to 3.The dissolved oxygen(DO)was controlled at 3.5–7 mg/L throughout the experiment.The biofilm reactor showed excellent nitritation performance after the inoculation of nitrifying sludge,with the hydraulic retention time being reduced from 24 to 7 hr.Above 90%nitrite accumulation ratio(NAR)was maintained in phase 1.Afterwards,nitratation occurred with the low NH_4^+–N concentration in the reactor.The improvement of NH_4^+–N concentration to 20–35 mg/L had a limited effect on the recovery of nitritation.However,nitritation recovered rapidly when sequencing batch feed mode was adopted in phase 4,with the effluent NH_4^+-N concentration above 7 mg/L.The improvement of ammonia oxidizing bacteria(AOB)activity and the combined inhibition effect of free ammonia(FA)and free nitrous acid(FNA)on the nitrite oxidizing bacteria(NOB)were two key factors for the rapid recovery of nitritation.Sludge activity was obtained in batch tests.The results of batch tests had a good relationship with the long term operation performance of the biofilm reactor.Zhaoming Zheng Zebing Li Jing Ma Jia Du Guanghui Chen Wei Bian Jun Li Baihang Zhao 2016Journal of Environmental Sciences2016,28,4:7

14Broadband perovskite quantum dot spectrometer beyond human visual resolution显示文摘The quantum dot spectrometer,fabricated by integrating different quantum dots with an image sensor to reconstruct the target spectrum from spectral-coupled measurements,is an emerging and promising hyperspectrometry technology with high resolution and a compact size.The spectral resolution and spectral range of quantum dot spectrometers have been limited by the spectral variety of the available quantum dots and the robustness of algorithmic reconstruction.Moreover,the spectrometer integration of quantum dots also suffers from inherent photoluminescence emission and poor batch-to-batch repeatability.In this work,we developed nonemissive in situ fabricated MA_(3)Bi_(2)X_(9) and Cs_(2)SnX_(6)(MA=CH_(3)NH_(3);X=Cl,Br,I)perovskite-quantum-dot-embedded films(PQDFs)with precisely tunable transmittance spectra for quantum dot spectrometer applications.The resulting PQDFs contain in situ fabricated perovskite nanocrystals with homogenous dispersion in a polymeric matrix,giving them advantageous features such as high transmittance efficiency and good batch-to-batch repeatability.By integrating a filter array of 361 kinds of PQDFs with a silicon-based photodetector array,we successfully demonstrated the construction of a perovskite quantum dot spectrometer combined with a compressive-sensing-based total-variation optimization algorithm.A spectral resolution of ~1.6 nm was achieved in the broadband of 250-1000 nm.The performance of the perovskite quantum dot spectrometer is well beyond that of human eyes in terms of both the spectral range and spectral resolution.This advancement will not only pave the way for using quantum dot spectrometers for practical applications but also significantly impact the development of artificial intelligence products,clinical treatment equipment,scientific instruments,etc.Xiaoxiu Zhu Liheng Bian Hao Fu Lingxue Wang Bingsuo Zou Qionghai Dai Jun Zhang Haizheng Zhong 2020Light(Science & Applications)2020,9,1:5
15Clinical effects of Xingnao Kaiqiao acupuncture on neurological impairment following cerebral infarction显示文摘BACKGROUND: Although the curative effects of acupuncture have been confirmed by various treatments of cerebral infarction, few studies have investigated when acupuncture can attain the best clinical effect. OBJECTIVE: Four different time points were selected for acupuncture treatment of cerebral infarction to evaluate the appropriate time course for Xingnao Kaiqiao therapy in terms of improved neurological function. DESIGN: Controlled observation. SETTING: Department of Traditional Chinese Medicine Rehabilitation and Physiotherapy of the Affiliated Hospital of Medical College of Chinese Armed Police Forces. PARTICIPANTS: A total of 120 inpatients with cerebral infarction of different stages, including 75 males and 45 females, aged 41-75 years, were selected from November 2005 to December 2006 at the Department