1Numerical study of the effects of local atmospheric circulations on a pollution event over Beijing–Tianjin–Hebei, China显示文摘Currently, the Chinese central government is considering plans to build a trilateral economic sphere in the Bohai Bay area, including Beijing, Tianjin and Hebei(BTH), where haze pollution frequently occurs. To achieve sustainable development, it is necessary to understand the physical mechanism of the haze pollution there. Therefore, the pollutant transport mechanisms of a haze event over the BTH region from 23 to 24 September 2011 were studied using the Weather Research and Forecasting model and the FLEXible-PARTicle dispersion model to understand the effects of the local atmospheric circulations and atmospheric boundary layer structure. Results suggested that the penetration by sea-breeze could strengthen the vertical dispersion by lifting up the planetary boundary layer height(PBLH) and carry the local pollutants to the downstream areas; in the early night, two elevated pollution layers(EPLs) may be generated over the mountain areas: the pollutants in the upper EPL at the altitude of 2–2.5 km were favored to disperse by long-range transport, while the lower EPL at the altitude of 1 km may serve as a reservoir, and the pollutants there could be transported downward and contribute to the surface air pollution.The intensity of the sea–land and mountain–valley breeze circulations played an important role in the vertical transport and distribution of pollutants. 2Advances in dielectric elastomer actuation technology显示文摘Dielectric elastomer actuators(DEAs) have attracted much interest over the past decades due to the inherent flexibility, large strain, high efficiency, high energy density, and fast response of the material, which are known as one of the most promising candidates for artificial muscle. In this paper, we first introduce the actuation principle and electromechanical modeling approaches of dielectric elastomers(DEs). Then, the performance of different DEs material and existing compliant electrodes that are widely utilized for DEAs are presented. We also highlight the compatibility of DEs, which is suitable for a variety of actuator designs and applications. 4A Multi-Scale Urban Atmospheric Dispersion Model for Emergency Management显示文摘To assist emergency management planning and prevention in case of hazardous chemical release into the atmosphere, especially in densely built-up regions with large populations, a multi-scale urban atmospheric dispersion model was established.Three numerical dispersion experiments, at horizontal resolutions of 10 m, 50 m and 3000 m, were performed to estimate the adverse effects of toxic chemical release in densely built-up areas. The multi-scale atmospheric dispersion model is composed of the Weather Forecasting and Research(WRF) model, the Open Source Field Operation and Manipulation software package, and a Lagrangian dispersion model. Quantification of the adverse health effects of these chemical release events are given by referring to the U.S. Environmental Protection Agency's Acute Exposure Guideline Levels. The wind fields of the urban-scale case, with 3 km horizontal resolution, were simulated by the Beijing Rapid Update Cycle system, which were utilized by the WRF model. The sub-domain-scale cases took advantage of the computational fluid dynamics method to explicitly consider the effects of buildings. 5Efficacy and safety of darbepoetin alfa injection replacing epoetin alfa injection for the treatment of renal anemia in Chinese hemodialysis patients:A randomized,open-label,parallel-group,noninferiority phase III trial显示文摘Background:This study was to explore the clinical efficacy and safety of darbepoetin alfa injection replacing epoetin alfa injection(recombinant human erythropoietin injection,rHuEPO)for the treatment of anemia associated with chronic kidney failure in Chinese patients undergoing hemodialysis.Method:This study was a multicenter,randomized,open-label,intergroup parallel control phase III noninferiority trial from April 19,2013 to September 9,2014 at 25 sites.In this study,the members of the darbepoetin alfa group underwent intravenous administration once per week or once every two weeks.The members of the control drug epoetin alfa group underwent intravenous administration two or three times per week.All subjects underwent epoetin alfa administration during the 8-week baseline period.After that,subjects were randomly assigned to the darbepoetin alfa group or epoetin alfa group.The noninferiority in the changes of the average Hb concentrations from the baseline to the end of the evaluation period(noninferiority threshold:-1.0 g/dl)was tested between the two treatments.The time-dependent hemoglobin(Hb)concentration and the maintenance rate of the target Hb concentration(the proportion of subjects with Hb concentrations between 10.0 and 12.0 g/dl)were also evaluated.Iron metabolism,including changes in the serum iron,total iron-binding