2OTUB1 promotes osteoblastic bone formation through stabilizing FGFR2显示文摘Bone homeostasis is maintained by the balance between osteoblastic bone formation and osteoclastic bone resorption.Dysregulation of this process leads to multiple diseases,including osteoporosis.However,the underlying molecular mechanisms are not fully understood.Here,we show that the global and conditional osteoblast knockout of a deubiquitinase Otub1 result in low bone mass and poor bone strength due to defects in osteogenic differentiation and mineralization.Mechanistically,the stability of FGFR2,a crucial regulator of osteogenesis,is maintained by OTUB1.OTUB1 attenuates the E3 ligase SMURF1-mediated FGFR2 ubiquitination by inhibiting SMURF1’s E2 binding.In the absence of OTUB1,FGFR2 is ubiquitinated excessively by SMURF1,followed by lysosomal degradation.Consistently,adeno-associated virus serotype 9(AAV9)-delivered FGFR2 in knee joints rescued the bone mass loss in osteoblast-specific Otub1-deleted mice.Moreover,Otub1 mRNA level was significantly downregulated in bones from osteoporotic mice,and restoring OTUB1 levels through an AAV9-delivered system in ovariectomy-induced osteoporotic mice attenuated osteopenia.Taken together,our results suggest that OTUB1 positively regulates osteogenic differentiation and mineralization in bone homeostasis by controlling FGFR2 stability,which provides an optical therapeutic strategy to alleviate osteoporosis.Qiong Zhu Yesheng Fu Chun-Ping Cui Yi Ding Zhikang Deng Chao Ning Fan Hu Chen Qiu Biyue Yu Xuemei Zhou Guan Yang Jiang Peng Weiguo Zou Cui Hua Liu Lingqiang Zhang 2023Signal Transduction and Targeted Therapy2023,8,5:1
3Novel polyzwitterion shell with adaptable surface chemistry engineered to enhance anti-fouling and intracellular imaging of detonation nanodiamonds under tumor pHe显示文摘In this work,zwitterionic polymer(polyzwitterion)coated nanodiamonds(ND)with tumorous pH responsiveness were prepared for efficient application of commercial NDS in tumor cells imaging.Poly(carboxybetaine methacrylate)was firstly grafted on the pristine NDS(PCBMA-@-NDs)by surface-initiated reversible addition-fragmentation chain transfer(RAFT)polymerization.To achieve pH responsiveness and activated interaction with tumor cells,PCBMA-@-NDs were subsequently modified with benzene sulfonamide(PCBSA-@-NDs)via one step carbodiimide chemistry.The surface polymer functionalization was characterized by FTIR,1H NMR and TGA,and PCBMA-@-NDs and PCBSA-@-NDs showed great dispersibility,enhanced fluorescence intensity and excellent antifouling property in contrast to pristine NDs.Moreover,PCBSA-@-NDs was able to switch zwitterionic surface(at pH 7.4)reversibly into positive charge(at pH 6.5)via protonated or deprotonated acylsulfonamide.As expected,PCBSA-@-NDS demonstrated higher cell affinity and better imaging performance than those of zwitterionic NDS at tumor slight acid environment,proven by fluorescence microscopy and flow cytometry.Overall,we provide a feasible and valuable strategy to construct smart NDS,thus encouraging the application of cost-effective fluorescence nanomater-ials in biomedical fields.Biyu ZHOU Junbo LI Binzhong LU Wenlan WU Leitao ZHANG Ju LIANG Junpeng YI Xin LI 2020Frontiers of Materials Science2020,14,4:0
4Smart City Development in China: One City One Policy显示文摘China is in a process of urbanization and is aiming at a type of people-centered urbanization.The main purpose of developing a 'smart city' is to help this type urbanization and to serve the people of the city.From 2012 to 2015,China has chosen more than300 cities or towns to be national pilot smart cities.These pilot smart cities are located in more than 30 provinces around China,which differ greatly in thousands ways.So we advocated One City One Policy.In 2012,MOHURD announced 90 cities as first batch of pilot smart cities.After three years,some pilot cities achieved great progress.This paper introduces five example cities(including town,district) as five different models of China s smart city development.They are:Guilin city;Yunlong demonstration zone;Panyu District;Yangling Agricultural Hi-tech Industries Demonstration Zone;Lecong town.This paper also introduces our standardization work on smart city field at present.Biyu Wan Rong Ma Weiru Zhou Guoqiang Zhang 2015ZTE Communications2015,13,4:0
5Synthesis of poly(ethylene glycol)-SS-poly(ε-caprolactone)-SS-poly(ethylene glycol)triblock copolymers via end-group conjugation and self-assembly for reductively responsive drug delivery显示文摘In this study,we describe a simple synthesis route to prepare triblock copolymers with disulfide-linkers,poly(ethylene glycol)-SS-poly(ε-caprolactone)-SS-poly(ethylene glycol)(PEG-SS-PCL-SS-PEG)for application in the reductively responsive release of doxorubicin(DOX).To synthesize PEG-SS-PCL-SS-PEG,two end-groups of PCL-diol were first modified with cystamine to introduce disulfide bonds and subsequently conjugated with PEG-NHS via carbodiimide chemistry.PEG-SS-PCL-SSPEG fabricated into polymeric micelles with stable structure and different nanoscale sizes via adjusting the PCL chain length,showing obvious reductive responsiveness and fast drug release of encapsulated DOX in the presence of glutathione(GSH).Moreover,DOX-loaded PEG-SS-PCL-SS-PEG micelles exhibited higher therapeutic efficacy than reduction-insensitive PEG-b-PCL micelles in vitro.Thus,end-groups conjugation is a simple and straightforward strategy to introduce intelligent responsiveness in biocompatible block copolymers and improve their therapeutic efficacy.Junbo LI Junting JIANG Biyu ZHOU Chaohuang NIU Wendi WANG Wenlan WU 2019Frontiers of Materials Science2019,13,4:0