of Traditional Chinese Medicine Rehabilitation and Physiotherapy in Affiliated Hospital of Medical College of Chinese Armed Police Forces. Diagnostic criteria: in accordance with 'main points of diagnosis on different cerebrovascular disease' secondly revised in the Second Cerebrovascular Disease Academic Meeting of Chinese Medicine Association in 1986. All accepted subjects provided confirmed consent, and the experiment received ethical permission from the hospital’s ethics committee. METHODS: ① Experiment grouping: All inpatients were divided into four groups with non-stochastic concurrent control method according to the disease course: Group I (onset within 7 hours), group Ⅱ (onset from 7 hours to 3 days), group Ⅲ (onset within 4-7 days), and group Ⅳ (onset within 21-180 days). On the basis of symptomatic treatment with western medicine, each group received Xingnao Kaiqiao therapy after onset within 7 hours, 7 hours to 3 days, 4 to 7days, and 21 to 180 days. ② The principal acupoints were Neiguan, Renzhong, and Sanyinjiao. ③ The auxiliary acupoints were Jiquan, Chize, and Weizhong. ④Acupuncture manipulations: initially, Neiguan (PC6, bilateral) was needled at 0.5-1.0 cun vertically with a reducing technique achieved by rotating-swirling and lifting-inserting for 1 minute. Subsequently, Renzhong (GV26) was needled at 0.3-0.5 cun obliquely towards the nasal septum, with a heavy bird-pecking method until eyeballs were moist or shed tears. Sanyinjiao (SP6, affected limb) was needled at 1.0-1.5 cun obliquely along the medial border of the tibia with a 45° angle between the needle and skin. The technique of reinforcing was achieved by a lifting-inserting method until the affected lower limb twitched three times. The auxiliary acupoint, Jiquan, was selected when the patient extended the affected upper limb abduction. Jiquan was along the downward channel, 1 cun from the quondam Jiquan (HT1) acupoint; the armpit hair was avoided. Jiquan (affected limb) was needled vertically at 1.0-1.5 cun. The technique of reduction was achieved by lifting-inserting until the affected upper limb twitched three times. Chize (LU5) was selected when the patient flexed the elbow at 120°. Chize was vertically needled at 1 cun with a lifting-inserting reducing technique until the affected forearm and fingers twitched three times. Weizhong (BL40) was selected when the patient’s leg was raised straight in a supine position. Weizhong was vertically needled at 0.5-1.0 cun with a lifting-inserting reducing technique until the affected lower limb twitched three times. ⑤ Acupuncture manipulations were achieved by the doctor who is an associate chief physician and doctor of acupuncture and moxibustion and has aptitude for the post’s specification. ⑥One course of treatment lasted for 10 days and each group was treated for two courses of treatment. ⑦Experiment evaluation: To apply neurological impairment scores for evaluating clinical curative effects before and after treatment with Xingnao Kaiqiao therapy on day 7, 14, and 21(range of 0-45; decreasing score indicates functional improvement).To evaluate clinical curative effect by clinical effective power:Basic recovery: equal to ormore than 81%; significant improvement: 56% to 81% (including 56%); Improvement: between 36% to 56% (including 36%); Slight Improvement: 11% to 36% (including 11%); No change: less than 11%; Aggravation (including death): negative value. Total effective rate was computed at the same time. MAIN OUTCOME MEASURES: The total effective rate and amelioration of neurological impairment of the patients after treatment. RESULTS: All 120 patients with cerebral infarction were included in the final analysis, without deletion. ① Comparison of the total effective rate: the total effective rate of group Ⅰ, group Ⅱ, group Ⅲ, and group Ⅳ was 97%, 93%, 93%, and 67%, respectively. There were significant differences of interclass synthetic curative effects (χ2 = 36.351 7, P < 0.01). ② Amelioration of neurological impairment: A decreased neurological impairment score was observed in group I on day 7, 14, and 21 after Xingnao Kaiqiao therapy. The differences between group IV and the remaining three groups were significant (P < 0.05). 