capacity,ferritin,transferrin saturation,and comparisons of the dose adjustments between the two groups during the treatment period were analyzed further.Adverse events(AEs)were also observed and compared,and the safety was analyzed between the two treatment groups.The conversion rate switching from epoetin alfa to darbepoetin alfa was also discussed.SAS?software version 9.2 was used to perform all statistical analyses.Descriptive statistics were used for all efficacy,safety,and demographic variable analyses,including for the primary efficacy indicators.Results:Four hundred and sixty-six patients were enrolled in this study,and ultimately 384 cases were analyzed for safety,including 267 cases in the darbepoetin alfa group and 117 cases in the epoetin alfa group.There were 211 cases in the per-protocol set,including 152 cases in the darbepoetin alfa group and 59 cases in the epoetin alfa group.The changes in the average Hb concentrations from the baseline to the end of the evaluation period were-0.07 and-0.15 g/dl in the darbepoetin alfa group and epoetin alfa group respectively.The difference between the two groups was 0.08 g/dl(95%confidence interval[CI]:-0.22 to 0.39),and the lower limit of the 95%CI was-0.22>-1.0 g/dl.The average Hb concentrations of the two groups were 10.88-11.43 g/dl(darbepoetin alfa)and 10.91-11.38 g/dl(epoetin alfa)during the study period of Weeks 0-28,with the maintenance rates of the target Hb concentration ranging within 71%-87%and 78%-95%in the darbepoetin alfa group and epoetin alfa group respectively.During the period of comparison between the two groups,the incidence of AEs in the darbepoetin alfa group was 61.42%,while in the epoetin alfa group it was 56.41%.All of the adverse events and reactions in the study were those commonly associated with hemodialysis.Conclusion:The overall efficacy and safety of darbepoetin alfa for the treatment of Chinese renal anemia patients undergoing hemodialysis are consistent with those of epoetin alfa.Bicheng Liu Nan Chen Jinghong Zhao Aiping Yin Xiongfei Wu Changying Xing Gengru Jiang Junzhou Fu Mei Wang Rong Wang Jianying Niu Ping Fu Zhaohui Ni Fanfan Hou Jiuyang Zhao Jing Chen Yuqing Chen Wei Shi Jianghua Chen Wenge Li Gang Xu Ling Zhong Wenhu Liu Guohua Ding Yuichiro Kondo Changhe Yue Changlin Mei 2022Chronic Diseases and Translational Medicine2022,8,2:1

7Darbepoetin alfa injection versus epoetin alfa injection for treating anemia of Chinese hemodialysis patients with chronic kidney failure:A randomized,open-label,parallel-group,non-inferiority Phase III trail显示文摘Background:Erythropoietin is a glycoprotein that mainly regulates erythropoiesis.In patients with chronic renal failure with anemia,darbepoetin alfa can stimulate erythropoiesis,correct anemia,and maintain hemoglobin levels.This study was designed to demonstrate the efficacy and safety of darbepoetin alfa injections as being not inferior to epoetin alfa injections(Recombinant Human Erythropoietin injection,rHuEPO)when maintaining hemoglobin(Hb)levels within the target range(10.0-12.0 g/dL)for the treatment of renal anemia.Methods:Ninety-five patients were enrolled in this study from April 15,2013 to April 10,2014 at 25 sites.In this study,patients(n=95)aged 18-70 years were randomized into a once per week intravenous darbepoetin alfa group(n=56)and a twice or three times per week intravenous epoetin alfa group(n=39)for 28 weeks,who had anemia with hemoglobin levels between 6 g/dL and 10 g/dL due to chronic kidney disease(CKD)and were undergoing hemodialysis or hemofiltration with ESA-naive(erythropoiesis stimulating agent-naive).The primary efficacy profile was the mean Hb level(the non-inferiority margin was-1.0 g/dL,week 21-28);the secondary efficacy profiles were the Hb increase rate(week 0-4),the target Hb achievement cumulative rate and time,the change trends of the Hb levels,and the target Hb maintenance ratio.Adverse events(AEs)were observed and compared,and the efficacy and safety were analyzed between the two treatment groups.Additionally,the frequencies of dose adjustments between the darbepoetin alfa and epoetin alfa groups were compared during the treatment period.SAS?software version 9.2 was used to perform all statistical analyses.Descriptive statistics were used for all efficacy,safety,and demographic variable analyses,including for the primary efficacy indicators.Results:The mean Hb level was 11.3 g/dL in the darbepoetin alfa group and 10.7 g/dL in the epoetin alfa group,respectively;the difference of the lower limits of the 95%confidence intervals(CI)between the two groups was 0.1 g/dL(>-1.0 g/dL),and non-inferiority was proven;the Hb levels started to increase in the first four weeks at a similar increase rate;no obvious differences were observed between the groups in the target Hb achievement cumulative rates,and the Hb levels as well as the target Hb level maintenance rate changed over time.The incidence of AEs was 62.5%in the darbepoetin alfa group and 76.9%in the epoetin alfa group.All the adverse events observed in the study were those commonly associated with hemodialysis.Conclusion:Darbepoetin alfa intravenously once per week can effectively increase Hb levels and maintain the target Hb levels well,which makes it not inferior to epoetin alfa intravenously twice or three times per week.Darbepoetin alfa shows an efficacy and safety comparable to epoetin alfa for the treatment of renal anemia.Nan Chen Changying Xing Jianying Niu Bicheng Liu Junzhou Fu Jiuyang Zhao Zhaohui Ni Mei Wang Wenhu Liu Jinghong Zhao Ling Zhong Xiongfei Wu Wenge Li Yuqing Chen Wei Shi Jianghua Chen Aiping Yin Ping Fu Rong Wang Gengru Jiang Fanfan Hou Guohua Ding Jing Chen Gang Xu Yuichiro Kondo Yuliang Su Changlin Mei 2022Chronic Diseases and Translational Medicine2022,8,1:1

11A novel immunogenomic signature to predict prognosis and reveal immune infiltration characteristics in pancreatic ductal adenocarcinoma显示文摘Background:The immune response in the tumor microenvironment(TME)plays a crucial role in cancer progression and recurrence.We aimed to develop an immune-related gene(IRG)signature to improve prognostic predictive power and reveal the immune infiltration characteristics of pancreatic ductal adenocarcinoma(PDAC).Methods:The Cancer Genome Atlas(TCGA)PDAC was used to construct a prognostic model as a training cohort.The International Cancer Genome Consortium(ICGC)and the Gene Expression Omnibus(GEO)databases were set as validation datasets.Prognostic genes were screened by using univariate Cox regression.Then,a novel optimal prognostic model was developed by using least absolute shrinkage and selection operator(LASSO)Cox regression.Cell type identification by estimating the relative subsets of RNA transcripts(CIBERSORT)and estimation of stromal and immune cells in malignant tumors using expression data(ESTIMATE)algorithms were used to characterize tumor immune infiltrating patterns.The tumor immune dysfunction and exclusion(TIDE)algorithm was used to predict immunotherapy responsiveness.Results:A prognostic signature based on five IRGs(MET,ERAP2,IL20RB,EREG,and SHC2)was constructed in TCGA-PDAC and comprehensively validated in ICGC and GEO cohorts.Multivariate Cox regression analysis demonstrated that this signature had an independent prognostic value.The area under the curve(AUC)values of the receiver operating characteristic(ROC)curve at 1,3,and 5 years of survival were 0.724,0.702,and 0.776,respectively.We further demonstrated that our signature has better prognostic performance than recently published ones and is superior to traditional clinical factors such as grade and tumor node metastasis classification(TNM)stage in predicting survival.Moreover,we found higher abundance of CD8+T cells and lower M2-like macrophages in the low-risk group of TCGA-PDAC,and predicted a higher proportion of immunotherapeutic responders in the low-risk group.Conclusions:We constructed an optimal prognostic model which had independent prognostic value and was comprehensively validated in external PDAC databases.Additionally,this five-genes signature could predict immune infiltration characteristics.Moreover,the signature helped stratify PDAC patients who might be more responsive to immunotherapy.Ang Li Bicheng Ye Fangnan Lin Yilin Wang Xiaye Miao Yanfang Jiang 2022Precision Clinical Medicine2022,5,2:1

13Bioprocess inspired formation of calcite mesocrystals by cation-mediated particle attachment mechanism显示文摘Calcite mesocrystals were proposed,and have been widely reported,to form in the presence of polymer additives via oriented assembly of nanoparticles.However,the formation mechanism and the role of polymer additives remain elusive.Here,inspired by the biomineralization process of sea urchin spine comprising magnesium calcite mesocrystals,we show that calcite mesocrystals could also be obtained via atachment of amorphous calcium carbonate(ACC)nanoparticles in the presence of inorganic zinc ions.Moreover,we demonstrate that zinc ions can induce the formation of temporarily stabilized amorphous nanoparticles of less than 20 nm at a significantly lower calcium carbonate concentration as compared to pure solution,which is energetically beneficial for the atachment and occlusion during calcite growth.The cation-mediated particle atachment crystallization significantly improves our understanding of mesocrystal formation mechanisms in biomineralization and offers new opportunities to bioprocess inspired inorganic ions regulated materials fabrication.Qihang Wang Bicheng Yuan Wenyang Huang Hang Ping Jingjing Xie Kun Wang Weimin Wang Zhaoyong Zou Zhengyi Fu 2023National Science Review2023,10,4:0