17The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 18Inhibition factors and Kinetic model for ammonium inhibition on the anammox process of the SNAD biofilm显示文摘The aim of the present work was to evaluate the anaerobic ammonium oxidation(anammox) activity of simultaneous partial nitrification, anammox and denitrification(SNAD) biofilm with different substrate concentrations and p H values. Kaldnes rings taken from the SNAD biofilm reactor were incubated in batch tests to determine the anammox activity. Haldane model was applied to investigate the ammonium inhibition on anammox process. As for nitrite inhibition, the NH_(4^-)^+N removal rate of anammox process remained 87.4% of the maximum rate with the NO_(2^-)^-N concentration of 100 mg/L. Based on the results of Haldane model, no obvious difference in kinetic coefficients was observed under high or low free ammonia(FA) conditions, indicating that ammonium rather than FA was the true inhibitor for anammox process of SNAD biofilm. With the p H value of 7.0, the r max, Ks and K_I of ammonium were 0.209 kg NO_(2^-)^-N/kg VSS/day, 9.5 mg/L and 422 mg/L, respectively. The suitable p H ranges for anammox process were 5.0 to 9.0. 19RELATIONSHIP BETWEEN CYCLIN G1 AND HUMAN PAPILLOMA VIRUS INFECTION IN CERVICAL INTRAEPITHELIAL NEOPLASIA AND CERVICAL CARCINOMA显示文摘Objective To evaluate the overexpression of cyclin G1 in cervical intraepithelial neoplasia (CIN) and cervical carcinoma, and the correlation between cyclin G1 and high-risk human papilloma virus (HPV) infection. Methods All of the specimens were obtained from the Department of Pathology of China-Japan Friendship Hospital from January 2000 to August 2004.We detected the expression of cyclin G1 with immunohistochemistry, HPV16/18 infection with in situ hybridization, and high-risk HPV infection with Hybrid capture system II (HC-II) in normal group (25 cases), CIN I (48 cases), CIN II (56 cases), CIN III (54 cases), and invasive cervical squamous-cell carcinoma (SCC, 31 cases). Results The positive rates of cyclin G1 expression in CIN(77.85%)and SCC cervical tissues (87.10%) were significantly higher than normal (8.00%, P<0.01), and the intensities of cyclin G1 expression in CIN(40.60%)and SCC cervical tissues (61.51%) were significantly higher than normal (2.72%, P<0.05). The positive rates and intensities of cyclin G1 expression increased gradually with the grade of cervical lesions. High-risk HPV infection rates were higher in CIN and SCC than normal groups (P<0.05). There was a positive correlation between cyclin G1 expression and high-risk HPV infection detected with HC-II (Kendall’s tau-b=0.316, 0.269, 0.352, and 0.474 in CIN I, CINII, CIN III, and SCC, respectively, P<0.05). Conclusions Cyclin G1 is overexpressed in CIN and SCC. Cyclin G1 may be a biomarker for detecting CIN and SCC. 20The Stereocomplex Formation and Phase Separation of PLLA/PDLA Blends with Different Optical Purities and Molecular Weights显示文摘In this study, the poly(L-lactide)/poly(D-lactide)(PLLA/PDLA) blends with different optical purities of PLLA and various molecular weights of PDLA are prepared by solution mixing, and the stereocomplex formation and phase separation behaviors of these blends are investigated. Results reveal that optical purity and molecular weight do not vary the crystal structure of PLA stereocomplex(sc) and homochiral crystallites(hc). As the optical purity increasing in the blends, the melting temperature of sc(Tsc) and the content of sc(ΔHsc) increased, while the melting temperature of hc(Thm) hardly changes, although the content of hc(ΔHhm) decreased gradually. The Tsc and ΔHsc are also enhanced as the molecular weight of PDLA reduces, and the ΔHhm reduces rapidly even though the Thm does not vary apparently. With lower optical purities of PLLA and higher molecular weights of PDLA, three types of crystals form in the blends, i.e., PLA sc, PLLA hc and PDLA hc. As molecular weight decreases and optical purity enhances, the crystal phase decreases to two(sc and PDLA hc), and one(sc) finally.