17Lymphatic vessel:origin,heterogeneity,biological functions,and therapeutic targets显示文摘Lymphatic vessels,comprising the secondary circulatory system in human body,play a multifaceted role in maintaining homeostasis among various tissues and organs.They are tasked with a serious of responsibilities,including the regulation of lymph absorption and transport,the orchestration of immune surveillance and responses.Lymphatic vessel development undergoes a series of sophisticated regulatory signaling pathways governing heterogeneous-origin cell populations stepwise to assemble into the highly specialized lymphatic vessel networks.Lymphangiogenesis,as defined by new lymphatic vessels sprouting from preexisting lymphatic vessels/embryonic veins,is the main developmental mechanism underlying the formation and expansion of lymphatic vessel networks in an embryo.However,abnormal lymphangiogenesis could be observed in many pathological conditions and has a close relationship with the development and progression of various diseases.Mechanistic studies have revealed a set of lymphangiogenic factors and cascades that may serve as the potential targets for regulating abnormal lymphangiogenesis,to further modulate the progression of diseases.Actually,an increasing number of clinical trials have demonstrated the promising interventions and showed the feasibility of currently available treatments for future clinical translation.Targeting lymphangiogenic promoters or inhibitors not only directly regulates abnormal lymphangiogenesis,but improves the efficacy of diverse treatments.In conclusion,we present a comprehensive overview of lymphatic vessel development and physiological functions,and describe the critical involvement of abnormal lymphangiogenesis in multiple diseases.Moreover,we summarize the targeting therapeutic values of abnormal lymphangiogenesis,providing novel perspectives for treatment strategy of multiple human diseases.Zhaoliang Hu Xushi Zhao Zhonghua Wu Bicheng Qu Minxian Yuan Yanan Xing Yongxi Song Zhenning Wang 2024Signal Transduction and Targeted Therapy2024,9,2:0
18Transcriptomic but not genomic variability confers phenotype of breast cancer stem cells显示文摘Background:Breast cancer stem cells(BCSCs)are considered responsible for cancer relapse and drug resistance.Understanding the identity of BCSCs may open new avenues in breast cancer therapy.Although several discoveries have been made on BCSC characterization,the factors critical to the origination of BCSCs are largely unclear.This study aimed to determine whether genomic mutations contribute to the acquisition of cancer stem-like phenotype and to investigate the genetic and transcriptional features of BCSCs.Methods:We detected potential BCSC phenotype-associated mutation hotspot regions by using whole-genome sequencing on parental cancer cells and derived serial-generation spheres in increasing order of BCSC frequency,and then performed target deep DNA sequencing at bulk-cell and single-cell levels.To identify the transcriptional program associated with BCSCs,bulk-cell and single-cell RNA sequencing was performed.Results:By using whole-genome sequencing of bulk cells,potential BCSC phenotype-associated mutation hotspot regions were detected.Validation by target deep DNA sequencing,at both bulk-cell and single-cell levels,revealed no genetic changes specifically associated with BCSC phenotype.Moreover,single-cell RNA sequencing showed profound transcriptomic variability in cancer cells at the single-cell level that predicted BCSC features.Notably,this transcriptomic variability was enriched during the transcription of 74 genes,revealed as BCSC markers.Breast cancer patients with a high risk of relapse exhibited higher expression levels of these BCSC markers than those with a low risk of relapse,thereby highlighting the clinical significance of predicting breast cancer prognosis with these BCSC markers.Conclusions:Transcriptomic variability,not genetic mutations,distinguishes BCSCs from non-BCSCs.The identified 74 BCSC markers have the potential of becoming novel targets for breast cancer therapy.Mengying Tong Ziqian Deng Mengying Yang Chang Xu Xiaolong Zhang Qingzheng Zhang Yuwei Liao Xiaodi Deng Dekang Lv Xuehong Zhang Yu Zhang Peiying Li Luyao Song Bicheng Wang Aisha Al-Dherasi Zhiguang Li Quentin Liu 2018Cancer Communications2018,38,1:0

20Modular crawling robots using soft pneumatic actuators显示文摘Crawling robots have elicited much attention in recent years due to their stable and efficient locomotion.In this work,several crawling robots are developed using two types of soft pneumatic actuators(SPAs),namely,an axial elongation SPA and a dual bending SPA.By constraining the deformation of the elastomeric chamber,the SPAs realize their prescribed motions,and the deformations subjected to pressures are characterized with numerical models.Experiments are performed for verification,and the results show good agreement.The SPAs are fabricated by casting and developed into crawling robots with 3D-printing connectors.Control schemes are presented,and crawling tests are performed.The speeds predicted by the numerical models agree well with the speeds in the experiments.Nianfeng WANG Bicheng CHEN Xiandong GE Xianmin ZHANG Wenbin WANG 2021Frontiers of Mechanical Engineering2021,16,1:0
